Résumé

l-Heptopyranoses are important components of bacterial polysaccharides and biological active secondary metabolites like septacidin (SEP), which represents a group of nucleoside antibiotics with antitumor, antifungal, and pain-relief activities. However, little is known about the formation mechanisms of those l-heptose moieties. In this study, we deciphered the biosynthetic pathway of the l,l-gluco-heptosamine moiety in SEPs by functional characterizing four genes and proposed that SepI initiates the process by oxidizing the 4'-hydroxyl of l-glycero-α-d-manno-heptose moiety of SEP-328 ( 2) to a keto group. Subsequently, SepJ (C5 epimerase) and SepA (C3 epimerase) shape the 4'-keto-l-heptopyranose moiety by sequential epimerization reactions. At the last step, an aminotransferase SepG installs the 4'-amino group of the l,l-gluco-heptosamine moiety to generate SEP-327 ( 3). An interesting phenomenon is that the SEP intermediates with 4'-keto-l-heptopyranose moieties exist as special bicyclic sugars with hemiacetal-hemiketal structures. Notably, l-pyranose is usually converted from d-pyranose by bifunctional C3/C5 epimerase. SepA is an unprecedented monofunctional l-pyranose C3 epimerase. Further in silico and experimental studies revealed that it represents an overlooked metal dependent-sugar epimerase family bearing vicinal oxygen chelate (VOC) architecture.

COVID-19 has been reported to have caused more than 286 million cases and 5.4 million deaths till date. COVID variants have appeared at regular intervals—alpha, beta, gamma, delta and now omicron. ‘Omicron’ is driving the current surge of cases in most countries including India and is poised to replace ‘delta’ the world over. This variant with more than 50 mutations is phylogenetically very diferent from other variants. The omicron variant spreads rapidly with an average doubling time of two days. The disease so far has been mild as compared with delta. Though previous infection and vaccination ofer little or no protection against infection with omicron, they do seem to partially protect against hospitalization and severe disease. Booster vaccinations have not made any notable impact on the spread of omicron and have further worsened global vaccine equity. The indirect consequences of omicron from lockdowns, restrictions, travel bans, economic losses, health care worker infections and overwhelming of health care facilities are likely to be enormous. The direct efects of omicron on children are expected to be mild like with the previous variants. However, the indirect efects on child mental, physical, and social health may be considerable owing to school closures, missed vaccinations, neglect of other diseases, etc. It is, therefore, imperative that governments take rational decisions to navigate the world through this latest crisis.

The unique characteristics of 2-dimensional hetero structure offers efficient gas sensing with high selectivity to identify gases from the interference gases which is quite difficult. In the present work, ZnO: SnO2Nano composite clusters (NCC) is prepared. A resistive metal oxide volatile organic compound (VOC) gas sensor is fabricated with nullifying the effect of humidity by increasing temperature optimally. A single-step SOL-GEL (SG) synthesis is used to prepare ZnO: SnO2 NCC with maximum Zn/Sn molar concentration ratio of 3. The morphological studies through Scanning Electron Microscopy (SEM), electrical properties due to oxygen vacancies and energy band variations of Nanocomposite are measured. The enhancement of gas sensor sensitivity due to highly mesoporous nature of the composite is observed. From the findings, the abundant mesopores in the range of 2 nm-14 nm and specific surface area of 54.2 m2 g?1 with the average crystal size of 14.236 nm, and polar surface area of the composite 25.9651Åis achieved. When compared to bare ZnO and SnO2 gas sensors, the present gas sensor offers the higher selectivity with enhanced performance due to the mesoporous structure. Fast repeatability rate of 2200 sec at 350?C to ethanol is attained and the overall selectivity of the sensor increased twice as 2.085. The NCC compound is tested firstly with micro organisms such as B. subtilis (B. S), Bacillus cereus (B. C), B. coagulans (B. C), Pseudonymous auriginosa (P. A) are considered for antimicrobial activity. From the findings, zinc stannate compound showed good efficacy towards B. cereus Gram positive and P.A gram-negative. A bacterial growth is arrested highly with B. cereus.

