RESUMO

Objective To investigate the application effect of remote ischemic preconditioning (RIPC) combined with controlled low central venous pressure (CLCVP) in hepatectomy. Methods A total of 80 patients who underwent elective partial hepatectomy in Yougchuan Hospital Affiliated to Chongqing Medical University from May 2021 to April 2022 were enrolled and divided into control group (group C), CLCVP group (group L), RIPC group (group R), and RIPC+CLCVP group (group RL) using a random number table, with 20 patients in each group. The patients in group L received CLCVP, those in group R received RIPC, and those in group RL received both CLCVP and RIPC. The patients were compared in terms of perioperative general status and the levels of tumor necrosis factor-α (TNFα), alanine aminotransferase (ALT), aspartate aminotransferase (AST), and total bilirubin on preoperative day 1(D0), postoperative day 1(D1), postoperative day 3(D3), postoperative day 5(D5), and postoperative day 7(D7). A one-way analysis of variance or a repeated measures analysis of variance was used for comparison of normally distributed continuous data between groups, and the Kruskal-Wallis H test was used for comparison of continuous data with skewed distribution between groups; the chi-square test was used for comparison of categorical data. Results Compared with group R, group RL had a significantly shorter time of operation ( H =14.278, P =0.015), a significantly lower total infusion volume ( H =24.175, P =0.001), and a significantly lower estimated blood loss ( H =45.625, P < 0.001). Group and time factors had significant interaction effects on TNFα, ALT, and AST in the four groups ( P < 0.001; P =0.010; P =0.012). Group RL had a significantly lower level of TNFα than group L on D1( P < 0.001) and D3( P < 0.001). Group RL had a significantly lower level of ALT than group L on D1( P =0.008) and D7( P < 0.001). Conclusion For patients undergoing hepatectomy, RIPC combined with CLCVP can effectively reduce intraoperative blood loss, provide a clear surgical field, and shorten the time of operation; meanwhile, it can also inhibit inflammatory response by reducing TNFα, but it cannot effectively alleviate hepatic ischemia-reperfusion injury after hepatectomy under the context of CLCVP.

Objective:To investigate the effects of Buyang Huanwutang on the expression of microtubule-associated protein-2(MAP-2), neurofilament-M(NF-M), and growth associated protein-43(GAP-43)in rat sciatic nerve after sciatic nerve transection and anastomosis. To explore the mechanism of Buyang Huanwutang promoting peripheral nerve regeneration. Method:SD rats were selected as the experimental subjects, and sciatic nerve transection model was selected as the experimental model. They were randomly divided into model group, sham operation group, Buyang Huanwutang group high, medium and low dose (29.6, 14.8, 7.4 g·kg<sup>-1</sup>)group, and mecobalamin (0.156 mg·kg<sup>-1</sup>)group, the model group and the sham operation group were given distilled water intragastric administration. After successful modeling, each group was treated with relevant drugs for 4 weeks. After 4 weeks, sciatic nerve function index(SFI), degree of inclined plate test and hematoxylin-eosin(HE)of sciatic nerve in each group were tested. The expression levels of MAP-2, NF-M, and GAP-43 at the sciatic nerve anastomosis site were detected by immunohistochemistry and Western blot. Result:Compared with sham operation group, the expression levels of SFI, inclined plate test, MAP-2, NF-M and GAP-43 in model group were significantly increased (<italic>P</italic><0.01). Compared with model group, the expression levels of SFI, inclined plate test, MAP-2, NF-M and GAP-43 in Buyang Huanwutang high, medium and low-dose groups were significantly increased (<italic>P</italic><0.05,<italic>P</italic><0.01). Conclusion:Buyang Huanwutang has a positive effect on nerve regeneration after sciatic nerve transection and anastomosis in rats.

Objective: To evaluate the impact of mean platelet volume (MPV) on prognosis in patients with ST-segment elevation myocardial infarction (STEMI). Methods: A total of 1012 consecutive STEMI patients received percutaneous coronary intervention (PCI) in our hospital from 2010-01-01 to 2014-10-31 were enrolled. Major adverse cardiovascular events (MACE) were recorded in 2015-10. Based on ROC curve measured optimal critical point of MPV in all-cause death, the patients were divided into Low MPV group and High MPV group. The incidence of MACE was compared between 2 groups, the impact of MPV on prognosis of PCI treated STEMI patients was studied by uni- and multivariate COX regression analysis. Results: By ROC curve, the optimal critical point of MPV in all-cause death was MPV≤9. 466 fl. MPV≤9. 466 fl in Low MPV group, n=549 and MPV>9. 466 fl in High MPV group, n=463. The patients were followed-up for 34 (12-69) months. Compared with Low MPV group, the patients in High MPV group had the higher incidences of all-cause death and cardiac death, P<0. 001 and P=0. 001. With adjusted multivariates, COX regression analysis showed that MPV was an independent risk factor of all-cause death (HR=1. 463, P<0. 001) and cardiac death (HR=1. 458, P<0. 001) in relevant patients. Conclusions: Increased MPV at admission had the predictive value for long-term prognosis in PCI treated STEMI patients.

