Résumé

Objective To evaluated the effect of Total Glucosides of Paeony (TGP)combined with urokinase for the patients with acute cerebral ischemic stroke.MethodsA total of 152 patients with ACIS were divided into the control group (77 patients) and the observation group (75 patients) by completely randomized grouping method. Patients in the control group were treated with Urokinase by intravenous, and the observation group were treated with TGP combined with Urokinase. All patients were treated with thrombolysis treatment with in the 24 h after the exclusion of cerebral hemorrhage by brain CT and coagulation mechanism disorder. In the following 14 days, they were given the treatment of anti platelet aggregation, lowering blood lipid, trophic nerve and improving circulation. The National Institutes of Health Stroke Scale(NIHSS), the modified Rankin scale(mRS) and the daily life ability index(Barthel) were measured.ResultsThere were 28 (37.3%) patients with the NIHSS scores decreasing 4 points in observation group and 12 (15.6%) in control group after treatment of 7 days,and there was significant difference between two groups (χ2=8.181,P=0.004). There were 68 (90.7%) patients with mRS score less than 4 points in observation group and 51 (15.6%) in control group after treatment of 14 days,and there was significant difference between two groups (χ2=11.945,P=0.001). The NIHSS scores in the observation group was lower than control group (8.3 ± 2.2vs. 9.9 ± 2.5;t=4.192,P<0.05). The mRS score in the observation group was lower than control group (2.3 ± 0.9vs. 2.9 ± 1.1;t=3.684,P<0.01), and the Barthel index in the observation group was higher than the control group (88.7 ± 16.2vs. 77.5 ± 15.2;t=4.401,P<0.01). ConclusionsThe TGP capsules combined with Urokinase intravenous thrombolysis could improve the neurological deficit, and the ability of daily life of ACIS patients.

Objective To study the effect of resveratrol on hepatitis B virus (HBV) and hepatic fibrosis.Methods The rat model of HBV infection was established by intraperitoneal injection of rAAV8-1.3HBV.After successfully establishing the modle, were randomly divided into four groups:negative control group, resveratrol low dose group, high dose group, positive control group, and five rats in each group.Giving a physiological saline, 20 mg/kg respectively resveratrol, 40 mg/kg, 0.1 mg/kg acyclovir via gastrogavage respectively.2、4、8、12 weeks later ,we detected the HBsAg, HBeAg, HBV-DNA via tail vein blooding.HSC-T6 cell model was established.Then rats were divided into two groups, control and resveratrol serum group.Each group was administrated with normal sodium and resveratrol via gastrogavage to make serum.The HSC-T6 viability was observed by AlamarBlue assay.The expression of Col I and TGP-β1 mRNA was determined by Realtime PCR.The expressions of Col I and TGP-β1 Protein was analyzed by WesternBlot.Results Using ELISA method to detect HBsAg、HBeAg、HBV-DNA and found that the expression of HBsAg and HBeAg of negative control group were significantly higher than the rest of the three groups (P<0.05) at the second and the fourth week.The highest expression of HBsAg and HBeAg were (4118 ±367) IU/mL、(160.2 ±56.90) NCU/mL at the second week.The expression of HBV-DNA of negative control group is significantly higher than the rest of the three groups (P<0.05) at the second week.AlamarBlue assay indicated that different concentration of serum of resveratrol can inhibit HSC-T6 proliferation.Compared with control group, serum with drug resveratrol significantly down-regulated the mRNA and protein expression of Col I and TGP-β1.Conclusion Resveratrol inhibits HBV and liver fibrosis by inhibiting type I collagen and TGF-β1.