Objective:To analyze and summarize the epidemiological and molecular characteristics of SARS-CoV-2 Delta variant, a variant of concern (VOC), in Henan Province in 2021 in order to provide a basis for epidemic prevention and control.Methods:According to the feedback of sequencing results from Chinese Center for Disease Control and Prevention, 111 patients infected with SARS-CoV-2 Delta VOC were selected from the Henan imported and local cases in 2021. Basic patient information was obtained from the pandemic website. The differences in age, gender, vaccination history, the number of vaccine doses and different clinical types were analyzed. Moreover, the differences in RT-qPCR results of ORF1 ab gene and N gene Ct values between cases of different genders and symptoms were analyzed statistically. Sequencing results of the nucleotide and S protein mutation sites were analyzed. Results:There was no significant difference in the gender distribution of 111 cases between different age groups (χ 2=2.217, P=0.529). There was also no significant difference in clinical types between patients with different vaccination history (χ 2=12.074, P=0.209). The Ct values of most SARS-CoV-2 nucleic acid-positive specimens were distributed in the lower range and the viral loads were higher. The difference in the Ct value of ORF1 ab gene between different gender groups was not statistically significant (χ 2=1.646, P=0.439), but were significantly different among asymptomatic, mild, normal, and severe cases (χ 2=13.257, P=0.039). There was no significant difference in N gene Ct value among cases of different genders or different symptoms (all P>0.05). The 111 patients in this study were mainly found through close-contact screening and full-staff nucleic acid screening and accounted for 62.2% (69 cases) of the total. The sequencing length coverage was basically greater than 99% (accounting for 90.1%, 100/111); the total number of nucleotide mutation sites was mostly in the range of 46-50 (86.4%, 89/103); the total number of S protein mutation sites was mostly 12 (82.5%, 85/103). The 103 Delta mutants all contained nine mutation sites, which were T19R, R158G, L452R, T478K, D614G, P681R, D950N, E156del and F157del, with a mutation rate of 100%. Conclusions:People were highly susceptible to the SARS-CoV-2 Delta in Henan Province in 2021. High viral load and increase in the ORF1 ab gene load would aggravate the clinical symptoms.

Résumé

A new variant of coronavirus B.1.1.529. appeared on the scene, when discovered by the researchers in South Africa on Nov 24.2021. It is a heavily mutated variant of coronavirus discovered thus far with over 50+ mutations with 32 mutations over the spike protein itself. Spike proteins help the virus to bind to the bodily receptors of humans to gain entry inside. In comparison to the delta variant, which had nine mutations, it means that Omicron has better chances of evading the host immunity and is also more transmissible. And rightly so, it has been declared as a variant of concern(VoC) by the WHO. The presence of S- gene is of the determinants for the detection of the virus. But omicron seems to have missed this gene- being called as S-gene dropout or S-gene target failure (S- spike glycoprotein)

Cigarette smoke is a major risk factor for several diseases, including chronic obstructive pulmonary and cardiovascular diseases. The toxicity of the cigarette smoke can be determined in vitro. The cytotoxicity test of the cigarette smoke is commonly conducted using the cigarette smoke condensate (CSC) and cigarette smoke extract (CSE). The CSC and CSE methods are well known for sampling of the particles and water-soluble compounds in the cigarette smoke, respectively. In this study, the CSC and CSE were analyzed by using a gas chromatography-mass spectrometry (GC-MS) system equipped with a wax column for separation of the volatile organic compounds. The cytotoxic effect of the CSC and CSE were evaluated thoroughly by comparing the analytical results of the CSC and CSE samples. The total concentration of the volatile organic compounds detected in the CSC sample was similar to that in the CSE sample based on the peak area. Except for the dimethyl sulfoxide solvent, nicotine had the highest concentration in the CSC sample, while acetonitrile had the highest concentration in the CSE sample. The compositions were as follows: (1) CSC sample: 55.8% nicotine, 18.0% nicotyrine, 3.20% 1,2,3-propanetriol, triacetate, 1.28% ethyl chloride, 1.22% phenol, etc. and (2) CSE sample: 18.7% acetonitrile, 18.0% acetone, 12.5% 2-hydroxy-2-methyl-propanenitrile, 8.98% nicotine, 5.86% nicotyrine, etc. In this manner, to accurately examine the cytotoxicity of the cigarette smoke using CSC or CSE, the components and their concentrations in the CSC and CSE samples should be considered.