Objective To explore the effect of the unilateral lumbar anesthesia by hypobaric bupivacaine and general anesthe‐sia in elderly hip replacement and their influence on the stress response .Methods Forty elderly patients undergoing hip replace‐ment in our hospital from December 2013 to November 2014 were selected and randomly divided into the unilateral lumbar anesthe‐sia group and general anesthesia group according to the random number table ,20 cases in each group .Venous blood was collected to determine the levels of blood sugar ,serum catecholamine(CA) and cortisol(CORT ) at 10 min before anesthesia(T0 ) ,end of surgery (T1 ) and at postoperative 24 h(T2 ) .MAP ,HR ,SpO2 ,intraoperative adverse reactions and postoperative related complications were recorded at each time point ,and the comparative analysis was performed .Results All patients successfully completed surgery and the anesthetic effect was satisfied .The occurrence rates of intraoperative hypotension ,hypertension and bradycardia in the unilateral lumbar anesthesia group were lower than those in the general anesthesia group ,the differences had statistical significances (P>0 .05) .HR and MAP at T1 in the two groups were reduced ,but the differences between the two groups were no statistical signifi‐cance(P>0 .05);the levels of GLU ,CORT and CA were increased ,moreover the differences between the two groups were statisti‐cally significant(P<0 .05) .SpO2 at T2 in the general anesthesia group was obviously lower than that in the unilateral lumbar anes‐thesia group ,while CORT was higher than that in the unilateral lumbar anesthesia group ,the differences were statistically signifi‐cant(P<0 .05) .The occurrence rates of postoperative nausea and vomiting ,sore throat ,dizziness ,drowsiness and pulmonary infec‐tion in the unilateral lumbar anesthesia group were lower than those in the general anesthesia group ,the differences were statistical‐ly significant(P<0 .05) .Conclusion The unilateral lumbar anesthesia by using hypobaric bupivacaine and general anesthesia all could offer better anesthetic effect .The unilateral lumbar anesthesia by using hypobaric bupivacaine can effectively regulate the stress response in elderly patients with hip replacement .

<p><b>OBJECTIVE</b>To study the autophagy activity between rat bone marrow stem cells (BMSCs) neural differentiation in order to explore the mechanism involve in this process.</p><p><b>METHODS</b>BMSCs were passed by 3 generation, then was induced with the revulsant 2% (DMSO) + 200 µmol/L (BHA), NSE expression was detected by immunocytochemical stain, the mRNA expression of autophagy associated genes L3B, Beclinl, Atg5, Atg7, Atg10 were detected by RT-PCR, the autophagy protein LC3B was examined by Western blot and flow cytometry analysis.</p><p><b>RESULTS</b>BMSCs were passed by 3 generation, the purity of BMSCs could reach more than 90%, the morphology of cells were like fibroblasts, after the revulsant 2% DMSO + 200 µmol/L BRA induced, cells were extended long neurites, like nerve cells, positive rate of NSE staining was (83±5) %, RT-PCR results showed that the expression of autophagy associated genes LC3B, Beclinl, Atg5, Atg7 Atg0 were rised after BMSCs neural differentiation, Western blot analysis showed that the LC3B-II protein expression was increased after neural differentiation and the MFI of L3B was highten by flow cytometry.</p><p><b>CONCLUSION</b>Autophagy is increased after rat BMSC neural differentiation.</p>