OBJECTIVE: To prepare the total alkaloids of Strychni Semen(TASS)-total glucosides of Paeony(TGP) gel and study its in vitro transdermal absorption of two alkaloids(strychnine and brucine) after the combination of TASS and TGP. METHODS: The excised abdominal skin of mice was used as the permeation model. Utilizing the modified Franz diffusion cell, the suitable receiving solution was elected to test the content of two alkaloids by HPLC, and thus the percutaneous rates and permeability coefficients were obtained. RESULTS: 20% Ethanol-normal saline was taken as receiving solution. With combination use of TASS and TGP, the penetration quantities of strychnine and brucine in different (1:1, 1:3, 1:6) gels were felled by 22.7%, 48.4%, 69.1% and 5.93%, 23.8%, 80.7% after 24 h. And with the increase of compatibility proportion, infiltration rate and skin retention rate also gradually reduced. CONCLUSION: The compatibility of TASS and TGP drug delivery can reduce the toxic ingredients through capacity, there is a"attenuated"effect, the best ratio is 1:6.

Introducción: la enfermedad hepática tóxica es un problema desconcertante debido al amplio número de sustancias capaces de inducir este tipo de reacciones. Datos epidemiológicos y estadísticos demuestran que la Mimosa púdica conocida como moriviví, sensitiva, adormidera es una de las plantas (objeto de abuso) más empleadas como adulterante en los pitillos de marihuana en pacientes drogadictos. Objetivos: evaluar el comportamiento de los parámetros bioquímicos y describir los hallazgos anatopatológicos en el hígado de ratas por efecto de la planta Mimosa púdica. Métodos: se realizó un estudio preclínico de hepatotoxicidad aguda con la planta Mimosa púdica en animales de experimentación. Se seleccionaron 25 ratas Sprague Dawley, cinco por cada grupo (control positivo, control y tres grupos experimentales) de experimentación. Se administró una dosis de 500, 1000 y 2000 miligramos por kilogramo de peso corporal de la decocción de la planta a los grupos experimentales respectivamente; tetracloruro de carbono a dosis de 3 mililitros por kilogramo de peso corporal al control positivo y agua estéril al control. Resultados: aumento de los niveles de transaminasas (TGP y TGO) en los grupos experimentales con respecto al grupo control. Las alteraciones anatomopatológicos observadas fueron: necrosis focal periportal, congestión de venas centrolobulillares y degeneración acidófila del hígado. Conclusiones: el consumo de la decocción de la planta Mimosa púdica afecta el hígado, lo que provoca alteraciones bioquímicas y anatomopatológicas, de probable importancia en pacientes drogadictos, aunque se requieren más estudios.


Introduction: toxic liver disease is a concerning problem due to the large number of substances that cause such reactions. Epidemiological and statistical data show that Mimosa (Moriviví), known as sensitive and opium poppy is one of the most used plants by drug-addict patients, as an adulterant in marihuana cigarettes drug. Objectives: to evaluate the behavior of biochemical parameters and describe pathological findings in the liver of rats as a result of Mimosa pudica plant. Methods: a preclinical study of acute hepatotoxicity with Mimosa pudica plant in experimental animals was performed. Twenty five Sprague Dawley rats, five per group (positive control, group control and three experimental) were selected for the study. A dose of 500, 1000 and 2000 mg administered per kilogram of body weight of the decoction of the plant to the experimental groups respectively; carbon tetrachloride at doses of 3 milliliters per kilogram of body weight to the positive control and sterile water to control. Results: the results showed an increase of levels of transaminases (SGOT and SGPT) in the experimental groups compared to the control group. The pathological changes observed were focal periportal necrosis, congestion and centrilobular veins acidophilic degeneration of the liver. Conclusions: the consumption of the decoction of the plant Mimosa pudica affects the liver, causing biochemical and pathological alteration of likely importance in drug patients, although further studies are required.