ABSTRACT The profile of volatile organic compounds, the glandular and non-glandular trichomes of Plectranthus ornatus, obtained by in vitro cultivation, was evaluated in plants grown in Murashide and Skoog medium supplemented with benylaminopurine at 4.5, 9.0, and 18.0 µM + naphthaleneacetic acid at 5.37 µM, kinetin at 4.7, 9.3 and 18.5 µM + naphthaleneacetic acid (5.37 µM) or Murashide and Skoog 0 medium (as a control). Scanning Electron Microscopy was performed on samples of the third leaf node of the 90 days old plants obtained from treatment with 4.5 or 9.0 µM benylaminopurine, and 4.7 or 9.3 µM kinetin. Headspace Solid Phase Micro-Extraction of the 30, 60 and 90 days old in vitro plants permitted to determinate by GC/MS the composition comprised of 62 compounds. The data were analyzed using Principal Component Analysis and Hierarchical Clustering Analysis and, the major constituents of these oils after treatment and aging were monoterpenes and sesquiterpenes. Morphoanatomical analysis of trichomes, by Scanning Electron Microscopy, enabled the identification of non-glandular trichomes and four types of glandular trichomes, which comprised capitate and peltate glandular trichomes that were distributed on both sides of the leaf. We observed that the regulators influenced qualitative and quantitative profiles of the volatile organic compounds and the number and distribution of hairs on the leaf surface.

The floral volatile chemicals of four Heliotropium (Boraginaceae) species endemic to Chile are described and their relationship with an Heliotropium phylogeny assessed. Twenty-two compounds were identified; the most prominent family of compounds in H. floridum, H. myosotifolium and H. stenophyllum were benzenoids, and in H. pycnophyllum prominence of benzenoids was shared with two monoterpenes, (E)-Ocymene and 1,8-cineol. The similarity between chemical mixtures of the species significantly decreased with increasing phylogenetic distance between them. Neighbor-joining analysis produced a tree topology with one clade comprised by H. myosotifolium and H. stenophyllum and an unresolved relationship between this clade and H. floridum and H. pycnophyllum; this topology bears a close relationship with molecular phylogeny proposed for the group. These preliminary results presented suggest that the genus Heliotropium complies with the condition of phylogenetically-related floral scent interspecific variation, a key condition in studies of plant-pollinator coevolution mediated by them.


Se describieron los volátiles florales de cuatro especies de Heliotropium (Boraginaceae) y se evaluó su relación con una filogenia del género. Se identificaron 22 compuestos; los bencenoides fueron los más abundantes en H. floridum, H. myosotifolium and H. stenophyllum, acompañados de dos monoterpenos, el (E)-Ocimeno y el 1,8-cineol, en H. pycnophyllum. La similitud entre los componentes de cada especie aumentó con la distancia filogenética entre ellas. El análisis “neighbor-joining” produjo un árbol con un clado que contenía a H. myosotifolium y H. stenophyllum y una relación no resuelta entre ese clado y H. floridum y H. pycnophyllum; esta topología guarda similitud con la hipótesis filogenética molecular propuesta para el grupo. Estos resultados preliminares sugieren que el género Heliotropium cumple con la condición de mostrar variación interespecífica en sus volátiles florales, condición básica para estudios de coevolución planta-polinizador mediada por ellos.
