RESUMO

AIM: To investigate the effects of curcumin on the proliferation and apoptosis and migration of human pterygium fibroblasts(HPF)in vitro.METHODS: A total of 7 cases of pterygium tissue removed at our hospital from November 24, 2021 to December 16, 2021 were collected. Then, primary fibroblasts were cultured in vitro and identified by immunofluorescence staining. HPF were treated with 0, 10, 20, 40, 80 and 160μmol/L curcumin containing equal amount of dimethyl sulfoxide for 24h, then the cell proliferation was detected by CCK8 assay. According to the results of CCK8, the cells were divided into control group, 20μmol/L curcumin group and 40μmol/L curcumin group, and the cells were treated with corresponding concentration of curcumin for 24h in each group. Flow cytometry was used to detect apoptosis, Transwell migration assay was used to detect cell migration, and real-time fluorescence quantitative polymerase chain reaction and Western blot were used to detect the expression of mRNA and protein of B-cell lymphoma-2 associated X protein(Bax), B-cell lymphoma-2(Bcl-2), Cyclin D1 and matrix metalloproteinase 2(MMP2).RESULTS: Compared with the control group, both 20μmol/L curcumin group and 40μmol/L curcumin group can inhibit the proliferation and migration of HPF and induce its apoptosis(all P<0.05). Compared with the control group, 20μmol/L curcumin group can down-regulate the mRNA expression of Cyclin D1 and MMP2, up-regulate the mRNA expression of Bax, and down-regulate the protein expression of Bcl-2(all P<0.05). Compared with the control group, 40μmol/L curcumin group can down-regulate the expression of mRNA and protein of Bcl-2, Cyclin D1 and MMP2, and up-regulate the expression of mRNA and protein of Bax(all P<0.05). Compared with 20 μmol/L curcumin group, the 40 μmol/L curcumin group can down-regulate the mRNA expression of MMP2, down-regulate the protein expression of Bcl-2, and up-regulate the mRNA and protein expression of Bax(all P<0.05).CONCLUSION: Curcumin can inhibit the proliferation of HPF by inhibiting the expression of Cyclin D1, induce the apoptosis of HPF by down-regulating Bcl-2 and up-regulating the expression of Bax, and inhibit the migration of HPF by down-regulating the expression of MMP2.

RESUMO

ObjectiveTo observe the effects of the water extracts of Trichosanthis Radix-Polygonati Rhizoma at different ratios on glucose and lipid metabolism in KKAy mice with spontaneous type 2 diabetes and explore the mechanism of the extract in alleviating insulin resistance based on phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt)/forkhead box O1 (FoxO1) signaling pathway. MethodThe 8-week-old C57BL/6J male mice were taken as the normal control group, and KKAy male mice of the same age were randomly assigned into a model group, a metformin group, Trichosanthis Radix-Polygonati Rhizoma groups at the ratios of 1∶1 (Trichosanthis Radix 30 g, Polygonati Rhizoma 30 g), 1∶3 (Trichosanthis Radix 15 g, Polygonati Rhizoma 45 g), and 1∶5 (Trichosanthis Radix 10 g, Polygonati Rhizoma 50 g) according to blood glucose level and body weight, with 6 mice in each group. The administration lasted for 8 weeks, and the body weight (BW) and fasting blood glucose (FBG) of mice were recorded at the same time points of the 2nd, 4th, 6th, and 8th weeks, respectively. Oral glucose tolerance test (OGTT) was performed at the 7th week. After drug administration, the serum levels of total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), and fasting insulin (FINS) were measured, and homeostasis model assessment-insulin resistance (HOMA-IR) index was calculated. The liver tissue samples were stained with hematylin-eosin (HE) and periodic acid-Schiff (PAS) for observation of the fat distribution and glycogen content. The protein levels of PI3K, Akt, p-Akt, FoxO1, and p-FoxO1 in the liver were determined by Western blot. ResultCompared with the normal group, the model group showed increased food intake, FBG, glucose tolerance, FINS, HOMA-IR, TC, TG, and LDL-C (P<0.01), and down-regulated protein levels of PI3K, Akt, phosphorylaison (p)-Akt, FoxO1, and p-FoxO1 in the liver (P<0.01). Compared with the model group, Trichosanthis Radix-Polygonati Rhizoma lowered FBG and HOMA-IR (P<0.05, P<0.01). In particular, the combination at the ratio of 1∶3 showed the best performance (P<0.01) comparable to metformin. Furthermore, Trichosanthis Radix-Polygonati Rhizoma at different ratios lowered blood glucose at different time points of OGTT (P<0.05) and TC and LDL-C (P<0.01). Additionally, the combination at the ratio of 1∶3 reduced TG (P<0.01). The liver of mice in the drug administration groups showed regular morphology, with few lipid droplets and rich glycogen. Western blot showed that Trichosanthis Radix-Polygonati Rhizoma up-regulated the protein levels of PI3K and p-Akt, down-regulated the protein level of FoxO1, and up-regulated the protein level of p-FoxO1 (P<0.05). ConclusionTrichosanthis Radix-Polygonati Rhizoma, especially at the ratio of 1∶3, lowered the FBG, TC, LDL-C, and HOMA-IR index, promoted liver glycogen synthesis, and reduced steatosis in KKAy mice, which may be related to the regulation of PI3K/Akt/FoxO1 signaling pathway in the liver.

