RÉSUMÉ

Coconut oil production is the eminent agro based industries in Tamil Nadu. The matured coconut water was considered as a waste. Coconut oil industries discharge considerable amount of waste coconut water having very high values of Biochemical Oxygen Demand (BOD) to the extent of 29,000 mgl-1, the wash water about 3,000 mg. and Total Solids (TS) of 5.45 ± 0.35%. Coconut water contains most nutrients required for microbial growth. The pH of coconut water is 6.1. The Total Suspended Solids in coconut water is found out as 0.0028 g/ml. The Total Dissolved Solids is 0.8014 g/ml, Total Solids is 0.8042 g/ml and Volatile Soluble Solids is 99.5 %. A laboratory scale batch type anaerobic digester (Air tight 2 litre plastic bottles) was set up for the experimental purpose for a retention time of 35 days. Coconut water, water and cow dung were mixed in different proportions (5 treatments). Each treatment was replicated 3 times. Biogas volume and biogas composition were measured at 5:30 pm every day. Remarkable volume of biogas production was noticed between 12th and 16th days. Maximum biogas volume of 976. 67 ml was noticed for the combination of 1:1 (Cow dung: Coconut water) with methane content of 53 %. For the combination of 2:1:1 (Cow dung: Coconut water: Water) the maximum volume of biogas production is 833.3ml with methane content of 31 %. For the combination of 1:1 (water: Cow Dung) the maximum volume of biogas produced is 133.3 ml with methane content of 48 %. The biogas produced from different ratios was tested for its flammability. It was seen that blue flame is produced and the flame is last for few seconds for all ratios but the maximum amount of flame was produced from the sample which contains maximum amount of Coconut water than other samples.

ABSTRACT A comparative study to determine variations in postprandial pH values along the gastrointestinal tract was performed among juveniles of Piaractus brachypomus (n = 21) and Schizodon fasciatus (n = 21) in rearing conditions. After fasting for 24 hours, the fish were fed three times a day to apparent satiety, with a mixed diet containing pelleted balanced food and chicken heart muscle (ratio 1:1). The evaluation was made before feeding and postprandial (eight hours), where the pH values of the stomach, in the anterior and in the middle portions of the intestine were estimated, using a pH electrode with a lancetype tip. Significant differences (p < 0.05) at the stomach and the middle portions of the intestine, according to the digestion time were observed in both species, while at the anterior portions of the intestine, only S. fasciatus showed significant differences (p < 0.05). In both species, an increase in stomach pH was observed in the first postprandial hour (2,39 to 4,78 in P. brachypomus, 4,47 to 4,73 in S. fasciatus), led by the effect of the ingested food, and the pH values were reduced at the end of the experiment. During the evaluation hours, intestinal pH values for P. brachypomus and S. fasciatus were between 6,68 and 7,47 and 6,86 and 7,60, respectively, increasing distally along the intestine. It is concluded that both species present a permanent stomach acidification, and that the anterior and middle portions of the intestine maintain a slightly neutral pH during digestion and assimilation of food.

RÉSUMÉ

Plastic products are widely used in various fields, and the discarded plastics in the environment can be degraded into microplastics (MPs) or even nanoplastics (NPs), which significantly increases the risk of organism exposure. MPs/NPs have been found in aquatic organisms, birds of prey, and even humans. The detection of plastic particles in biological samples is more complicated than that in environmental samples. Biological samples are mainly composed of various organic substances such as proteins and lipids, which makes the pretreatment process particularly critical. Effective detection and accurate quantification of MPs/NPs are crucial to health risk assessment. In this paper, the exposure levels and composition of MPs/NPs in different tissues and organs of the human body, aquatic organisms, and birds of prey were reviewed. The digestion of biological samples (acids, bases, enzymes, and hydrogen peroxide digestion protocols) and MPs/NPs identification methods (spectroscopy and chromatography) were compared and their advantages and disadvantages were assessed, providing a methodological basis for plastic exposure risk assessment and pollution control.

Based on the traditional medical theory, the theory of"ear-brain-liver-spleen correlation"was proposed by linking the ear with the brain, liver and spleen. Then,by combining the Chinese medical etiology of Functional Dyspepsia(FD)and the corresponding modern medical mechanism,we realized that the close connection between the ear, brain, liver and spleen is the theoretical basis for treating FD from the ear.In recent years,a new type of vagus nerve stimulation therapy, auricular concha eletro-acupuncture(ACEA),has been developed to treat FD by regulating the liver and spleen through the regulation of the brain, relieving liver depression and strengthening spleen and qi,thus treating FD.In terms of modern medicine, this is related to its ability to reduce anxiety and depression, reduce pain perception, enhance gastrointestinal motility and reduce central and peripheral inflammation by regulating the activity of the corresponding brain functional areas.By exploring the application of auricular concha eletro-acupuncture in FD on the basis of the"ear-brain-liver-spleen correlation",we will better understand the unique role of auricular stimulation in regulating the brain and the blood of the liver and spleen, and promote the inheritance, innovation and development of TCM theory.

