Objective:To observe and evaluate the safety and efficacy of anti-vascular endothelial growth factor (VEGF) in the treatment of eyes with macular edema (ME) secondary to branch retinal vein occlusion (BRVO) in Lhasa, Tibet.Methods:A retrospective case series. From September 2018 to January 2022, a total of 41 patients (41 eyes) with BRVO-ME, who were diagnosed in Department of Ophthalmology of Tibet Autonomous Region People’s Hospital, were included in this study. There were 21 eyes in 21 males and 20 eyes in 20 females. The median age was 53 (31,75) years. There were 24 patients with hypertension (58.8%, 24/41). Best corrected visual acuity (BCVA), ocular pressure, fundus color photography and optical coherence tomography (OCT) were performed in all eyes. The BCVA was performed using the international standard logarithmic visual acuity chart, which was converted into logarithm of the minimum angle of resolution (logMAR) BCVA for record. The foveal macular thickness (CMT) was measured by OCT. All eyes were treated with intravitreous injection of anti-VEGF drugs, once a month, among which 23 eyes (56.1%, 23/41) received intravitreous injection of ranibizumab (IVR), and 18 eyes (43.9%, 18/41) received intravitreous injection of conbercept (IVC), and were grouped accordingly. There was no significant difference in age ( Z=-0.447), gender composition ( Z=-0.485), logMAR BCVA ( t=-1.591), intraocular pressure ( t=-0.167) and CMT ( t=-1.290) between two groups ( P>0.05). During the follow-up, the same devices and methods were used at baseline to perform relevant examinations, and the changes of BCVA, intraocular pressure, CMT and new cardiovascular and cerebrovascular events were compared between baseline and the last follow-up. logMAR BCVA, intraocular pressure and CMT were compared between baseline and last follow-up using Student t test. The comparison of injection times and follow-up time between IVR group and IVC group was conducted by Mann-Whitney U test. Results:At baseline, logMAR BCVA, intraocular pressure, and CMT were 0.852±0.431, (12.5±2.5) mm Hg (1 mm Hg= 0.133 kPa), and (578.1±191.1) μm, respectively. At the last follow-up, the number of anti-VEGF drug treatments was (2.7±1.2) times; logMAR BCVA and CMT were 0.488±0.366 and (207.4±108.7) μm, respectively, with CMT > 250 μm in 14 eyes (34.1%, 14/41). Compared with baseline, BCVA ( t=4.129) and CMT ( t=-0.713) were significantly improved, with statistical significance ( P<0.001). The injection times of IVR group and IVC group were (2.6±0.9) and (3.0±1.5) times, respectively. There were no significant differences in the number of injection times ( t=-1.275), logMAR BCVA ( t=-0.492), intraocular pressure ( t=0.351) and CMT ( t=-1.783) between the two groups ( P>0.05). No new hypertension, cardiovascular and cerebrovascular events occurred in all patients during follow-up. At the last follow-up, there were no eye complications related to treatment modalities and drugs. Conclusion:Short-term anti-VEGF treatment can improve the visual acuity of BRVO secondary ME patients and alleviate ME in Lhasa, Tibet. The safety and efficacy of ranibizumab and conbercept were similar.

Anthocyanidin reductase (ANR) is one of the key enzyme in the flavonoid biosynthetic pathway, and its catalytic activity is important for the synthesis of plant anthocyanin. In this study, specific primers were designed according to the transcriptome data of Lonicera japonica Thunb., and the CDS, gDNA and promoter sequences of ANR genes from Lonicera japonica Thunb. and Lonicera japonica Thunb. var. chinensis (Wats.) Bak. were cloned. The results showed that the CDS sequences of LjANR and rLjANR were 1 002 bp, the gDNA sequences were 2 017 and 2 026 bp respectively, and the promoter sequences were 1 170 and 1 164 bp respectively. LjANR and rLjANR both contain 6 exons and 5 introns, which have the same length of exons and large differences in introns. The promoter sequences both contain a large number of light response, hormone response and abiotic stress response elements. Bioinformatics analysis showed that both LjANR and rLjANR encoded 333 amino acids and were predicted to be stable hydrophobic proteins without transmembrane segments and signal peptides. The secondary structures of LjANR and rLjANR were predicted to be mainly consisted of α-helix and random coil. Sequence alignment and phylogenetic analysis showed that LjANR and rLjANR had high homology with Actinidia chinensis var. chinensis, Camellia sinensis and Camellia oleifera, and were closely related to them. The expression levels of LjANR and rLjANR were the highest in flower buds and the lowest in roots. The expression patterns at different flowering stages were similar, with higher expression levels in S1 and S2 stages and then gradually decreased until reaching the lowest level in S4 stage, after a slow increase in S5 stage, the expression levels decreased again. The expression levels of ANR genes in the two varieties showed significant differences in roots, S2 and S5 stages, while the differences in stems, flower buds, S1, S3 and S6 stages were extremely significant. The prokaryotic expression vector pET-32a-LjANR was constructed for protein expression. The target protein was successfully expressed of about 59 kD. This study lays a foundation for further study on the function of ANR gene and provides theoretical guidance for breeding new varieties of Lonicera japonica Thunb.

