RESUMO

Mogrosides and steroid saponins are tetracyclic triterpenoids found in. Squalene synthase (SQS) and cycloartenol synthase (CAS) are key enzymes in triterpenoid and steroid biosynthesis. In this study, full-length cDNAs ofandwere cloned by a rapid amplification of cDNA-ends with polymerase chain reaction (RACE-PCR) approach. ThecDNA has a 1254 bp open reading frame (ORF) encoding 417 amino acids, and thecDNA contains a 2298 bp ORF encoding 765 amino acids. Bioinformatic analysis showed that the deduced SgSQS protein has two transmembrane regions in the C-terminal. Bothandhave significantly higher levels in fruits than in other tissues, suggesting that steroids and mogrosides are competitors for the same precursors in fruits. Combinedprediction and subcellular localization, experiments in tobacco indicated that SgSQS was probably in the cytoplasm or on the cytoskeleton, and SgCAS was likely located in the nucleus or cytosol. These results will provide a foundation for further study ofandgene functions in, and may facilitate improvements in mogroside content in fruit by regulating gene expression.

RESUMO

The 5-HTreceptor agonist 8-hydroxy-2-[di--propylamino] tetralin (8-OH-DPAT) promotes ejaculation of male rats, whereas dapoxetine delays this process. However, the gene expression profile of the brain at ejaculation following administrationof these two compounds has not been fully elucidated. In the present study, a transcriptomic BodyMap was generated by conducting mRNA-Seq on brain samples of male Sprague-Dawley rats. The study included four groups: pre-copulatory control (CK) group, ejaculation (EJ) group, 0.5 mg/kg 8-OH-DPAT-ejaculation group (DPAT), and 60 mg/kg dapoxetine-ejaculation (DAP) group. The resulting analysis generated an average of approximately 47 million sequence reads. Significant differences in the gene expression profiles of the aforementioned groups were observed in the EJ (257 genes), DPAT (349 genes) and the DAP (207 genes) compared with the control rats. The results indicate that the expression ofandwas significantly different after treatment with 8-OH-DPAT, whereas the expression ofwas significantly different after treatment with dapoxetine. Other genes, such as,and, exhibited significant differences in expression after the two treatments and are related to bladder cancer, renal cell carcinoma and sexual addiction. The present study reveals the basic pattern of gene expression that was activated at ejaculation in the presence of 8-OH-DPAT or dapoxetine, providing preliminary gene expression information during rat ejaculation.

CYP450 plays an essential role in the development and growth of the fruits of. However, little is known about thegene in. Here, based on transcriptome data, a full-length cDNA sequence ofwas cloned by reverse transcriptase-polymerase chain reaction (RT-PCR) and rapid-amplification of cDNA ends (RACE) strategies.is 1677 bp in length (GenBank accession No. AEM42985.1) and contains a complete open reading frame (ORF) of 1422 bp. The deduced protein was composed of 473 amino acids, the molecular weight is 54.01 kDa, the theoretical isoelectric point (PI) is 8.8, and the protein was predicted to possess cytochrome P450 domains.gene was highly expressed in root, diploid fruit and fruit treated with hormone and pollination. At 10 days after treatment with pollination and hormones, the expression of Sghad the highest level and then decreased over time, which was consistent with the development of fruits of. Hormonal treatment could significantly induce the expression of. These results provide a reference for regulation of fruit development and the use of parthenocarpy to generate seedless fruit, and provide a scientific basis for the production of growth regulator application agents.

RESUMO

Abstract: Total RNA was isolated from Siraitia grosvenorii fruit by the method of modified Trizol, according to S. grosvenorii fruit characteristics of rich phenols, polysaccharide, oil and proteins. The OD260/280, OD260/230, RNA integrity (RIN) and yield of the total RNA with this method were 2.01, 2.02, 9.50 and 260 mirog.g-1, respectively. The open reading frame (ORF) of dehydroascorbate reductase (DHAR), named as SgDHAR, was cloned by rapid amplification of cDNA ends (RACE) and RT-PCR method from S. grosvenorii. The GenBank accession number for this gene is KC907731. The SgDHAR gene contains a full-length cDNA of 1,252 bp including ORF of 819 bp and encodes a predicted protein of 272 amino acids. The molecular mass is 30.217 7 kD and the isoelectric point is 8.76. Homology comparison showed that it shared 87% nucleotide sequence homology with Cucumis sativus. Expression patterns using qRT-PCR analysis showed that SgDHAR was mainly expressed in fruit and stem, followed by flower, and was lowest in root, while the expression level was 6.83 times in triploid. T than that in diploid. Therefore, SgDHAR gene may be involved in abortion of triploid seedless S. grosvenorii.

RESUMO

<p><b>OBJECTIVE</b>To establish the growth curve of Siraitia grosvenorii and analyze the correlation between seed and growth of fruit.</p><p><b>METHOD</b>The growth curve fitting function was applied for the study of correlation between seed and fruit growth.</p><p><b>RESULT</b>The significant positive correlation existed between seed and horizontal diameter x vertical diameter, not between seed and flesh weight of single fruit.</p><p><b>CONCLUSION</b>The growth curve is a reciprocal function, and seed is one of major factors to influencing the size of fruits and the shape of fruit.</p>

RESUMO

<p><b>UNLABELLED</b>To explore the condition of CA storage for fruit of Siraitia grosvenorii.</p><p><b>METHOD</b>The CA storage of 5% CO2, 15% CO2, 30% O2 under normal temperature was applied in the experiments, and the results were compared.</p><p><b>RESULT</b>The results showed that CA storage condition could alleviate the reduction of weight loss and water content, the best CA storage condition were 5% CO2, which alleviates the reduction of firmness and increases of soluble solid content during the 45 day period.</p>

<p><b>OBJECTIVE</b>To analyze the relationship between the net photosynthetic rate and the quality of Siraitia grosvenorii.</p><p><b>METHOD</b>Taken two species of S. grosvenorii: Yongqing No. 1 and Yehong No. 1 as the experimental material, CI-310 portable photosynthesis system was used to test the net photosynthetic rate in the late growth period; HPLC was used to determine momordica-glycosides V.</p><p><b>RESULT</b>Light compensation point of Yongqing No. 1 was lower than that of Yehong No. 1 and the light saturation point was identical, the weight of dried fruit and the content of momordica-glycosides V of Yongqing No. 1 were higher than those of Yehong No. 1 at the late growth period.</p><p><b>CONCLUSION</b>The net photosynthetic rate can be used as an important subservient index to assess the quality of S. grosvenorii.</p>