RÉSUMÉ

Inductively coupled plasma mass spectrometry (ICP-MS) was applied to determine the concentrations of lead (Pb), cadmium (Cd) and arsenic (As) in Lindera aggregata (Sims) Kosterm. The physiologically based extraction test (PBET) digestion in vitro/Caco-2 cell model was established to investigate the bioaccessible contents of Pb, Cd and As in decoction of Lindera aggregata (Sims) Kosterm. The target-organ toxicity dose modification of HI method (TTD) was used to evaluate the cumulative risk caused by the combined exposure of the total levels of Pb, Cd and As in Lindera aggregata (Sims) Kosterm. and the bioaccessible contents in the decoction. The results showed that the total contents of Pb, Cd and As in 4 batches of samples were in the range of 2.901-3.872, 1.299-1.800 and 0.062-0.216 mg·kg-1, respectively. After transportation by Cacco-2 cells, the bioaccessible contents of Pb, Cd, and As in the decoction were in the range of 0.045-0.080, 0.070-0.112 and 0.004-0.018 mg·kg-1. The results of risk assessment showed that calculated by the total amounts of heavy metals in the Lindera aggregata (Sims) Kosterm., for the end points of nervous system, the cumulative risks of co-exposure of heavy metals in 3 batches of samples were of concern. After decoction and transportation by Caco-2 cells, for the end points of cardiovascular system, blood, nervous system, kidney and testis, the TTD modification of HI values of all batches of samples were less than 1, and the health risks were acceptable. The study provided methodology basis for a more objective assessment of the health risks of heavy metals and harmful elements in traditional Chinese medicine and for a more scientific limit standard of heavy metals and harmful elements.

Introduction: Biomedical waste (BMW) generated is a matter of great concern as it consists of high levels of hazardous waste. The lack of segregation and disposal policy in India leads to use of such infectious waste in day-to-day life [1]. Lack of awareness about segregation of Biomedical Waste (BMW) at source is a major cause for Hospital Acquired Infection (HAI) and increased risk of contamination of hospital and external environment. Materials & Methods: To address the problem, we decided to train interns and health care workers to increase their awareness about BMW segregation and thereby reduce HAI. The program was run in 3 phases. In the 1st phase need assessment was done as to how important and effective this program would be in controlling HAI. The 2nd phase comprised of identifying the stake holders in the program who will play the key role in implementing the program effectively. The 3rd phase of Biomedical Segregation Program BMWSP focused on the outcomes as enumerated in the Kirkpatrick model [2], viz., participants’ reaction, learning, change in behavior and results. Measuring the impact of the program on the learners at these four levels revealed the effectiveness of the program. Results: To overcome the shortfalls in the Kirkpatrick model we used Logic model which took care of the instructional design. Logic model is better suitable for evaluating BMWSP as it considers inter-relation between constituents in a sequence and their effect on the process as well as product providing the necessary roadmap from the planned work (inputs and activities) to the intended results (outputs, outcomes and impact). It is a sequence of events connecting each other starting with Resources / Inputs, Activities, Outputs, Outcomes and Impact. The model helped medical interns and dental students to understand the importance for BMWSP and also made a significant difference in its implementation. Conclusion: The program helped in making the health care staff more aware about BMWSP. It also increased their knowledge, and helped in implementation of the program. This will significantly reduce HAI and overall benefit the society.

RÉSUMÉ

Anaerobic digestion is another important anaerobic catabolism pathway besides lactic acid and ethanol fermentation, which is of great significance for recycling resources, maintaining the ecological balance, optimizing the energy structure, alleviating the energy crisis, and promoting the implementation of the "Carbon Peaking and Carbon Neutrality" strategy. However, such an important metabolic process has not been involved in the current textbooks and teaching of biochemistry courses, making the teaching system incomplete. The anaerobic digestion process involves many reactions and complex metabolic pathways. In order to improve the students' understanding to this process, we created a full chart of the whole anaerobic digestion process based on systemic literature review and integrated it into the classroom teaching through the BOPPPS teaching mode. It was found that the classroom teaching assisted by this metabolic chart could help students establish the structural framework of the anaerobic digestion process and enrich the knowledge system of metabolism, achieving a good teaching effect. This paper introduces the content of the metabolic pathways of anaerobic digestion and the design of the teaching process, which would facilitate the teaching reforms and perfection of textbooks for related courses, such as Biochemistry, Environmental Engineering Microbiology and New Energy Engineering.

