RESUMO

Objective:To screen anti-EGFR components from Aconitum soongaricum Stapf. and investigate their inhibitory effect on HEK293 cells and EGFR/HEK293 high expression cell lines. Methods:A two-dimensional online system (EGFR/CMC-HPLC-MS) was built to screen anti-EGFR active compounds from Aconitum soongaricum Stapf.;molecular docking assay was performed to determine the binding affinity of the components to EGFR;MTT was used to confirm the inhibition effect of the screened compounds on HEK293 cells and EGFR/HEK293 high expression cell lines.Results:The screening results showed that aconitine,12-epi-napellin and songorine were the active components acting on EGFR like gefitinib; molecular docking assay showed the targeting components acting on EGFR in the similar manner with gefitinib used as a positive control drug; MTT assay confirmed the screened components had inhibitory effects on HEK293 cells and EGFR/HEK293 high expression cell lines in a dose-dependent manner within the dose range of 0.4- 50.0 μmol·L-1. Conclusion:The screening results are apparently consistent with the pharmacological effect. The online EGFR/CMC-HPLC-MS method has a good application prospect in the rapid screening of the active compounds in the complex system of traditional Chinese medicines,and fully demonstrates such advantages of cell membrane chromatography as simple and rapid with high efficiency.

RESUMO

Objective To set up the quality control standard for Leaf of Chinese Holly; To provide basis for the utilization and development of Leaf of Chinese Holly.Methods Ten batches of Leaf of Chinese Holly samples from different habitats were collected, and the properties were described. The crosscutting and powder of leaf was under microscopic identification. Chlorogenic acid was set as reference substance to conduct thin-layer identification. Moisture, total ash, and acid-insoluble ash of the 10 batches were detected. HPLC was used to detect chlorogenic acid in Leaf of Chinese Holly.Results The properties and microscopic identification were described. Thin-layer chromatography method for chlorogenic acid in Leaf of Chinese Holly was formulated. Check items temporarily required that the moisture in Leaf of Chinese Holly should be less than 13%, total ash less than 13%, and acid-insoluble ash less than 5%. Content determination method for chlorogenic acid in Leaf of Chinese Holly was confirmed. It temporarily required that the content of chlorogenic acid should be more than 0.60%.Conclusion The established method can be used for the formulation of quality control of Leaf of Chinese Holly.

OBJECTIVE:To establish mathematical model of liquid volume added of TCM medicinal broth decoction machine to accurately calculate liquid volume added in the process of medicinal herb decocting,so as to guarantee the quality of medicinal herb decocting. METHODS:The water absorption rate of representative TCM decoction piece with high use frequency were deter-mined,and cluster analysis of water absorption rate of TCM decoction piece was conducted according to closely related index as density,size,shape,moisture. TCM decoction piece with similar water absorption rate were bracketed together,so that of single ingredient TCM decoction piece can be estimated by water absorption of representative TCM decoction piece;the quantity of water evaporation and liquid extrusion were determined among different types of decoction machine (powered by electric and gas);ac-cording to the above parameters,mathematical model of liquid volume of TCM medicinal broth prepared by different types of de-coction machine had been established,and validated with TCM formula. RESULTS:Factors that affected the liquid volume added included the water absorption of each ingredient,the quantity of water evaporation and extrusion function. The mathematical model was liquid volume added=water absorption of each ingredient × quality of decoction piece+the quantity of water evaporation+re-quired amount of liquid-parameters of extrusion function×total weight of decoction piece;in validation test,the percentage of the practical amount of liquid to required amount was within ±5%. CONCLUSIONS:Established model can promote the accuracy li-quid volume added and guarantee the quality of TCM decoction when using TCM decoction machine.

OBJECTIVE: To probe into the utilization characteristics and the consumption of oral traditional Chinese medicinal decoctions in our hospital.METHODS: The department of internal medicine in our hospital was subdivided into 8 special divisions.6 physicians of senior professional title(associated professor or above) from each subdivision were randomly selected for analysis of the prescriptions of oral traditional Chinese medicinal decoctions prescribed by the 48 physicians.The ingredients and their dosages in the prescriptions were recorded to database and analyzed by Weka data mining software.RESULTS: The drug use frequency of no less than 5% was noted in 68 kinds of oral traditional Chinese medicinal decoctions,with the drugs for reinforcing deficiency showing the highest drug use frequency,followed by drug for clearing heat,drugs for blood-activating and stasis-dissolving,drugs for eliminating sputum and relieving cough and asthma etc.The drug use frequency of no less than 1% was noted in 193 kinds,of which,160(83%) whose doses exceeded the specified limit value specified in Chinese Pharmacopeia,and in 52 kinds(27%),the proportion of prescriptions involving overdosing was no less than 50%.CONCLUSION: The concerned departments should attached great importance to the widespread phenomenon that the dosages of oral traditional Chinese medicinal decoctions in our hospital exceed the specified dosages in Chinese Pharmacopeia.
