BACKGROUND:Grape seed extract can inhibit chondrocyte apoptosis and aging,and improve osteoarthritis.However,the effects of grape seed extract on the apoptosis of chondrocytes in the growth plate and tibial growth are still unclear. OBJECTIVE:To investigate the effect of grape seed extract on interleukin-1β-induced apoptosis in rat growth plate cells and on tibial bone growth. METHODS:(1)Cell experiment:Growth plate chondrocytes from Sprague-Dawley rats were isolated,cultured,and identified.The cells were then randomly divided into control group,model group,grape seed extract group,miR-138-5p NC group and miR-138-5p inhibitor group.In the model group,20 ng/mL interleukin-1β was used to induce apoptosis in rat growth plate chondrocytes.In the grape seed extract group,20 ng/mL interleukin 1β was added along with 10 μmol/L grape seed extract solution for 48 hours.Cells in the miR-138-5p NC and inhibitor groups were transfected with 5 nmol/L miR-138-5p NC and 5 nmol/L miR-138-5p inhibitor for 12 hours,respectively,followed by addition of 20 ng/mL interleukin-1β.qRT-PCR was used to detect miR-138-5p and caspase-3 expression.Luciferase reporter assay was used to analyze the relationship between miR-138-5p and caspase-3 targeting.Cell counting kit-8 was used to detect cell proliferation activity.Flow cytometry was used to detect cell apoptosis.Caspase-3 and Bcl-2 protein expressions were detected by western blot.(2)Animal experiment:The animals were divided into normal control group,grape seed extract group and miR-138-5p inhibitor group.The effects of grape seed extract on epiphyseal closure and tibial growth of the tibial plateau in rats were observed. RESULTS AND CONCLUSION:miR-138-5p had a targeting relationship with caspase-3.Compared with the control group,cell proliferation was significantly reduced,apoptosis was significantly increased(P<0.01),miR-138-5p,Bcl-2 expression was reduced(P<0.01),and caspase-3 expression was increased(P<0.01)in the model group.Compared with the mod group,the grape seed extract group showed a significant increase in cell proliferation,a significant decrease in apoptosis(P<0.01),an increase in miR-138-5p and Bcl-2 expression(P<0.01)and a decrease in caspase-3 expression(P<0.01).Compared with the grape seed extract group,the miR-138-5p inhibitor group showed a significant decrease in cell proliferation,a significant increase in apoptosis(P<0.01),a decrease in miR-138-5p and Bcl-2 expression(P<0.05 or P<0.01),and an increase in caspase-3 expression(P<0.05).Intragastric administration of grape seed extract could delay epiphyseal closure of the tibial plateau and promote tibial bone growth in rats,whereas miR-138-5p inhibitor intervention inhibited the effect of grape seed extract on tibial bone growth in rats.To conclude,grape seed extract can inhibit apoptosis of rat growth plate chondrocytes through regulating the miR-138-5p/caspase-3 pathway,improve epiphyseal closure of rat tibial plateau and promote tibial bone growth.

BACKGROUND:Grape seed extract has been shown to be effective in inhibiting the growth of androgen-dependent tumors(e.g.,breast cancer),and thus grape seed extract could theoretically inhibit epiphyseal closure induced by estrogen in late adolescence. OBJECTIVE:To screen the effects of grape seed extract on apoptosis of growth plate chondrocytes and epiphyseal closure in rats. METHODS:(1)In vitro experiment:Growth plate chondrocytes from rat large tibia and femur at logarithmic growth stage were obtained and cultured in groups:normal control group,model control group(adding 17β-estradiol to induce apoptosis),positive control group(adding letrozole and 17β-estradiol),grape seed extract group(adding 17β-estradiol and 10 μg/mL grape seed extract),Caspase-9 inhibitor group(adding 17β-estradiol and Caspase-9 inhibitor),Caspase-9 agonist group(adding 17β-estradiol and Caspase-9 agonist).Cell apoptosis was detected by flow cytometry after 48 hours of culture.(2)In vivo experiment:Thirty 3-month-old Sprague-Dawley rats were randomly divided into model control group,positive control group and low-,medium-and high-dose groups,with five rats in each group.All rats were injected subcutaneously with 17β-estradiol(3 times per week)to establish epiphyseal closure models,followed by intragastric administration of letrozole in positive control group and 0.05,0.2 and 0.8 g/kg grape seed extract in low-,medium-and high-dose groups,respectively,once a day until over 2/3 of the epiphyseal plate in the model control group was closed.The length of the tibia was then observed.Another 18 Sprague-Dawley rats were randomly divided into model control group,positive control group,and medium-dose group,with 6 rats in each group,treated as above for 1.5 continuous months.The expression of Caspase-9 protein in rat growth plate cartilage was detected by western blot. RESULTS AND CONCLUSION:(1)In vitro experiment:17β-estradiol could induce apoptosis in growth plate chondrocytes,and letrozole,grape seed extract,and caspase-9 inhibitors could all inhibit apoptosis in growth plate chondrocytes.(2)In vivo experiment:When more than 2/3 of the epiphyseal plate in the model control group was closed,the number of rats with epiphysis closure in the positive control and medium-dose groups was less than that in the model control group(P<0.05),and the tibial length was longer than that in the model control group(P<0.05),and the Caspase-9 protein expression in the tibial growth plate was lower than that in the model control group(P<0.05).To conclude,the appropriate dose of grape seed extract can effectively inhibit the apoptosis of growth plate chondrocytes and delay epiphyseal closure,which has the potential to promote bone growth.

