Résumé

BACKGROUND: A recent day is one of a rare drug plant, which use in traditional medicine a long time ago. Therefore based on nature resource of traditional medicine, on the base of evaluating pharmacological and biological action to develop a new drug of plant origin is important not only for treatment, but also has a economic significance. This plant has profound medicinal use and is a proved antipyretic, analgesic anti inflammatory and anti-cancer. No detail report was found in literature to evaluate renal damage experimentally in rats. The present study was hence designed to determine protective effect (Saposhnikovia divaricate (Turcz) Schischk) kanamycin-induced nephrotoxicy in rats. In addition, we attempted to test and compare the possible action of Saposhnikovia divaricate (Turcz) Schischk) kanamycin-induced nephrotoxicity in rats. MATERIALS AND METHODS: Experimental animals A dry extract of SDS is root was prepared by the lyophilization method and used in the study. Threemonth old Wistar albino rats of either sex weighing 150-250 g were used for the study. The animals were placed at random and allocated to treatment groups in polypropylene cages with paddy husk as bedding. Animals were housed at a temperature of 24 ± 2⁰C and relative humidity of 30-70%. A 12/12 h light and dark cycle was followed. All animals were fed on standard balanced diet and provided with water ad libitum. All the experimental procedures and protocols used in the study were reviewed and approved by the Bio-Medical Ethical Committee of Mongolian National University of Mongolia. Biochemical estimation At the end of experimental period, rats were anaesthetized with ether. The pathological model of kidney we use lipid peroxidation in the process of kidney inflammation kidney tissue, blood serum, erythrocyte MDA of membrane amount on 3, 7, 14 day by using spectrophotometer apparatus of “Shimadzu” firm of Japan and measured absorption at 535 nm. Statistical Analysis: Statistical analysis of data was performed by SPSS 16.0 program and analyzed statistically using criteria of Student t test. RESULTS: When we studied effect of extract of SDS preparation to condition of lipid peroxidation activation in the process of kidney inflammation by the indices of MDA which contains in blood plasma and erythrocyte membrane, MDA amount in renal tissue, amount of MDA decreased and it showed effect of decreasing lipid peroxidation MDA of plasma. CONCLUSION: The extract of Saposhnikoviadivaricata (Turcz) Schischk) has action of protecting kidney and decreasing lipid per oxidation.

Aims: This study aims to determine the antioxidant enzyme EC-SOD polymorphisms in Egyptian patients with type 2 diabetes mellitus with and without retinopathy, and its association with other biochemical changes to assess whether decreased SOD activity is associated with the development of diabetic complications. Study Design: Cross-sectional study. Place and Duration of Study: Department of Medicine Al-Hussein University Hospital, Cairo, Egypt, between May 2010 and April 2011. Methodology: The present study investigated the relationship among diabetes mellitus, lipid profiles, SOD activity, ESR, and CRP in the blood of 40 patients with type 2 diabetes with and without retinopathy and 20 healthy control subjects. The mean age of the diabetic patients was similar to that of control. The mean duration of the disease was 3.53 ± 1.17 years (1–5 years) in patients with type 2 diabetes without retinopathy and 18.7 ± 3.1 with retinopathy. Also we studied Arg213Gly dimorphism of the EC-SOD gene in type 2 diabetic patients with and without retinopathy and control persons using the PCR technique. Results: Superoxide dismutase (SOD) level was significantly decreased in diabetics and more markedly decrease in those with retinopathy. Total cholesterol (CHOL), triglyceride (TG) and low-density lipoprotein-cholesterol (LDL-c) levels were significantly increased in diabetics and more markedly increased in those with retinopathy compared with the control level. Genotype distribution of the EC-SOD in diabetic patients with and without retinopathy differed from normal individuals, as the argentines-to-glycine amino acid substitution was higher in diabetic patients compared with the normal individuals. Conclusion: The present study revealed that hyperglycemia produced marked oxidation impact as evidenced by a significant increase in lipid profile, lipid per oxidation products, as well as a significant decrease in the total SOD activity. Moreover, it showed that the genotype distribution of the EC-SOD was differed as the arginine-to-glycine amino acid substitution was higher in diabetic patients with and without retinopathy compared with the control individuals. This report emphasizes the important role of superoxide dismutase and its genotype distribution in diabetic patients with and without retinopathy, and hence the need for antioxidant supplements to delay the severity of diabetic retinopathy.

Résumé

To investigate the protective effect of the leaf extract of Andrographis paniculata (Ap) against ethanol induced liver toxicity in male albino rats. The liver toxicity was induced by the administration of ethanol to the animals at the optimum dosage of 7.9g/kg body wt., orally for 45 days. After induction of liver toxicity the aqueous plant extract of A.p was administered to the animal 250 mg/kg body wt., for 45 days. The liver toxicity and protective effect of the plant extract was assessed by the estimation of liver marker enzymes, antioxidant enzymes and liver histopathological studies. The ethanol induced animals the liver marker enzymes like ALT, AST, ALP and Bilirubin were significantly elevated (P<0.001) when compared to the normal animals. After administration of aqueous extract of A.p the elevated levels of marker enzymes were significantly decreased (P<0.001). The antioxidant enzymes were decreased significantly in ethanol induced animals after administration of plant extract the decreased levels were increased significantly (P<0.001). The aqueous leaf extract of A.paniculata could protect the liver against ethanol induced liver toxicity by possibly reducing the rate of lipid peroxidation and increasing the antioxidant defense mechanism in rats.
