Objective: To investigate the impact of the extracts of Gac fruit parts (peel, pulp, seed, and aril) on the cell viability and angiogenesis markers of human retinal pigment epithelial (ARPE-19) cells under high glucose conditions. Methods: The effect of the extracts of Gac fruit peel, pulp, seed and aril on the ARPE-19 cells was determined using MTT viability assay, Trypan blue dye and morphological changes were observed using light microscopy. Enzyme-linked immunosorbent-based assay was performed to evaluate the effect of Gac fruit parts on the reactive oxygen species (ROS), vascular endothelial growth factor (VEGF) and pigmented epithelium-derived factor (PEDF) secretions. Results: High glucose (HG) at 30 mmol/L increased ARPE-19 cell viability and ROS and VEGF secretions. While, the exposure of ARPE-19 cells in high glucose condition to Gac fruit extracts led to inhibition of cell viability, induced morphological changes, decreased ROS and VEGF secretions, and increased PEDF level. Gac pulp, seed, and aril at 1 000 μg/mL showed significant inhibition activities [(7.5 ± 5.1)%, (2.7 ± 0.5)%, (3.2 ± 1.1)%, respectively] against HG-induced ARPE-19 cell viability. The findings also demonstrated that Gac aril at 250 μg/mL significantly decreased ROS and VEGF levels [(40.6 ± 3.3) pg/mL, (107.4 ± 48.3) pg/mL, respectively] compared to ROS [(71.7 ± 2.9) pg/mL] and VEGF [(606.9 ± 81.1) pg/mL] in HG untreated cells. Moreover, 250 μg/mL of Gac peel dramatically increased PEDF level [(18.2 ± 0.3) ng/mL] compared to that in HG untreated cells [(0.48 ± 0.39) ng/mL]. Conclusions: This study indicates that the extracts of Gac peel, pulp, seed and aril reduced cell viability, minimized ROS generations and showed angiogenic activities. Therefore, our findings open new insights into the potentiality of Gac fruit against HG-related diabetic retinopathy disease.

Abstract Gac, Momordica cochinchinensis (Lour.) Spreng., Cucurbitaceae, is an indigenous South Asian edible fruit and has been used therapeutically in Traditional Chinese Medicine. Previous studies have shown that M. cochinchinensis seed (Momordicae Semen) has various pharmaceutical properties such as antioxidant and anti-ulcer effects as well as contains secondary metabolites with potential anticancer activities such as triterpenoids and saponins. However, its biological activities in cancer have not yet been investigated. In this study, we found that its ethanol extract reduced cell proliferation in four human lung cancer cell lines, A549, H1264, H1299 and Calu-6. Phytochemical investigation of the ethanol extract was carried out, and resulted in isolation of two major saponins, which were identified as gypsogenin 3-O-β-d-galactopyranosyl(1 → 2)-[α-l-rhamnopyranosyl(1 → 3)]-β-d-glucuronopyranoside (1) and quillaic acid 3-O-β-d-galactopyranosyl(1 → 2)-[α-l-rhamnopyranosyl(1 → 3)]-β-d-glucuronopyranoside (2). Treatment with these isolated compounds (1 and 2) decreased cel1 proliferation in all human lung cancer cell lines tested. In addition, the compounds attenuated primary lung endothelial cell proliferation. Taken together, these findings suggest M. cochinchinensis seeds have antiproliferative activity on human lung cancer cells as well as angiostatic effect on lung endothelial cells.

Objective To analyze the phylogenetic and mutation sites of IRa (trnV-GAC to trnR-ACG) sequence among 10 Dendrobium Sw. samples, to understand the similarities and differences of nine genes in this sequence, and to explore the system evolution and DNA barcode in species of Dendrobium Sw. Methods The improved kit method was used to extract total DNA and design primers, PCR method and T-vector cloning transformation were used to amplify the region of IRa and the sequences were obtained after the amplified fragment sequences were blasted in NCBI database and splicing. Genetic structures were drawn, and variable sites, genetic distance and phylogenetic relationship among 10 sequences were analyzed. Results The lengths of sequences were about 7 800 bp and 55 variable sites were found. The numbers of mutation points distributeed on trnV-GAC, rrn16, trnI-GAU, trnA-UGC, orf42, rrn23, and intergenic region were 1, 8, 12, 6, 1, 9, and 18, respectively. These 10 samples were clustered to four latitude-dependent groups based on IRa sequences. Conclusion A set of methods on sequence IRa is established. Phylogenetic and mutation point analysis indicate that there is a rich and relatively concentrated distribution variation loci among the research sequences. And it will provide a good theoretical basis on the further study of DNA barcodes, authentic assessment and identification of Dendrobium Sw. germplasm resources.

