Objective:To investigate the value of fibroblast growth factor 2 (FGF-2) and microRNA-206 (miR-206) in predicting postoperative delayed union of closed tibial shaft fractures.Methods:The clinical data of 136 patients who underwent closed tibial shaft fracture surgery in Hospital of the 80 th Group Army of Chinese People's Liberation Army Ground Forces from May 2018 to May 2021 were retrospectively analyzed. Eighty-six patients who had delayed union of closed tibial shaft fractures were included in the observation group, and fifty patients who had normal union of closed tibial shaft fractures were included in the control group. Serum FGF-2 level was measured using the enzyme-linked immunosorbent assay, and serum miR-206 expression was detected using the real-time fluorescence polymerase chain reaction. The relationship between FGF-2 expression and miR-206 expression and closed tibial shaft fractures was analyzed. Results:At 1 day, 1, and 4 weeks after surgery, serum FGF-2 level was significantly lower in the observation group than the control group [(14.24 ± 2.15) ng/L vs. (20.36 ± 3.42) ng/L, (21.38 ± 3.27) ng/L vs. (30.45 ± 4.29) ng/L, (23.59 ± 4.36) ng/L vs. (36.67 ± 4.51) ng/L, t = 7.42, 8.42, 16.66, all P < 0.001]. Serum FGF-2 level gradually increased with time in each group. At 1 day after surgery, serum miR-206 expression was significantly lower in the observation group than the control group ( t = 7.50, P < 0.001). At 4 weeks after surgery, serum miR-206 expression was significantly higher in the observation group than the control group ( t = 17.24, P < 0.001). At 1 week after surgery, there was no significant difference in serum miR-206 expression between the two groups ( P > 0.05). Univariate analysis results showed that postoperative infection, FGF-2, and miR-206 were closely related to the delayed union of closed tibial shaft fractures after surgery (all P < 0.05). Multivariate logistic regression analysis results showed that postoperative infection ( OR = 1.93, 95% CI: 1.20-3.07), FGF-2 ( OR = 2.10, 95% CI: 1.31-3.36), miR-206 ( OR = 2.30, 95% CI: 1.35-3.89) were independent risk factors for delayed union of closed tibial shaft fractures after surgery (all P < 0.05). The receiver operating characteristic (ROC) curves plotting serum FGF-2 level and serum miR-206 expression after closed tibial shaft fractures showed that at 4 weeks after surgery, the optimal cut-off value of FGF-2 for predicting delayed union of closed tibial shaft fractures was 29.83 ng/L, with the area under the curve, sensitivity, and specificity of 0.76 (95% CI: 1.23-3.25), 79.34%, and 68.82%, respectively; at 4 weeks after surgery, the optimal cut-off value of miR-206 for predicting delayed union of closed tibial shaft fractures was 0.63, with the area under the curve, sensitivity and specificity of 0.72 (95% CI: 1.10-2.45), 75.33%, and 67.25%, respectively. The area under the curve, the sensitivity, and specificity of combined use of FGF-2 and miR-206 in predicting delayed union of closed tibial shaft fractures were 0.81 (95% CI: 1.35-3.26), sensitivity and specificity were 83.45% and 67.36% respectively. Conclusion:The decrease in serum FGF-2 level and the increase in serum miR-206 expression at 4 weeks after surgery are independent risk factors for delayed union of closed tibial shaft fractures. Combined use of FGF-2 and miR-206 can better predict the delayed union of closed tibial shaft fractures.