Objective to investigate the effect of TGP assisted treatment of SLE on the expression of CD4+CD25+ T in patient peripheral blood .Methods flow cytometry was used to detect the peripheral blood CD4+ CD25+ T cells in healthy group ,routine group and TGP group .Results The expression rate of CD4+CD25+ T cells in SLE patients was (6 .15 ± 1 .21)% ,and that of the healthy controls was (12 .30 ± 1 .78)% .The expression rate of CD4+CD25+ T cells in active SLE patients and healthy controls were significantly different (t=22 .03 ,P<0 .05) .In the routine group ,the expression rate of peripheral blood CD4+CD25+ T cells before and after treatment were significantly different (t= 12 .30 ,P<0 .05);in the TGP group ,the expression rate of peripheral blood CD4+ CD25+ T cells before and after treatment were significantly different ,too (t=16 .68 ,P<0 .05) .The expression rate of periph‐eral blood CD4+CD25+ T cells in routine group and TGP group were (9 .34 ± 1 .37)% and (11 .49 ± 1 .14)% respectively ,and the difference was statistically significant (t=6 .46 ,P<0 .05) .Conclusion The expression rate of CD4+ CD25+ T cells in active SLE patients was significantly lower than that of healthy controls ;the expression rate of CD4+CD25+ T cells in SLE patients increased significantly after treatment .The TGP treatment may work on CD4+CD25+ T cells .

OBJECTIVE: To investigate total glucosides paeony (TGP) on the expression phosphorylated extracellelar signal regulated kinase 1/2(p-ERK1/2), Toll-like receptors 4(TLR4) and Toll-like receptors 9(TLR9) in rats with nonalcoholic fatty liver disease (NAFLD) induced by fructose and high-fat feed. METHODS: SD rats were divided into normal group and test groups. The rats in test groups were fed with fructose and high-fat feed for 10 weeks totally to induce the test model. After 6 weeks the model was established, the rats were divided into four groups randomly, the model group (NAFLD), the metformin group (Met, 200 mg · kg-1), the low-dose TGP group(TGP-L, 100 mg · kg-1) and the high-dose TGP group(TGP-H, 200 mg · kg-1) (n=10). Four weeks later all the rats were killed and checked the indexes such as serum fasting blood glucose(FBG), fasting insulin(Fins), insulin sensitivity index (ISI), total cholesterol (TC), low-density lipoprotein (LDL-C), high-density lipoprotein (HDL-C), triglyceride (TG), free fatty acids (FFA), alanine aminotransferase (ALT), aspartate aminotransferase (AST), glutathione S-transferase (GST) and liver index. The expression p-ERK1/2, TLR4 and TLR9 were inspected by Western-blot. RESULTS: Compared with the normal group, the rats in test groups were with the high levels serum FBG, insulin, TC, LDL-C, TG, FFA, ALT, AST, GST, liver index, p-ERK1/2, TLR4 and TLR9 (P 0.05). CONCLUSION: By downregulating the expression ERK1/2, TLR4 and TRL9, TGP can improve abnormal glucose and lipid metabolism and insulin resistance, enhance insulin sensitivity and ameliorate liver function in rats with NAFLD induced by fructose and high-fat feed.

La coledocolitiasis puede ocurrir hasta en el 15% de los casos de colelitiasis. En la era moderna de la colecistectomía laparoscópica se ha planteado un debate sobre el manejo de la coledocolitiasis previo a la realización del procedimiento quirúrgico. Varios estudios han investigado el uso de criterios preoperatorios, como las pruebas de función hepática, los hallazgos ecográficos y la clínica del paciente, para predecir el riesgo de cálculos en el colédoco. Los resultados varían un poco entre los estudios y de ellos se han desprendido diferentes esquemas y algoritmos para orientar el manejo, dentro del cual se incluyen técnicas tradicionales de cirugía abierta y las endoscópicas y laparoscópicas de más reciente aparición.


Choledocholithiasis may occur in up to 15% of cases of cholelithiasis. A debate has been posed in the modern era of laparoscopic cholecystectomy on how to handle choledocholithiasis before conducting the surgical procedure. Several studies have analyzed the use of preoperative criteria, such as liver function tests, ultrasound and clinical findings of the patient, in order to predict the presence of gallstones in the common bile duct. Results vary between studies and different regimes and algorithms to guide management have derived from them, including traditional open surgery techniques and most recent endoscopic and laparoscopic techniques.

