Résumé

Objective To investigate the protective effects of the total bakkenolides from P.tricholobus on improving hypoxia tolerance in mice. Methods Mice normobaric pressure hypoxia model and oxygen glucose deprivation model in PC12 cells were established, and the effects of PTB on survival time, serum lactate dehydrogenase (LDH) activity and malondialdehyde (MDA) content, brain and heart superoxide dismutase (SOD) and reduced glutathione (GSH) activities, brain tissue pathological changes and cell survival were observed. Results The total bakkenolides from P.tricholobus had prolonged the survival time of mice in confined spaces, increased the activity of SOD and GSH, reduced the production of lipid peroxidation, decreased the degree of anaerobic glycolysis, protected the structure and function of neural cells, and improved the survival rate of OGD-treated cells. Conclusion The total bakkenolides from P.tricholobus could promote the hypoxia tolerance in mice which might be related to scavenging oxygen free radicals, inhibiting lipid peroxidation reaction and protecting the structures and functions of nerve cells.

Résumé

A quantitative analysis of bakkenolide D in the different parts of Petasites japonicus and Farfugium japonicum was performed by HPLC. A gradient HPLC elution system with a mobile phase consisting of water:acetonitrile solution (20:80 to 0:100 for 45 min) was followed and an INNO C₁₈ column was used for the chromatographic separation. The injection volume, flow rate, and UV detection were 10 µL, 1 mL/min, and 290 nm, respectively. Results show that both species showed the highest amount of bakkenolide D in the roots being 107.203 and 166.103 mg/g for P. japonicas and F. japonicum, respectively. Content analysis on the different parts of both plants displayed remarkably lower values which ranged from 0.403 – 4.419 and 7.252 – 32.614 mg/g for P. japonicas and F. japonicum, respectively. The results show that the roots of both plants are rich in bakkenolide D showing a promising use in the development of nutraceuticals and industrial application of the compound.

Résumé

A new sesquiterpene, bakkenolide-Ⅵa (1), was isolated from the rhizome of Petasites japonicas (Sieb. et Zucc.) Maxim. The structure was characterized on the basis of various NMR (1H, 13C, 1H-1H COSY, HMQC, HMBC and NOESY) and mass spectrometry data. Bakkenolide-Ⅵa showed potent cerebral hypoxia-ischemia protective activity in mice subjected to decapitation through prolonging the survival time and gasping time. It also exhibited a protective activity against hypoxia injury in PC12 cells in anaerobic culture by inhibition of lactate dehydrogenase (LDH) release.

La rinitis alérgica es una patología frecuente, de alto impacto social, y alteración de la calidad de vida, lo que conlleva al cambio frecuente de la medicación, y al uso de más de un medicamento, sin el control adecuado de la sintomatología. Se evaluó la evolución de los síntomas clínicos en 927 pacientes con historia de rinitis alérgica perenne y estacional sintomática, sin indicación de antibioticoterapia en las dos semanas previas al tratamiento con Extracto estandarizado de hojas de Petasites Hybridus Ze339 (Tesalin®). Los síntomas de la rinitis alérgica fueron registrados en la visitas a sus médicos los días 0, 7, 14 y 28 después de iniciar el tratamiento. Los pacientes con Extracto estandarizado de hojas de Petasites Hybridus Ze339 (Tesalin®) una tableta dos o tres veces al día durante 28 días, se permitió cualquier otra medicación concomitante excepto antimicrobianos. El análisis de efectividad y tolerancia fue realizado por el médico y paciente. Para el médico, el porcentaje de pacientes que se encontraban totalmente libres de síntomas, o éstos eran muy leves,fue de 94.71%, mientras que para los pacientes fue de 95.90%, con una excelente correlación entre ambas opiniones. La tolerancia fue buena para el médico (90.70%) y para el paciente (94,47%) (p 0.95). El extracto estandarizado de hojas de Petasites Hybridus Ze339 (Tesalin®) resultó efectivo y seguro en el tratamiento de los síntomas de la rinitis alérgica no complicada