RESUMO

ObjectiveTo investigate the potential mechanism of Xiao Chaihutang (XCHT) in the treatment of Alzheimer's disease (AD) based on network pharmacology and bioinformatics. MethodThe active components of XCHT and corresponding targets were retrieved from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), and the differentially expressed genes related to AD were searched from Gene Expression Omnibus (GEO). Thereby, the common targets of XCHT and AD were yielded, followed by Gene Ontology (GO) term enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the common targets. The component-target network and protein–protein interaction (PPI) network were constructed. Furthermore, amyloid β-protein (Aβ)1-40 was used to induce AD in PC12 cells and then the AD cells were intervened with XCHT. Afterward, cell viability was detected by Cell Counting Kit-8 (CCK-8) assay and cell morphology was observed based on 4',6-diamidino-2-phenylindole (DAPI) staining. Cell membrane potential was determined and apoptosis was detected by flow cytometry, and cellular immunofluorescence detects the expression of B-cell lymphoma-2 (Bcl-2)/Bcl-2-related X protein (Bax). Moreover, immunofluorescence assay was performed. ResultA total of 190 active components and 41 anti-AD targets of XCHT were screened out. The key components included mairin, quercetin, berberine, protoporphyrin, 24-ethylcholest-4-en-3-one, and β-D-ribofuranoside, and the core targets were sigma non-opioid intracellular receptor 1 (SIGMAR1), checkpoint kinase 1 (CHEK1), protein tyrosine phosphatase non-receptor type 6 (PTPN6), protein kinase C(PRKCH), inhibitor of nuclear factor kappa B kinase subunit beta (IKBKB), cathepsin D (CTSD), cysteine aspartate protease-3 (Caspase-3), Bax, and Bcl-2-like protein 1 (Bcl-2L1). The anti-AD targets of XCHT were involved in 302 GO terms (P < 0.05), particularly the regulation of neuronal cell apoptosis, and 73 KEGG pathways (P<0.05). The major pathways and biological processes included the apoptosis pathway, virus infection pathway, lipid and atherosclerosis pathway, and cancer-related pathways. In the in vitro experiment, the model group demonstrated the decrease in cell survival rate (P<0.05), increase in apoptosis rate (P<0.05), and down-regulation of mitochondrial membrane potential and Bcl-2/Bax ratio compared with the blank control. Compared with the model group, XCFT group showed the increase in cell survival rate (P<0.05), decrease in apoptosis rate (P<0.05), and up-regulation of mitochondrial membrane potential and Bcl-2/Bax ratio. ConclusionBased on network pharmacology, this study reveals the multi-component, multi-target, and multi-pathway characteristics of XCHT in the treatment of AD, laying a foundation for further research on the material basis and mechanism of this prescription.

RESUMO

Small cell lung cancer (SCLC), as a pathological type with high malignancy, presents a high risk of early brain metastasis. Prophylactic cranial irradiation (PCI) can reduce the risk of brain metastasis in patients with SCLC, however, the incidence of brain structural and functional damage caused by PCI is high, and their clinical symptoms are not typical. The existing treatment methods can relieve some clinical symptoms, but can not effectively reverse the process of brain injury, which reduces the survival benefit of patients. In-depth understanding the mechanisms of PCI neurotoxicity and exploring effective strategies for ptevention and treatment are of great value in improving prognosis of SCLC.