UDP-glucose: flavonoid 3-O-glucosyltransferase (UF3GT) uses flavones, dihydroflavonol or anthocyanin as the acceptor and uridine 5′-diphosphate-sugar as the donor to catalyze the production of flavonoid 3-O-glycoside compounds. Based on sequence homology and transcriptome data, we screened and cloned a UF3GT gene named CtUF3GT (GenBank No. OM948976) from safflower. Biological information analysis demonstrate that CtUF3GT has highly conserved PSPG motif. The open reading frame of CtUF3GT is 1 446 bp, encoding 481 amino acids, with a presumed molecular weight of 52.36 kD and a theoretical isoelectric point of 5.33. Multiple sequence alignment indicate that CtUF3GT has a high homology with UF3GT from Asteraceae, and phylogenetic analysis showed that CtUF3GT clusters with functional identified UF3GTs from other species. The purified recombinant protein glucosylated kaempferol and quercetin to biosynthesis of kaempferol 3-O-glucoside and quercetin 3-O-glucoside, respectively. And CtUF3GT prefered to use kaempferol as substrate. qRT-PCR analysis showed that the UF3GT gene was most highly expressed in flowers, followed by leaves, with very low expression in bracts and stems, and no expression in roots. The expression of UF3GT gene showed a trend of increasing and then decreasing at different stages of flower development. The expression of CtUF3GT gene in safflower with different flower color was highly significant (P < 0.01) at S1, S2, S5, S6 and S7 stages of flower development, in which the expression of CtUF3GT in white safflower was 5.3 and 3.1 times higher than that in red safflower at S6 and S7 stages. This study lays the foundation for further exploring the role of CtUF3GT in the mechanism of safflower flavonoid secondary metabolite biosynthesis and accumulation.

RESUMO

A total of 33 pesticides have been banned from Chinese medicinal materials and decoction pieces(plants) according to Chinese Pharmacopoeia(2020 edition). According to the chemical structures, they are mainly divided into seven categories: organophosphorus compounds, organochlorines, carbamates, amidines, sulfonylureas, phenylpyrazoles, and ethers. These banned pesticides exhibit neurotoxicity, reproductive toxicity, immune system toxicity, teratogenicity, carcinogenesis, and mutagenesis, seriously damaging human and animal health. They affect not only the quality and safety of traditional Chinese medicines and resulting products, but also their competitiveness in the international market. Due to the numerous varieties of traditional Chinese medicines and their complex substrates, it is necessary to establish a universal and highly sensitive method for pesticide residue detection. This review systematically summarized the residual status, toxicity, and analytical methods of banned pesticides in traditional Chinese medicines, and forecasted the prospects of different analytical techniques, so as to provide reference for further safety and risk assessment of banned pesticide residues in traditional Chinese medicines, thus ensuring the safe production of traditional Chinese medicines.

In the present study, the composition and content of pulmonary surfactant (PS) were analyzed to explore the hypoxia adaptation mechanism in plateau zokors (Myospalax baileyi) and plateau pikas (Ochotona curzoniae). 36 plateau zokors and plateau pikas were trapped alive at the Laji Mountain in Guide County, Qinghai Province (at the altitude of about 3 600 m), and 36 Sprague-Dawley (SD) rats were purchased from the experimental animal center of Lanzhou University (at the altitude of about 1 500 m). All animals were lavaged after laboratory anesthesia, the blood in lung tissues was fully washed out and the lung tissues were then taken out to obtain the bronchoalveolar lavage fluid by bronchoalveolar lavage. The composition and content of phospholipids in the PS of three different kinds of animals were analyzed by using high performance liquid chromatography; the protein composition, content and types in the PS were analyzed by G-250 Coomassie brilliant blue method, polyacrylamide gel electrophoresis (PAGE) and mass spectrometry; the dissolved oxygen in the PS solutions were determined by using dissolved oxygen electrode. The results showed that the total contents of phospholipids in the PS were successively increased among plateau zokors, plateau pikas and SD rats (P 0.05). The relative content of PSe had no significant differences between plateau zokors and plateau pikas (P > 0.05), but both were significantly higher than that of SD rats (P < 0.01). The serum albumin (SA) was identified in the PS of three kinds of animals, including homologous tetramer protein containing heme, which is composed of hemoglobin β subunit, in plateau zokors and plateau pikas. Immunoglobulin (Ig) heavy chain was found in PS of plateau zokors and SD rats. The content of Ig heavy chain in plateau zokor was significantly higher than that in SD rats (P < 0.01), and the content of protein containing heme was significantly higher than that in plateau pikas (P < 0.05). The amount of dissolved oxygen was successively decreased in the PS among plateau zokors, plateau pikas and SD rats (P < 0.01), but it was significantly higher than that in saline (P < 0.01). These results suggest that the total content of proteins in the PS of plateau zokors and plateau pikas was significantly higher, while the total content of phospholipids was significantly decreased. There were high content of homologous tetramer protein containing heme in the PS of plateau zokors and plateau pikas. The relative content of DPPC, the main component of phospholipids, was significantly increased in plateau zokors. The changes of PS component and content improve the adaptability of the two plateau animals in hypoxia environment.

