RESUMO

[ ABSTRACT] AIM: To observe the effect of circadian rhythms on wound healing of mouse corneal epithelium. METHODS:The C57BL/6 male mice were used in the study.A part of corneal epithelium (2 mm in diameter) was struck off by a golf-like knife to form a round wound area.The dynamics of epithelial healing in the wound area were ob-served under microscope with fluorescein staining.In addition, with related antibodies and DAPI, the dynamic changes of the neutrophils, platelets and dividing cells were also investigated.RESULTS:The healing rates in LL group (12 h light/12 h light) and DD group (12 h dark/12 h dark) were obviously slower than that in LD group (12 h light/12 h dark), mainly showing delayed re-epithelialization, decreased epithelial cells, increased diameter of blood vessel, and delayed re-cruitment of neutrophils and platelets, but more cell number.CONCLUSION:Disruption of circadian rhythms significant-ly inhibits the wound healing of corneal epithelium, mainly through delaying the inflammation and re-epithelialization, but aggravating the inflammatory responses.

RESUMO

Objectives To investigate the expression of matrix metalloproteinase-9 (MMP-9) and its tissue inhibitor (TIMP-1) in cutaneous squamous cell carcinomas (SCC) and the relationship between the expression and invasive growth and metastasis of SCC. Methods Immunohistochemical method of streptavidin-peroxidase (SP) was used to detect the expression of MMP-9 and TIMP-1 proteins on the paraffin embedded sections of 65 patients with cutaneous SCC and 5 cases of normal epidermis. The immunohistochemical results were analyzed quantitatively using an image analysis system. Results MMP-9 and TIMP-1 proteins were diffusely expressed on the tumor nests and the mesenchymal cells around the nests, while in normal epidermis MMP-9 and TIMP-1 were weakly positive. The expression level of MMP-9 protein and the ratio of MMP-9/TIMP-1 were higher in aggressive SCC group than those SCC in situ group (t = 2.33 and 2.36, respectively, P

RESUMO

AIM: To investigate quantitatively the tubulin expression of basal cells in patients with Alzheimer's disease(AD) and vascular dementia(VD) and evaluate its significance in diagnosing Alzheimer's disease. METHODS: ?-tubulin of basal cells was examined using LSAB immunohistochemical technique, and analyzed qualitatively and quantitatively by image analyzer. RESULTS: The integrated absorbance (I A ,261.43?25.21) and the average absorbance (A A ,1.89?0.14) in the AD group were significantly higher than those (I A ,120.55?19.71 and A A , 0.85?0.14, respectively)in the VD group ( P

RESUMO

Objective To explore the roles of Bcl-2, Bax, Bcl-xl proteins in the carcinogenesis of basal cell carcinoma(BCC). Methods Forty-six BCC cases were divided into 2 groups (21 in non-aggressive BCC group and 26 in aggressive BCC group) according to their pathological features. Bcl-2, Bax and Bcl-xl were evaluated in formalin fixed, paraffin embedded specimens by immunohistochemical methods and analyzed by image-analysis methods to obtain average optical density (AOD) of each protein for each case. The AOD data of 3 proteins and Bcl-2/Bax ratios were then compared between the 2 BCC groups. Results The level of Bcl-2 expression and Bcl-2/Bax ratio of non-aggressive BCC group were higher than those of aggressive group,(P=0.005 and 0.044, respectively). For the expression level of Bcl-xl and Bax, there was no statistically difference between the two groups (P=0.097 and 0.979, respectively). Conclusion Bcl-2, Bax,Bcl-xl proteins might participate in the carcinogenesis and development of BCC.

RESUMO

Objective To observe the distributions of epidermal growth factor receptor(EGFR),transforming growth factor ?(TGF ? 1,? 2,? 3)and thyroid transcription factor 1(TTF 1)in developing lungs,and to discuss their functions and relationships each other. Methods The expression of EGFR and TGF ? was examined in mouse fetal lungs by immunohistochemistry.TIGER image analysiser was used in TTF 1 quantitative study. Results 1.In the early developmental phase,EGFR localized in the epithelium of airway surface,TTF 1 was expressed predominantly in the nuclei of distal lung buds and TGF ? was expressed in the bronchioles with localization patterns.2.At the late developmental phase,the positive reactions of EGFR and TTF 1 were mainly detected in alveolar cells.TGF ? expressed in mesenchymal at high levels.3 The average optical density(A A )of TTF 1 in the distal epithelial cells increased gradually during the development,while their volume(VOL)and volume integrated optical density(VI A )all decreased.Conclusion\ The functions of EGFR,TGF ? and TTF 1 might be respectively different at different developmental stages.They not only stimulate the process of branching morphogenesis,but also modulate epithelial maturation and differentiation.\;[
