Fine particulate matter (PM2 5) with diameters of less than 2.5 |xm can lead to many diseases in human.According to the results of epidemiological studies, PM25 can injury human reproductive system, hut the mechanism of reproductive injury is rarely reported.This study aimed to investigate the protective effect and mechanism of curcumin (CRC) on short-term exposure to PM2 5-induced rat uterine injury.Fifty Sprague-Dawley rats were randomly divided into control group, P.\l2 5 exposure group, PM15 +CRC-L group, PM2 5+CRC-H group and PM2 5 + VE group, which all were administered for 30 days.After a short-term exposure, PM2 s induced atrophy of endometrial epithelial cells and structural damage of endometrial glands and stromal cells.At the same time, these conditions were improved after administered with curcumin and VE.The results of TUNEL detection showed that the cell apoptosis rate (48.81 ±8.27)% in PM2 s exposure group was significantly higher than that in control group (P5+CRC-H group (P<0.05).Compared to control group, the levels of reactive oxygen species (ROS) were significantly higher (P< 0.05) and the level of total antioxidant capacity (T-AOC) was significantly lower (P<0.05) in PM15 exposure group.However, curcumin and VE could attenuate PM2 5- induced oxidative stress in rat uterus.CRC and VE inhibited proteins phosphorylation of ASK 1, JNK, p38 and activation of caspase-3 induced by PM25(P<0.05).Moreover, curcumin significantly inhibited the protein expression levels of p-ASKl, p-JNK, p-p38 and cleaved caspase-3 induced by PM25.In conclusion, curcumin attenuates short-term exposure to PIV12 5-induced uterine injury by inhibiting the signal pathway of JNK-p38-caspase-3 mediated by ASK 1.

Objective To explore the reproductive toxicity of 2,4-D butylate to the testis in male mice.Methods Forty-eight ICR male mice were randomly divided into four groups : the control group, and three 2,4-D butylate experimental groups (10, 20, 40 mg/kg), 12 mice in each group.2,4-D butylate was intragastrically administered once a day and six days per week for five weeks .At the end of the exposure, the activities of total antioxidant capacity (T-AOC), Na+ K+-ATPase, Ca+ + Mg+ +-ATPase, lactate dehydrogenase (LDH) and succinate dehydrogenase (SDH) in testis homogenate were measured by spectrophotometry .Results The activity of T-AOC was gradually decreased with the increase of doses, with a significant difference between the high dose group and other groups .The activities of LDH in the moderate and high dose groups were significantly lower than those of the low dose group and control group , and there was a significant difference between the high dose group and moderate dose group .The activities of SDH in the testis was gradually decreased with the increase of the 2,4-D butylate dose, showing significant differences between the high dose group and the moderate dose and control groups , and between the high and moderate dose groups and the low dose group . The activities of Na +K +-ATPase in the moderate and high dose groups were significantly lower than that of the control and low dose group.The activities of Ca++Mg++-ATPase was significantly lower in the experimental groups than that in the control group.Conclusion Exposure to 2,4-D butylate has certain toxic effect on the testicular tissue in male mice .

Objective:Through animal experiments to observe the behavior and serum malondialdehyde(MDA)and total antioxidative capacity(T-AOC)content after electro-acupuncture "Baihui,Fengfu,Shenshu,Xuanzhong" Point of Alzheimer, s disease in rat.Method:Select forty Healthy adult Wistar rats,the Wistar rats were randomly divided into four groups,model group,electro-acupuncture group,western medicine group,electro-acupuncture and western medicine group.Abdominal cavity injection of D-galactose in order to prepare the Alzheimer, s disease model rats.Electro-acupuncture group:acupuncture Baihui,Shenshu, Fengfu,Xuanzhong.Western medicine group:Piracetan intragastric administration one times a day.Every other day give the fixed and crawl for ten minutes.Electro-acupuncture and western medicine group:Accupuncture treatment and drug treatment are above all.Model group:Every day give the same fixed and crawl as electro-acupuncture group.Behavior and serum MDA,T-AOC levels tested after treatment.Results:electro-acupuncture group,electro-acupuncture and western medicine group could extend the incubation period,reduce the number of errors(P