RÉSUMÉ

Objective:To investigate the stability of α-synuclein (α-Syn) aggregates formed by incubation with blood plasma of patients with Parkinson's disease (PD).Methods:Peripheral blood samples were collected from 10 patients diagnosed as having PD in our hospital from June 2020 to December 2020 and 10 healthy control subjects (HC) at the same time period. The 1 mg recombinant human α-Syn was dissolved in 140 μL 0.01 mol/L phosphate buffer solution (PBS), and then, incubated with plasma from HC and PD patients and PBS at 37 ℃ for 7 d (HC group, PD group and PBS group). Preformed fiber (PFF) group was used for subsequent experiment with 10 μg PFF. After digestion with different concentrations of trypsin (concentration ratios of trypsin/α-Syn=1:80, 1:40, and 1:20) and protease K (PK, 1.0, 1.5, and 2.0 μg/mL), the differences of α-Syn levels before and after digestion were detected by Western blotting.Results:(1) Effect of trypsin on PFF digestion: PFF gradually decreased with the increase of trypsin doses; when trypsin/α-Syn ratio=1:20, α-Syn aggregates with molecular weight greater than 35 000 were almost completely digested, and its digestion was significantly different as compared with that in other concentration ratios ( P<0.05). (2) Effect of trypsin on digestion of α-Syn aggregates: at the relative molecular weight<25 000, when the concentration ratio of trypsin/α-Syn=1:20, as compared with HC and PD groups, the PBS group had significantly more obvious decrease ( P<0.05). At the relative molecular weight 35 000-40 000, the α-Syn levels in PBS, HC and PD groups were significantly decreased as compared with those before digestion at all concentration ratios; as compared with HC and PD groups, the PBS group had significantly more obvious decrease ( P<0.05). At molecular weight>40 000, α-Syn decreased significantly in PD group only when the concentration ratio of trypsin/α-Syn=1:20, and the degrees of digestion was PBS group>HC group>PD group, with significant differences among groups ( P<0.05). (3) Effect of PK on PFF digestion: when PK concentration was 1.0, 1.5 or 2.0 μg/mL, α-Syn was basically digested at relative molecular weight>35 000, and α-Syn monomer was reduced and small fragment appeared at relative molecular weight<25 000; as compared with negative controls (0 μg/mL PK), these changes in groups of PK concentration of 1.0, 1.5 or 2.0 μg/mL were significantly different ( P<0.05). (4) Effect of PK on digestion of α-Syn aggregates: at relative molecular weight<25 000, α-Syn in PBS group was digested into smaller fragments while α-Syn in HC and PD groups was not digested; significant differences was noted among groups at the same concentration ( P<0.05). At relative molecular weight of 35 000-40 000, with the increase of PK concentration, the amount of α-Syn dimer in PBS group decreased (increased digestibility), that in HC group increased, however, that in PD group did not change obviously; significant difference was noted at the same concentration among the three groups ( P<0.05). At relative molecular weight>40 000, with the increase of PK concentration, α-Syn in PBS, HC and PD groups decreased to a certain extent, and significant difference was noted among groups at the same concentration ( P<0.05). Conclusion:The stability of α-Syn aggregates formed by incubation with plasma from PD patients is higher than that formed by incubation with HC plasma and PBS.

There were significant differences in the positive rate of the liver, kidney tissues between MD-VF-Auto SEM method and plankton gene multiplex PCR system (P<0.05), as well as the total positive rate of cases. However, no significant differences were found in the positive rates of lung tissues (P>0.05).@*CONCLUSIONS@#MD-VF-Auto SEM method is more sensitive than plankton gene multiplex PCR system in diatom test. But the plankton gene multiplex PCR system can also detect plankton other than diatoms. Combination of the two methods can provide a more reliable basis for the diagnosis of drowning.

