Résumé

To investigate the effect of ginseng neutral polysaccharide on gut microbiota composition and diversity as well as the therapeutic effect for antibiotic associated diarrhea( AAD) in mice. The water-soluble ginseng neutral polysaccharide( WGPN) was purified from water-soluble ginseng polysaccharides( WGP) by DEAE-sepharose fast flow column,which was obtained from the roots of Panax ginseng. AAD mice were induced by gastric gavage with lincomycin hydrochloride,followed by administration of normal saline( natural recovery group,NR) or WGPN( WGPN group) for one week. Body weight changes,psychosis and diarrhea status were observed and assessed. 12 h after the last administration,histological observation of ileum and 16 S rRNA high throughput sequencing analysis of intestinal contents were conducted to identify the effects of WGPN on AAD mice. The results showed that WGPN could alleviate the symptoms of diarrhea in mice,decrease the inflammation and edema of ileum,and increase the length of intestinal villi. As compared to NR mice,WGPN could increase the relative abundance of Lactobacillus,and significantly decrease the relative abundance of Bacteroides,Streptococcus,Ochrobactrum and Pseudomonas at the genus level. In conclusion,WGPN could improve the gut microecology by recovering the ileum structure and improving the diversity and composition of the gut microbiota in AAD mice.

Objective: To clarify the structure features of polysaccharides from Chrysanthemum morifolium (PCM) and to study their activities against tumor cells and NF-κB. Methods: Six homogeneous neutral polysaccharides were obtained from three kinds of C. morifolium (Hangju, Huaiju, and Boju) flowers by successive hot water extraction, followed by ethanol precipitation, ion-exchange chromatography, and gel permeation chromatography. Their primary structures were characterized by HPGPC, IR, GC, and GC-MS analyses. Their bioactivities were examined by MTT assay using PANC-1 and LO2 cells. In addition, NF-κB signaling activation in PANC-1 and LO2 cells treated by polysaccharides were also measured. Results: The weight-average molecular mass of the six PCM, CMTA0S1, CMTA0S3, CMJA0S1, CMJA0S2, CMBA0S1, and CMBA0S3 was 7.523 × 104, 7.80 × 103, 7.80 × 104, 1.04 × 104, 5.79 × 104, and 1.35 × 104, respectively. CMTA0S1, CMJA0S1, and CMBA0S1 mainly contained galactose (Gal), arabinose (Ara) and glucose (Glc) residues in molar ratio of 1.23:1.00:0.20, 2.18:1.00:0.53, and 3.30:1.00:0.75, while CMTA0S3, CMJA0S2, and CMBA0S3 mainly contained Gal, Ara, Glc, and mannose (Man) residues in molar ratio of 0.73:1.00:0.40:0.21, 1.39:1.00:0.84: 0.55, and 1.19:1.00:0.48:0.19. Methylation analysis indicated that six PCM primarily consisted of T-arabinofuranosyl, 1, 5-arabinofuranosyl, 1, 4-galactopyranosyl, 1, 3, 6-galactopyranosyl, and 1, 4-glucopyranosyl residues. The biological activity study suggested that all the PCM could inhibit the growth of PANC-1 cells. Among them the inhibitory rates of CMTA0S3 and CMJA0S2 were at most to 70% with concentration-effect relationship. The NF-κB inhibition test indicated that only the crude polysaccharide CMBA had strong immunosuppressive activity, and homogeneous polysaccharides CMTA0S1 and CMJA0S1 showed potential immunostimulation. Conclusion: The six homogeneous polysaccharides share similar structures and inhibition on PANC-1 cells growth. Meanwhile they also may regulate the NF-κB activation.

Objective To isolate water-soluble neutral polysaccharides(NTPS) and acidic polysaccharides(ATPS) from tea,investigate their hypoglycemic effects,and elucidate the relationship between structure and activity.Methods Weak base macroporous anion-exchange resin D315 was used to isolate two kinds of tea polysaccharides NTPS and ATPS based on electric charge of tea polysaccharides.Their monosaccharide compositions were analyzed by gas chromatography and ion chromatography.NTPS and ATPS were further isolated by DEAESepharose FF.Molecular weights of the main fractions were analyzed by HPGPC.The model of diabetic mellitus was established by ip alloxan in mice.The blood samples were collected with tail vein puncture to determine blood glucose.The hypoglycemic activity between neutral and acidic polysaccharides was compared after ig tea polysaccharides for 12 d successively.Results Both NTPS and ATPS were(heteropolysaccharides.) NTPS was mainly composed of neutral polysaccharide and its main monopolysaccharide was D-galactose.NTPS contained 6.82% galacturonic acid.Whereas,ATPS was mainly composed of acidic polysaccharide and contained 33.02% D-galactose,whose neutral polysaccharide was mostly(L-)rhamnose,D-arabinose and D-galactose. The molecular weights of both two kinds of tea polysaccharides were less than 3?10~4.Continuous ig administration of NTPS and ATPS [200 and 400 mg/(kg?d)] for 12 d was found to depress glucose increases in alloxan-dependent diabetic model mice.Conclusion Two different structures of tea polysaccharides NTPS and ATPS are isolated by anion-exchange resin D315 firstly.Both tea polysaccharides show good hypoglycemic effect.
