RESUMO

Long non-coding RNAs (lncRNAs) are members of RNA that are structurally similar to mRNA. They cannot encode proteins because they do not have a conserved open reading frame. LncRNAs were once regarded as abnormalities or noises or without any biological function after gene transcription. With the further development of research, it has been found that it can participate in normal or abnormal biological processes as an important regulator. LncRNAs are closely related to the development of nervous system function, metabolic disorders and tumors. LncRNAs abnormally expressed in cervical cancer participate in the regulation of various biological processes of cervical cancer by inhibiting or promoting tumors. This article reviews the recent reports on the abnormal regulation, molecular regulation mechanism and potential clinical application of lncRNAs in cervical cancer.

RESUMO

To adapt to the system reformation of medical laboratory science from five academic years to four academic years and to meet the new professional technology-oriented requirements, the medical laboratory science institute of Guangdong Medical College has carried out a comprehensive reform of curriculum system. This paper has analyzed the current problems in the school medical ex-amination and explored the curriculum system reform from three respects such as adjusting curriculum by restructuring and integrating programs, implementing modular teaching to build its characteristics and strengthening practice teaching.And it has also explored the full assessment mode by optimizing the traditional one-stop assessment.

RESUMO

This paper is aimed to present a research on fusion protein of human tumor necrosis factor-alpha (hTNF-alpha), matrix metalloproteinase 1 (MMP1), and foldon sequence using the methord of gene engineering. We transformed the recombinant plasmid, which contains the DNA sequences of hTNF-alpha, MMP1, and foldon sequence, into Rosetta2, and successfully induced the fusion protein to express under given conditions by isopropyl beta-D-1-Thiogalactopyranoside (IPTG). Then we purified the expression product through a glutathione S-transferase (GST) resin and collected the interested protein. This research may lay the groundwork for scientific research and clinical application.

RESUMO

Objective To observe the effects of physical agents therapy on serum hs-CRP, TNF-α andadiponectin in patients with cerebral infarction and the possible underlying mechanisms. Methods Sixty patientswith cerebral infarction were randomly and equally divided into two groups: 30 cases were treated with physical a-gents therapy ( physical therapy group) , and 30 with drugs only ( drug treated group). Thirty normal subjectsserved as the control group. The level of hs-CRP in the serum was determined by latex agglutination reaction, TNF-and adiponectin were determined by using ELISA before and after therapy. Results The levels of serum hs-CRP and TNF-α of patients with cerebral infarction before therapy were much higher than those of the control group,but adiponectin was significantly lower than those of the control group( P < 0.01 ). After therapy, the levels of ser-um hs-CRP and TNF-α were decreased and adiponectin was increased significantly in both treated groups ( P <0.01 ). Comparison with two treated groups showed that the levels of hs-CRP and TNF-α were lower and adiponec-tin was obviously higher in physical agents therapy group than those in the drug treated group ( P < 0.05 ). Con-clusion The patients with cerebral infarction have low level of serum adiponectin. Physical therapy might exertbeneficial effects on patients with cerebral infarction by the decreasing serum hs-CRP and TNF-α, as well as by ele-vating adiponectin.

OBJECTIVE@#To observe the suppressive effects of immunostimulatory DNA sequences (ISS) in conjunction with dermato phalloides farinae allergen (Df) on nasal cavity inflammation in rats of experimental allergic rhinitis (AR).@*METHOD@#Thirty-two rats were divided into 4 groups: group ISS(A), group ISS+ Df (B), group Df (C) and group normal saline (D). Rats in groups A,B and C were sensitized and challenged with Df. Blood samples were obtained every week for six weeks, Df specific IgE was measured by ELISA. The nasal mucosa were studied pathologically. The levels of serum interferon gamma (IFN-gamma), IL-12, IL-4 and IL-5 were determined by ELISA.@*RESULT@#The serum level of sIgE in group B was significantly lower than that in group C in six weeks, but that in group A was not significantly different to that in group C. The mean levels of serum IFN-gamma and IL-12 in A, B group were significantly higher than in C group (P < 0.01). But IL-4 and IL-5 level was much lower (P < 0.01).@*CONCLUSION@#ISS+ Df have inhibited effectiveness on allergic nasal cavity inflammation in rats and its action time is 6 weeks.

