RESUMO

Objective To study the effects and possible mechanism of Huoluo Jiangu Capsule (HJC) on adjuvant arthritis rats. Methods The arthritic model of rats induced by Freund’s complete adjuvant was used in experiment. The effect of HJC on swelling degree of feet, weight of thymuse and spleen of AA rats were observed. The serum activity of SOD, GSH-Px and content of NO, MDA were determined. Results HJC can relieve swelling degree of feet and weight of spleen of AA rats, increase serum SOD, GSH-Px and decrease serum NO, MDA of AA rats. Conclusion HJC has good effet of cleaning up the oxygen free radicals and anti-inflammatory.

RESUMO

Objective To observe effect of Huoluojiangu capsule (HJC) on analgesic experiments. Methods The 3 kinds of analgesic experiments of whirling body, electric irritation and hot board were respectively adopted in mice. 60 mice were randomly divided into 6 groups in every analgesic experiment:A group(blank control), normal saline 0.25 mL/10 g. B group (positive control), diclofenac sodium 3.5 mg/kg;C group (positive control), pethidine 15 mg/kg, D,E and F group were respectively as low,mid and high dose of HJC groups (HJC 14, 21, 28 g/kg). Afterward, effects of HJC on ache induced by chemistry, electricity and heat were respectively observed at different time after pretreatments. Results Low, mid and high dose of HJC groups can all inhibit reaction of whirling body by chemical irritation. Compared with blank control group, low dose of group had evidently effect, P 0.05), but it had stronger analgesic effect than positive groups (B and C groups) at 180 min, P

RESUMO

Objective To observe the effect of Huoluojiangu Capsule(HJC) on anti-inflammatory experiment.Methods The anti-inflammatory experimental models were established in three kinds of rats with pleuritis,the swellen sole of the foot and the PGE_2 content in inflammatory tissues,respectively.Results The swollen reaction in the sole of foot in rats induced by egg white was lessoned with the treatment of low,middle and high dosage of HJC.Compared with the blank control group,the treatment is effective in low dosage group and evidently effective in groups of middle and high dosage after 30 min(P0.05),respectively.Compared with the blank control group,the effect of HJC on the PGE_2 content in the inflammatory tissues of middle and high dosage groups was significant(P

RESUMO

AIM: To evaluate association of genotype and phenotype of CYP2D6 and the application of oligonucleotide microarray hybridization genetic testing in CYP2D6 multiple alleles analyses. METHODS: Two hundred forty-two healthy volunteers were recruited, and a 60 mg oral dose of dextromethorphan (DXT) was administered to each for assessment of the DXT metabolic ratio . A 20 ml blood sample was also collected for DNA isolation and testing. CYP2D6 alleles *2--*11; *17 and multiple CYP2D6 gene copies were tested by allele-specific PCR and/or the affymetrix CYP450 gene chip assay. Five of the CYP2D6 alleles (*3, *4, *6, *7, and *9) were evaluated by both PCR and the CYP450 gene chip assay. RESULTS: The CYP2D6 genotype was more informative and reflective in CYP2D6 enzyme expression than a phenotype. Genetic tests for the CYP2D6 *3, *4, *6, *7 and *9 alleles showed a high degree of concordance between the CYP450 gene chip and AS-PCR methods. CONCLUSION: Oligonucleotide microarray hybridization is a promising and reliable approach for detecting multiple alleles at gene loci.

RESUMO

AIM To investigate the protective action of onychin aganist the growth inhibition of endothelial cell injured by menadione and its mechanism. METHODS The injured model was established by endothelial cell treated with menadione.Protective effect of onychin aganist growth inhibition of injured endothelial cell was determined by MTT assay and cell counting method; NO concentration in the medium was determined by nitrate reductase assay; eNOS and phosph ERK1/2 protein levels were determined by Western blot. RESULT Onychin significantly decreased the growth inhibitory rate of injured endothelial cells,increased NO concentration in the medium and eNOS activity and up regulated phosph ERK1/2 expressing. CONCLUSION Onychin has protective action and against the growth inhibition of endothelial cell injured by menadione may be via NO and ERK1/2 signal pathway.

The growth inhibitory rate of endothelial cell was determined by MTT assay; NO - 2/NO - 3 concentration in the medium was determined by nitrate reductase assay; eNOS and caveolin-1 protein levels were determined by Western blot. RESULTS: Onychin significantly decreased the growth inhibitory rate of endothelial cells injured by menadione, increased NO - 2/NO - 3 concentration in the medium and eNOS activity and up-regulated caveolin-1 expression. CONCLUSION: Onychin possesses a protective effect against endothelial cell injury induced by menadione via caveolin-1/eNOS pathway.
