Résumé

<p><b>OBJECTIVE</b>To prepare the 4 candidate vaccine strains of H5N1 avian influenza virus isolated in China.</p><p><b>METHODS</b>Recombinant viruses were rescued using reverse genetics. Neuraminidase (NA) and hemagglutinin (HA) segments of the A/Xinjiang/1/2006, A/Guangxi/1/2009, A/Hubei/1/2010, and A/Guangdong/1/2011 viruses were amplified by RT-PCR. Multibasic amino acid cleavage site of HA was removed and ligated into the pCIpolI vector for virus rescue. The recombinant viruses were evaluated by trypsin dependent assays. Their embryonate survival and antigenicity were compared with those of the respective wild-type viruses.</p><p><b>RESULTS</b>The 4 recombinant viruses showed similar antigenicity compared with wild-type viruses, chicken embryo survival and trypsin-dependent characteristics.</p><p><b>CONCLUSION</b>The 4 recombinant viruses rescued using reverse genetics meet the criteria for classification of low pathogenic avian influenza strains, thus supporting the use of them for the development of seeds and production of pre-pandemic vaccines.</p>

Se evaluó la crioconservación a - 70 ºC con 9 variantes de crioprotectores como método de conservación de las cepas de Leptospira interrogans, utilizadas como antígenos en la vacuna antileptospirósica vax-SPIRALâ. La viabilidad se evaluó periódicamente según el rendimiento celular alcanzado por las cepas recién descongeladas en medio EMJH. La estabilidad de la virulencia fue estimada en hámster. La antigenicidad antes y después de la crioconservación fue comparada mediante microaglutinación frente a antisueros de referencia. Solo el empleo de dimetilsulfóxido a 2,5 y 5 % o leche descremada 5 % como agentes crioprotectores permitió una rápida recuperación de las 3 cepas tras 7 meses de crioconservación sin afectación de su virulencia y antigenicidad, mientras el glicerol tuvo un efecto variable sobre el grado de recuperación de las cepas evaluadas. Los resultados obtenidos demuestran la factibilidad de la crioconservación a - 70 ºC con un apropiado agente crioprotector como método de conservación de cepas vacunales de Leptospira.


The goal of this study was to evaluate the cryopreservation at -70ºC with the use of 9 different cryoprotectants as a preservation method for the strains of Leptospira interrogans used as antigens in the leptospiral vaccine vax-SPIRALâ. Viability was systematically evaluated according to the cell yield achieved by the just thawed strains in EMJH medium. Virulence stability was estimated in Hamsters and antigenicity was evaluated by microscopic agglutination test using reference antisera before and after cryopreservation. Only the use of 2,5% and 5% dimethyl sulfoxide or 5% skim milk allowed a quick recovery of the three strains without virulence or antigenicity depletions after 7 months of cryopreservation while glycerol had a variable effect on the degree of recovery of each strain. The results showed the feasibility of the cryopreservation at -70 °C with the use of a suitable cryoprotectant as a preservation method for vaccine strains of Leptospira.