Allergic rhinitis is a common condition, high social impact, and impaired quality of life, which leads to frequent change of medication, and the use of more than one drug, without adequate control of symptoms. We evaluated the evolution of clinical symptoms in 927 patients with a history of seasonal allergic rhinitis and symptomatic, with no indication of antibiotic therapy in the two weeks before, that receiving standarized extract of leaves of Petasites Hybridus Ze339 (Tesalin®). The symptoms of allergicrhinitis were recorded in the visits to their doctors on days 0, 7, 14 and 28 after starting treatment. Patients were treated with standarized extract of leaves of Petasites Hybridus Ze339 (Tesalin®) one tablet two or three times a day for 28 days, allowed any concomitant medication except antibiotics. The effectiveness and tolerance analysis was performed by both the physician and patient. For physicians, the percentage of patients who were completely free of symptoms it was very mild, was of 94.71%, while for the patients was 95.90%, with an excellent correlation between both. Tolerance was good for the doctor (90.70%) and patients (94.47%) (P 0.95). The standarized extract of leaves of Petasites Hybridus Ze339 (Tesalin®) is effective and safe in treating the symptoms of allergic rhinitis is not complicated

Résumé

Objective: To isolate and identify the chemical constituents in the rhizome of the Petasites ja ponicus. Methods: Dried rhizome of Petasites ja ponicus were subjected to extraction with EtOH, and the chemical constituents in the extract were isolated and their structures were identified by physicochemical properties and spectral analysis. Results: Five compounds were obtained and identified from the rhizome of Petasites ja ponicus, inlcuding as β-sitoterol (I), ergosterol (II), bakkenolide-B (III), bakkenolide-D (IV) and furanofukinol (V). Conclusion: Compound (II) has been obtained from Petasites japonicus for the first time.

The methanol extract from the leaves of Petasites japonicus Maxim (PJ) was studied for its (anti-)mutagenic effect with the SOS chromotest and reverse mutation assay. The (anti-)carcinogenic effects were evaluated by the cytotoxicity on human cancer line cells and by the function and the expression of gap junctions in rat liver epithelial cell. PJ extracts significantly decreased spontaneous beta-galactosidase activity and beta-galactosidase activity induced by a mutagen, ICR, in Salmonella (S.) typhimurium TA 1535/pSK 1002. All doses of the extract (0.08-100 mg/plate) decreased the reversion frequency induced by benzo (alpha)pyrene (BaP) in S. typhimurium TA 98. It decreased not only the spontaneous reversion frequency but also that induced by BaP in S. typhimurium TA 100. PJ extract showed greater cytotoxic effects on human stomach, colon and uterus cancer cells than on other cancer cell types and normal rat liver epithelial cells. Dye transfers though gap junctions were significantly increased by PJ extracts at concentrations greater than 200 microg/mL and the inhibition of dye transfer by 12-O-tetradecanoylphorobol-13-acetate (TPA) was obstructed in all concentrations of PJ. PJ significantly increased the numbers of gap junction protein connexin 43, and increased the protein expression decreased by TPA in a dose-dependent manner. Based on these findings, PJ is suggested to contain antimutagenic and anticarcionogenic compounds.

Objective:To study the chemical constituents in the root of Petasites tricholobus Franch.and evaluate their anti-inflammatory activity.Methods: The primary extraction was done with 95% ethanol and subsequently with other agents including ethyl acetate.The compounds extracted with ethyl acetate were isolated by silica gel column chromatography and purified by means of recrystallization.The structures of the compounds were elucidated by employing chemical and spectral methods.The anti-inflammatory activities of the compounds were evaluated through their inhibitory effect on the trachea contraction of guinea pig induced by histamine in vitro.Results: Six compounds were isolated from the root of Petasites tricholobus Franch.,namely,homofukinolide(1),bakkenolide-B(2),stigmasterol(3),and ?-sitosterol(4),bakkenolide-D(5),and ?-sitosterol-3-O-?-D-glucopyranoside(6).Bakkenolide-B and bakkenolide-D demonstrated inhibitory effect on the trachea contraction induced by histamine in vitro.Conclusion: Homofukinolide,?-sitosterol,and ?-sitosterol3-O-?-D-glucopyranoside are isolated for the first time from Petasites tricholobus Franch..Bakkenolide-B and-D have obvious anti-histamine activities.
