Objective To investigate the role of miR-199a-3p in children with severe mycoplasmal pneumonia (MP) and to study its effect on Th17 cells. Methods Sixty children with severe MP (severe group), 50 children with recovery MP (recovery group) and 40 healthy children (control group) were enrolled. Venous blood samples were collected from all subjects, and the expression of miR-199a-3p in the peripheral blood leukocytes was detected by RT-PCR. The levels of IL-17a, IL-13, GM-CSF and IL-6 in serum were determined by enzyme-linked immunosorbent assay (ELISA). The effect of miR-199a-3p on the differentiation of Th17 cells was investigated by inducing differentiation of Th17 cells and transfecting miR-199a-3p mimics or inhibitors in vitro. Results Compared with the control group, the expression of miR-199a-3p in peripheral blood leukocytes of children with severe MP was significantly decreased (P<0.05). The levels of IL-17a, IL-13, GM-CSF and IL-6 in serum were significantly higher than those in the control group (all P<0.05). High expression of miR-199a-3p inhibited the differentiation of Th17 cells in vitro. Conclusions The expression of miR-199a-3p is decreased in the pathogenesis of MP. Th17 cells are the target cells of miR-199a-3p, and their differentiation is regulated by miR-199a-3p. The high expression of miR-199a-3p significantly inhibites the differentiation of Th17 cells.

Résumé

We developed a method to identify serological humoral immunodominant proteinic antigen of Mycoplasma hyopneumoniae (Mhp). After constructing the recombinant plasmid pGEX-6P-1-mhp366 and transforming it into Escherichia coli BL21(DE3), the recombinant GST-Mhp366 protein was expressed successfully. The lysates of the recombinant GST-Mhp366 and genetic engineering GST were added into glutathione coated plates and reacted with 17 positive sera or 13 negative sera. Meanwhile, the optimization of experimental conditions, including coated antigen, blocking buffer, dilutions of sera and second antibody were determined. The optimal concentration of the coated antigen was the original bacteria lysates without dilution, and the optimal blocking buffer contained 10% FBS and 2.5% skim milk in PBS. Besides, the working concentration of serum samples and the HRP-tagged rabbit anti-pig IgG secondary antibody were 1:500 and 1:40 000, respectively. Thus, an indirect ELISA was established for identification of immunodominant protein antigens of Mhp. Meanwhile, this method was confirmed by the identified serological humoral immunodominant proteinic antigen Mhp156 and Mhp364. This method can be used for identification of the candidate vaccine antigens on a genome-wide scale. Furthermore, it can lay the foundation for identifying the candidate vaccine antigens through colostra and the nasal mucosal secretions.

Small ubiquitin-related modifier(SUMO) is a small molecule that is structurally similar to ubiquitin.SUMOylation has emerged as an important post-translational modification.It profoundly influences the function of proteins,including the regulation of protein-protein interaction,the activity of protein transcription,DNA damage repair,gene regulation and biological rhythm.In recent years,evidence suggests that the SUMOylation plays crucial roles in lung.By binding to target proteins,SUMO is associated with pulmonary biological function and disease.

Mycoplasmal pneumonia is an important disease in the pig industry. Due to the controversial role of Mycoplasma hyorhinis in this disease, confirmation of the presence of this bacterium and the identification of its roles in respiratory disease remain major challenges. The objectives of this study were to evaluate the presence of M. hyorhinis in early cases of mycoplasmal pneumonia and to determine the usefulness of fluorescent in situ hybridization (FISH) for the diagnosis of respiratory mycoplasmosis in naturally infected pigs. Ninety M. hyopneumoniae and/or M. hyorhinis-infected lung tissue samples based on diagnostic mosaic (DM) were used. The average age of the animals was 116 and 57 days (P<0.01) for groups 1 (positive-M. hyopneumoniae only) and 2 (positive-M. hyorhinis only), respectively. These findings suggest that development of lesions caused by M. hyorhinis occurs earlier than for M. hyopneumoniae. Using the DM as the gold standard, the sensitivity and specificity of FISH for M. hyopneumoniae were 75 and 100%, respectively, and were 40 and 73.3% for the immunohistochemistry (IHC). The sensitivity and specificity of FISH for M. hyorhinis were 76.7 and 100%, respectively. These findings demonstrate that FISH can be a useful tool for diagnosing mycoplasmosis. Viral antigens (PCV2 or influenza A) were detected in 53.3% (16/30) of the samples in group 2 (M. hyorhinis-PCR positive) and 13.3% (4/30) of the samples in group 1 (M. hyopneumoniae-PCR positive). This finding indicates that the association of M. hyorhinis and viral infection in nursery pigs likely starts due to a viral immunosuppressive condition.(AU)