Objective To determine the influence of total glucosides of paeony (TGP) on bone density, protein level of skeletal muscle and relative cytokine of osteoporotic rats. Methods Six-month-old SD rats were shamed as control (8 rats) or were ovariectomized (24 rats). AIM: To investigate the effects of TGP on immunologic changes associated with CJ-S131-induced SLE-like model in mice. METHODS: SLE-like syndrome was induced by injection of CJ-S131 and CFA. Various doses of TGP were given by gavage once daily for 28 days. RESULTS: The levels of IgG anti single-stranded DNA(ssDNA) and histone autoantibodies were increased in sera four weeks after injection. The proliferation response of splencytes to ConA and LPS and the production of IL-1 were also increased. TGP(50,100,200 mg·kg-1·d-1, ig) inhibited the aboved changes of SLE-like mouse model partly or completely. CONCLUSION: The SLE mouse model was similar to idiopathic SLE in humans. TGP has modulating effects on alternations of immunity of SLE-like mouse model.

Objective To prepare totel glucosides in paeony(TGP) pellets by extrusion-spheronization and to optimize the formulation of TGP sustained-release pellets coated with Eudragit RS 100.Methods The TGP sustained-release pellets were prepared by mini Glatt fluidized bed coating method.The factors to influence the drug release behaviors and their properties were evaluated.Results The optimal TGP sustained-release pellet was shown significant sustained-releasing at the loading weight of copolymers of 6% and the plasticizer concentration of 10%.And the curves of cumulative drug release were fit for Peppas and Higuchi equation.Conclusion The pellets showed an obviously sustained-release effect.

Résumé

The inhibitory effects of total glucosides of Moutan Cortex (TGM) and to talglucosides of Paeony Lactiflora (TGP) on osmotic hemolysis and oxidative hemolysis induced by hydrogen peroxide were studied. TGM at lower concentration and TGP could inhibit osmotic hemolysis significcantly, but higher concentration (100mg ? L-1) of TGM had a hemolytic activity. TGM and TGP could inhibit hemolysis induced by H2O2 significantly,but TGM was more effective than TGP. TGM .TGP and Vit E all could inhibit lipid peroxidation and glutathione depletion, induced by H2O2, on erythrocytes.

In long-term toxicity test, 50, 1000 and 2000mg/kg?d-1 ig on 30~90 consecutive days to rats and 280 and 560mg/kg?d-1 ig on 90 consecutive days to dogs respectively, revealed no significant toxicity in general physical signs, body weight, appearance. behavious, routine urine test, routine blood test, liver and kidney function tests, histological examinations of important organs, blood biochemistry tests and electrocardiogram, except that the platelet numbers of both animals increased.

Résumé

The total glucosides of paeony root ( TCP ) extracted from paeony root of Boxian county, Anhui province was studied on the immunoregulatory effects in mice.The delayed hypersensitivity (DH) was increased by TGP at dose of 200mg/kg/d ig for 8d in the normal 6l5 mouse. Both depressed DH & enhanced DH induced by cyclophosph-amid ( Cy ) were restored by TGP ( 5 mg/kg/d ip x 5 - 8 d) in ICR mice. The decreased hemolysin antibody formation of spleen induced by Cy in BALB/c mice immunized with SRBC was raised by TGP ( 5mg /kg/d ip x 4d) . The phagocytic activity of ICR mice peritoneal macrophage on chiken RBC . In addittion, it was considerably Stimulated by using ip TGP at dose of 40mg/kg/d for 3d. The results suggested that TGP might possess the immunoregulatory effects in mice.

Total glucosides of paeony (TGP, 50 mg/kg 7d, ig) could enhance the episode duration of slow-wave sleep (SWS) in normal rats, and restore the sleep parameters in the insomniac rats induced by caffeine ( 1 2. 5 mg/kg 7 d,ip)nearly to the normal level. It (50 mg/kg 3 d,ig) also increased significantly thetotal time of SWS and paradoxical sleep (PS) in the swimming rats (water temperature 25?1℃, swimming time 30 min). These results suggest that TGP probably improve the sleep of rats under the different states.