Résumé

A new species of Moenkhausia is described from the rio Machado drainage, Amazon basin, Brazil. It is diagnosed from congeners by its color pattern, consisting of the concentration of chromatophores on the anterior portion of body scales, the horizontally elongate blotch on caudal peduncle, a bright golden coloration of the dorsal portion of eye when alive, and a dark line crossing the eye horizontally. The new species has variable morphology regarding trunk lateral-line canals. Most fully grown individuals do not have enclosed bony tube in many lateral line scales, resembling early developmental stages of tube formation of other species. This paedomorphic condition is interpreted as a result of developmental truncation. Such evolutionary process may have been responsible for the presence of distinct levels of trunk lateral line reductions in small characids. Variation in this feature is common, even between the sides of the same individual. We reassert that the degree of trunk lateral-line tube development must be used with care in taxonomic and phylogenetic studies, because reductions in the laterosensory system may constitute parallel loss in the Characidae. We suggest the new species to be categorized Near Threatened due to the restricted geographical distribution and continuing decline in habitat quality.(AU)


Uma espécie nova de Moenkhausia é descrita da drenagem do rio Machado, bacia Amazônica, Brasil. É diagnosticada das congêneres pelo padrão de coloração, que consiste na concentração de cromatóforos na porção anterior das escamas do corpo, em uma mancha horizontalmente alongada no pedúnculo caudal, na coloração dourada brilhante da porção dorsal do olho quando vivo e na faixa escura que atravessa o olho horizontalmente. A nova espécie apresenta variação na morfologia do canal da linha lateral do corpo. A maioria dos indivíduos totalmente desenvolvidos não possuem tubo ósseo fechado em muitas escamas da linha lateral, assemelhando-se aos estágios iniciais do desenvolvimento da formação do tubo de outras espécies. Essa condição pedomórfica é interpretada como resultado do truncamento do desenvolvimento. Tal processo evolutivo pode ter sido responsável pelos diferentes níveis de redução do canal sensorial de pequenos caracídeos. A variação neste caráter é comum, até entre os lados do mesmo indivíduo. Por isso, reafirmamos que o grau de desenvolvimento do canal sensorial do corpo deve ser usado com cuidado em estudos taxonômicos e filogenéticos, porque reduções no sistema látero-sensorial podem significar perdas paralelas em Characidae. Sugerimos que a espécie nova seja categorizada como Quase Ameaçada devido à distribuição geográfica restrita e ao declínio contínuo da qualidade do habitat.(AU)

Résumé

A new species of miniature fish of the characid genus Priocharax is described from a small lake near the rio Ipixuna, rio Purus drainage, Amazonas State, Brazil. It is distinguished from all congeners except P. pygmaeus by the lower number of teeth on the maxilla and dentary. It differs from P. pygmaeus by the presence of two postcleithra and 22-27 branched anal-fin rays (vs absence and 19-22). The new species is further distinguished from other species of Priocharax by a combination of characters involving the number of pelvic-fin rays and branched anal-fin rays, the number of postcleithra, the shape of postcleithrum 3, and the absence of the claustrum. Molecular evidence based on COI sequences of all valid species of Priocharax also corroborates the validity of this new species.(AU)


Uma nova espécie de peixe miniatura do gênero de caracídeo Priocharax é descrita de um pequeno lago próximo ao rio Ipixuna, drenagem do rio Purus, estados do Amazonas, Brasil. Ela difere de todas as congêneres, exceto P. pygmaeus pelo menor número de dentes no maxilar e dentário. Distingue-se de P. pygmaeus pela presença de dois pós-cleitros e 22-27 raios ramificados na nadadeira anal (vs ausência e 19-22). A nova espécie também difere de outras congêneres por uma combinação de caracteres como número de raios das nadadeiras pélvica e anal, número de pós-cleitros e formato do pós-cleitro 3, e ausência de claustrum. Evidências moleculares com base em sequências do gene COI de todas as espécies válidas também corrobora a validade da nova espécie.(AU)