RÉSUMÉ

Abstract To investigate structure-property relationship of polymer-based curcumin solid dispersion (SD), three acrylic polymers were used to formulate curcumin SD by solvent evaporation method. Curcumin Eudragit EPO SD (cur@EPO), curcumin Eudragit RS PO SD (cur@RSPO) and curcumin Eudragit RL PO SD (cur@RLPO) showed deep red, golden orange and reddish orange color, respectively. Cur@RSPO entrapped 15.42 wt% of curcumin followed by cur@RL PO and cur@EPO. FTIR spectra indicated that in cur@EPO, curcumin may transfer hydrogen to the dimethylaminoethyl methacrylate group and thus change its color to red. In contrast, curcumin may form hydrogen bonding with Eudragit RS PO and Eudragit RL. Curcumin exists in amorphous state in three SDs as proved by differential scanning calorimetry and X-Ray diffraction measurement. In vitro digestion presented that lower pH value in simulated gastric fluid (SGF) stimulates the curcumin release from cur@EPO while permeability influences the release profile in other two SDs. When in simulated intestinal fluid (SIF), first order release model governs the release behaviors of all three SDs which showed sustained release pattern. Our results are helpful to elucidate how structure of polymer may impact on the major properties of curcumin contained SD and will be promising to broaden its therapeutic applications.

ABSTRACT Biogas recovery systems contribute to the mitigation of greenhouse gases, besides adding economic value to the anaerobic decomposition of organic matter. This research aims to analyze the energy potentials and economic feasibility of implementing a landfill biogas recovery system (scenario 1) and an anaerobic digestion plant (scenario 2) for the Baturité Massif Consortium in the state of Ceará, Brazil. To that end, a population projection followed by an estimate of biogas production were made using the LandGEM software. For the biogas recovery system of the landfill, an optimum power of 460 kW and a maximum energy generation of 3,223,680 kWh/yr were obtained, whereas the anaerobic digestion plant resulted a power and energy of 252 kW and 1,986,754.86 kWh respectively. According to the economic analysis, both projects are unfeasible, but the price of the energy unit generated (kWh) is 42.42% higher for the anaerobic digestion plant than for the biogas recovery system of the landfill. The methodology applied can be used for preliminary studies in other consortiums, contributing to a better waste management and energy use of biogas systems in the country.

RÉSUMÉ

Abstract This study evaluates the efficiency of acid extraction and total digestion to determine the presence of metals in geopropolis produced by the stingless bee Melipona scutellaris. Geopropolis samples were collected at five meliponaries in the city and in the metropolitan region of Salvador, Bahia State, Brazil. The sample treatment methods comprised acid extraction and total digestion. The Inductively Coupled Plasma Optical Emission Spectrometry (ICP OES) technique was used to quantify Cd, Cu, Cr, Pb, and Zn. Geopropolis samples submitted to both digestion methods showed statistical differences. For both methods, Cr and Zn showed the highest concentrations, while those of Cd were the lowest. The Cr concentration for determination by total digestion was 37.53 mg/kg, while for acid extraction it was 32.90 mg/kg. For Zn, the concentration was 17.65 mg/kg and 8.85 mg/kg for total digestion and acid extraction, respectively. The total digestion method showed the highest concentrations of the metals evaluated; however, acid extraction (USEPA 3050b) is a more straightforward procedure for metal evaluation in geopropolis samples and presented values that support the use of geopropolis as a bioindicator. The acid extraction method USEPA 3050b, in combination with detection using ICP OES, showed efficiency in analyses carried out to determine metals in geopropolis.

ABSTRACT The objective of this study was to analyze the chemical composition and bioaccessibility of phenolic compounds as well as their antioxidant capabilities of organic peppermint leaves after each phase of simulated digestion. Moisture was determined until a constant weight was obtained in an oven at 105 °C; ash was determined after sample calcination in a muffle furnace at 550 °C. The protein concentration was determined by the Macro-Kjeldahl method and lipid content by hot-extraction in a Soxhlet apparatus. Carbohydrates were calculated from differences and energy values based on the Atwater conversion factors. Total phenolic, flavonoid content, and antioxidant activity were determined by spectrophotometry. A four-step procedure was used for in vitro digestion. Organic peppermint was found to contain the following: 78% moisture, 1.7% ash, 1.5% lipids, 0.3% proteins, 17.7% carbohydrates, and a total of 85.5 kcal/100 g. Values of 705 mg GAE/100 g of phenolic, 918 mg QE/100 g of flavonoids, and 58.8 mg/g of vitamin C were also measured. It was discovered that total phenolics had the highest bioaccessible fraction relative to flavonoids; the salivary phase was identified as that with the highest release of these compounds and thus the phase in which peppermint showed significant antioxidant activity (1509 μmol TEAC/100g). This study demonstrated that organic peppermint has a high content of phenolic compounds that can be extracted from the alimentary matrix in the salivary and intestinal phases of the digestive system. Because of the antioxidant activity of these compounds, the use of this aromatic plant as seasonings and spices is relevant.