Objective To study the relationship between serum lipids level and bone mineral density( BMD)in postmenopausal OSAHS patients.Methods One hundred and twelve postmenopausal OSAHS patients were divided into osteoporosis group(n=35)with a T value<-2.5,osteopenia group(n=39)with a T value=-2.5-1.0,and normal BMD group(n=38)woth a T value> -1.0,36postmenopausal OSAHS-free patients served as a control group and their BMD was detected. The relationship between blood lipids level and BMD was analyzed.Results The serum TC,TG and LDL-C levels were significantly higher in normal BMD group,osteopenia group and osteoporosis group than in control group(P<0.05).The serum TG and LDL-C levels were significantly higher while the serum phosphorus level was significantly lower in osteopenia group and osteoporosis group than in normal BMD group (P <0.05).Pearson correlation analysis showed that the serum ALP and LDL-C levels were negatively related with the BMD of lumbar vertebrae 1-4(r=-0.145,P=0.041;r=-0.331,P=0.000)while the serum phosphorus and TG levels were positively related with the BMD of double femoral neck(r=0.290,P=0.000;r=0.085,P=0.031;r=0.310,P=0.000;r=0.160,P=0.029).Conclusion The serum TG and LDLC levels are related with the BMD in postmenopausal OSAHS patients.Serum blood lipids level is an indicator for the early diagnosis and treatment of osteoporosis.

Background@#Fibroblasts were the main seed cells in the studies of tissue engineering of the pelvic floor ligament. Basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) were widely studied but at various concentrations. This study aimed to optimize the concentrations of combined bFGF and EGF by evaluating their effects on proliferation and collagen secretion of fibroblasts.@*Methods@#Fibroblasts were differentiated from rat adipose mesenchymal stem cells (ADSCs). Flow cytometry and immunohistochemistry were used for cell identification. The growth factors were applied at concentrations of 0, 1, 10, and 100 ng/ml as three groups: (1) bFGF alone, (2) EGF alone, and (3) bFGF mixed with EGF. Cell proliferation was evaluated by Cell Counting Kit-8 assays. Expression of Type I and III collagen (Col-I and Col-III) mRNAs was evaluated by real-time quantitative reverse transcription-polymerase chain reaction. Statistical analysis was performed with SPSS software and GraphPad Prism using one-way analysis of variance and multiple t-test.@*Results@#ADSCs were successfully isolated from rat adipose tissue as identified by expression of typical surface markers CD29, CD44, CD90, and CD45 in flow cytometry. Fibroblasts induced from ADSC, compared with ADSCs, were with higher mRNA expression levels of Col I and Col III (F = 1.29, P = 0.0390). bFGF, EGF, and the mixture of bFGF with EGF can enhanced fibroblasts proliferation, and the concentration of 10 ng/ml of the mixture of bFGF with EGF displayed most effectively (all P < 0.05). The expression levels of Col-I and Col-III mRNAs in fibroblasts displayed significant increases in the 10 ng/ml bFGF combined with EGF group (all P < 0.05).@*Conclusions@#The optimal concentration of both bFGF and EGF to promote cell proliferation and collagen expression in fibroblasts was 10 ng/ml at which fibroblasts grew faster and secreted more Type I and III collagens into the extracellular matrix, which might contribute to the stability of the pelvic floor microenvironment.