Recently, some scholars suggested that it is important to keep a stablehemodynamic state and prevent the stress responses in geriatric patients undergoing total hip replacement [THR]. We conducted this randomized prospective study to observe anesthetic potency of unilateral spinal anesthesia and stress response to it in geriatric patients during THR. We compared the effect of unilateral spinal and bilateral spinal on inhibition of stress response through measuring Norepinephrine [NE], epinephrine [E] and cortisol [CORT]. Plasma concentrations of NE, E and CORT were determined in blood samples using ELISA [enzyme-linked immunosorbent assays] at three time points: To [prior to anesthesia] T1 [at the time point of skin closure], T2 [twenty-four hours after the operation]. Sixty patients were randomly divided into two groups: group A [unilateral spinal anesthesia] and group B [conventional bilateral spinal anesthesia]. 7.5tymg of hypobaric bupivacaine were injected into subarachnoid cavity at group A and 12mg hypobaric bupivacaine were given at group B. The onset time of sensory and motor block, loss of pinprick sensation, degree of motor block, regression of sensory and motor blocks and hemodynamic changes were also recorded. These data were used to evaluate anesthetic potency of spinal anesthesia. The results of this experiment show that unilateral spinal anesthesia can provide restriction of sensory and motor block, minimize the incidence of hypotension and prevent the stress responses undergoing THR. It is optimal anesthesia procedure for geriatric patients by rapid subarachnoid injection of small doses of bupivacaine

<p><b>OBJECTIVE</b>To construct inducible lentiviral vector containing human Notch1 intracellular domain (NICD) gene and enhanced green fluorescent protein (EGFP), and to study its expression in PC12 cells.</p><p><b>METHODS</b>NICD cDNA was amplified by RT-PCR from human placenta tissue. EGFP gene was amplified by PCR from pEGFP-C1. Both NICD and EGFP were cloned into pcDNA 3.1 (+) plasmid to form pcDNA3.1-Notch1-EGFP. Then the Notch1-EGFP fragment was separated and cloned into pLVX-Tight-puro to form pLVX-Notch1-EGFP. The lentivirus were packaged and harvested, which were used to infect PC12 cells. After antibody selection for 2 weeks, the PC12 cells were induced by doxycycline (Dox). The expression of Notch1-EGFP was detected by fluorescence microscope and flow cytometry.</p><p><b>RESULTS</b>The recombinant inducible lentiviral vectors (pLVX-Notch1-EGFP) were success fully constructed. The EGFP positive cell percentage was over 90% in transfected PC12 cells after 500 ng/ml Dox induction for 36 h. The expression of Notch1 was posited correlated to the Dox concentration. The expression of Notch1 increased with the duration of Dox induction, which got the peak at 36 h after Dox induction.</p><p><b>CONCLUSION</b>The recombinant inducible lentiviral vectors containing Notch1 and EGFP gene are successfully constructed, which provides an effective and simple method to regulate the expression of Notch1 in PC12 cells.</p>

<p><b>OBJECTIVE</b>To find out the perceived stress in general public during prevalence of severe acute respiratory syndrome (SARS) and its impact on health behavior.</p><p><b>METHODS</b>A retrospective survey was conducted in Guangzhou, Hangzhou, and Taiyuan according to the epidemic situations of SARS, and 2532 subjects were randomly selected from constructive industry, school, and commercial business and residents in urban and rural areas. The perceive stress was measured by Chinese perceived stress scale (CPSS), and health related behavior during SARS was tested by uniform and self-made questionnaire. EpiData 2.0 was used for data management and CPSS value was calculated according to answer to 14 questions contained in the scale. Health risk stress among different population group and health related behavior among low, medium and high stress state were analyzed by SPSS 11.5.</p><p><b>RESULTS</b>2424 subjects were involved in the survey. The CPSS value was measured from 0 - 49 (22.7 +/- 6.8), M = 24.0. 39.3% (953/2379) subjects were under the health risk stress. The health related behaviors such as washing hands, opening the window for air, keeping away from others when cough and sneeze, doing exercises etc were reduced with the stress increased. Logistic regression indicated that compared with the persons with the thoughts of nothing serious of SARS, without any dread of SARS, and knowing nothing about prevention of SARS, the perceived stress was significantly related with perceiving of the thread to certain extent (beta = 0.41, Wald chi(2) = 4.84, P = 0.03), worrying little about the epidemic (beta = 0.50, Wald chi(2) = 6.69, P = 0.01), worrying about it to certain extent (beta = 1.39, Wald chi(2) = 48.59, P = 0.00) and scared so much (beta = 1.77, Wald chi(2) = 53.59, P = 0.00), and knowing little about the prevention (beta = 0.74, Wald chi(2) = 4.48, P = 0.03), knowing something about prevention (beta = -0.98, Wald chi(2) = 8.29, P = 0.00) and knowing the prevention very well (beta = -1.18, Wald chi(2) = 10.66, P = 0.00).</p><p><b>CONCLUSION</b>The adoption of health related behaviors declined with increase of perceived stress. Opening connection to authority and government, enhancing the awareness of outburst affairs among general public and providing positive social support may be effective ways to reduce the population perceived stress.</p>

