RESUMO

OBJECTIVETo investigate the changes of subgroups of peripheral blood T lymphocytes in patients with severe acute respiratory syndrome (SARS) and its clinical significance.

METHODSSubgroups of blood T lymphocytes in 93 patients with SARS were detected by flow cytometer. The results detected in 64 normal subjects and 50 patients with AIDS served as controls.

RESULTSThe numbers of CD(3)(+), CD(4)(+), and CD(8)(+) lymphocytes all significantly decreased in acute phase of patients with SARS [(722 +/- 533)/microliter, (438 +/- 353)/microliter, (307 +/- 217)/microliter] compared with those in normal controls [(1527 +/- 470)/microliter, (787 +/- 257)/microliter, (633 +/- 280)/microliter, all P <0.01], which was different from what we observed in patients with AIDS who had decreased CD(4)(+) [(296 +/- 298)/microliter] but increased CD(8)(+) [(818 +/- 566)/microliter] counts. The counts of CD(3)(+), CD(4)(+), and CD(8)(+) lymphocytes decreased more apparently in patients with severe SARS. All the five patients who died had CD(4)(+) counts less than 200/microliter. As the patients' condition improved, CD(3)(+), CD(4)(+), and CD(8)(+) counts gradually returned to normal ranges.

CONCLUSIONThe damage of cellular immunity is probably an important mechanism of pathogenesis of SARS.

RESUMO

OBJECTIVETo make a comparative study of HLA-DQA1 and HLA-DRB1 allele frequencies in the cases of endometriosis and adeonmyosis.

METHODSThe allelic types of HLA-DQA1 and HLA-DRB1 were detected by polymerase chain reaction-sequence specific primers (PCR-SSP) technique in 51 cases of endometriosis, 45 cases of adenomyosis, and 44 normal individuals as the control.

RESULTSThe frequencies of HLA-DQA1*0401(7.8%, 10.0%) were significantly increased in the endometriosis group and the adenomyosis group (Pc=0.03, Pc=0.01), and the frequencies of HLA-DQA1*0301(8.8%, 5.6%) were significantly decreased in these two groups (Pc=0.00, Pc=0.00).There was no significant difference between the frequencies of HLA-DQA1 and HLA-DRB1 of endometriosis and adenomyosis.

CONCLUSIONThe results indicate that HLA-DQA1*0301 and *0401 alleles are associated with both endometriosis and adenomyosis, and there is perhaps common mechanism involved in both endometriosis and adenomyosis based on HLA-DQA1 and HLA-DRB1 allele frequencies.

RESUMO

Objective Vessel injury provokes the release of angiotensin II that influences the proliferation and migration of vascular smooth muscle cells (VSMCs). T lymphocytes produced, interleukin 10 is of immunoregulatory function. The purpose of this study is to determine the effects of recombinant human interleukin 10 (rhIL 10) on the proliferation of isolated rat vascular smooth muscle cell and the activity of p44/p42 mitogen activated protein kinase (MAPK) promoted by angiotensin II. Methods Rat aortic VSMCs were cultured and treated with rhIL 10 or angiotensin II, and then co treated with rhIL 10 and angiotensin II. The proliferation was quantified by colormetric assay. The p44/p42 MAPK activity was evaluated by the immunobloting technique using anti p44/p42 phospho MAPK antibody. Results Compared with control group, angiotensin II stimulated significantly VSMCs proliferation(1 311?0 201 vs 0 781?0 236, P 0 05), but significantly inhibited VSMCs proliferation induced by angiotensin II at a dose of 1,10,100 ng/ml (0 984?0 172 , 0 932?0 134 , 0 784?0 097 vs 1 311?0 201, P

RESUMO

Objective:To study the HLA-DPB1 allele polymorphism in Ewenki from Inner Mongolian.Methods:HLA-DPB1 allele polymorphism in normal Ewenki were determined by PCR with sequencing-based-typing(SBT).Results:20 HLA-DPB1 alleles were observed and compared with other ethnic groups, the allele frequency of HLA-DPB1*02012(24.4%) and DPB1*0402(22.6%),DPB1*0401(20.2%),DPB1*0501(10.7%) are highest, while others are lower. Conclusion:The distributions of HLA-DPB1 alleles frequencies in normal Ewenki from Inner Mongolia has a unique style. It is most important to further study anthropology and related to illness in Ewenki nationality.

RESUMO

AIM: To determine the effects of recombinant human interleukin-10(rhIL-10) on proliferation of vascular smooth muscle cells(VSMC) by TNF-? and PDGF-BB and neointimal hyperplasia after rat carotid arterial injury.METHODS: Rat aortic VSMC was cultured and treated with rhIL-10 with or without tumor necrosis factor-?(TNF-?) and platelet-derived growth factor-BB (PDGF-BB), respectively.Proliferation of VSMC was quantified by colormetric assay.Cell cycle analysis was performed by flow cytomertry.SD rats were treated with recombinant human IL-10(rhIL-10) for 3 days after carotid arteries injury.Neointima to media area ratio at the site of arterial injury was measured at 28 days after balloon injury.RESULTS: Compared to control, both TNF-? and PDGF-BB stimulated VSMC proliferation. rhIL-10 alone had no effect on VSMC growth. With TNF-? or PDGF-BB stimulation,rhIL-10,at dose as low as 10 ?g/L,inhibited VSMC growth( P

RESUMO

AIM: To investigate the association between HBV infection and HLA-DPB1 gene in population of Guangzhou Chinese. METHODS: 58 unrelated patients (test positive of HbsAg,HBeAg,HbcAb) and 75 unrelated healthy control individuals were typed by sequencing based typing (SBT) method in their HLA-DPB1 gene. RESULTS: The phenotype frequencies of HLA-DPB1 alleles of patients and control have no significant difference. CONCLUSION: These results indicate that there is no association between HLA-DPB1 gene and HBV infection.