Objective To construct prokaryotic expression vector of γ-aminobutyric acid type C receptor ρ2 (GABACR ρ2) gene,induce the expression of the recombination protein GABACR ρ2,and transfect the protein into SH-SY5Y cell line,achieve the transient expression of the recombination protein GABACR ρ2 in SH-SY5Y cell line.Methods The ρ2-Tat gene was inserted into plasmid pET30 to construct the recombinant plasmid pET-ρ2-GFP-Tat.The expression of GABACR ρ2 was detected by Western blot.The histidine-tagged recombination protein ρ2 was purified through immobilized Ni2+ absorption chromatographic column and the purity of GABACR ρ2 protein was detected by sodium dodecyl sulfate polyacrylamide gel elec trophoresis.The transduction and cellular localization of the GABAC R ρ2 was observed by fluorescence microscope.The SH-SY5Y cells were divided into normal group and hypoxia low glucose group.The cells in normal group were divided into normal control group which cultured in high glucose medium and without protein transfection and normal transfection group which cultured in high glucose medium and with protein transfection;the cells in hypoxia low glucose group were divided into treatment group which cultured under hypoxia low glucose condition and treatment transfection group which cultured under hypoxia low glucose and with protein transfection;the viability of SH-SY5Y cells in each group was evaluated by cell counting Kit-8.Results The recombinant plasmid pET30-ρ2-Tat was constructed successfully.The purified recombination protein ρ2-Tat successfully crossed the cytomembrane and transfected the SH-SY5Y cells.The viability of cells in normal control group,normal transfection group,treatment group and treatment transfection group was(100.0 ± 6.9)%,(89.3 ± 3.6)%,(51.4 ± 3.6)%and (66.1 ± 8.5) % respectively.There was no statistic difference in the cell viability between the normal control group and normal transfection group(P >0.05);the cell viability in treatment group was significantly lower than that in the normal control group (P < 0.01);the cell viability in treatment transfection group was significantly higher than that in the treatment group (P < 0.01).Conclusion The recombination protein ρ2 can through the membrane under the effect of Tat transduction peptide and can successfully establish a SH-SY5Y cell lines which transiently express recombinant protein ρ2,which provide a new research method for the study of ρ2 subunit function.

Objective To explore the relationship between the expression of nerve growth factor (NGF) in bladder tissue of Sprague-Dawley rats with diabetes mellitus and the changes of bladder structure and urodynamics.Methods Sixty-three Sprague-Dawley rats were selected and divided into diabetic group (n =33) and control group (n =30).The diabetes mellitus model was made by injecting of steptozotocin (STZ) into the tail vein of rats in the diabetic group;the rats in control group were injected the same volume of citrate buffer solution.The 24 h urinary volume was detected by using metabolic cage;the function of bladder was detected by using Medlab bio-signal collection system;the expression of NGF in the bladder tissue of rats was detected by Western blot;the bladder structure was observed by using light microscope and transmission electron microscope (TEM).Results Four,eight and twelve weeks after the success of diabetic rat model,the level of blood glucose of rats in diabetic group was significantly higher than those in the control group (P < 0.05);the body weight of rats in diabetic group was significantly lower than that in the control group (P < 0.05).The 24 h urinary volume of rats in diabetic group was significantly increased compared with control group (P < 0.05).The maximum capacity of bladder and residual urine volume of bladder and elasticity of bladder of rats in diabetic group were significantly higher than those in the control group (P < 0.05);the maximum bladder pressure of rats in diabetic group was significantly lower than that in the control group (P < 0.05);and the changes of all above indexes of rats in diabetic group were time-dependent(P < 0.05).The expression of NGF in bladder tissues of rats in diabetic group was significantly lower than that in the control group (P < 0.05),and the change of NGF expression in diabetic group was time-dependent (P < O.05).In diabetic group,the bladder smooth muscle was hypertrophic,the organization of smooth muscle was diverse and lose,and the muscle bundle fracture was observed;in control group,the bladder smooth muscle arranged orderly,and the shape was consistent,the structure of fascicle was compact.The condensation and margination of nuclear chromatin and mitochondria swelling were observed in diabetic group;while the bladder tissues in control group showed homogeneous nuclear and mitochondria without fracture and swelling.Conclusion The decrease of the expression of NGF in bladder tissues of diabetic rats can decrease the systolic function of bladder and change the morphology of detrusor and these changes show a time dependence.

