RESUMO

ObjectiveTo determine the difference of serum neutralizing antibody levels in healthy persons following the vaccination of inactivated SARS-CoV-2 vaccines. MethodsHealth care workers that received inactivated SARS-CoV-2 vaccines were enrolled in Sichuan provincial people's hospital from January to December 2021. All participants were classified into four groups according to the number and time of vaccination, which were groups of 28 days after the second dose, 180 days after the second dose, 28 days after the third dose and 150 days after the third dose. Serum neutralizing antibody was quantitatively detected by chemiluminescence method. Furthermore, the serum neutralizing antibody levels were compared within and between groups by gender, age and body mass index(BMI). ResultsA total of 349 participants were enrolled in this study, including 113 males and 236 females. The positive rates of neutralizing antibody in the groups of 28 days after the second dose, 180 days after the second dose, 28 days after the third dose and 150 days after the third dose were 74.0%, 25.7%, 100.0% and 100.0%, respectively. In the four groups, neutralizing antibody levels were 10.38 (5.76, 24.00) AU·mL-1, 4.18 (3.00, 6.18) AU·mL-1, 41.18 (25.80, 116.21) AU·mL-1 and 33.33 (18.09, 69.12) AU·mL-1, respectively. The positive rate of neutralizing antibody significantly differed across the groups (P<0.001). Furthermore, neutralizing antibody level in the third dose groups were significantly higher than that in the second dose groups (P<0.001). Neutralization antibody level in young people (<45 years old) was significantly higher than that in middle-aged and elderly people (≥45 years old) in the groups of 180 days after the second dose and 28 days after the third dose (P<0.05). Additionally, neutralization antibody level in normal weight participants was significantly higher than that in overweight and obese participants in the group of 28 days after the second dose (P<0.05). However, no significant difference was found in all groups by gender (P>0.05). ConclusionCompared with two doses of inactivated SARS-CoV-2 vaccine, three doses can significantly induce higher neutralizing antibody and stronger immune protection. Age and BMI have certain effect on the neutralizing antibody.

OBJECTIVE To investigate the relationship between state trait anxiety and behavioral status in patients with sudden deafness. METHODS From April 2015 to June 2016, 140 cases of hospitalized patients with sudden hearing loss were selected as the observation group, and 140 healthy volunteers in our hospital for medical examination were selected as the control group. Average data, state trait anxiety, quality of life, and behavior survey were investigated in the two groups and correlation analysis was conducted. RESULTS The state trait anxiety score of the observation group was 51.20±4.19 points, and the control group was 34.40±3.49 points, and the state trait anxiety score of the observation group was significantly higher than that of the control group(t=12.949, P<0.05). The scores of physical, compulsive, interpersonal sensitivity, hostility and depression in the observation group were significantly higher than those in the control group(P<0.05). The problem solving and help scores in the observation group were significantly lower than that of the control group, while the fantasy and self accusation scores was significantly higher than that of the control group(P<0.05). In the observation group, Pearson correlation analysis shows anxiety scores were significantly correlated to the dimensions scores of quality of life, fantasy and self accusation scores(P<0.05), and were significantly negative correlated to the problem solving and help scores (P<0.05). CONCLUSION The patients with sudden deafness is accompanied by state trait anxiety and fantasy, self accusation and other bad behavior, it can seriously affect the patient's quality of life and form vicious circle, so the nursing intervention should be strengthened in clinical practice.

AIM: To investigate the role of fluoxetine in the hippocampal synaptic plasticity in chronic unpre-dictable mild stress (CUMS) depression rats and its effect on mTOR and autophagy signaling pathways.METHODS:Male Sprague-Dawley rats (n =60) were randomly divided into normal control group, CUMS group and fluoxetine group. The CUMS rat model was established through CUMS combined with solitary raising, and fluoxetine (20 mg? kg -1? d -1 ) was administered via intragastric gavage.The changes of body weight, the ratio of sugar intake and the results of the behav-ioral test were recorded to identify the modeling.Moreover, the expression of synaptic plasticity-related proteins glial fibril-lary acidic protein (GFAP) and synaptophysin (SYP), apoptosis-related proteins Bcl-2 and caspase-3, mTOR signaling proteins mTOR and 4EBP1, and autophagy-related proteins beclin 1 and LC3 were examined by RT-PCR and Western blot. RESULTS: Compared with control group, the body weight, sucrose intake, and total distance and intermediate residence time in the open field test were significantly decreased in CUMS group.The results of RT-PCR and Western blotting showed that the mRNA and protein levels of SYP and GFAP in CUMS group were significantly down-regulated compared with con-trol group.The expression of Bcl-2 in CUMS group was downregulated, while the protein level of cleaved caspase-3 in-creased.Decreased phosphorylation levels of mTOR and its downstream target molecule 4EBP1 were observed in CUMS group.Besides, the autophagy-related proteins beclin 1 and LC3 were significantly upregulated at mRNA and protein lev-els.All these results(upregulation or downregulation) were attenuated by the treatment with fluoxetine, and the difference was statistically significant.CONCLUSION: Fluoxetine might improve hippocampal synaptic plasticity and alleviate symp-toms of depression by supressing apoptosis/autophagy signaling pathways and upregulating mTOR signaling pathway.