<p><b>OBJECTIVE</b>To screen the exon12 mutation of pot1 gene in cultured human carcinoma cell strains (lines).</p><p><b>METHODS</b>The chromosomal DNA was extracted from 27 cultured carcinoma cell strains (lines). The exon 12 of pot1 gene was amplified by PCR, and the product was purified and screened. The screening results were compared with the data of GenBank and NCBI and the exon 12 mutations in cultured human carcinoma cell strains (lines) analyzed.</p><p><b>RESULTS</b>The exon12 sequence of pot1 could be specifically amplified using the designed primers. Direct sequence analysis of the PCR products after purification showed that 4 of the 5 carcinoma cell lines of the female genital system such as Hela and HO8910-PM cells shared the same transition (G17722-->C) in exon12 of human pot1 gene resulting in a conversion of G1385-->C in the cDNA and amino acid change of Leu454-->Phe in the translated polypeptide. The rest of the 23 cell strains (lines) from different origins showed no such mutation.</p><p><b>CONCLUSION</b>The exon12 (17,722 bp) is a mutant region specific for female genital system tumor.</p>

RESUMO

OBJECTIVE: To study the mechanisms of the antitumor and immunoregulation functions of polyporus polysaccharide (PPS). METHODS: The production of nitric oxide (NO), the activity and mRNA expression of inducible nitric oxide synthase (iNOS) in peritoneal macrophages of mice administered with different dose of PPS were observed by Griess reaction, fluorimetry assay and RT-PCR, respectively. RESULTS: PPS could elevate the iNOS activity with dose-dependence and stimulate the iNOS mRNA expression of peritoneal macrophages in mice. CONCLUSION: The regulation of PPS on the production of NO in peritoneal macrophages of mice may occur at transcriptional level of iNOS. This indicates that the mechanism of PPS's antitumor and immunoregulation functions may be related to increasing NO output of macrophages through stimulating iNOS's denovo synthesis.

RESUMO

To adapt the developing requirement of clinical laboratory diagnostics,the Analysis Technique of Molecular Biology was set up for Grade 2003 in laboratory medicine in our college.The students showed great interests and enthusiasm in this subject,especially in laboratory course.The studentsof Grade 2003 made a better score than those of Grade 2002 in the final Test of Basic Experimental Skills,which included the scores of basic operation,experiment theory and technical skill.These results showed that setting up the Analysis Technique of Molecular Biology could improve student's ability in experimental skills.

RESUMO

Object To investigate the mechanisms of action of lentinan (LTN) activated mouse peritoneal macrophages to produce nitric oxide (NO). Methods The effects of LTN on NO output and intracellular glutathione (GSH) in mouse peritoneal macrophages and the correlation of them were studied. Results (1) LTN can significantly increase production of NO in mouse peritoneal macrophages, decrease level of intracellular GSH followed increase of NO production. (2) The above effect can be blocked efficiently by the NO production inhibitors. (3) NO production can be inhibited by GSH lowering drugs. Conclusion LTN increase NO production with depletion of intracellular GSH in the activated mouse peritoneal macrophages. It suggested that intracellular GSH plays an important role in regulation of NO production and protection of host cells from cytotoxic attack induced by NO.

Objective To investigate the effects of procyanidin on apoptosis and caspase-8 activation in HeLa cell induced by TNF-?. Methods Flow cytometry was used to analyze the apoptosis. Aim To inhibit the expression of human protection of telomere gene hPOT1 in HeLa cells using vector based on RNA interference(RNAi) technique and to investigate its effects on the activation of Caspase-3 in HeLa cell.Methods Three recombinant plasmids(our group constructed before) containing different hPOT1 target sequences(pmU6-shRNA1,pmU6-shRNA2 and pmU6-shRNA3)were transfected into HeLa cells by liposomal.The expression inhibition of hPOT1 was detected by RT-PCR and EMSA.The expression and activation of Caspase-3 were inspected by RT-PCR,Western blot analysis and Caspase-3 kit.Results After 48 hour′s transfecting,hPOT1 mRNA and protein level in HeLa cells reduced and the expression of Caspase-3 mRNA had no change.Caspase-3 zymogen decreased but its activity increased.Conclusions hPOT1 expression in HeLa cells can be significantly inhibited by using siRNAvectored RNAi and the down-regulation of hPOT1 expression can activate Caspase-3.