RESUMO

Objective To analyse the transcriptome sequencing results of Eucommia ulmoides calli under light and dark culture condition, and verify the expression level of the related genes of flavonoid biosynthesis from transcriptome sequencing results. Methods The transcriptome high-throughput sequencing of E. ulmoides calli was performed by using the Illumina HiSeqTM 2500 sequencing platform, and de novo assembly of the transcriptome sequencing results was finished by Trinity software. The sequencing results of Unigenes were compared with the databases of NR, Swiss-Prot, GO, COG, KOG, KEGG and Pfam by BLAST software. The gene expression abundance was estimated by FPKM value. qRT-PCR was used to verify the expression level of the related genes of flavonoid biosynthesis. Results About 13.00 Giga base pairs (Gbp) clean data (more than 6.02 Gbp, respectively) were obtained, and de novo assembly generated 62 030 Unigenes with an average length of 736.40 bp. A total of 25 167 Unigenes were annotated to Nr. Totally 4 794 Unigenes and their associated enzymes (enzyme commission numbers) were annotated to 82 KEGG pathway. There were 1 986 genes identified as significantly and differentially expressed genes between the two calli under light and dark culture. Among them, 1 139 (57.35%) were up-regulated and 847 (42.65%) were down-regulated in the calli under light culture. Metabolic pathway analysis revealed that seven Unigenes were predicted to be responsible for the flavonoid biosynthesis, six Unigenes of which were up-regulated in the calli under light culture, encoding chalcone isomerase (EC 5.5.1.6), chalcome synthase (EC 2.3.1.74), flavonoid 3’-monooxygenase (EC1.14.13.21), trans-cinnamate 4-monooxygenase (EC 1.14.13.11), Flavonol synthase (EC 1.14.11.23), shikimate-O-hydroxycinnamoyl transferase (EC 2.3.1.133). One Unigene was down-regulated in the calli under light culture, which encoded leucocyanidin oxygenase (EC 1.14.11.19). qRT-PCR analysis showed that the expression of the seven genes related with flavonoid biosynthesis was coincident with the transcriptome high-throughput sequencing results. Conclusion The white light (12 000 lx, 16 h light and 8 h dark) could improve the production capacity of some metabolic intermediate of flavonoid biosynthesis pathway of E. ulmoides calli.

RESUMO

This paper systematically reviews the latest and relevant literatures and policy documents on the in-tegrated health services in Canada in recent years. Therefore, it summarizes the practice and mode of integrated serv-ice delivery in Ontario, Alberta and Quebec wherein the integration among health organization, health service team, and a series of health services are included. The contributing factors and impeding factors ( the barriers) of organiza-tional integration and specific integration strategy were summarized. Finally, according to the actual practical situa-tion, it is proposed that China should adhere to the government-led approach in promoting the integration of health services, and give a full play to the positive role of the market mechanism. Through strengthening the network man-agement and group service of primary health services, emphasis will be put on health services of population groups and specific diseases. Therefore, integration will be regarded as a strategic priority, increasing incentives and boos-ting promotion of nursing personnel on the process of Integrated Service Delivery, building the health information sys-tem that is conducive to integration in order to continuously advance Hierarchical Diagnosis and bridge the fragmented service system. This will help in providing residents with personalized, convenient, comprehensive, and continuous health services.

RESUMO

Objective To investigate the role of T helper 17 cells/interleukin?17(Th17/IL?17) axis in the occurrence of vaginal candidiasis in mice. Methods A total of 120 female BALB/c mice were randomly and equally divided into Ei, En, Ci and Cn groups. Three days before vaginal inoculation, estrogen (Ei and En)groups and control(Ci and Cn)groups received subcutaneous injection of 0.05 mg estradiol and 0.1 ml sterilized soybean oil at the hind legs, respectively, and then the hormone treatment continued every other day until the end of experiment. Infected(Ei and Ci)groups and noninfected(En and Cn) groups were inoculated intravaginally with 10μl(5 × 104 conidia)of Candida albicans suspension and 10μl of sterilized phosphate?buffered saline, respectively. Ten mice were randomly selected from each group and sacrificed on day 3, 7 and 14 after inoculation. The intact vagina tissues were resected and then frozen in liquid nitrogen or embedded in paraffin. Real?time fluorescence?based quantitative PCR(qRT?PCR)and immunofluorescent staining were performed to measure mRNA expression and immunofluorescence intensities of retinoic acid?related orphan receptorγt(RORγt), RORα and IL?17, respectively. Western blot analysis was conducted to determine protein expression of RORγt and IL?17. Results Laser scanning confocal microscopy showed that RORγt, RORα and IL?17 immunofluorescence was mainly located at inflammatory cells of the lamina propria and blood vessels in En and Cn groups, at mucosal epithelium, adherent hyphae, and inflammatory cells of the lamina propria and blood vessels in Ci group, and at mucosal epithelium, vaginal canal and endocytosed hyphae, and inflammatory cells of the lamina propria and blood vessels in Ei group. qRT?PCR and immunofluorescent staining uncovered that mRNA expression and immunofluorescence intensities of RORγt, RORα and IL?17 were significantly higher in En, Ci and Ei groups than in Cn group at the same time points(all P0.05). Moreover, RORγt and IL?17 protein expression in Ci and Ei groups was obviously up?regulated on day 7 after inoculation (RORγt: F = 13.137, P < 0.001; IL?17: F = 11.182, P < 0.001), but was not increased further on day 14. Conclusion Vaginal candida infection can up?regulate the expression of RORγt, RORα and IL?17, suggesting that Th17/IL?17 axis may be involved in the occurrence of vaginal candidiasis in BALB/c mice.