<p><b>OBJECTIVE</b>To explore the mechanism of "preventive acupuncture and moxibustion" for regulating hypothalamic function in ovariectomy rats.</p><p><b>METHODS</b>Thirty female SD rats, aged 3.5 months, were randomly divided into normal group, sham operation group, ovariectomy model group, preventive acupuncture group and preventive moxibustion group. After "preventive acupuncture and moxibustion" were previously given to the preventive acupuncture group and the preventive moxibustion group for one month, respectively. At the same time, ovarietomy was made in the preventive acupuncture group, the preventive moxibustion group and the ovariectomy model group. For the sham operation group, only a little fat around the ovary was removed without ovariectomy, and nothing was not applied to the normal group. Immunohistochemical and in situs hybridization methods were used to investigate changes of expressions of ER-alpha and ER-alpha mRNA in paraventricular nucleus, supraoptic nucleus and arcuate nucleus.</p><p><b>RESULTS</b>In the model group the expressions of ER-alpha and ER-alpha mRNA in paraventricular nucleus and the expression of ER-alpha mRNA in supraoptic nucleus increased significantly (P<0.01), but the expression of ER-alpha dropped significantly in both supraoptic nucleus and arcuate nucleus (P<0.01). After preconditioning with acupuncture or moxibustion, the expression of ER-alpha mRNA in paraventricular nucleus significantly reduced (P<0.05), and the expression of ER-alpha had no significant change (P>0.05); the expression of ER-alpha in supraoptic nucleus and arcuate nucleus significantly increased (P<0.01 or P<0.05), and the expression of ER-alpha mRNA in paraventricular nucleus had no significant change (P>0.05).</p><p><b>CONCLUSION</b>"Preventive acupuncture and moxibustion" at "Guanyuan" (CV 4) have an effect on the expressions of ER-alpha and ER-alpha mRNA in different hypothalamic nuclei in ovariectomy rats, which possibly is one of the ways for modulating the hypothalamic activity.</p>

<p><b>OBJECTIVE</b>To discuss the influence of the rhizome of Cibotium barametz on the heamorheology index in mice with adjuvant arthritis and to compare the effect of raw medicinals with that of the processed ones.</p><p><b>METHOD</b>Mice was injected with Freund's complete adjuvant on the rihgt behind foot to make model of adjuvant arthritis (AA). Hydroxyacrbamide tablets were orally administrated by mice with AA to make model of AA due to deficiency in the kidney (DK-AA). And then we determined the heamorheology index of the normal group, positive control group, AA group, DK-AA group and medicinals-treated groups.</p><p><b>RESULT</b>In the groups of AA, and DK-AA, the heamorheology index, such as high shearing, middle shearing, low shearing, plasma viscosity, whole blood reduction viscosity, erythrocyte aggregation exponent, erythrocyte degeneration exponent, sedimentation, sedimentation equation K value, erythrocyte rigidity exponent, erythrocyte electrophoresis time, casson viscosity, casson yield stress, increased significantly. After treated with Cibotium barametz, the heamorheology index except red blood count, packed cell volume, fibrinogen decreased obviously to get normal.</p><p><b>CONCLUSION</b>Rhizome of Cibotium barametz could promote heamorheology in mice with AA and DK-AA to exhibit effect of promoting blood circulation and remove blood stasis. The medicinal rhizomes processed with sand have the effect enhanced.</p>

<p><b>OBJECTIVE</b>To explore the situation of smoking behavior among the students of middle school in Beijing, Hangzhou, Wuhan and Urumchi and to analyze the relationship between smoking behavior and several unhealthy behaviors together with psychological troubles to provide evidence in developing an early intervention plan.</p><p><b>METHODS</b>The National Health Education Institute (NHEI) of Chinese Center for Disease Control and Prevention (CDC) provided relevant data on all middle schools in the 4 cities and then U.S. CDC randomly sampled 100 common middle schools from them with a special sampling process. The core questionnaire developed by the experts from WHO and other countries was used in the survey among 9015 sampled students.</p><p><b>RESULTS</b>Among all the sampled students, 29.4% of them had ever attempted cigarettes smoking while 6.6% of them tried tobacco in the 30 days before survey, 27.0% of the students with smoking behavior began smoking at the age of 9 or younger, 31.8% had learned how to refuse smoking from school education. The students with smoking behavior were more likely to drink alcohol, use drugs, bully others, be injured, miss classes, and have some psychological troubles than those without smoking behavior.</p><p><b>CONCLUSION</b>There were increasing trends noticed on the incidence of attempt and smoking cigarettes. Smoking was closely related to other unhealthy behaviors and psychological troubles. Comprehensive education activities on "no-smoking" should be implemented as early as possible among adolescents, as well as to promote training on life skills.</p>