OBJECTIVE@#To explore the role of small nuclear noncoding RNA 7SK in embryonic stem cell (ESCs) proliferation and the value of 7SK as a target for early diagnosis and treatment for primordial dwarfism (PD).@*METHODS@#ESC line R1 was transfected with the CRISPR/Cas9 system, and sequencing of the PCR product and glycerol gradient analysis were performed to identify novel 7SK deletion mutations. A lentivirus system was used to knock down cyclin-dependent kinase 9 (CDK9) in clones with 7SK deletion mutations, and the effect of CDK9 knockdown on the protein level of cell division cycle 6 (CDC6) was analyzed with Western blotting.@*RESULTS@#We identified a novel deletion mutation of 7SK at 128-179 nt in the ESCs, which resulted in deficiency of cell proliferation. 7SK truncation at 128-179 nt significantly reduced the protein expressions of La-related protein 7 (LARP7) and CDC6.@*CONCLUSIONS@#7SK truncation at 128-179 nt can significantly impair proliferation of ESCs by downregulating CDC6. 7SK is a key regulator of proliferation and mediates the growth of ESCs through a mechanism dependent on CDK9 activity, suggesting the value of 7SK truncation at 128-179 nt as a potential target for early diagnosis and treatment of PD.

Résumé

Sampling truncation, i. e. sampling ends before signal decays to zero, introduces wiggle-like artifacts that impair the quality of nuclear magnetic resonance ( NMR) spectra. In multidimensional ( mD) NMR experiments, the first few hundred or less data points are commonly sampled in the indirect dimension, and the truncation is unavoidable. Apodization can suppress truncation artifacts with cost of line-broadening. Linear prediction is also beneficial to the reduction of truncation artifacts. The worst situation is in the constant time ( CT) type experiments, where the signal doesn ' t decay at all in the CT evolution dimension. In this contribution we proposed that, although iterative soft thresholding ( IST) was rarely used due to the difficult parameter tuning, it was particularly suitable to suppress the truncation artifacts in the CT type NMR experiments. The simulation and experiments were performed to show the performance of this method. And the processing result was compared with a method proposed recently.

Objective Nonparametric maximum likelihood estimate(NPMLE)and Breslow-Fleming-Harrington estimate(BFH)are extremely sensitive to small risk set for left truncated and right censored data,this study aims to develop estimation methods to improve the estimation accuracy and compare the existing methods.Methods We introduced the NPMLE,weighted NPMLE,conditional NPMLE,BFH and a new weighted BFH estimate.Simulation studies were carried out to compare five methods via the integrated absolute error(IAE) and integrated average width(IAW).Results The IAE of NPMLE,BFH,weighted NPMLE,weighted BFH and conditional NPMLE is ascending in turn;The IAW of weighted BFH is the lowest and NPMLE is the largest,BFH,conditional NPMLE and weighted NPMLE is reversed under different censored rate.Conclusion According to the results of simulation and example,weighted BFH and weighted NPMLE is recommended in turn when the risk set is small.Otherwise,the results of five methods would be consistent.

PURPOSE: A new compressed sensing technique by iterative truncation of small transformed coefficients (ITSC) is proposed for fast cardiac CINE MRI. MATERIALS AND METHODS: The proposed reconstruction is composed of two processes: truncation of the small transformed coefficients in the r-f domain, and restoration of the measured data in the k-t domain. The two processes are sequentially applied iteratively until the reconstructed images converge, with the assumption that the cardiac CINE images are inherently sparse in the r-f domain. A novel sampling strategy to reduce the normalized mean square error of the reconstructed images is proposed. RESULTS: The technique shows the least normalized mean square error among the four methods under comparison (zero filling, view sharing, k-t FOCUSS, and ITSC). Application of ITSC for multi-slice cardiac CINE imaging was tested with the number of slices of 2 to 8 in a single breath-hold, to demonstrate the clinical usefulness of the technique. CONCLUSIONS: Reconstructed images with the compression factors of 3-4 appear very close to the images without compression. Furthermore the proposed algorithm is computationally efficient and is stable without using matrix inversion during the reconstruction.