The study was conducted in Vijayapura district of Karnataka during 2019-20 to analyse the constraints and suggestions of fruit crop growers. Forty each grape, lime and pomegranate growers were selected by use of simple random sampling technique. Thus, making a total sample size of 120. Personal interview method was used to collect data and appropriate statistical tools were applied to analyse the data. The findings revealed that, with respect to the constraints faced by fruit crop growers in fruit crop production, production constraints were lack of irrigation facilities (87.50%) followed by high cost of inputs (73.33%). Major financial constraints were non-availability of credit at right time (73.33%) followed by inadequate credit (70.83%). High wages (70.00%) and non-availability of labourer (61.67%) were the main issues relating to labour constraints. Regarding marketing constraints, fluctuating price (87.17%) followed by delayed cash payment and exploitation by middlemen (77.50%) were the major constraints. The suggestions offered by the fruit crop growers are provision for irrigation facility (85.00%) followed by provision of timely and adequate market information (76.67%), protection from exploitation by middlemen (75.00%). Provision for suitable market infrastructure, timely and adequate credit, timely and adequate cash payment, continuous power supply, interest free credit and longer repayment period for credit were other suggestions offered to improve the returns from fruit crop cultivation.

Abstract The use of lactic acid bacteria (LAB) in foods as biocontrol agents against foodborne pathogens has become increasingly known. Under the premise that controlling the adhesion of microorganisms to food contact surfaces is an essential step for meeting the goals of food processing, the aim of this work was to investigate the inhibitory and anti-biofilm effectiveness of Lactobacillus rhamnosus GG (ATCC 53103) and Lactobacillus casei (ATCC 393) against Escherichia coli O157:H7, Salmonella enterica and Listeria monocytogenes. Lactobacillus strains (108UFCCFU/ml) and pathogens (104UFCCFU/ml) were evaluated to monitor LAB anti-adhesive and antibiofilm effect, in two main scenarios: (i) co-adhesion and (ii) pathogen incorporation to stainless steel surfaces with a protective biofilm of Lactobacillus cells. In (i) the predominant effect was observed in L. rhamnosus against S. enterica and L. monocytogenes, whereas in (ii) both LAB significantly reduced the number of pathogenic adherent cells. The effect of pre-established LAB biofilms was more successful in displacing the three pathogens than when they were evaluated under co-adhesion. These findings show that both LAB can be considered good candidates to prevent or inhibit the adhesion and colonization of L. monocytogenes, S. enterica and E. coli O157:H7 on surfaces and conditions of relevance for juice processing industries, offering alternatives for improving the safety and quality of fruit-based products.
Resumen Existe un creciente interés en el uso de bacterias ácido lácticas (BAL) como agentes de biocontrol frente a patógenos de transmisión alimentaria. Bajo la premisa de que el control de la adhesión de microorganismos a superficies de contacto con alimentos es el paso esencial para evitar su contaminación, el objetivo de este trabajo fue investigar la efectividad inhibitoria y antibiofilm de Lactobacillus rhamnosus GG (ATCC 53103) y Lactobacillus casei (ATCC 393) frente a Escherichia coli O157:H7, Salmonella enterica y Listeria monocytogenes. A fin de cumplir con el objetivo propuesto, las cepas de Lactobacillus (108UFCUFC/ml) y los patógenos (104UFCUFC/ml) se ensayaron en 2 escenarios: (1) coadhesión, y (2) incorporación de los patógenos a las superficies de acero inoxidable con un biofilm preformado de Lactobacillus. En (1), el efecto predominante se observó con L. rhamnosus frente a S. enterica y L. monocytogenes, mientras que en (2), ambas BAL redujeron significativamente el número de células patógenas adheridas. En función de estos resultados, concluimos que el efecto de un biofilm preformado de ambas BAL fue más exitoso en el desplazamiento de los 3 patógenos que en coadhesión. Ambas BAL pueden considerarse buenas candidatas para mitigar la adhesión y colonización de L. monocytogenes, S. enterica y E. coli O157:H7 en superficies en condiciones de relevancia para la industria procesadora de jugos, y, de esta manera, ofrecer alternativas para mejorar la seguridad y calidad de los alimentos a base de frutas.

