RÉSUMÉ

Objective To find a new method for quality control of polysaccharides by establishing the fingerprint profiles of polysaccharides extracted from Guifu Dihuang Wan. Methods The polysaccharides extracted from Guifu Dihuang Wan were hydro-lyzed partially under appropriate hydrolysis condition with acid,and the fingerprint profiles of the hydrolyzates was obtained by HPLC. Microsoft Excel 2002 was applied for calculating the similarity of fingerprint profiles,and informatton analysis of these profiles was performed by a software for evaluating fingerprint profiles of traditional Chinese medicine. Results Fingerprint profiles of the hydroly-zates of polysaccharides from 10 of Guifu Dihuang Wan products,produced by different manufacturers,were obtained. It was observed that the similarity of eight products was greater than 0.99,while the similarity of the other two was 0.92 and 0.94,respectively. Con-clusion This method is technically feasible for the quality control of polysaccharides from Guifu Dihuang Wan.

The aim of the study was to establish the finger printing profile and evaluation of in vitro antidiabetic potential of C. obtusa. In vitro antidiabetic activity was carried out according to the method adopted by Miller, 1959. HPTLC studies were carried out using CAMAG HPTLC system equipped with Linomat 5 applicator, TLC scanner 3 and Win cats-4 software for the active fractionation of aqueous-methanolic leaf extracts of C. obtusa. Among the various plant parts analyzed, leaf exhibited efficient inhibitory activity against α-amylase and α-glucosidase enzymes. Therefore, the leaf extract was further fractionated using various solvent systems petroleum ether, chloroform, ethylacetate, butanol and water and were subjected to in vitro antidiabetic activity. Among the fractions analyzed chloroform fraction exhibited remarkable antidiabetic activity by inhibiting α-amylase and α- glucosidase enzymes. Furthermore, the active leaf fractions were analyzed with HPTLC to develop fingerprint profiles and these fractions revealed the presence of 13 and 22 major spots of alkaloids and flavonoids respectively with different Rf values. The results of the present study thus claim the folkloric usage of the plant in diabetic related maladies.

RÉSUMÉ

OBJECTIVE:To study the HPLC fingerprint profiles of crude and processed Semen Cassiae so as to provide basis for the establishment of chromatographic fingerprint of Semen Cassiae.METHODS: The samples of Semen Cassiae were hydrolyzed in the 1.5 mol?L-1 hydrochloride acid and then reflux extracted with chloroform.The HPLC analysis was run in gradient elution.The whole information of single-wavelength chromatographic fingerprint,two-dimension information data at full wavelength 220 nm~550 nm and the data of 18 fingerprint peaks compared with chrysophanol were treated by cluster analysis.RESULTS:It was observed that the contents of several chemical components were higher in the crude or processed Semen Cassiae than in chrysophanol.There were differences between the crude and the processed Semen Cassiae in HPLC fingerprint profiles though they were of good similarity.CONCLUSION: The HPLC fingerprints of liposoluble constituents were obtained with good resolution under the experimental chromatographic system.Three pure chromatographic peaks of chrysophanol,emodin and physcion have been identified.More reliable cluster result can be obtained from two-dimension information data at full wavelength 220 nm~550 nm than from the whole information of single-wavelength chromatographic fingerprint or from the information of chromatographic fingerprint of characteristic peaks.