Aim To investigate the protective effects of Angelica sinensis polysaccharides on bone marrow and spleen injury in SD rats induced by X-ray radiation.Methods Forty male SD rats were randomly divided into blank group,model group,low dose group of Angelica polysaccharide (63.5 mg · kg-1),Angelica polysaccharide middle dose group (127 mg · kg-1),and Angelica polysaccharide high dose group (254 mg · kg-1).The medicine group was treated daily with different concentrations of Angelica polysaccharide 2 mL gavage once,blank group and model group with an equal amount of distilled water instead.After a continuous administration of 7 d,X-ray irradiation was performed on rats except the blank group,with continuous exposure of 2 d,and the total radiation was at a dose of 6 Gy,then 3 d post the last radiation,the rats were sacrificed after anesthesia by bloodletting.The general quality of life,bone marrow nucleated cells,blood routine,spleen index,serum interferon gamma (IFN-γ)and interleukin-4 (IL-4) contents in each group of rats were detected.Pathological changes of spleen were observed under microscope.Results Compared with the blank group,the quality of life,bone marrow nucleated cells of rats in the model group,the number of white blood cells,spleen index,INF-γ and IL-4 expression significantly decreased,but the number of erythrocytes and hemoglobin increased significantly,and the difference was statistically significant (P < 0.05).HE staining showed the decrease in splenic corpuscle atrophy,germinal center,white pulp and red pulp structural disorder,and the number of lymphocytes.Compared with the model group,Angelica polysaccharide group's survival quality,bone marrow nucleated cell number,the number of white blood cell,spleen index,INF-γand IL-4 expression markedly increased,but the number of erythrocytes and hemoglobin decreased significantly,and the difference was statistically significant (P < 0.05).HE staining showed that pathological injury of spleen was alleviated,spleen body was visible,germinal center,red pulp and white pulp boundaries were still visible,and the number of lymphocytes increased.Conclusions Angelica sinensis polysaccharide can protect the bone marrow and spleen of SD rats induced by radiation,and the mechanism may be achieved by reducing the damage to hematopoietic cells and immune cells in the bone marrow and spleen,and replenishing blood and body fluid body.

Objective To evaluate the efficacy of adenosine triphosphate ( ATP ) bioluminescence test on quality monitoring of flexible endoscope reprocessing. Methods The patient-used gastroscopes were randomly assigned to two groups after standard reprocessing by one nurse. The group A were assessed by ATP test followed by microbiological surveillance test after cleaning and disinfection; and the gastroscopes in group B were tested in a reverse order. The biological load after endoscopic cleaning and disinfection were compared between ATP test and microbiological surveillance, and the correlation of the two tests on quality assessing of endoscopic cleanliness and disinfection were analyzed. Results A total of 51 samples were valid. The qualified rate of cleaning was 45. 1% ( 23/51) detected by ATP test, and the qualified rate of disinfection was 92. 2% ( 47/51) detected by microbiological surveillance. There was no statistical difference in cleaning qualification rate ( 50. 0% VS 40. 0%,χ2=0. 515, P=0. 473) and disinfection qualification rate ( 96. 2% VS 88. 0%,χ2=0. 316, P=0. 574) between the two groups. The logarithmic value of relative light unit ( RLU) of ATP test after cleaning was higher than the value after disinfection ( 1. 68 ± 0. 50 VS 0. 88 ±0. 49, t=2. 335,P=0. 024) . For gastroscopes disinfected appropriately, the RLU logarithmic value of ATP test after cleaning ( t=2. 505, P=0. 016) and disinfection ( t=2. 485, P=0. 016) were lower than that of disqualification. The correlation between the value of ATP test and the colony forming units of microbiological surveillance after cleaning was not statistically significant ( r=0. 199, P=0. 171 ) , but there was a low correlation between the two monitoring results after disinfection (r=0. 391, P=0. 005). There was a low correlation between the value of ATP test after cleaning and the colony forming units of microbiological surveillance after disinfection ( r=0. 301, P=0. 032) . Conclusion ATP bioluminescence test can be used to evaluate the trend of endoscopic biological load change and the quality of flexible endoscope reprocessing.

<p><b>OBJECTIVE</b>To establish a mouse model bearing human choriocarcinoma xenograft in severe combined immuno-deficient (SCID) beige mice and investigate the disease course and biological behaviors of the tumors.</p><p><b>METHODS</b>Human choriocarcinoma JAR cells were injected in female SCID beige mice (3-5 weeks old) either subcutaneously (group A, n=6) or via the tail vein (group B, n=6). Morphological studies, radioactive immunoassay, in vivo tumor imaging and histopathological examinations were performed to confirm JAR cell engraftment at the subcutaneous injection site and in the lungs of the mice.</p><p><b>RESULTS</b>On day 28 after tumor cell inoculation, the mice in group A showed palpable subcutaneous nodules, and HE staining revealed morphological features of the nodules consistent with choriocarcinoma cells; in vivo imaging in group B showed single or multiple solid tumor masses in the lungs, and tissue biopsy examination demonstrated varying degrees of tumor cell infiltration. Compared with the control mice, peripheral blood β-HCG levels in both groups A and B increased significantly on day 14 after cell inoculation (P<0.05), and the increment was more conspicuous in group B (P<0.05).</p><p><b>CONCLUSION</b>Mouse models bearing human choriocarcinoma xenograft can be successfully established by injecting JAR cells either subcutaneously or via the tail vein to mimic the characteristics of epithelial solid tumors and lung metastasis of human choriocarcinoma.</p>