SUMMARY: The potential anti-inflammatory and antifibrotic activity of polyphenolic extracts of blueberry and grape was evaluated in a mouse model of lung damage induced by subcutaneous administration of bleomycin. The results of testing the polyphenolic extracts on two different systemic administration variants of bleomycin (intraperitoneal and subcutaneous) were compared. It was found that regardless of the method of bleomycin administration, indirect cross-acute and subacute damage to the pulmonary system was observed. Both patterns exhibited the same prevalence and severity. The administration of polyphenolic extracts of blueberry and grape to mice resulted in a significant decrease in theseverity of acute and subacute patterns of lung damage, suggesting their protective properties for the microcirculatory bed and a pronounced anti-inflammatory effect.
La potencial actividad antiinflamatoria y antifibrótica de los extractos polifenólicos de arándano y uva se evaluó en un modelo de daño pulmonar en ratón inducido por la administración subcutánea de bleomicina. Se compararon los resultados de las pruebas de los extractos polifenólicos en dos variantes diferentes de administración sistémica de bleomicina (intraperitoneal y subcutánea). Se encontró que, independientemente del método de administración de bleomicina, se observaba daño indirecto cruzado, agudo y subagudo al sistema pulmonar. Ambos patrones exhibieron la misma prevalencia y gravedad. La administración de extractos polifenólicos de arándano y uva a ratones dio como resultado una disminución significativa en la gravedad de los patrones agudos y subagudos de daño pulmonar, lo que sugiere sus propiedades protectoras del lecho micro- circulatorio y un efecto antiinflamatorio pronunciado.

The present investigation was conducted at College of Horticulture, Rajendranagar, during the year 2023. The experiment was laid out in completely randomized design. The research experiment was conducted to evaluate qualitative parameters of different juice varieties of grape. The results revealed that maximum TSS (21.05 ?B) and brix/acid ratio (42.96) were recorded in T1 – H-516. Maximum pH was recorded in T6 – Manjari Medika (4.03) and highest titrable acidity was recorded in T5 – Gulabi X Bangalore Purple (0.71%). Reducing sugars were highest in T1 – H-516 (17.24%), whereas total sugars were maximum in T4 – Concord (28.56%). Juice recovery was recorded maximum in T6 – Manjari Medika (71.80 %).

Micronutrient deficiencies play a significant role in impacting the yield and quality of grapes in vineyards. To address the deficiencies, a field study was conducted during the summer, of 2022 by using foliar micronutrient mixtures in three different concentrations (0.5%, 1.0%, and 1.5%), with one spray during pre-fruit set stage and twice at pre-fruit set/berry growth stages. The study aimed to investigate the effects of foliar micronutrient application on the biochemical quality characteristics of the Muscat Hamburg grapevine cultivar. The results of the study showed that the application of micronutrients spray showed a beneficial impact on all the yield parameters, such as number of bunches per vine (ranging from 28 to 44), the yield per vine (varied from 5.56 to 6.55 kg per vine), with overall yield range between 20.7 to 24.2 t ha-1. The biochemical properties of the berries, including sugars (reducing, non-reducing, and total sugars), juice pH and content, TSS (Total Soluble Solids), Titrable acidity, and berry firmness, were found to be high in grapevines sprayed at 1.0% twice. Results showed that total sugars ranged from 15.2% to 10.7%, with a juice pH of 3.47 to 3.94, juice content of 83.5 to 95.5%, and TSS from 15.6 to 17.40º Brix. Regarding the Titrable acidity and firmness of the berries, the outcomes showed a range of 0.92% to 1.14% for acidity and 9.4 to 12.9 N for firmness. Findings from the study clearly showed that applying 1.0% micronutrients twice at fruit-set and berry stages increased yield and improved quality indices of Paneer grapes.

El Pisco es el destilado del Perú, elaborado a partir de mostos recientemente fermentados con uvas criollas denominadas "pisqueras". Las levaduras son los microorganismos clave en la fermentación y el uso de cepas nativas seleccionadas presenta ventajas competitivas para la tipicidad del producto, así como también para la estandarización del producto y el control microbiológico del proceso. El objetivo fue identificar y seleccionar las cepas de levaduras nativas para la producción del Pisco de uva Quebranta aisladas de procesos productivos de Pisco en el valle de Ica. Para ello, se emplearon técnicas microbiológicas y moleculares mediante el análisis de ITS1-5.8S-ITS2 PCR-RFLP para la identidad taxonómica. La evaluación de las cepas para producir Pisco consistió en el análisis fisicoquímico y organoléptico del destilado obtenido con las cepas seleccionadas. Se evaluaron 3 aislados para la producción de Pisco identificados como Saccharomyces cerevisiae: UNA SC - 25, UNA SC - 49 y UNA SC - 54, de los cuales la cepa UNA SC-49 destacó por mostrar aptitudes enológicas diferentes a las otras cepas. Este trabajo constituye el primer registro de levaduras nativas del Perú para mostos procedentes de uva Quebranta.
El Pisco stands as Peru's distilled spirit, crafted from recently fermented musts derived from native grape varieties known as "pisqueras". Yeasts serve as decisive microorganisms in the fermentation process, and the utilization of selected native strains confers competitive advantages for product typicity, standardization, and microbiological control within the production process. The aim was to identify and select native yeast strains for Quebranta grape Pisco production, isolated from Pisco production processes in the Ica Valley. Microbiological and molecular techniques were employed, utilizing ITS1-5.8S-ITS2 PCR-RFLP analysis for taxonomic identification. The assessment of yeast strains for Pisco production involved the physicochemical and organoleptic analysis of the distilled product obtained using the selected strains. Three isolates were evaluated for Pisco production, identified as Saccharomyces cerevisiae: UNA SC - 25, UNA SC - 49, and UNA SC - 54. Among these, the UNA SC-49 strain stood out due to displaying oenological characteristics distinct from the other strains. This work is the first documentation of native yeasts in Peru for musts derived from Quebranta grapes.