Objective To investigate the protective effect of Ca 2+ channal blocker(Verapamil)on the testis following testicular torsion in rats.Methods Forty healthy male Sprague-Danley rats were randomly divided equally into five groups:group A(sham operation),group B1(the left testis underwent 720? positive torsion,2 h later detorsion was performed),group B2(the process as group B1 except at 30 min before detorsion,intraperitoneal injection of physiological saline was performed),group B3(the process as group B1 except at 30 min before detorsion,intraperitoneal injection of 2 mg/kg Verapami was performed),group B4(the left testis underwent 720? positive torsion,and 2 h later the left testis was excised).Five days after operation,the bilateral testes(the right testis in group B4)were extracted and nitric oxide(NO),nitric oxide synthase activity(NOS),total antioxygen capacity(T-AOC)were measured.Results After detorsion,NO and NOS activity increased greatly,T-AOC decreased significantly in bilateral testes,but more obvious in the torsive testis;Verapamil could inhibit the changes.Conclusion The excessive production of NO and the decreased T-AOC may be the main causes of the contralateral testicular injury after unilateral testicular torsion.Verapamil can reduce the testicular injury in bilateral testes after testicular torsion/detorsion in rats.The resection of the torsive testis has protective effect on the contralateral one.

Aim To investigate the protective effects and mechanisms of a 5-lipoxygenase (5-LO) inhibitor zileuton on the acute lung injury (ALI) induced by focal cerebral ischemia-reperfusion injury (CIRI) in rats.Methods The right middle cerebral artery in rats were occluded by inserting a thread through internal carotid artery for 2 h,and then reperfused for 24 h.Zileuton(10,50 mg?kg-1) was orally administered 2h before ischemia and 0,5,10 h after reperfusion.Morphological changes of the lung tissue were observed by hematoxylin-eosin (HE)staining technique. Lung wet/dry weight ratios were detected to observe the water content of the lung. The mRNA of cysteinyl leukotrienes recepor1 (CysLTR1) was detected by RT-PCR. The content of Leukotriene B4 (LTB4) in serum was measured by enzyme linked immunosorbent assay (ELISA) technique. The expression of TNF-? protein was measured by immunohistochemical technique. The activities of MPO, T-AOC, and Na+,K+-ATPase from the lung tissue were measured by biochemical method.Results With the use of zileuton 10, 50 mg?kg-1,the pathological changes of the lung were alleviated. and the wet/dry weight in the lung tissue was reduced(P

AIM: To study the effect of total flavonoids of Pongamia pinnata roots(PRF) on ulcerative colitis(UC) induced by dextran sulphate sodium(DSS) in mice and the possible mechanism. METHODS: Balb/c mice were fed with 4% DSS solution for 7 d to induce ulcerative colitis and treated with sulfasalazine(SASP) as positive control.The clinical symptom and the lesion of colonic mucosa were observed and the levels of T-AOC、MDA、NO and the activity of NOS in colonic tissue were tested. RESULTS: Compared with the model control,the clinical symptom and the lesion of colonic mucosa in treatment group were remarkably improved after administration of PRF in large or medium dose.In the two groups,the decrease in the levels of MDA,NO and the activity of NOS,and the increase in the levels of F-AOC could be found.(P0.05). CONCLUSION: The results indicate that significant effect of PRF on DSS-induced UC may attributae to reduction in free radical,depression of lipid peroxidation and enhancement of antioxidant capability.

Objective: To study the effect of free radical injury induced by Soman in rats and protection by Se/Zn. [WT5FZ]Methods: [WT5BZ]40 male rats were randomly divided into 4 groups, i.e., negative group, positive group, Se group and Zn group according to weights. The serum, cerebrum and liver VE content, nitrogen oxide synthase (NOS activity and T AOC (total antioxidative capacity) were determined. Results: The VE content and T AOC decreased markedly, and NOS activities increased significantly after Soman intoxication. The changes were less significant in Se/Zn supplemented group. Conclusion: The mechanism of free radical injury is indicated in Soman intoxication and Se/Zn supplementation has significant protection.
