Résumé

In the present study, siderophore production capacity of various diarrhoeagenic E coli strains was tested on quantitative basis. The collected strains were classified under enteropathogenic E coli, enterotoxigenic E coli and entero-invasive E coli groups by using appropriate techniques. A few reference . enterotoxigenic E coli strains were also included in this study. The confirmation of various properties of enteropathogenic E coli, enterotoxigenic E coli and entero-invasive E coli strains was carried out by standard procedures. Several non-pathogenic E coli strains were also included in the study to assess their siderophore producing capacity. The results presented in the study showed that, the phenolate type of siderophore, that is enterochelin was predominantly produced by majority of these E coli strains. Only 30% of enteropathogenic E coli and 33% of enterotoxigenic E coli strains failed to produce any detectable level of enterochelin. On the other hand, only 50% of enteropathogenic E coli strains and all the entro-invasive E coli (100%) showed the positive aerobactin that is hydroxamate type of siderophore production ability. None of the enterotoxigenic E coli and non-pathogenic strains produced aerobactin.

Bacterial production of siderophores may involve specific genes related to nonribosomal peptide and polyketide biosynthesis, which have not been fully identified in the genome of Xylella fastidiosa strain 9a5c. However, a search for siderophore-related genes in strain 9a5c indicated five membrane receptors, including siderophore, ferrichrome-iron and hemin receptors. All these biomolecules are thought to be associated with iron transport and utilization. Eighty isolates obtained from citrus orchards containing trees that developed citrus variegated chlorosis (CVC) were screened for siderophore production. The results demonstrated that only 10 of the isolates did not produce siderophores. Additional strains obtained from coffee, almond, mulberry, elm, ragweed, periwinkle and grape also infected by X. fastidiosa were also shown by the chromeazurol bioassay to produce siderophores. In order to correlate siderophore production with the presence of siderophore-related genes, a polymerase chain reaction (PCR) was developed using specific primers for the catechol-type ferric enterobactin receptor (pfeA) and the hydroxamate-type ferrisiderophore receptor (fiuA) genes of strain 9a5c. The PCR results confirmed our hypothesis by demonstrating that amplification products were detected in all strains except for those isolates that did not produce siderophores.

Enteroinvasive "Escherichia coli" strains (EIEC) of different serotypes isolated from patients with acute diarrhea were examined for the ability to produce siderophores and iron-regulated outer membrane proteins (IROMP). For iron starvation cultures were grown at 37ºC in LB supplied with 200(micro)M of (alpha)-(alpha)'dypirydil. All strains produced enterobactin and twelve (40(per cent)) produced aerobactin. The strains showed IROMP varying from 67-82 kDa. Proteins were either induced or stimulated by the iron starvation. Differences were observed in the electrophoretic profile among the serotypes, originating 5 electrophoretic profiles. All serotypes expressed proteins of 82kDa (FepA) and 76 kDa (IutA) (except serotype O28ac:H(-) that did not produce the 76 kDa protein). Several strains (O29:H(-), O144:H(-), 0152:H(-), and O167:H(-)) expressed IutA in the outer membrane, in the abscence of aerobactin production. Additionally to well characterized proteins (FepA and IutA), we found two IROMP of unknown function in some serotypes: a 71 kDa protein was detected in three profiles and a 67 kDa protein was present in serotype O152:H(-). Moreover, two bands (39 and 43 kDa) which were not iron-regulated bound specifically to human lactoferrin