Background: The edible green algae Caulerpa racemosa (Forsskal) J. Agardh (Caulerpaceae), also known as "sea grape", is an excellent source of phenolic compounds known for their activity to reduce free radicals. Objectives: The research aims to evaluate the total phenolic content and antioxidant activity of C. racemosa (70% ethanol extracts) obtained from different extraction methods, such as maceration, Soxhlet, and ultrasound. Methods: Total phenolics of the extracts were determined by the colorimetry method using the Folin-Ciocalteu reagent. Total phenol content was expressed as mg gallic acid equivalent (GAE) per g extract. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical was used to assess the antioxidant activity of the extracts. Results: the ethanol extract of C. racemosa obtained from the ultrasonic methods had the highest phenolic content (39.38 mg GAE/g) compared to other extraction methods (Soxhlet was 36.88 mg GAE/g and maceration was 22.05 mg GAE/g). The IC50 value against DPPH of the C. racemosa ethanol extracts using ultrasonic, Soxhlet, and maceration was 352.95, 365.73, and 375.84 µg/mL, respectively. Conclusions: the variation of the extraction methods affected the total phenolics content of C. racemosa ethanol extracts and their antioxidant activity. We reported here the potential of C. racemosa extracts as an antioxidant raw material from marine plants for medicinal, nutraceutical, cosmetics, and food products; however, more research is needed.
Antecedentes: El alga verde comestible Caulerpa racemosa (Forsskal) J. Agardh (Caulerpaceae), también conocida como "uva de mar", es una excelente fuente de compuestos fenólicos conocidos por su actividad para reducir los radicales libres. Objetivos: La investigación pretende evaluar el contenido fenólico total y la actividad antioxidante de C. racemosa (extractos de etanol al 70%) obtenidos a partir de diferentes métodos de extracción, como maceración, Soxhlet y ultrasonido. Métodos: Los fenoles totales de los extractos se determinaron por el método colorimétrico utilizando el reactivo de Folin-Ciocalteu. El contenido total de fenoles se expresó como mg de ácido gálico equivalente (GAE) por g de extracto. Para evaluar la actividad antioxidante de los extractos se utilizó el radical libre 2,2-difenil-1-picrilhidrazilo (DPPH). Resultados: el extracto etanólico de C. racemosa obtenido por ultrasonido presentó el mayor contenido fenólico (39,38 mg GAE/g) en comparación con otros métodos de extracción (Soxhlet fue de 36,88 mg GAE/g y maceración fue de 22,05 mg GAE/g). El valor IC50 frente a DPPH de los extractos etanólicos de C. racemosa mediante ultrasonido, Soxhlet y maceración fue de 352,95, 365,73 y 375,84 µg/mL, respectivamente. Conclusiones: la variación de los métodos de extracción afectó al contenido total de fenoles de los extractos etanólicos de C. racemosa y a su actividad antioxidante. Aquí reportamos el potencial de los extractos de C. racemosa como materia prima antioxidante a partir de plantas marinas para productos medicinales, nutracéuticos, cosméticos y alimenticios; sin embargo, se necesita más investigación.

Abstract The increase in life expectancy has led to a higher incidence of osteoporosis, characterized by an imbalance in bone remodeling. Several drugs are used for its treatment, but most promote undesirable side effects. The present investigation evaluated the effects of two low concentrations of grape seed extract (GSE) rich in proanthocyanidins on MC3T3-E1 osteoblastic cells. The cells were cultured in an osteogenic medium and divided into control (C), 0.1 µg/mL GSE (GSE0.1), and 1.0 µg/mL GSE (GSE1.0) groups to evaluate cell morphology, adhesion, and proliferation, in situ alkaline phosphatase (ALP) detection, mineralization and immunolocalization of osteopontin (OPN). The data obtained were analyzed by statistical tests for a significance of 5%. Cell morphology was maintained with both GSE concentrations, whereas cell adhesion significantly increased within three days in all groups. Cell proliferation increased significantly at seven days of culture, followed by a significant decrease in all experimental periods, with no statistical difference among them. In situ detection of ALP and mineralization increased with time, but within each period, no statistical differences among groups were observed. The expression of osteopontin was distributed regularly with more intensity after 24 hours in the GSE0.1 group. After three days, OPN expression was more intense in the control group, followed by GSE0.1 and GSE1.0 groups. Data obtained suggest that low concentrations of GSE do not affect the morphology and may stimulate the functional activity of osteoblastic cells.
Resumo O aumento da expectativa de vida tem levado a uma maior incidência de osteoporose, caracterizada por um desequilíbrio na remodelação óssea. Vários medicamentos são utilizados para o seu tratamento, contudo, a maioria promove efeitos colaterais indesejáveis. A presente investigação avaliou os efeitos de duas baixas concentrações de extrato de semente de uva (GSE) rico em proantocianidinas em células osteoblásticas MC3T3-E1. As células foram cultivadas em meio osteogênico e divididas em grupos controle (C), 0,1 µg/mL de GSE (GSE0.1) e 1,0 µg/mL de GSE (GSE1.0) para avaliar morfologia, adesão e proliferação celular, detecção in situ de fosfatase alcalina (ALP), mineralização e imunolocalização da proteína osteopontina (OPN). Os dados obtidos foram analisados por testes estatísticos para um nível de significância de 5%. A proliferação celular aumentou significativamente aos sete dias de cultura, seguido de uma diminuição significativa em todos os períodos experimentais, sem diferença estatística entre eles. A detecção in situ de ALP e mineralização aumentou com o tempo, mas dentro de cada período não foram observadas diferenças estatísticas entre os grupos. A morfologia celular foi mantida com ambas as concentrações de GSE, enquanto a adesão celular aumentou significativamente aos três dias em todos os grupos. A expressão de osteopontina distribuiu-se regularmente com maior intensidade após 24 horas no grupo GSE0.1. Após três dias, a expressão de OPN foi mais intensa no grupo controle, seguida pelos grupos GSE0.1 e GSE1.0. Os dados obtidos sugerem que baixas concentrações de GSE não afetam a morfologia e podem estimular a atividade funcional das células osteoblásticas.