Résumé

It is necessary predict the effect of aquifer stresses in surface water and wetlands and consider the mutual effects that are produced by the conjunctive use of surface water and groundwater. This was originally made with very simple idealized analytical methods. The next development was the application of finite differences or finite elements numerical models, but poses problems when the model has to be run many times to analyze different management alternatives. When aquifer behavior is linear, as in confined, semiconfined, or unconfined aquifers with not too large changes in its saturated thickness, it is possible to apply the superposition strategy through influence functions. That has simplified significantly modeling and improved the effectiveness of management models. However, for large models, long modeling periods and a large number of alternatives, it is needed to handle and store many influence functions and to consider and store all the previous stresses. In that case, the eigenvalue method can be a more appropriated option. This approach solves the spatially discretized flow equation explicitly and continuously in time, obtaining modal orthogonal components through very simple explicit state equations in function of time. To reduce the computational load, the simulation can be simplified with appropriate truncation using only dominant modes of the components at the expense of a small error. Efficient methods have been developed to get the modal components as well as to perform truncation with limited errors.

Objective To investigate the relationship between truncating mutations of APC gene and sporadic colorectal cancer,and analyze the feasibility of non-radioactive protein truncation test (PTT) in the detection of mutations of APC gene.Methods Ninety-six samples of sporadic colorectal cancer tissues ( including 44 patients with colonic cancer and 52 patients with rectal cancer) were obtained from the General Hospital of Ningxia Medical University from September 2008 to September 2010.The mutation cluster region of the APC gene was screened using digital PTT labeled with fluorescent Lys-t-RNA,with a polymerase chain reaction fragment amplified from genomic DNA serving as a tenplate for in vitro translation.The occurrence of gene mutation was determined according to the emergence of truncated peptides.The mutation cluster region of the APC gene in 46 samples of colorectal cancer tissues was analyzed by direct sequencing.The detection rates of the 2 methods were compared by chi-square test.Results Thirteen (26%) truncated peptides were detected in the 50 samples of colorectal cancer tissues.The mutation type of 4 samples is nonsense mutation,which resulted in emergence of truncated gene products.Eleven (24%) truncated peptides were detected in the 46 samples of colorectal cancer tissues.There was no significant difference in the detection rates between PTT and direct sequencing ( x2 =0.033,P > 0.05 ).Conclusions Truncating mutations of APC gene are common alterations in sporadic colorectal cancer in the Chinese.Digital PTT labeled with fluorescent Lys-t-RNA is rapid and high-sensitive in screening gene mutations.

Maximal oxygen uptake (VO<sub>2</sub>max) is an important determinant of health-related physical fitness. In 2006, the Japan Ministry of Health, Labour and Welfare (JMHLW) officially declared a standard reference value and reference interval of VO<sub>2</sub>max. However, these values were established on the basis of a systematic review of reports published in Western countries and were not based on actual VO<sub>2</sub>max data of the Japanese population. Therefore, we conducted a study entitled “The study on a minimum zone of VO<sub>2</sub>max as one of the determinants of health-related physical fitness in Japan” from 2004 to 2006 as a project of the Japanese Society of Physical Fitness and Sports Medicine (JSPFSM). In addition, we collected published VO<sub>2</sub>max data of the Japanese population from the JSPEFM website. In the present study, we attempted to determine the reference interval of VO<sub>2</sub>max with regard to age, gender, and different methods of exercise. Further, we established a cut-off value of VO<sub>2</sub>max for determining metabolic syndrome (MS).1. Reference interval of VO<sub>2</sub>maxFor both men and women, 325 and 364 values for the treadmill exercise, and 1175 and 2178 values for the cycle ergometer exercise, respectively, were collected. This data revealed a balanced distribution of VO<sub>2</sub>max with regard to age. Data that satisfied the VO<sub>2</sub>max criterion were used for the analysis. These data were regressed to age on gender and methods of exercise. The percentage of VO<sub>2</sub>max was calculated using the following equation: %VO<sub>2</sub>max = measured VO<sub>2</sub>max × 100/age-estimated VO<sub>2</sub>max. The iterative truncation method was used to calculate the reference interval of VO<sub>2</sub>max (70%∼130% VO<sub>2</sub>max) from the crude data of %VO<sub>2</sub>max, and then converted to actual VO<sub>2</sub>max. Thus, the reference interval of VO<sub>2</sub>max for healthy Japanese was determined with regard to age, gender, and different methods of exercise.2. Cut-off value of VO<sub>2</sub>max for determining MSUsing the VO<sub>2</sub>max data of subjects with body mass index (BMI) of ≧25kg/m<sup>2</sup> and ≧2 MS risk factors, and the data of subjects with normal BMI without any risk factors, we calculated sensitivity and specificity. The cut-off value was determined using the receiver operating characteristic curve. This cut-off value was defined as the critical value of VO<sub>2</sub>max that should be maintained to avoid MS and remain healthy.

