Résumé

ABSTRACT Purpose: Glucose is a major energy resource for tumor cell survival and growth, and its influx into cells is mainly carried out by facilitative glucose transporters (GLUTs). Sodium - dependent glucose transporters (SGLTs) have been highlighted as playing important roles in diabetic treatment. However, their potential roles in cancer remain unclear. We examined expression patterns of SGLTs in tumor tissues together with conventional pathological variables to determine prognostic significance in patients with renal cell carcinoma (RCC). Materials and Methods: Nephrectomy specimens were obtained from 68 patients. GLUT - 1, - 2 and SGLT - 1, - 2 expression in tumor and adjacent normal tissues were analyzed by immunohistochemical staining, and intensity was quantified using an image analyzer. Results: The four glucose transporters evaluated were broadly distributed in tumor tissues as well as throughout the normal parenchyma. There was no significant correlation between transporter expression and conventional pathological variables. However, increased SGLT - 2 expression was significantly associated with shorter overall survival (p < 0.01), regardless of metastatic status. Conclusions: We propose possible prognostic significance of SGLT - 2 expression in human RCC. Given that glucose is a major energy resource for tumor cells and that glucose transport is largely mediated by SGLT, SGLT - 2 may serve as a possible therapeutic target in RCC.

Résumé

Abstract The aim of this study was to evaluate the immunoexpression of glucose transporters 1 (GLUT-1) and 3 (GLUT-3) in keratocystic odontogenic tumors associated with Gorlin syndrome (SKOTs) and non-syndromic keratocystic odontogenic tumors (NSKOTs), and to establish correlations with the angiogenic index. Seventeen primary NSKOTs, seven recurrent NSKOTs, and 17 SKOTs were selected for the study. The percentage of immunopositive cells for GLUT-1 and GLUT-3 in the epithelial component of the tumors was assessed. The angiogenic index was determined by microvessel count. The results were analyzed statistically using the nonparametric Kruskal-Wallis test and Spearman’s correlation test. High epithelial immunoexpression of GLUT-1 was observed in most tumors (p = 0.360). There was a higher frequency of negative cases for GLUT-3 in all groups. The few GLUT-3-positive tumors exhibited low expression of this protein in epithelial cells. No significant difference in the angiogenic index was observed between groups (p = 0.778). GLUT-1 expression did not correlate significantly with the angiogenic index (p > 0.05). The results suggest that the more aggressive biological behavior of SKOTs when compared to NSKOTs may not be related to GLUT-1 or GLUT-3 expression. GLUT-1 may play an important role in glucose uptake by epithelial cells of KOTs and this process is unlikely related to the angiogenic index. GLUT-1 could be a potential target for future development of therapeutic strategies for KOTs.

Résumé

BACKGROUND: The glucose transporters (GLUTs) exhibit different tissue-specific expression. This study aimed to investigate the types of GLUTs expressed in human granulosa cells (GCs) obtained from women with polycystic ovary syndrome (PCOS) and their relationship with insulin resistance (IR) and the outcomes of in vitro maturation (IVM) of immature oocytes. METHODS: Expression of GLUTs was evaluated in GCs from women with PCOS with or without IR. Thirty-six women with PCOS undergoing an IVM program were included. Differential gene expression between the insulin sensitive (IS) and IR group was measured by reverse transcription polymerase chain reaction. RESULTS: Expression of GLUTs 1, 3, 5, 8, and 13 was constitutive, whereas expression of GLUTs 2 and 7 was not observed in human GCs. The remaining GLUTs, 4, 6, 9, 10, 11, and 12, were differentially expressed among patients according to metabolic status, such as insulin sensitivity. A higher number of GCs from patients with IR (92%) expressed GLUT6 than GCs from IS PCOS patients (46.3%). Logistic regression showed that expression of GLUTs 9, 11, and 12 correlates with rates of IVM at 48 hours, fertilization, and implantation, respectively. CONCLUSION: This is the first report describing the expression pattern of all 13 members of the GLUT family in human GCs. Results of the present study suggest that patients' insulin sensitivity regulates GLUT expression in GCs in PCOS patients, and this may control oocyte quality for IVM and subsequent processes such as fertilization and implantation in patients taking part in an in vitro fertilization program.

Résumé

Glucose and related hexoses are very important metabolic substrates. Their most important function is to provide quick fuel for most organisms in all three kingdoms because they are the first substrate for energy production in the form of ATP through glycolysis and the subsequent metabolic pathways. In this paper we review the current information about how glucose and related hexoses are transported across biological membranes to carry out their function either as a metabolic molecule or as energy store in marine invertebrate organisms. In these animals, there are two sugar transport systems that are mediated by the sodium/solute symporter family proteins (SGLT) and the major facilitative super-family proteins (GLUT). The most studied sugar transporters in marine invertebrates are involved with dietary sugar uptake, such as SGLT1, SGLT4, GLUT2 and GLUT5, however more studies need to be done to extend the knowledge about these and other sugar transporters involved in metabolic processes.

Résumé

Objective To evaluate the expression of the GLUT-1 glucose transporter in borderline and malignant epithelial neoplasms of the ovary. Methods Histologic sections of tumor tissues from 100 epithelial neoplasms of the ovary were stained for GLUT-1 using polyclonal GLUT-1 antibody and the labeled streptavidin procedure. Chi-square analysis was used to assess if their expression was associated with clinicopathoiogic characteristics of the tumors. Results Normal ovarian surface epithelial cell were either negative or weakly positive. The intensity of the stain was significantly stronger in malignant than borderline tumors (P<0.001). In malignant neoplasms, the intensity of the stain for GLUT-1 increased with histology. Conclusion Overexpression of the GLUT-1 transporter is associated with the histology and grade of the minors. These data suggest that the expression of this transporter may be closely related to the malignant transformation of epithelial ovarian tumors.

Résumé

Objective: To explore the changes of hypoxia-inducible factor 1α(HIF-1α), glucose transporter protein-1(GLUT-1), and matrix metalloproteinase-2 (MMP-2) in hypoxic human lung adenocarcinoma A549 cells. Methods: The human lung adenocarcinoma A549 cells were cultured in normoxic and CoCl2-induced hypoxic conditions respectively. MTT assay was used to test the effect of hypoxia on the proliferation of lung cancer cells, and Transwell assay to determine the invasive capacity of hypoxic cells. Reverse transcription-polymerase chain reaction analyses (RT-PCR) was used to detect the mRNA expression of HIF-1α, GLUT-1 and MMP-2 under hypoxia. The protein levels of HIF-1α, GLUT-1, and MMP-2 were measured with immunohistochemical staining and western blot at different time points after hypoxia. Results: The proliferative and invasive capabilities of lung cancer cells increased under hypoxia condition. HIF-1α, GLUT-1, and MMP-2 were expressed at certain levels in A549 cells under normoxic condition. After hypoxia exposure, the mRNA and protein expression of the three proteins increased in a time-dependent manner. The difference was significant between hypoxia group and normal control group (P <0.05). Conclusion: Hypoxia induced by CoCl2 up-regulates the expression of HIF-1α, GLUT-1 and MMP-2 in lung adenocarcinoma A549 cells and stimulates the proliferation and metastasis of lung cancer cells, thus further promotes their tolerance to hypo-xia.