Abstract Background: Although it is possible to preserve the aerial part of cassava in the form of silage, as demonstrated in previous studies, its low dry matter content can result in undesirable fermentation and increased effluent losses during silage, leading to a reduction in the final quality of the silage. A possible way to get around this problem would consist in mixing this silage with dehydrated by-products from the agroindustry. Objective: To evaluate the addition of dehydrated grape marc (DGM) to a silage of aerial parts of cassava (Manihot esculenta, Crantz) on the chemical composition, degradability and in vitro gas production of silage. Methods: A completely randomized experimental design was used, with two treatments: (I) silage of the aerial part of cassava without additive; (II) silage of the aerial part of cassava added with 10% dehydrated grape marc (DGM) in the natural matter, as an additive. After 30 days of fermentation, silages were evaluated to estimate the contents of dry matter (DM), crude protein (CP), ether extract (EE), neutral detergent fiber (NDF), total digestible nutrients (TDN), as well as pH, in vitro degradability, and cumulative gas production by semi-automatic in vitro gas production. Results: Addition of DGM did not affect CP, EE, NDF, nor TDN levels in silage. The DGM, however, promoted an increase in DM content (32.61 vs 30.31%) and a reduction in pH (4.00 vs 4.75) of the silage. The silage that received DGM presented higher degradability coefficients of soluble and potentially degradable fractions, which led to higher values of potential and effective degradability. Similarly, DGM promoted a reduction in particle colonization time (6.74 vs 10.01 h) and increased cumulative gas production (62.03 vs 57.82 mL/g DM). Conclusion: Addition of 10% dehydrated grape marc in the silage of aerial parts of cassava can be useful to reduce pH and increase dry matter contents of the silage.
Resumen Antecedentes: Si bien es posible preservar la parte aérea de la yuca en forma de ensilaje, como se demostró en estudios anteriores, el bajo contenido de materia seca puede resultar en fermentaciones indeseables y mayores pérdidas de efluentes durante el ensilaje, levando a una reducción en la calidad final del ensilaje. Una de las formas de solucionar este problema sería el ensilaje mixto de la parte aérea de la yuca con subproductos deshidratados de la agroindustria. Objetivo: Evaluar el efecto de la adición de bagazo de uva deshidratado (BUD) a la parte aérea del ensilaje de yuca (Manihot esculenta, Crantz) sobre la composición química, degradabilidad y producción de gas in vitro del ensilado. Métodos: El diseño experimental utilizado fue completamente al azar y los tratamientos consistieron en: (I) ensilado de brotes de yuca sin aditivos; (II) ensilaje de la parte aérea de yuca con 10% de BUD en materia natural, como aditivo. Luego de 30 días de fermentación, los ensilajes fueron evaluados para materia seca (MS), proteína cruda (PC), extracto de éter (EE), fibra detergente neutra (FDN), nutrientes digestibles totales (NDT), pH, degradabilidad in vitro y producción acumulada de gas mediante la técnica de producción de gas in vitro semiautomática. Resultados: La adición de BUD no cambió los contenidos de PB, EE, FDN y NDT del ensilaje. Sin embargo, BUD promovió un aumento en el contenido de MS (32,61 vs 30,31%) y una reducción en el pH (4,00 vs 4,75) del ensilaje. El ensilado que recibió la BUD mostró mayores coeficientes de degradabilidad de las fracciones solubles y potencialmente degradables, lo que resultó en mayores valores de degradabilidad potencial y efectiva. De manera similar, BUD promovió una reducción en el tiempo de colonización de partículas (6,74 vs 10,01 h) y un aumento en la producción acumulada de gas (62,03 vs 57,82 mL/g MS). Uma das formas de contornar esse problema seria a silagem mista da parte aérea da mandioca com subprodutos desidratados da agroindústria. Objetivo: Avaliar o efeito da adição do bagaço desidratado de uva (BDU) na ensilagem da parte aérea de mandioca (Manihot esculenta, Crantz) sobre a composição química, degradabilidade e produção de gases in vitro da silagem. Métodos: O delineamento experimental utilizado foi inteiramente casualizado e os tratamentos constituíram-se por: (I) silagem da parte aérea da mandioca sem aditivo; (II) silagem da parte aérea da mandioca com 10% de bagaço desidratado de uva (BDU) na matéria natural, como aditivo. Após 30 dias de fermentação, as silagens foram avaliadas quanto aos teores de matéria seca (MS), proteína bruta (PB), extrato etéreo (EE), fibra em detergente neutro (FDN), nutrientes digestíveis totais (NDT), pH, degradabilidade in vitro e produção cumulativa de gases por meio da técnica semiautomática de produção de gases in vitro. Resultados: A adição do BDU não alterou os teores de PB, EE, FDN e NDT da silagem. No entanto, o BDU promoveu aumento no teor de MS (32,61 vs 30,31%) e redução no pH (4,00 vs 4,75) da silagem. A silagem que recebeu o BDU apresentou maiores coeficientes de degradabilidade das frações solúvel e potencialmente degradável, o que acarretou em maiores valores de degradabilidade potencial e efetiva. Do mesmo modo, o BDU promoveu uma redução no tempo de colonização da partícula (6,74 vs 10,01 h) e um aumento na produção cumulativa de gás (62,03 vs 57,82 mL/g MS). SUMMARY: An experimental morphological and morphometric study of the antifibrotic function of blueberry and grape extracts was carried out on a model of lung injury in mice induced by intraperitoneal administration of bleomycin. During intraperitoneal administration of bleomycin to mice, acute and subacute damage to the pulmonary system was noted. Both patterns had the same prevalence and severity. The administration of polyphenolic extracts of blueberry and grape to mice showed a significant reduction in the severity of the acute and subacute pattern of lung injury. Blueberry and grape extracts reduce the acute phase of damage to the microvasculature, enhance phagocytic function, have an anti-inflammatory effect, reducing the degree of lymphohistiocytic infiltration and locoregional foci of residual inflammatory effects.
Se realizó un estudio experimental morfológico y morfométrico de la función antifibrótica de extractos de arándano y uva en un modelo de lesión pulmonar en ratones inducida por la administración intraperitoneal de bleomicina. Durante la administración intraperitoneal de bleomicina a ratones, se observaron daños agudos y subagudos en el sistema pulmonar. Ambos patrones tuvieron la misma prevalencia y severidad. La administración de extractos polifenólicos de arándano y uva a ratones mostró una reducción significativa en la severidad del patrón agudo y subagudo de lesión pulmonar. Los extractos de arándano y uva reducen la fase aguda del daño a la microvasculatura, mejoran la función fagocítica, tienen un efecto antiinflamatorio, reducen el grado de infiltración linfohistiocítica y los focos locorregionales de efectos inflamatorios residuales.