Objective To identify pre-operative demographic and urodynamic (UD) parameters related to overactive bladder (OAB) symptoms of pre-operation and persistence of after pelvic organ prolapse (POP) repair.Methods From Jan 1,2010 to Oct 31,2012,this perspective study examined demographic and UD data of 175 patients undergoing POP surgery.Pre-and post-operative urinary distress inventory 6 (UDI-6) scores for frequency and urge urinary incontinence (UUI) were analyzed,and correlations between scores and pre-operative UD data were also analyzed.Results (1)Surgery resulted in a improvement of frequency (71.0%,71/100) and UUI (69.2%,63/91) at 12-24 months follow-up.(2)Pre-operative UD parameters:first desire of bladder <100 ml,6.3% (11/175); detrusor overactivity,14.9% (26/175); post-void residual ≥50 ml,11.4% (20/175); maximal flow rate (Qmax),(16±7) ml/s.(3)Comparison of pre-operative UD datas between patients with OAB symptoms and without,with OAB symptoms group had more patients of type Ⅲ stress urinary incontinence [5% (5/91) versus 0 (0/84),P=0.022].(4)Qmax was higher in improvement in UUI group than that in persistent UUI after POP repair [(17±7) m]/s versus (13±5) ml/s,P<0.01].Conclusions POP repair significantly reduces OAB symptoms; however,there had not exact predictor for symptoms of persistent frequency,UUI after POP repair.Persistent UUI symptom may be related to Qmax of pre-operative UD.

<p><b>OBJECTIVE</b>To study if the expression of maspin protein in respiratory epithelial cells was downregulated through IκB kinase-α (IKKα)-controlled mechanism in an Aspergillus fumigatus-induced model in rat.</p><p><b>METHODS</b>Inactivated Aspergillus fumigatus hyphae (AFH) suspension was used to induce respiratory epithelial cells (REC) cultured in vitro in rat, with PBS buffer as control. By RT-PCR, the mRNA expression of maspin was quantified, and by immunocytochemistry, the expression of maspin and IKKα in REC was observed. Furthermore AFH (from level 1 to level 3) suspension was prepared to induce REC. Then Western blot hybridization technique was used to detect the expression of maspin and IKKα protein. All data were processed by analysis of variance and two-variable correlation analysis.</p><p><b>RESULTS</b>By RT-PCR, a statistically significant (t = 2.463, P < 0.05) expression of maspin mRNA was found (0.128 ± 0.059 in AFH group, 2.972 ± 0.353 in control group). By Immunocytochemistry, the difference of maspin protein color in different groups was shown statistically significant in integrated scoring (t = 3.721, P < 0.05, weak positive in AFH group, moderately positive in control group). While in IKKα color study, the difference between the two groups was also statistically significant (t = 6.825, P < 0.05) in integrated scoring, with a moderate positive in AFH group and weak positive in control group. By Western Blot hybridization, grayscale ratio of maspin and β-actin was 0.912 ± 0.023 in control group, 0.607 ± 0.030, 0.476 ± 0.019, 0.416 ± 0.017 in AFH 1-3 groups, respectively, with a statistically significant difference within the four groups (F = 281.91, P < 0.05); While grayscale ratio of IKKα and β-actin was 0.624 ± 0.012 in control group, 0.739 ± 0.020, 0.778 ± 0.010, 0.927 ± 0.017, respectively, in AFH 1-3 groups; with a statistically significant difference within the four groups(F = 200.91, P < 0.05). Moreover, the difference between any two groups from both AFH group (including subgroup 1, 2 and 3) and control group was statistically significant. Two-variable correlation analysis indicated a negative correlation between expression of maspin and IKKα (r = -0.911, P < 0.05).</p><p><b>CONCLUSIONS</b>Induced by Aspergillus fumigatus, the rat respiratory epithelia might upregulate the expression of IKKα with a downregulated expression of maspin protein.</p>

Through random sampling,a total of 300 residents of Beijing Changping Ming Tombs Town were selected for home-based questionnaires.The smoking rate of interviewed residents was 21.4% (53/248).The rate in men (42/125,33.6% ) was bigber than that in women ( 11/123,8.9% ).The rate in those with higher education was lower than that in those with lower educational level ( x2 =27.12,P < 0.05 ).The population awareness of tobacco hazards was 80.6% (200/248).Awareness in smokers was lower than the average awareness of ex-smokers and nonsmokers ( x2 =5.07,P < 0.05 ).The higher the education level,the higher the awareness of tobacco health hazards ( x2 ≈ 19.72,P < 0.05 ).There were differences in awareness among different age groups (x2 =13.37,P < 0.05 ).