<p><b>OBJECTIVE</b>To describe the changes of health related behavior and influencing factor during and after the prevalence of severe acute respiratory syndrome (SARS), as to providing evidence for inducing public health related behavior to cope with the emergent infectious public health hazards.</p><p><b>METHODS</b>According to the epidemic strength of SARS, Guangzhou, Taiyuan, Hangzhou were selected to be the research areas, and convenience sampling was used to identify 200 city residents, 200 country residents and 400 occupational populations from each area. The uniform and self-made questionnaire was carried out by about 2400 residents.</p><p><b>RESULTS</b>All 2424 subjects completed the questionnaire. The correlation coefficient of scores of health related behavior during and after SARS was 0.782 (P < 0.01). And 39.3% subjects were considered as under the health risked stress by CPSS. Multiple linear regression indicated that the scores of health related behavior were significantly related to the perceived stress, demographic characteristics and recognition of SARS.</p><p><b>CONCLUSIONS</b>The public health related behavior should be developing in the SARS, and part of health related behavior had been kept after SARS and part been decreased. Behavior change should depends on the prevalence of the disease, cultural heritage and behavior variability. More attention should be paid to induce the public psychology and behavior and enhance the degree of recognition on related knowledge through health education as to relieving the stress in populations when we conduct to cope with the emergent public health hazards.</p>

Objective To analyze and sort the character of symptom and pathology of 223 HIV/AIDS patients in Yunnan Provine. Methods Sort the clinical periods into groups subject to 223 HIV/AIDS patients who voluntarily accept the consultations and treatments for the first try in Yunnan province, then observe their tongue-appearance accordingly. Results By separately observing the color, shape and aspect of the tongue and tongue-fur which belongs to patient during AIDS period and asymptomatic period, discovered that the pathological tongue-fur appearance of all clinical periods trends to greasy, thereinto, in period of AIDS, 47.45% were white-greasy, 16.06% were yellow-greasy, 8.02% were yellow and thin. In terms of color of tongue, 46.71% of the total samples were light pink and white, 32.12% were red, and 21.17% were purple and gore. In period of asymptomatic, most of the tongue-appearance was light pink and thin white, which obviously exceeded the AIDS period. It indicates that the higher level of immunity, the more normal appearance of tongue can be expected. Group A (200≤CD4

<p><b>OBJECTIVE</b>To provide data on alcohol consumption and drug use among middle-school students aged 13-15 in 4 cities of China, and to provide evidence for developing intervention strategies on adolescents alcohol and drug use.</p><p><b>METHODS</b>Standardized sample selection process of two-stage cluster-sampling was used in middle-school students in Beijing, Hangzhou, Wuhan and Urumchi. A self-administered questionnaire survey was conducted in Sept. 2003 and data was analyzed by Epi Info software.</p><p><b>RESULTS</b>Among 7344 students from grade 1 to 3, 36.5% had tasted while 14.4% had drunk alcohol in the past 30 days. 9.9% had experienced drunkness, 5.1% had been in trouble because of drinking, and 1.6% had ever used illegal drugs. Significant differences had been found in all the cities. Higher graders, older students and boys had higher rates of alcohol and addictive drug use than low graders, younger students and girls. 51.9% had been taught on take alcohol safety and another 27.6% on skills of rejecting alcohol, during the past school year.</p><p><b>CONCLUSIONS</b>The current situation of alcohol and addictive drug use among Chinese middle-school students aged 13-15 seemed to be quite critical, suggesting that it is necessary to carry out relevant health education in accordance with different characteristics in area, gender and age of the students.</p>