OBJECTIVE@#To examine the anti-inflammatory effect of grape seed extract (GSE) in animal and cellular models and explore its mechanism of action.@*METHODS@#This study determined the inhibitory effect of GSE on macrophage inflammation and Th1 and Th17 polarization in vitro. Based on the in vitro results, the effects and mechanisms of GSE on multiple sclerosis (MS)-experimental autoimmune encephalomyelitis (EAE) mice model were further explored. The C57BL/6 mice were intragastrically administered with 50 mg/kg of GSE once a day from the 3rd day to the 27th day after immunization. The activation of microglia, the polarization of Th1 and Th17 and the inflammatory factors such as tumor necrosis factor- α (TNF- α), interleukin-1 β (IL-1 β), IL-6, IL-12, IL-17 and interferon-γ (IFN-γ) secreted by them were detected in vitro and in vivo by flow cytometry, enzyme linked immunosorbent assay (ELISA), immunofluorescence staining and Western blot, respectively.@*RESULTS@#GSE reduced the secretion of TNF-α, IL-1 β and IL-6 in bone marrow-derived macrophages stimulated by lipopolysaccharide (P<0.01), inhibited the secretion of TNF-α, IL-1 β, IL-6, IL-12, IL-17 and IFN-γ in spleen cells of EAE mice immunized for 9 days (P<0.05 or P<0.01), and reduced the differentiation of Th1 and Th17 mediated by CD3 and CD28 factors (P<0.01). GSE significantly improved the clinical symptoms of EAE mice, and inhibited spinal cord demyelination and inflammatory cell infiltration. Peripherally, GSE downregulated the expression of toll-like-receptor 4 (TLR4) and Rho-associated kinase (ROCKII, P<0.05 or P<0.01), and inhibited the secretion of inflammatory factors (P<0.01 or P<0.05). In the central nervous system, GSE inhibited the infiltration of CD45+CD11b+ and CD45+CD4+ cells, and weakened the differentiation of Th1 and Th17 (P<0.05). Moreover, it reduced the secretion of inflammatory factors (P<0.01), and prevented the activation of microglia (P<0.05).@*CONCLUSION@#GSE had a beneficial effect on the pathogenesis and progression of EAE by inhibiting inflammatory response as a potential drug and strategy for the treatment of MS.