ObjectiveTo investigate the clinicopathological features and prognosis of natural killer (NK)/T cell lymphoma and to analyze its relationship with Epstein-barr virus(EBV). MethodsTotally, 36 cases of cutaneous NK/T cell lymphoma were collected from 2000 to 2010 at the Department of Pathology, Peking University Health Science Center, and classified into primary and secondary groups according to whether there is evidence of extracutaneous involvement within 6 months after diagnosis. Clinicopathological features were analyzed and Epstein-barr virus (EBV) was detected. ResultsOf these 36 cases, 13 (36.1%) were classified as primary cutaneous NK/T cell lymphoma, 20 (55.6%) as secondary, and 3 (8.3%) remained unclassified because of the lack of clinical data. Males were more likely to develop both primary and secondary cutaneous NK/T cell lymphoma than females, but there was no striking difference in sex ratio between the patients with primary and secondary lymphoma (P > 0.05 ). Compared with the patients with primary cutaneous NK/T cell lymphoma, those with secondary cutaneous NK/T cell lymphoma showed a younger median age at onset(43.5 vs. 54 years, P < 0.05), higher prevalence of B symptoms(including fever, night sweat, body weight loss) and multiple skin lesions (P < 0.05 and 0.01, respectively). EBV was positive in 92.3% (12/13) of the primary lymphoma cases and 85%(17/20) of the secondary lymphoma cases. Moreover, the median survival was 8 months in all the cutaneous NK/T cell lymphoma cases, and was significantly shorter in secondary cases than in the primary cases(6 vs. 18 months, x2 = 6.074, P < 0.05). ConclusionsCutaneous NK/T cell lymphoma is an EBV-associated, clinica]ly aggressive disease entity. Patients with primary cutaneous NI/T cell lymphoma seem to have an older age at onset and a better prognosis as compared with those with secondary cutaneous NK/T cell lymphoma.

In a search for natural products that could be used to control schistosomiasis, nineteen extracts of eleven medicinal plants from eastern China have been tested for molluscicidal activity against snail Oncomelania hupensis. The n-butanol fraction of the fresh leaf from Buddleja lindleyana Fortune, Buddlejaceae, showed significant activity against the snails. Statistical analysis revealed that the LC50 and LC90 values for the n-butanol fraction were 39.91 mg L-1 and 59.28 mg L-1 for 48 h, respectively. Otherwise, the LC50 values for the n-butanol fraction to zebrafish was 403.24 mg L-1 for 48 h. Therefore, the n-butanol fraction of the fresh leaf from B. lindleyana may be a potent and safe molluscicides.


Na busca por produtos naturais que podem ser utilizados para controle da esquistossomose, dezenove extratos de onze plantas medicinais do leste da China foram testados para atividade moluscicida contra o caramujo Oncomelania hupensis. A fração n-butanol das folhas frescas de Buddleja lindleyana Fortune, Buddlejaceae, mostrou atividade significativa contra os caracóis. A análise estatística revelou que os valores de CL50 e CL90 para a fração n-butanol foram 39,91 mg L-1 e 59,28 mg L-1 por 48 h, respectivamente. Por outro lado, a CL50 para a fração n-butanol para peixe-zebra foi 403,24 mg L-1 por 48 h. Portanto, a fração n-butanol das folhas frescas de B. lindleyana poderá vir a ser um moluscicidas potente e seguro.