A pneumonia micoplásmica causada por bactérias do gênero Mycoplasma é uma enfermidade de grande importância para indústria suinícola, sendo ainda controverso o papel desempenhado por Mycoplasma hyorhinis nessa doença. A confirmação da presença dessas bactérias bem como a identificação de seus papéis em doenças respiratórias continua sendo um grande desafio. Os objetivos desse estudo foram comparar diferentes técnicas, em especial a de hibridização fluorescente in situ (FISH), para diagnóstico de micoplasmoses respiratória em suínos naturalmente infectados e avaliar a presença do M. hyorhinis em casos precoces de pneumonia micoplásmica. Foram utilizadas 90 amostras de tecido pulmonar infectado para cada um ou ambos os agentes (M. hyopneumoniae e M. hyorhinis) determinados pelo mosaico de diagnóstico (sinais clínicos, lesões macroscópicas e microscópicas e pela PCR). No grupo de animais positivos pela PCR apenas para M. hyorhinis (Grupo 2) a média da idade foi de 57,32 dias e no grupo apenas positivo para M. hyopneumoniae (Grupo 1) a média foi de 116,31 dias (P<0,01). Estes achados sugerem que a colonização e o aparecimento de lesões causadas pelo M. hyorhinis seja mais precoce do que aquelas causadas pelo M. hyopneumoniae. As alterações microscópicas foram estatisticamente (P<0,01) mais intensas no grupo 1 do que no grupo 2. Usando o mosaico de diagnóstico como padrão ouro, a sensibilidade e especificidade na FISH para M. hyopneumoniae foi de 75 e 100%, respectivamente, e 40 e 73,3%, na imuno-histoquímica. A sensibilidade e especificidade da FISH para M. hyorhinis foi de 76,7 e 100%. Esses valores demonstram que a FISH pode ser uma ferramenta útil para diagnóstico de micoplasmoses. Foi detectada a presença de agentes virais (PCV2 ou influenza) em 53,3% das amostras do grupo 2 (M. hyorhinis) e em 13,3% das amostras do grupo 1 (M. hyopneumoniae).(AU)

Objective To investigate the epidemic characteristics of mycoplasmal pneumonia outbreak in a middle school, and to provide suggestions for outbreak prevention and control.Methods Principle and method of field epidemiology was used and,speciously,questionnaire investigation was carried out for the outbreak of mycoplasmal pneumonia.Outbreak characteristics and corresponding emergency intervention were discussed.Results There was an outbreak of mycoplasmal pneumonia in 2 classes of the middle school,and the attack rate was 42.50%.The attack rate of other classes was 6.75%.Timely identification and diagnosis of the disease,and comprehensive control to prevent the spread of the epidemic,brought the disease under control.Conclusion The outbreak of mycoplasmal pneumoniae infection is associated with the close contact in daily activities.Emergency intervention can prevent the further spread of the disease.

BACKGROUND: The incidence of mycoplasmal pneumonia is predominantly at childhood and early adulthood, but in adults, its incidence is low and its symptoms and physical findings are nonspecific. The definite diagnosis of M. pneumoniae pneumonia can be made by sputum culture, but requires several weeks for positive results, and the early diagnosis must initially be based on the serologic tests and appropriate clinical findings. Thus, we evaluated the clinical aspects of M. pneumoniae pneumonia in the adults patients. METHOD: Among the admitted patients due to pneumonia, the definite diagnosis is anti-M. pneumoniae antibody titer of > 1:40 and a single cold agglutinin titer of > 1:64. The presumptive diagnosis is anti-M. pneumoniae antibody titer of > 1:40 or a single cold agglutinin titer of > 1:64 and the clinical characteristics or chest X-ray findings are compatible with M. pneumoniae pneumonia. We studied the age and sex distribution, seasonal distribution, clinical symptoms, physical findings, serologic test, chest X-ray findings, treatment and its progression. RESULTS: 1) The age distribution was even and the ratio of male to female was 1:1. 2) The monthly distribution was most common in January(16.7%) and the seasonal distribution in autumn and winter(autumn: 30%, winter: 33.3%). 3) The cold agglutinin titers were higher than 1:64 in 12 cases(40%), and reached the peak level around 2 weeks from onset and antimycoplasma antibody titers were higher than 1:160 in 5 cases(16.7%). 4) On the chest X-ray, pulmonary infiltration was noted in 28 cases(93.3%) among 30 cases and right lower lobe involvement was the most common(33.3%) and both lower lobe involvement was noted in 7 cases(23.3%). 5) The mean treatment duration was most common(33.3%) in 1 week to 2 weeks after admission and 26 cases(86.7%) were improved within 4 weeks. 6) On admission, there was fever(> or =38.9degreesC) in 17 cases(56.7%), and the fever subsided in 12 cases(70%) within 3 days after treatment using erythromycin. CONCLUSION: The mycoplasmal pneumonia in adults shows milder clinical patterns than that in childhood and can be completely recovered without complication by early diagnosis and treatment.

Résumé

Objective To improve diagnostic ability of mycoplasmal pneumonia in children.Methods The clinical data and X-ray findings of 563 cases with mycoplasmal pneumonia who hospitalized during 1999~2000 were retrospectively analyzed.Results Obvious abnormalities were seen on the chest films,which were in sharp contrast with the mild clinical respiratory symptoms.Erythromycin produced very effective results.Chest X-ray findings of mycoplasmal pneumonia were as follows:Pulmonary interstitial infiltration(417 cases,74%),parenchymatous infiltration(104 cases,18%),and mixed pattern (42 cases,8%),of them,pleurisy(63 cases,11%)and the hila lymph node enlargement (105 cases,19%).Conclusion The X-ray findings of mycoplasmal pneumonia are variant,a correct diagnosis can be made if one combines X-ray findings closely with laboratory results.