The biodiversity of the agroecosystem leads to changes in the Floristic composition, diversity, and abundance of weed species in crop and orchard farms. The present study is considered the first attempt to categorize, evaluate, and document the diversity of weed populations among different grape farms in Tayma, Tabuk region, Saudi Arabia. The results of ANOVA showed that the number of weed species varied significantly across the six grape farms. During six grape farms under study, 30 weed species belonging to thirteen families and 28 genera were recorded. In total, twelve weed species belong to the Poaceae family with a rate of 40%. While the other families are represented either by one or two species with rates of 3.3% and 6.7%, respectively. Biogeographic origins analysis showed that the Tropical area represented 23.3% of total weed flora, followed by Saharo Arabian (16.7%), Mediterannean-Euro Siberian-Irano-Turanian (13.3%) and American (10.0%). Therophytes were the dominant life forms with a rate of 66.7%, followed by Hemicryptophyte (20.0%), Geophyte (6.7%), and Chamaephyte (6.7%). A total of 16 and 14 weed species belong to the annual and perennial life span, respectively. Also, the grass and herb habits represented 40% and 60% of 30 weed species, respectively. According to the abundance score, the most common weed species of those grape farms in the region study are Cynodon dactylon, Desmostachya bipinnata, and Setaria verticillata, they belong to the Poaceae family. Based on the presence and absence of weed species in grape farms, the Principal Component Analyses (PCA) show that the grape farms and weed species (into two groups) are distinguished mainly along the first two components. Also, positive correlations were observed among most grape farms, according to PCA. Generally, more studies on the ecological aspects and floristic composition of weed species in grape farms are also needed.

Aims: Local varieties of “Hönüsü” and “Horozkaras?” red grapes have been evaluated for the production of red wine and characterized33 for their chemical and sensory characteristics. Study Design: This research was initiated by the Food microbiology researcher and Applications Unit of Fermentation Laboratory. Place and Duration of Study: Laboratory of Fermentation of the Food Engineering Department in Gaziantep University, October 2019 to May 2021. Methodology: All wines were produced by a standard procedure of vinification. Three types of red wines were produced from combinations of red grapes. Ten phenolics were quantitatively detected in the red wines during processing. Brix, alcohol, pH and free SO2 contents were also detected. Results: The results showed that the type of technology affects significantly (p<0.05) the level of phenolic compounds formed during processing. Horozkaras?” red grape contributed to the highest amount of phenolic compounds in red wines. Gaziantep wine provides valuable information about the production of red wine from Gaziantep red grapes. Conclusion: The phenolic compounds of red wines were significantly (p<0.05) higher than that of musts. Many of the remarkable features of the phenolic profiles and Brix of grape varieties could help us to characterize Gaziantep wines. The mixture of must from “Hönüsü” and “Horozkaras?” red grapes with 7:3 ratio contributes suitable sugar and phenolic compounds for red wine. The results from this study provide valuable information about the red wine produced from the ancient grape varieties of the Southeast region.

El objetivo del estudio es determinar el efecto de distintas concentraciones de un gel experimental a base de extracto etanólico de semillas de uva (Vitis vinifera L) sobre la resistencia a la adhesión de resina nanohíbrida. Se obtuvo el extracto etanólico a base de Semillas de Uva (EESU) por maceración en alcohol al 70 % durante 45 días. Se reconstituyó el principio activo de las semillas de uva en forma de polvo mezclando 200 ml de agua ultrapura, 2 g. de Carbopol 940, 20 g. de glicerol, 20g. de propilenglicol, 1g. de benzoato de sodio, 1.5 de trietanolamina, obteniendo un gel con un pH de 14, en concentraciones de 5 %, 10 % y 15 %. La sustancia madre fue sometida a cromatografía en capa fina HPLC para identificar sus componentes químicos hallando compuestos fenólicos (ácido Gálico), taninos y en menor cantidad flavonoides. No existieron diferencias significativas entre los grupos control y experimentales cuya tracción se realizó inmediatamente al tratamiento de aclaramiento dental, a los 7 días y 14 días. El extracto etanólico de semillas de uva (Vitis Vinífera L) en concentraciones de 5 %, 10 % y 15 % tuvieron el mismo efecto sobre la resistencia a la adhesión de resina nanohíbrida.
The objective of the study is to determine the effect of different concentrations of an experimental gel based on ethanolic extract of grape seeds (Vitis vinifera L) on the resistance to adhesion of nanohybrid resin The ethanolic extract based on Grape Seeds (EESU) by maceration in 70 % alcohol for 45 days. The active principle of grape seeds was reconstituted in powder form by mixing 200 ml of ultrapure water, 2 g. of Carbopol 940, 20 g. glycerol, 20g. of propylene glycol, 1g. of sodium benzoate, 1.5 of triethanolamine, obtaining a gel with a pH of 14, in concentrations of 5 %, 10 % and 15 %. The base substance was subjected to HPLC thin-layer chromatography to identify its chemical components, finding phenolic compounds (Gallic acid), tannins and, to a lesser extent, flavonoids. There were no significant differences between the control and experimental groups whose traction was performed immediately after dental whitening treatment, at 7 days and 14 days. The ethanolic extract of grape seeds (Vitis vinifera L) in concentrations of 5 %, 10 % and 15 % had the same effect on the resistance to the adhesion of nanohybrid resin.