In Amazonia, chromium is mainly used in the leather and wood industries. It is responsible for many health problems, because of its toxicity. These industries remove chromium waste by various processes, such as adsorption. This work shows the results of Cr(VI) adsorption by commercial granular activated carbon (GAC) as adsorbent from diluted solutions, and batch systems with controlled pH. The functional groups on the CAG surface was carried out by the Boehm method. In addition, effect of pH on the Cr(VI) adsorption, adsorption equilibrium, and kinetic were studied under experimental conditions (pH = 6, MA = 6g, for 90min.). On the GAC surface, carboxylic groups were found to be in higher concentrations (MAS=0,43 mmol/gCAG), which increase the Cr(VI) adsorption, principally in acidic pH values. The adsorption capacity is dependent on the pH of the solution, due to its influence on the surface properties of the CAG and different ionic forms of the Cr(VI) solutions. The adsorption equilibrium data was adjusted satisfactorily by the Langmuir isotherm (R²=0,988), favorable type. From kinetics adsorption of 20 mg/L and 5 mg/L, the results were compatible with the national legislation (Res. nº 357/05). Therefore, the experimental system using (CAG) was efficient in removing the Cr(VI) from liquid streams containing low concentrations of the metal.


Na Amazônia o cromo é empregado principalmente na indústria de couro e de madeira, sendo responsável por vários problemas de saúde porque é tóxico para os seres vivos. A remoção de cromo de efluentes industriais é feita por meio de diversos processos como a adsorção. Este trabalho mostra os resultados da adsorção de Cr(VI) por carvão ativado granular comercial (CAG) como adsorvente de soluções diluídas empregando um sistema de adsorção batelada com controle de pH. Os grupos funcionais da superfície do CAG foram determinados pelo método de Boehm. Além disso, o efeito do pH na adsorção de Cr(VI), o equilíbrio e a cinética de adsorção foram estudados nas condições experimentais (pH = 6, MA = 6g, tempo de adsorção 90min.). Na superfície do CAG, os grupos carboxílicos foram determinados em maior concentração (MAS=0,43 mmol/gCAG), estes, presentes em concentrações elevadas aumentam a adsorção do metal, principalmente em valores de pH ácidos. A capacidade de adsorção é dependente do pH da solução, devido a sua influência nas propriedades de superfície do CAG e nas diferentes formas iônicas das soluções de Cr(VI). Os dados de equilíbrio da adsorção foram ajustados satisfatoriamente pela isoterma de Langmuir (R²=0,988), tipo favorável. A partir da cinética de adsorção a 5mg/L e 20mg/L, os resultados obtidos foram compatíveis com o valor limite preconizado na legislação nacional (Res. nº 357/05). Portanto, para o sistema experimental utilizando CAG foi eficiente na remoção de Cr(VI) a partir de correntes líquidas contendo baixas concentrações do metal.

Background: Mosquitoes and pigs play important roles in maintaining and increasing the Japanese Encephalitis (JE) virus in nature and which is then transmitted to humans. Thus, surveillance of the JE infection frequency in the pig population may predict the human JE cases. \r\n', u'Objectives: The study aimed to determine IgG antibody against the JE virus in the pig population in Hanam province \r\n', u'Subjects and methods: The study included 1791 pig serum samples collected from 3 districts of Hanam province from Apr 2006 to Mar 2007. GAC-ELISA technique was used to determine the JE virus infection in the swine population.\r\n', u'Results: The average positive rate in pig population was 34.9 % (626/1791); with the highest frequency occurring in the summer (37.7%- 84.0 %), co-incident with the JE season in Northern Vietnam. On the contrary, in winter JE case are rare, frequency of IgG antibody against JE virus in the swine population was low, ranging from 9.2% to 22.0.%. \r\n', u'Conclusions: These results have shown the ecologically close relationship between the amplification of the JE virus in the swine population, vector and JE cases in northern Vietnam. \r\n', u'

PURPOSE: In order to evaluate the effects of GAC, which is the combination of active hexose correlated compound (AHCC) and genistein combined polysaccharide (GCP), we investigated the changes in the biochemical profiles and the quality of life of prostate cancer patients with androgen suppression after the administration of GAC. MATERIALS AND METHODS: Thirty two eligible metastatic prostate cancer patients between the ages of 54 and 84 were enrolled in this study, and they were supplemented with 5g GAC per day (n=23) or placebo (n=9) for a 6 months period. Blood and urine sample analysis were taken and the quality of life (QoL) was assessed using the Visual Analogue Scale (VAS) and the Functional Assessment of Cancer Therapy Scale Questionnaire (FACT-G) at baseline and at post intervention (after 3 and 6 months). RESULTS: Twenty six patients (n=18 in the GAC group and n=8 in the placebo group) completed the 6 months intervention. No statistically significant adverse events were reported by the study participants. GAC had no significant effect on the serum biochemical parameters. However, all 7 GAC-treated hypercholesterolemic patients had their cholesterol level decreased after 3 months treatment (p<0.02). Results of Comet assay showed significant decreases in tail moment (p<0.009) and tail length (p<0.004) at 6 months compared to baseline for the GAC group. Although the results of the VAS were inconsistent, the score for physical well-being was increased in GAC group on the FACT-G analysis (p<0.05 between baseline and 3 months, respectively). CONCLUSIONS: Oral administration of GAC 5g per day for 6 months showed a decrease in DNA damage of blood lymphocytes and in the total serum cholesterol level in hypercholesterolemic patients without any significant influences on the serum biochemical parameters of the metastatic prostate cancer patients. Further studies on the role of GAC are necessary to clarify the advantage of GAC supplementation in prostate cancer patients with androgen suppression.