Objective To reduce information loss in the refining processing of photoplethysmographic (PPG) pulse waveform. Method A novel PPG pulse waveform refining processing technique, named truncation/extrapolation technique was proposed. The key point of the proposed technique was to preserve the shape pattern of the most significant parts of the waveform at the expense of the distortion of the insignificant part. Thus, as compared with the conventional time-normalization method, the proposed technique ensured the accumulation coherent of the signal energy could be. Result Waveform analysis and comparison of experimental data between two methods verified the effectiveness of the proposed method in maintaining the most significant time domain features of the pulse waveform over a relatively wide range of the heart rate change, especially the shape and width of the dominant peak and the position of the dicrotic notch of the first derivative of PPG(dPPG). Conclusion The proposed method reduces the information loss in PPG pulse waveform refining processing, thus helps further processing of the PPG signal using some advanced signal processing techniques, such as waveform modeling and pattern recognition.

PURPOSE: Recent discovery of BRCA1 and BRCA2 genes has made it possible to perform presymptomatic diagnosis in hereditary breast/ovarian cancer families. We have previously reported germline mutations of the BRCA1 gene in Korean hereditary breast/ovarian cancer families. In that study two out of 13 families were found to have germline mutations in BRCA1 gene. One was a nonsense mutation in codon 1815, and the other was a frameshift mutation due to 2 base-pair deletion in codon 1701 of BRCA1 gene. This study was intended to identify germline mutations of the BRCA2 gene in Korean breast/ovarian cancer families. MATERIALS AND METHODS: Peripheral blood DNA was obtained from 10 breast cancer patients registered at the Korean Hereditary Tumor Registry with positive family history of breast and/or ovarian cancer. Exons 11 and 27 of the BRCA2 gene(together accounting for 50% of the coding region of the BRCA2 gene) were amplified by polymerase chain reaction(PCR) and screened for mutations by in vitro transcription/translation method. For confirmation of the mutations, automatic sequencing of the PCR products displaying abnormal truncated protein bands was perfomed. RESULT: We identified an abnormal truncated protein in the exon 11 of the BRCA2 gene from a member of hereditary breast cancer family, SNU-B4. Sequencing analysis revealed a 4 bp deletion in codons 1248-49 of the exon 11, resulting in frameshift that led to premature stop codon and truncation of the protein product. CONCLUSION: We have identified a germline mutation from a Korean hereditary breast cancer family. So far only one case of the same mutation has been registered in Database of BRCA2 mutation (BIC) by a commercial genetic diagnosis company, Myriad Genetics, Inc. Identification of the germline mutation in BRCA2 gene should aid in the accurate presymptomatic diagnosis of the at-risk members in this family.

Résumé

Damage caused to rice production by coleopteran insects like rice weevil (Sitophilus oryzae), a stored grain insect pest and rice hispa (Dicladispa armigera), a pest of the growing plant is quite high. In order to combat the damage, generation of insect resistant transgenic rice plant was considered desirable. CryIIIA endotoxin of Bacillus thuringiensis var. tenebrionis, a 65 kDa protein toxic to coleopteran insects, figured as the candidate gene product. Thus, the cryIIIA gene was isolated from a local isolate of Bacillus thuringiensis var. tenebrionis. The gene was tailored at the N-terminal end to its minimal size by using a synthetic ATG codon which replaced the first codon next to ATG of threonine to proline. This modification did not affect the functional property of the gene product.A chimeric construct of the modified cryIIIA gene was developed containing CaMV35S promoter and nos terminator for plant expression. The expressibility of the cryIIIA gene in indica rice was judged through test for transient expression in indica rice protoplasts.