Objectives: The present study was designed to examine the efficacy of Cissus quadrangularis paste on fracture healing in artificially induced fractured rabbits. Methods: Fifteen rabbits were separated into three groups namely A, B, and C. Veldt grape paste was applied in groups B and C (Treatment Group) by close reduction and open reduction methods of fracture management; respectively, while group A was kept as control. The blood parameter and fracture healing properties in all animals have been monitored and examined routinely during the study period. Results: Both treated groups revealed lower serum calcium levels (SCL) than the control group after 24 hours of fracture that became within the normal range on the 14th day. Fracture healing in the treated groups has been commenced more rapidly than the control group with complete bridging of discontinuity by a distinct osseous callus in the fracture line on day 7 and complete effacing of fracture line on day 14. Conclusion: We did not find any type of anomalousness, clinical deviations, and alteration of serum calcium level on the 14th day of the fracture in treated animals hence Veldt Grape paste could be readily applicable to the management of the fracture in animals.
Objetivos: avaliar a eficácia da pasta de Cissus quadrangularis na consolidação de fraturas em coelhos fraturados artificialmente. Metodos: quinze coelhos foram separados em três grupos (A, B e C). A pasta foi aplicada nos grupos B e C (Grupo de Tratamento) pelos métodos de redução fechada e redução aberta de gerenciamento de fraturas; respectivamente. O grupo A foi mantido como controle. O parâmetro sanguíneo e as propriedades de cicatrização de fraturas em todos os animais foram monitorados e examinados, rotineiramente, durante o período do estudo. Resultados: ambos os grupos tratados revelaram níveis séricos de cálcio (SCL) mais baixos do que o grupo controle, após 24 horas de fratura que se tornou normal no 14º dia. A cicatrização da fratura nos grupos tratados foi iniciada mais rapidamente do que o grupo controle, com ponte completa de descontinuidade por um calo ósseo distinto na linha de fratura no dia 7 e apagamento completo da linha de fratura no dia 14. Conclusao: não encontramos nenhum tipo de anomalia, desvios clínicos e alteração do nível sérico de cálcio no 14º dia da fratura nos animais tratados, portanto, a pasta Veldt Grape pode ser aplicável ao manejo da fratura em animais.

Abstract Grape seed oil, which is usually extracted with highly toxic organic solvents that are harmful to human health, is produced from tons of grape pomace waste, generated during winemaking. Sometimes, this waste is used to make compost or is burnt, which causes environmental contamination. The functional qualities, antioxidant capacity (AC), α-tocopherol and total phenolic compounds content (TPC) of Black Borgoña (Vitis labrusca) grape seed oil, extracted by supercritical CO2, were evaluated. The high content of linoleic acid (ω-6) and monounsaturated fatty acids contributed to the beneficial effect on the functional quality indices, which were 0.20, 0.23, 11.80 for IA, IT and H:H, respectively. In addition, a POV of 6.23 ± 0.08 milliequivalents of peroxide/kg oil and an anisidine index of 2.70 ± 0.05 indicated a good quality oil. Also, a high concentration of a-tocopherol (9.82 ± 0.02 mg/100 g oil) and a high TPC ("4.14 ± 3.24 mg GAE/kg oil) were obtained. This study demonstrated that supercritical CO2 extraction is a suitable method for the delivery of a high-quality grape seed oil.
Resumen El aceite de semilla de uva que generalmente se extrae con disolventes orgánicos altamente tóxicos y perjudiciales para la salud humana, se produce a partir de toneladas de residuos de orujo de uva, generados durante la elaboración del vino. A veces, estos residuos se utilizan para hacer compost o se queman, lo que provoca la contaminación del medio ambiente. Se evaluaron las cualidades funcionales, la capacidad antioxidante (AC), el contenido de a-tocoferol y los compuestos fenólicos totales (TPC) del aceite de semilla de uva Borgoña Negra (Vitis labrusca), extraído mediante CO2 supercrítico. El alto contenido de ácido linoleico (ω-6) y de ácidos grasos monoinsaturados contribuyó al efecto beneficioso sobre los índices de calidad funcional que fueron de 0.20, 0.23, ''.80 para IA, IT y H:H, respectivamente. Además, un POV de 6.23 ± 0.08 miliequivalentes de peróxido/ kg de aceite y un índice de anisidina de 2.70 ± 0.05 indicaban una buena calidad del aceite. También se obtuvo una alta concentración de α-tocoferol (9.82 ± 0.02 mg/100 g de aceite) y un alto TPC ("4.14 ± 3.24 mg de GAE/ kg de aceite). Este estudio demostró que la extracción con CO2 supercrítico es un método adecuado para obtener un aceite de semilla de uva de alta calidad.

O alto teor de ácido linoleico (ω-6) e ácidos graxos monoinsaturados contribuiu para o efeito benéfico sobre os índices de qualidade funcional que foram 0.20, 0.23, 11.80 para IA, IT e H:H, respectivamente.
