Résumé

OBJECTIVE To explore the role of P38MAPK inhibitor on the soft palate reconstruction of the rats with chronic intermittent hypoxia. METHODS The animals were divided into normal control group, hypoxia control group and SB203580+hypoxia group (every group 20 rats). After 5 weeks, the expressions of p38MAPK and p-p38MAPK protein on the soft palate of the rats were detected with immunohistochemical techniques and western blot. RESULTS Compared with the normal control group, the soft palate tissue thickness of the hypoxia group were increased most obviously; Levels of p38MAPK were increased in hypoxia control group; Compared with the hypoxia control group, the levels of p-p38MAPK group were decreased in SB203580+hypoxia group. CONCLUSION p38MAPK may play important roles in the soft palate reconstruction of the rats with chronic intermittent hypoxia.

Objective To confirm the expression of Aquaporin2,3,4(AQP2,3,4,water channel protein) in rats'endolymphatic sacs(ES)and kidneys and to investigate and to compare the effects of anti-diuretic hormone(AVP) and DDAVP([deamino-Cys1,D-Arg8]-Vasopressin,V2-receptor agonist)on the expression of AQP2 in rats' ESs and kidneys.Methods 30 healthy Swards white rats were divided into the negative control,AVP group and dDAVP group respectively.The animals were cardiacally perfused.The temporal bones and kidneys were taken out and sectioned by means of the paraffin-embedded technique.The sections of ESs were labeled with fluorescent antibody by immunohistochemical method,and the kidneys' with avidin-biotin-peroxidase complex method(ABC).The expressions of AQP-2 were confirmed in the ESs of the rats while the different effects of the AVP and DDAVP on the ESs and the kidneys were observed.The slides used were analyzed by the image-analyzer and the subsequent data were statistically studied.Results In the cytomembrane and cytoplasm of ESs' epithelia,the constant and clear fluorescent reaction could be observed in the normal control group with the first antibody of AQP2.Significant feeble fluorescent reaction of the first antibody of AQP-2 was revealed in AVP group and DDAVP group.In comparison with the control group,significant stain was found in AVP group and DDAVP group.Noted were also lower gray(P

Objective:To study the expression and significance of cyclinD 1 in 45 cases of transitional cell carcinoma of bladder.Methods:The expression of cyclinD 1 was detectde by immunohistochemistry(SP).Results: Expression of cyclinD 1 was found in 55.56% of TCCs and none of controls,and 78.57% of T a~T 1,11.76%of T 2~T 4.Approximately,85% of G 1,46.67% of G 2 and 10% of G 3 with the progress of tumors clinical stages and grades ,and the positive rates of cyclinD 1 expression decreased. There was no significant correlation between cyclinD 1 expression and number of lesions and postoperative reccurance.Conclusions: CyclinD 1 overexpression can play an important role in the early stage of bladder tumorgenesis.There was significant relativity between cyclinD 1 expression and the biological behaviour of bladder TCCs.

PURPOSE: p53 gene is a well-known suppressor gene of tumor. And the p53 protein expression in immunohistochemical staining acts a role as a prognostic indicator in prostate and bladder tumor. However, the validity of p53 protein expression was not determined as an independent factor of prognosis in renal cell carcinoma(RCC). Furthermore, there were not enough studies about whether the expression of p53 protein plays a role as a prognostic indicator in RCC or not. Therefore, we evaluate the prognostic significance of p53 protein expression in RCC. MATERIALS AND METHODS: To evaluate the prognostic significance of p53 immunohistochemical staining in RCC, paraffin embedded specimens taken from 84 patients with RCC were studied from January 1982 to April 1997, retrospectively. In addition to the prognostic significance of p53 immunohistochemical staining, correlation between p53 immunohistochemical staining and pT category, nuclear grade, and histologic type was studied. RESULTS: The expression rate of p53 protein was 19.0%(16/84). The most important prognostic indicator of RCC was pT category(relative risk=2.19, p=0.007). And the second important prognostic indicator was the expression of p53 protein(relative risk=1.82, p=0.017). Nuclear grade had prognostic significance(p=0.027) but was not an independent prognostic indicator. The expression of p53 protein did not correlate with pT category, nuclear grade or histologic pattern. CONCLUSIONS: We have shown that the expression of p53 protein in RCC is the second most important prognostic indicator. In conclusion p53 immunohistochemical staining can be used to predict the prognosis in patients with localized RCC after radical nephrectomy.