RESUMO

<p><b>OBJECTIVE</b>To provide accurate data on health related behaviors and protective factors among students in middle schools in China, for developing priorities, programs and policies on health education.</p><p><b>METHODS</b>We used a standard scientific sample selection process developed by American Center for Disease Control and Prevention (CDC) to conduct the questionnaire survey among middle schools from four cities--Beijing, Hangzhou, Wuhan and Urumchi.</p><p><b>RESULTS</b>Data were found as: 3.2% of students are overweight; 25.3% of students rarely washing hands before eating at school, 20.5% of the students had seriously injured in the past 12 months, 30.4% of male students having had physical fighting, 17.1% of the students having serious attempted suicide, 29.7% of the students ever tried or experimented cigarette smoking, 13.0% having drunk from alcohol and 14.5% having been offered or selling drugs during the past 30 days, 78.9% were in an insufficient amount of physical activity and only 14.3% often used seat belt when riding in a car.</p><p><b>CONCLUSIONS</b>There were many problems on health related behaviors among middle school students in these four cities, especially on hygiene, physical activities, psychological situation, smoking and drinking etc. It is absolutely necessary to develop health education for children and adolescence to promote their healthy behaviors and lifestyle.</p>

RESUMO

Objective To investigate the feasibility of laparoscopic operations in treating large ovarian cysts.Methods A total of 38 patients with large ovarian cysts underwent laparoscopic cyst cystectomy or adnexectomy on the affected side from December 2002 to October 2005.The 2nd or 3rd trocar(usually on the tumor side)was inserted into the cyst,the fluid in which was drained with a suction unit.The cyst was then removed after being elevated toward the incision,which was extended to 20 mm.Results Large ovarian cysts in the 38 cases were all removed successfully,including cystectomy in 28 cases and adnexectomy in 10 cases respectively.No complication occurred.The follow-up period was 1-24 months(mean:10 months)for 35 cases.No abnormal findings in pelvic examination and B ultrasonography were observed.Conclusions Laparoscopic operation is a safe and feasible therapeutic method for large ovarian cysts.

RESUMO

Objective To investigate the efficacy of conservative treatment with laparoscopy for patients with ectopic pregnancy.Methods From January 2000 to December 2005,76 patients with ectopic pregnancy received conservative treatment by laparoscopy in our hospital.The data of the patients were analyzed retrospectively.Results In this series,fenestration of the Fallopian tube was performed on 42 cases,salpingostomy on 21,fimbrial expression on 8,and segmental resection and end-to-end anastomosis on 5.Salpingography showed that the Fallopian tube was patent after the operation in all of the patients.The mean operation time was(58.5?19.4) minutes(ranged from 35 to 105).The serum level of ?-hCG decreased to a normal range in 3 weeks postoperation.Follow-up was available in 61 cases for 1 year,during which 35 of the cases(57.4%,35/61) had intrauterine pregnancy;2(3.3%) had spontaneous abortion,and 2(3.3%) showed ectopic pregnancy;the other 22(36.1%) had no pregnancy.Conclusions Conservative treatment by laparoscopy is safe and effective for ectopic pregnancy.To avoid repeated ectopic pregnancy,?-hCG monitoring after the operation is necessary.

<p><b>AIM</b>To investigate alterations of smooth muscle cells and collagen fibers in corpus cavernosum following cavernous neurectomy and its relation to the expression of transforming growth factor-beta1 (TGF-beta1).</p><p><b>METHODS</b>Ten adult male SD rats (neurectomy group) were subject to a bilateral cavernous nerve (CN) resection aseptically under an operating microscope, with 6 sham-operated rats as the control. Fifteen weeks after the operation, the penile specimens were collected and prepared for quantitative-analyzing of ratio of smooth muscle to collagen fibers in corpus cavernosum with confocal microscopy, and for detecting the expression of TGF-beta1 by RT-PCR and western-blot.</p><p><b>RESULTS</b>Smooth muscle cells that show red color after fluorescent-labeling with tetramethylrhodamine isothiocyanate-phalloidin and collagen fibers that produce green autofluorescence after paraformaldehyde fixation were clearly identified under the confocal microscope. Quantification of fluorescent intensity showed that the ratio of smooth muscle to collagen fibers in corpus cavernosum in neurectomy group was 0.265 +/- 0.125, which was significantly lower than that in sham-operated group (0.760 +/- 0.196, P<0.01). RT-PCR and western-blot analyses revealed a significantly higher expression of TGF-beta1 in the penile tissues of the neurectomy animals than that in sham-operated group.</p><p><b>CONCLUSION</b>Bilateral ablation of CN can lead to fibrosis of corpus cavernosum, which may be related to an increased expression of TGF-beta1 induced by hypoxia in cavernous tissue after denervation.</p>
