Résumé

Objective To investigate the content change of aconitine in Radix Aconiti Kusnezoffii in different boiling time. Methods The chromatographic procedure was carried out with methanol-water-chloroform-diethylamide (70∶30∶3∶0.2) as mobile phase at a flow rate of 1.0 mL/min, the column temperature at 30 ℃, and UV detector at 240 nm. Results There was a good linear relationship of aconitine within 0.08-0.8 μg, r=0.999 8 (n=6), and the average recovery rate was 97.16%. The content of aconitine increased gradually in 0-2 h, then gradually decreased. And the reduction was evident particularly during 2-4 hours. Conclusion HPLC method is rapid, simple, accurate, reproducible and reliable for determining aconitine. The content of aconitine can be decreased effectively by boiling 4 hours.

Since ancient times in China, decocting times have been adjusted to improving objective medicinal effects. In medical texts, decocting times are dictated by the amount of water that is reduced. And in this report, we surveyed rhubarb (Chinese, Da-huang ; Japanese, Daio) formulation decocting times described in the earliest Shang-han-lun and Jin-gui-yao-lue texts, from the amount of water changed. We found that rhubarb decocting times could be divided into four 10-, 20-, 30- and 50-minute groups when rhubarb is decocted with other crude drugs, while when rhubarb is put in water after other crude drugs, decocting times could be divided into four 1-, 5-, 10- and 20-minute groups. Next, when we prepared rhubarb decoctions using unprocessed rhubarb and processed rhubarb, and compared change in the principal compounds eluted into those decoctions, we found that 80% or more of the principal compounds were eluted into a decoction when either unprocessed or processed rhubarb were decocted for 30 minutes. It was therefore clear that, in ancient times, rhubarb formulations were decocted for 10 to 30 minutes so that not all of the principle compounds would elute, or given 50 minutes to allow all the principle compounds to elute. Meanwhile, it was also clear that rhubarb was put in water1to 20 minutes before decocting finished, to adjust the amount of principle compound eluted.

Objective To analysis the effective ingredients in Huanglianjiedu decoction under different decocting conditions. Methods The detecting method of HPLC was established and the ingredients in Huanglianjiedu decoction under different decocting time and times were analyzed. Result Water- soluble glycoside of gardenoside and baicalin reached balance of dissolution within 20 min and their dissolution rate were about 82% and 60% respectively. Water-soluble alkaloids of berberine and palmatine reached balance of dissolution within 30 min and their dissolution rate were about 45%. The dissolution rate of wogonin was 31% at 60 min. The dissolution rate of Crocin-Ⅰ reached top at 20 min and fell down after 20 min. Decocting times for fat-soluble constitutes was more important than water-soluble constitutes. Conclusion The decocting condition has effect on the dissolution rate and stability of the ingredients in Huanglianjiedu decoction. The decocting condition is very important to the quality control of decoction.