Methods One hundred non-small-cell lung cancer patients who underwent pulmonary lobectomy or segmentectomy in thoracic surgery department from April 2012 to January 2014 were assigned into the control group , while the experiment group were paired patients from February 2014 to December 2014. We applied ACBT to the experiment group for 5 days after surgical resection and afterward compared the two groups in view of incidence of respiratory complications . Result The incidences of pulmonary infection, atelectasis and hypoxemia in the experiment group were all significantly lower than those in control group (P<0.05). Conclusion ACBT strengthens endurance and force of patients′respiratory muscle group, and therefore reduces postoperative pulmonary complications.

RESUMO

Objective To observe the effect ofGuanxin-Shutongcapsule in the treatment of arterial elasticity on patients with hypertension. Methods The hypertension patients who met the inclusion criteria were divided into treatment group (50 cases) and control group (52 cases). The control group was treated with antihypertensive drugs to control blood pressure within the normal range. The treatment group was treated withGuanxin-Shutongcapsule on the basis of the control group. All were given 8 weeks treatment. The main artery elastic parameters were meansured by the carotid-femoral pulse wave velocity (C-FPWV) and cervical-dorsal arterial pulse wave velocity (C-DPWV). The immune turbidimetric method was employed to enhance for the determination of high sensitive C reactive protein (hs-CRP); and radioimmunoassay was used to assess the serum IL-6, TNF-a, triglyceride (TG) and total cholesterol (TC). The blood pressure was monitored during the treatment. Results After the treatment, the level of hs-CRP (2.83 ± 1.35 mg/Lvs. 3.65 ± 1.38 mg/L,t=6.357), TNF-α (0.16 ± 0.08 mg/Lvs. 0.28 ± 0.07 mg/L,t=18.213), C-FPWV (13.85 ± 1.86 m/svs. 15.34 ± 1.78 m/s,t=6.524), C-DPWV (11.98 ± 1.45 m/svs. 12.87 ± 1.48 m/s,t=7.152) in treatment group was significantly lower than those in the control group (P<0.01).ConclusionGuanxin-Shutong capsule by inhibiting systemic inflammation, reducing and reversing atherosclerosis, and improving the arterial elasticity and blood pressure.

RESUMO

Objective To explore the quality of life and its influencing factors among permanent colostomy patients. Methods Totally 219 permanent colostomy patients were recruited by convenient sampling method from January 2013 to December 2014 and investigated by Stoma Quality of life-Chinese Version, Stoma self-efficacy Scale, Stoma Self-care Scale-general version , and demographic questionnaire. Data were analyzed by SPSS 17.0. Results The score of Stoma-QoL-C among permanent colostomy patients was (54.86±12.17) points, which showed a dynamic V-type change as time went by. The quality of life of patients with colostomy for 1 to 3 years was especially low (50.46±13.77) points, P<0.01. The influencing factors of quality of life among permanent colostomy patients included self-efficacy, body image change, family members′acceptance of stoma except their spouses and self-care ability, while self-efficacy was the most important influencing factor. Conclusions The enterostomal nurses should pay more attention to the quality of life of the patients who have had colostomy for 1 to 3 years, enhance the patients self-efficacy and self-care ability, and help the patients to accept their body image changes and their family members′acceptance of stoma to improve their quality of life.

RESUMO

Objective To explore the designing and delivery strategies of nursing interventions, in order to improve the design level and delivery effect. Methods We expounded the idea through litera-ture review and the author′s practical experience in research work. Results The adoption and design of nursing intervention were discussed from literature review, theoretical framework, meta- analysis, and expert opinions. Strategies were recommended for effective delivery of nursing intervention, including integrity of in-tervention, compliance of participants, and randomization of grouping. Conclusions The above strategies should be insisted to design and carry out nursing intervention research, in order to make the intervention applicable, standardized and easy to generalize in clinic.
