Abstract@#Introduction: Arterial and venous thromboses contribute to significant morbidity and mortality rate, thus an antithrombotic agent is needed for prevention and treatment of thrombosis. Ajwa dates (Phoenix dactylifera L.) reportedly contain a high level of salicylic acid which is a compound responsible for anticoagulation via antagonism of vitamin K. The present study was designed to assess coagulation activities in human plasma treated with Ajwa date extracts in vitro. Methods: Platelet-poor plasma samples from 27 donors were treated with ethanol crude date extract (ET) or aqueous crude date extract (AQ) of Ajwa dates at different concentrations to generate the following seven test groups from each donor: control (normal saline), ET-I (0.1 g/mL), ET-II (0.5 g/mL), ET-III (1.0 g/mL), AQ-I (0.1 g/ mL), AQ-II (0.5 g/mL) and AQ-III (1.0 g/mL). In vitro coagulation activities of Ajwa dates were assessed based on prothrombin time (PT, an assessment of extrinsic coagulation pathway), activated partial thromboplastin time (APTT, an assessment of the intrinsic coagulation pathway), and thrombin time (TT, an evaluation of level and function of fibrinogen). Results: A very significant prolongation of PT, APTT and TT were observed for the ET-II and ET-III groups and very significant prolongation of PT and TT was observed for the AQ-II and AQ-III groups. Significant prolongation of TT was observed in the AQ-I group. Conclusion: In conclusion, Ajwa date extracts had an anticoagulation effect on human plasma.

Objective To compare coagulation data measured with domestic produced and imported coagulation testing solutions on SD rat and human by testing prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time(TT), fibrinogen(FIB).Methods Blood samples were obtained from SPF SD rat and human .Domestic produced and imported coagulation testing solutions were applied to test PT , APTT, TT, and FIB.Results Compared to rat data measured with imported coagulation testing solution , data measured with domestic produced coagulation testing solution of PT, APTT, FIB was significantly higher (P<0.05), while, data of TT was statistically lower(P<0.05), and there was no obvious difference in human blood coagulation .Conclusion The data measured with different coagulation testing solution varies on SD rat , so the laboratories are required to establish reference data according to different products .

Objective To study the effect of Tibet medicine of Herba Lamiophlomis rotata on promoting hemostasis and blood coagulation and to explore its mechanism. Methods High dose(2 g/kg),middle dose(1 g/kg) and low dose (0.5 g/kg) of the aqueous extract from Herba Lamiophlomis rotata(HLRE)were given to mice by gastric gavage for 3 days,and then bleeding time (BT),clotting time(CT) and platelet count(PLC)were determined. Different doses of HLRE(3,1.5,0.75 g/kg)were administered by gastric gavage to rats for 14 days,and then blood samples were collected from common carotid artery for the determination of prothrombin time (PT),activated partial thromboplastin time (APTT),thrombin time(TT),fibrinogen(FIB),hepatic function and blood lipid indexes. Results Compared to the blank control group,the bleeding time and clotting time were obviously shortened in 2g/kg HLRE and 1g/kg HLRE groups and positive control group,but the difference of PLC was insignificant. PT and APTT values in all of the treatment groups did not differ from those in the blank control group. However, TT values were obviously shortened in 3 g/kg HLRE group,FIB and Albumen(Alb)values increased,the aminotransferase and blood lipid values tended to decrease. Conclusion The aqueous extract from Herba Lamiophlomis rotata has an effect on promoting hemostasis and blood coagulation ,and its mechanisms may be related to the increase of FIB and Alb contents and the shortening of TT value.
