Objective To investigate the value of nuclear matrix protein 22(NMP22),cytokeratin 20(CK20) and survivin mRNA in the diagnosis of bladder cancer.Methods The research enrolled 107 cases with hematuresis or irritation sign of bladder,who were divided into the control group(44 cases) and the observed group(63 cases).Besides,45 health volunteers were chose as the health group.The urine level of NMP22 was detected by ELISA,and CK20 and survivin mRNA by RT-PCR to evaluate the value in the diagnosis of bladder tumor The sensitivities and specificities of NMP22,CK20 and survivin mRNA were compared and analyzed.Results The urine level of NMP22 in the observed group(37.92 U/mL) was obviously higher than that of health volunteers (4.31 U/mL) and the control group(7.04 U/mL).The difference was significant(P＜0.05).The sensitivity and specificity of NMP22 were 82.54％ and 61.36％ respectively,which was unrelated with the tumor stage.The sensitivities and specificities of CK20 and survivin mRNA were 83.70％,63.64％ and 85.71％,90.91％ respectively,which was positively related with the tumor stage.The sensitivities of CK20 and survivin mRNA in the T2-T4 subgroup were higher than that T1s-T1 subgroup.And there was statistic difference(P＜0.05).Conclusion There are significant evidences that the detections of NMP22,CK20 and survivin mRNA as non-invasive measures could be better methods and higher sensitivity and specificity in diagnosing bladder cancer.

Objective To study the mierovessel density (MVD) marked by CD105 and expression of survivin gene in pathologic scar, and to investigate the function of oncogenes in tumorigenic process and abnormal scarring. Methods In situ hybridization and immunohistoehemical methods (SP technique) were used to detect survivin mRNA, CD105-MVD in 40 pathologic scar cases, 20 normotrophic scar cases, and 20 normal skin cases, respectively. The relationship between the expression of survivin mRNA and CD105-MVD in 40 pathologic scar cases were analyzed. Results Compared with normotrophic scar and normal skin cases, the expression of survivin mRNA, CD105-MVD were significant difference in pathologic scar cases (P<0.05). In pathologic scar tissue, the expression of survivin had significant correlation with MVD marked by CD105 (P<0.05). Conclusion survivin is up-regulated in pathologic scar, which may contribute to the vascular formation.

Objective To evaluate the expression of Srvivin mRNA in the occurrence and progress of prostate cancer.Methods The inhibitor of apoptosis gene Survivin mRNA in 45 prostate cancer cases and 30 benign prostatic hyperplasia(BPH) cases were detected by hybridization in situ technique.Results The average optical density of prostate cancer groups(0.4232±0.0085)was signifi cantly higher than BPH groups (0.3303±0.0834) (P=0.001 ).Among the different pathological grades [G1(0.3401±0.0474), G2 (0.4270±0.0074), G3 (0.4560±0.0883)],the difference was statistically significant in the expression of Survivin mRNA (P= 0.004).It was found that there was positive correlation between the transfer of lymph (0.4557±0.7921)and the expression of Survivin mRNA in prostate cancer(P=0.005 ).Conclusion Survivin mRNA is overexpression in prostate cancer.It presumed that Survivin participate in genesis,development of prostate cancer and closely correlated with prognosis.

Objective To evaluate the expression and significance of survivin and PCNA in human colorectal carcinoma. Methods Using in situ hybridization and immunohistochemistry(S-P method) respectively to detect survivin mRNA, and survivin and PCNA proteins in 64 cases of colorectal carcinoma. Objective:To study the change of the survivin mRNA and protein of BEP-2D cells during its malignant transformation.Methods:Normal BEP-2D cell and BEP-2D cells treated by cigarette smoke condensate(CSC)for 15 weeks(P-15),25 weeks(P-25)and 38 weeks(P-38)were respectively chosen to study the survivin gene and protein by RT-PCR(retro-translation PCR,RT-PCR)and immunohistochemical method.Results:The survivin mRNA was found in BEP-2D cells of P-15,P-25 and P-38,respectively with 0.56,0.80,and 0.81,but not in the normal BEP-2D cell.The survivin protein was found in the normal BEP-2D cell,but not in BEP-2D cells of P-15,P-25 and P-38.The levels of survivin protein expressian were different between BEP-2D cells of P-15,P-25 and P-38 and normal BEP-2D cell,with significant difference between P-15 BEP-2D cell and normal BEP-2D cell(P
