RESUMO

Background: Cardiovascular diseases (CVD) are the leading cause of death throughout world population each and every year. Focus on dyslipidemia management is urgently required in India to halt the rising tide of CVD. The purpose of diabetic dyslipidemia study is a record based one, to find out the effect of Rosuvastatin plus Fenofibrate, in adult Type 2 diabetes with dyslipidemia, with high TGL/HDL ratio in Lipid profiles, in a tertiary care hospital in the Union territory of Puducherry.Methods: There were 101 patients hospital records were analysed in which male were 45 and females were 56. The various biochemical parameters like serum Total Cholesterol, HDL, LDL, TGL, Non-HDL, TCL/HDL Ratio and TGL/HDL ratio reports were collected before and after 12-weeks of Rosuvastatin 10 mg with Fenofibrate 145 mg combination, for the treatment period once daily for their lipid-lowering therapy.Results: The combination therapies of Rosuvastatin plus Fenofibrate were safe and feasible to achieve more TG goal and proved that has predominately decreased the elevated lipid profiles from the medical resources of our record based study. The use of combination medications of rosuvastatin (10mg) plus Fenofibrate (145mg) is often needed to effectively treat the lipid triad, by the potency of rosuvastatin to lower LDL-C and Fenofibrates effectiveness in lowering TG in treating mixed diabetic dyslipidemia.Conclusions: After Rosuvastatin (10mg) plus Fenofibrate (145mg), the lipid profile data proved that the importance of TGL/HDL ratio apart from the TCL/HDL ratio, for good lipid control in diabetic dyslipidemic patients.

RESUMO

Fertility enhancement effect of ethanol extract of leaf of Melastoma malabathricum was observed in male albino rats. The relative weight of the testis and epididymis were increased. The epididymal sperm count, motility and sperm abnormality were increased significantly in treated rats. There was an increase in serum protein, albumin, globulin, urea, creatinine and the activity of liver marker enzymes (SGOT, SGPT and ALP) levels of treated rats. The activities of serum antioxidants (CAT, SOD. GPX, GST and GRD) in leaf extract treated rats were increased. The results of the hormonal assay showed that increased serum levels of LH and testosterone but decreased in the serum levels of FSH and estrogen compared to control. The results of fertility test indicated that the treated adult male rats increased the number of female’s impregnation. In addition, the number of implantations and the number of viable fetuses were also increased. The results of the present study concluded that, ethanol extract of leaf of Melastoma malabathricum enhanced sperm concentration, motility and testosterone which might produce positive result in the male fertility.

The effects of theophylline (a phosphodiesterase inhibitor) and cAMP on 17α, 20ß-dihydroxy-4-pregnen-3-one-induced germinal vesicle breakdown was investigated in vitro in catfish (Clarias batrachus) oocytes. Folliculated oocytes incubated with 17α, 20ß-dihydroxy-4-pregnen-3-one at the concentration of 1 μg/ml induced 93·2 ± 2·23% germinal vesicle breakdown. When the oocytes were prestimulated with 17α,20ß-dihydroxy- 4-pregnen-3-one for 6 h and then treated with different concentrations of theophylline, there was a significant drop in the frequency of germinal vesicle breakdown at the concentrations 2·0, 1·5 and 1·0 mM. However, theophylline was found to be incapable of inhibiting germinal vesicle breakdown at its lowest concentration (0·5 inM). In the time course study, significant inhibition of germinal vesicle breakdown was recorded when 1 mM theophylline was added up to 30 h of 17α,20ß-dihydroxy-4-pregnen-3-one Stimulation but the inhibitory effect of theophylline gradually (time dependent manner) declined if the stimulatory time of 17α,20ß-dihydroxy-4-pregnen-3-one was increased. A similar inhibition of germinal vesicle breakdown was also recorded with various concentrations of cAMP. Except 0·5 mM, all the higher concentrations of cAMP significantly inhibited 17α,20ß-dihydroxy- 4-pregnen-3-one induced germinal vesicle breakdown.

The effect of inhibitors in the oocyte maturation of Clarias batrachus, was investigated in vitro using actinomycin D and cycloheximide. Full-grown immature oocytes incubated with salmon gonadotropin (SG-G100) and 17α, 20ß-dihydroxy-4-pregnen-3-one at the concentration of 1 μg/ml induced 86·0 ± 1·2% and 91·3 ± 2·4% of germinal vesicle breakdown, respectively. When the oocytes were incubated with SG-G100 (1 μg/ml) + different concentrations of actinomycin D or cycloheximide, a significant drop in the frequency of germinal vesicle breakdown was observed. Thus, gonadotropin-induced maturation was inhibited by both transcriptional and translational inhibitors. When the oocytes were incubated with 17α, 20ß-dihydroxy-4-pregnen-3-one (1μg/ml) + different concentrations of cycloheximide, a significant inhibition of germinal vesicle breakdown was recorded. However, the maturation was not inhibited when the oocytes were incubated in the presence of 17α, 20ß-dihydroxy-4-pregnen-3-one (1 μg/ml) and different concentrations of actinomycin D. This suggests that mRNA synthesis is not obligatory for 17α, 20β- dihydroxy-4-pregnen-3-one induced oocyte maturation. Based on the time course experiment, it was observed that the inhibition of maturation in cycloheximide treated oocytes extends up to 12 h after which the effect becomes slowly subdued.
