Résumé

Cinnamon ( Cinnamomum verum J. Presl) is a well - known medicinal plant considered as an effective treatment for neurological disorders based on Persian medicine . The aim of the present study was assessing the effect of cinnamon oil, cinnamic acid, and cinnamaldehyde, on the in vitro model of Parkinson's disease (PD). Cinnamon oil, prepared in sesame oil, was phytochemically analyzed using high performance liquid chromatography (HPLC). Pheochromocytoma - 12 (PC - 12) cells were treated with 1 - methyl - 4 - phenyl - 1,2,3,6 - tetrahydropyridine (MPTP) as an in vitro model of neurodegeneration in PD. Cell viability, activity of caspase enzymes, and formation of reactive oxygen sp ecies (ROS) were evaluated. MPTP significantly decreased cell viability and increased Casp activity, as well as ROS formation. Cinnamon oil and cinnamic acid at 200 µg/m L could significantly reverse MPTP - induced abnormalities in PC - 12 cells including Casp activity and ROS formation. Our study supports the beneficial effect of cinnamon oil in neurodegeneration. Furt her investigations are needed to clarify the mechanisms and main active components.


La canela ( Cinnamomum verum J. Presl) es una planta medicinal m uy conocida, y considerada como un tratamiento efectivo para patologías neurológicas según la medicina persa. El objetivo de este estudio fue evaluar el efecto del aceite de canela, el ácido cinámico, y el cinamaldehído, en un modelo in vitro de la enferme dad de Parkinson (PD). El aceite de canela, preparado en aceite de sésamo, fue analizado fitoquímicamente usando cromatografía líquida de alta eficacia (HPLC). Se trataron células con feocromocitoma - 12 (P - 12) usando 1 - metil - 4 - fenil - 1,2,3,6 - tetrahidropiridi na (MPTP) como un modelo in vitro de neurodegeneración en PD. Se evaluó la viabilidad celular, actividad de enzimas caspasa, y formación de especies reactivas del oxígeno (ROS). El tratamiento con MPTP disminuyó significativamente la viabilidad celular y a umentó la actividad casp, así la formación de ROS. Aceite de canela y ácido cinámico a 200 µg/mL podría revertir significativamente las anormalidades inducidas por MPTP en células PC - 12, incluyendo la actividad casp y la formación de ROS. Nuestro estudio e ntrega sustento sobre los efectos benéficos del aceite de canela en la neurodegeneración. Se requiere más investigación para clarificar los mecanismos y los principales componentes activos.

The liver as an organ is important for the metabolism of drugs and toxins. However, it is not immune from environmental insults. Exposure of liver cells to carbon tetrachloride (CCl4) results in the generation of tricholoromethyl radicals, which induce liver toxicity. This study aims at investigating the ameliorative effect of the cinnamon aqueous extract (CAE) against CCl4-induced hepatotoxicity in male albino rats. Hepatotoxicity was induced in rats through the intraperitoneal administration of 0.5 mL kg-1body weight of CCl4. The analyses of the results obtained showed significant reduction in the levels of serum biochemical markers for 400 and 600 mg kg-1bw of CAE protected rats as compared with CCl4group. In addition, CAE administration reversed liver tissue damaged via increased antioxidants markers. Histopathological examination of CAE treatment on rats showed improved changes to the liver damage caused by CCl4 with no evidence of steatosis and inflammation. This result hence suggests that CAE has marked hepatoprotective and healing activities against CCl4-induced liver damage and could serve as a suitable candidate in drug discovery for the treatment of liver toxicity.

Résumé

The main objective of this study was to investigate the effects of high fat fructose [HF/HFr] diet on the onset of the characteristics of the metabolic syndrome and the levels of some adipocytokines in growing male and female rats. Also we aimed to study the possible protective effects of cinnamon [CN] against [HF/HFr] diet induced-metabolic disturbances. Forty eight growing Sprague-Dawley rats were randomly assigned into 4 groups [12 rats/group; 6 males + 6 females] and were fed for a period of 6 weeks the following diets [i] basal diet, control [C] [ii] basal diet containing 20 g CN/kg diet [C+CN], [iii] high fat diet [15% beef tallow + 5% corn oil] combined with fructose [13% W/V]in drinking water [HF/HFr], [iv] HF/HFr diet containing 20 g CN/kg diet [HF/HFr+CN], HF/HFr diet resulted in elevated levels of fasting blood glucose, serum insulin, and homeostasis model assessment [HOMA] index along with impaired glucose tolerance, as well as elevated serum triacylglycerols [TG], total cholesterol [TC], low-density lipoprotein cholesterol [LDL-C], resistin, leptin, tumor necrosis factor - alpha [TNF-alpha] and total oxidant capacity [TOC] with a decrease in high-density lipoprotein cholesterol [HDL-C] and total antioxidant capacity [TAC] compared to control. These deleterious effects were more pronounced in male and female rats. Cinnamon reinstated most of the altered measured parameters. So HF/HFr diet developed characteristics of metabolic syndrome in growing male and female rats. Females were protected against some metabolic disturbances of HF/HFr diet. The inclusion of cinnamon in the diet was effective in modulating some aspects of metabolic parameters associated with metabolic syndrome