Objective To analyze the influence of hyponatremia on hospitalization days and hospital mortality of hospitalized patients with chronic heart failure. Methods All data were collected from 2465 patients with chronic heart failure in our hospital between January 1980 and August 2007. According to the admission serum sodium, all the patients were divided into two groups: low serum sodium group (Na+<135 mmol/L) and normal serum sodium group (Na+≥135retool/L). Clinical characteristics, therapeutic conditions and the influence of hyponatremia on prognosis were analyzed between the two groups. Results There were 618 patients in low serum sodium group among 2465 cases. Patients with hyponatremia were more likely to have lower systolic blood pressure and lower left ventricular ejection fraction. Lower admission serum sodium was related with higher concentration of serum angiotensin, aldosterone and higher activity of serum renin. Patients in low serum sodium group had significantly longer hospitalization days (Z=-4.026, P<0.01) and higher rate of hospital mortality (χ2=76.935, P<0.01) than patients in normal serum sodium group. Multivariate logistic regression indicated that lower admission serum sodium was an independent risk factor for hospitalization days (b= 0.928, P<0.01) and hospital mortality (OR=0.928, P<0.01). There was a negative correlation between admission serum sodium and hospitalization days (r=-0.132, P<0.01), and the rate of hospital mortality increased by 24.7for each 3 mmol/L decrease in admission serum sodium level. Conclusions Hyponatremia in hospitalized patients with chronic heart failure is relatively common and is associated with poorer heart function, higher activity of neural hormone and more severe prognosis. It is important to prevent hyponatremia for shortening hospitalization days and improving the prognosis of patients with chronic heart failure.

<p><b>OBJECTIVE</b>To analyze the impact of renal dysfunction on survival in hospitalized chronic heart failure (CHF) patients.</p><p><b>METHODS</b>In this retrospective analysis, we collected all clinical data from eligible patients hospitalized in the second hospital of Tianjin Medical University between Jan 1980 and Aug 2007. CHF patients were divided into three groups according to glomerular filtration rate (GFR): A, normal renal function; B, mild renal dysfunction; C, renal dysfunction. Patients in group C were further divided into three subgroups according to hospitalization year: D, 1980.01 - 1989.12; E, 1990.01 - 1999.12; F, 2000.01 - 2007.08.</p><p><b>RESULTS</b>Renal dysfunction was found in 714 patients. Compared with group A (n = 817) and group B (n = 928), patients in group C were older, had worse heart function and major medications included nitrates, diuretics and digitalis. From 1980 to 2007, use of Angiotensin II receptor antagonist, beta-blocker, statins significantly increased and the in-hospital mortality significantly decreased in group C patients. Percent of angiotensin converting enzyme inhibitor (ACEI) use was the highest in 1990s. The hospital stay was significantly longer and all cause in-hospital mortality was significantly higher in group C compared to group A and group B (all P < 0.01). After adjustment for other risk factors by multivariate analysis, renal dysfunction was an independent risk factor of in-hospital all cause mortality. Patients faced 16.7% higher risk of all cause in-hospital mortality for every 10 mlxmin(-1) x1.73 m(-2) decrease in GFR.</p><p><b>CONCLUSIONS</b>The incidence of renal dysfunction was high in CHF patients. The hospital stay was longer, in-hospital all-cause mortality was higher in CHF patients with renal dysfunction compared to CHF patients without or with mild renal dysfunction. Renal dysfunction was an independent risk factor for all-cause in-hospital mortality. Increased use of ACEI, ARB, beta-blocker and statins might be responsible for reduced in-hospital mortality in CHF with renal dysfunction patients in recent years.</p>

<p><b>OBJECTIVE</b>To observe the effect of sulphur dioxide (SO2) inhalation on the pathogenesis of allergic rhinitis (AR) in mice, and investigate the toxic effect of SO2 on respiratory tract mucosa.</p><p><b>METHODS</b>Fifty male Kunming strain mice were randomly allocated to five groups, i. e., control group, AR group and AR plus different concentrations of SO2 group (three sub-groups). Nasal cavity irrigating solution was gathered from nasal cavity, and blood from the orbital venous sinus after anesthesia The concentrations of IL-5 and IL-13 in the peripheral serum and nasal cavity irrigating solution were measured by ELISA. The number of eosinophils (EOS) per square millimeter in sinus mucosa was calculated by hematoxylin-eosin. The expression of SP-D in nasal mucosa was analyzed by immunohistochemistry technique.</p><p><b>RESULTS</b>With increasing concentrations of SO2, the levels of IL-5 and IL-13 in the peripheral serum, and the density of Eos in sinus mucosa increased simultaneously. A positive correlation existed between the concentration of inhaled SO2 and the elevation of both IL-5, IL-13 and Eos infiltration in nasal mucosa. The coefficient correlation relatively were 0.894, 0.874, 0.894, 0.891 and 0.870 (P <0.01). The expression of SP-D in 56 mg/m3 and 112 mg/m3 SO2 groups was higher, while it was lower in 168 mg/m3 SO2 group (P < 0. 001).</p><p><b>CONCLUSIONS</b>The study showed that sulphur dioxide inhalation facilitates the onset of allergic rhinitis in mice. SO2-related Th2-derived cytokines as well as the infiltration of EOS in nasal mucosa help to aggravate the development of allergic rhinitis.</p>