Objective To investigate the effect of cell death by HIV-1 infection on gene 90K/Mac-2BP by RNA interference (RNAi) in U937 cell line.Methods We used human monocyte-macrophage cell line U937 as the cell model.Cells were infected by HIV-1 ( R5-tropic) 5 days, and then stained by PE-Annexin V and PerCP-7-AAD.90K/Mac-2BP in U937 cell line was knocked down , and these cells were infected by HIV-1 for 5 days.Then, cells were stained by PE-AnnexinV and PerCP-7-AAD.Apoptosis were examined upon flow cytometry .Results The percentages of Annexin V+cells without 90K/Mac-2BP knock-down were (16.27 ±0.30)% by HIV-1 infection.The percentages of them with 90K/Mac-2BP knock-down were (31.26 ±0.35)%, (25.76 ±0.30)%, (23.69 ±0.33)% respectively.The increase of cell apoptosis rate for HIV-1-infected U937 cells by 90K/Mac-2BP siRNA transfection was significantly greater than that for HIV-1-infected untreated cells (P﹤0.01).Conclusion The apoptosis of HIV-1-infected U937 cells was regulated by the expression of 90K/Mac-2BP.

Objective To study the inhibition activity of volatile oil from Mappianthus iodoies on SPC-A-1 and BEL-7402 cancer cells.Methods The volatile oil in Mappianthus iodoie was extracted by SFE-CO2.MTT assay was employed to test the antitumor effect of volatile oil from Mappianthus iodoies in two kinds of malignant tumor cell lines,with IC50 applied to evaluate the degree of inhibition activity.Results When the dose of volatile oil from Mappianthus iodoies was 200 μg/ml,the inhibition ratios of the tumor cell was in excess of 50%,the IC50 was 169.54,695.21 μg/ml respectively.Conclusion Volatile oil from Mappianthus iodoies extracted by SFE-CO2 has obvious inhibition activity on SPC-A-1 and BEL-7402 cancer cells.

RESUMO

Objective To study the antitumor components from an actinomycete strain(N2010-37)of bottom mud in Zhanjiang Mangrove,South China Sea.Methods The components were isolated and purified by chromatographic techniques and recrystallization,and the structures were identified by spectral methods together with physicochemical analyses.The antitumor effects of these components were tested in vitro by MTT method.Results Three compounds were identified including two anthrones and one novel lactone.They are(3S,4R,7R,8R,9S)-3,8-dihydroxy-4,7,9-trimethyl-2,6-cyclononanediolacetone(1),2-hydroxy-l-methoxy-3-methylanthraquinone(2),and 1,6,8-thihydroxy-3-methyl-anthraquinone(3).Conclusion Compound 1 is a new compound,and compounds 1 and 3 show the favorablecytotoxic activities against human chronic granulocytic leukemia cell line K562 strain by MTT method in vitro.

OBJECTIVE@#To investigate the relation between expression of vascular endothelial growth factor (VEGF)/ proliferating cell nuclear antigen (PCNA) and multi-slice spiral computer tomography (MSCT) perfusion imaging.@*METHODS@#Sixty-one patients who underwent CT perfusion scan by 16-slice spiral CT were examined. Among them,22 were brought into our research after surgery. The corresponding layers of tumor tissue specimens to the layer of CT perfusion scan were selected to determine the expression of CD34,VEGF, and PCNA. Spearman correlation analysis was used to determine the relation between differentiation of non-small cell lung cancer (NSCLC), the expression of CD34,VEGF/PCNA, and CT perfusion parameters.@*RESULTS@#There was a lot of heterogeneity in VEGF and PCNA expression of NSCLC.The degree of differentiation had positive correlation with the uncomplete lumen of the surrounding area CD34-MVD and the expression of PCNA and VEGF (P<0.05).There were positive correlations between the uncomplete lumen of the surrounding area CD34-MVD and expression of VEGF and PCNA, respectively (both P<0.05). Blood flow (BF), blood volume (BV),and peak enhancement image (PEI) decreased with the decreasing differentiation of NSCLC (P<0.05). The total CD34-MVD showed a positive correlation with PEI (P<0.05),and the uncomplete lumen of the surrounding area CD34-MVD showed a negative correlation with BF,BV, and PEI (all P<0.05). The PCNA expression showed a negative correlation with BF,BV, and PEI (P<0.05).@*CONCLUSION@#PCNA and VEGF expression in NSCLC regulates angiogenesis and proliferation at the same time. Perfusion parameters reflect the expression of microvascular architecture phenotype, and exactly evaluate the malignant degree of tumor.

OBJECTIVE@#To investigate the expression of fibroblast growth factor-2 (FGF-2) and osteopontin (OPN) in non-small cell lung cancer (NSCLC) tissues and analyze the correlation between FGF-2 and OPN.@*METHODS@#Immunohistochemical SP method was used to detect the expression of FGF-2 and OPN in 76 patients with NSCLC and 15 normal lung tissues. The effect of FGF-2 on OPN expression at mRNA and protein level in A549 cell was examined by RT-PCR and Western blot.@*RESULTS@#The positive expression of FGF-2 (65.8%) and OPN (60.5%) in the NSCLC tissues was significantly higher than that in the normal lung tissues (13.3% and 0, respectively ) (P0.05).A positive correlation was found between the expression of FGF-2 and OPN in NSCLC (r=0.552,P<0.01). The expression of OPN protein and mRNA was up-regulated by FGF-2 in A549 cells.@*CONCLUSION@#The overexpression of FGF-2 and OPN is related to the metastasis and invasion of NSCLC.FGF-2 may promote the metastasis and invasion of NSCLC depending on the upregulation of OPN expression.
