RESUMO

Objective To examine the predictive value of serum soluble E-selectin ( sE-selectin) levels for high risk plaques as detected by coronary computed tomography angiography ( CCTA).Methods This was a cross-sectional study.CCTA was performed for each patient.Serum levels of sE-Selectin, sVCAM-1,sICAM-1 and sP-Selectin were measured by flow fluorescence immunomicrobead assay ( FFIA). ROC curve and multivariate analysis were used to determine the predictive value for the presence of high risk plaques.Results The high risk plaques group showed higher sICAM-1 and sE-selectin levels.The multivariate Logistic analysis showed that male (OR=2.131, P=0.019), old age (OR=1.053, P=0.002), hyperlipidemia (OR=1.960, P=0.022), high level of apoB (OR=2.896, P=0.004) and high level of sE-Selectin(OR=1.975, P=0.008) were independently associated with the presence of high risk plaques.The AUC of the multivariate model for high risk plaques was 0.722.The sensitivity, specificity, the positive predictive value and the negative predictive value were 55.3%, 78.3%, 71.8% and 64.4%respectively.Conclusions High sE-selectin levels were associated with the presence of high risk plaques in asymptomatic populations at low to intermediate Framingham risk .sE-selectin can play an important role in plaque screening, as detected by coronary CTA.

RESUMO

Objective To investigate the value of combinedevaluation of serum lipoprotein associated phospholipase A2 (LP-PLA2) and cystatin C (CysC) on systemic lupus erythematosus (SLE) patients with lupus erythematosus.Methods A total of 177 patients with SLE were enrolled in the General Hospital of People's Liberation Army from June 2000 to February 2017,87 cases in lupus group (SLE group),90 cases in lupus nephritis group (LN group),60 cases in healthy control group.The concentration of LP-PLA2 and CysC were measured.The ROC curves was established for estimation of the cutoff values of two indexes in SLE patients with LN,and the diagnostic efficacy of different diagnostic criteria was compared.Results The LP-PLA2 and CysC concentration in the LN group were higher than those in the SLE group (Z=-2.512,-5.688,all P<0.05).The cutoff value of LP-PLA2 was 245.5 U/L and the cutoff value of CysC was 1.235 mg/L on the risk assessment of in SLE patients with LN.When using the parallel method for combined LP-PLA2 and CysC to evaluate the risk of LN,the sensitivity and negative predictive value would be significantly improved (x2 =9.461,4.377,all P< 0.05),the efficiency was better.Conclusion Parallel assessment of serum LP-PLA2 and CysC have clinical value on assessment of the risk of LN in SLE patients.

<p><b>OBJECTIVE</b>To investigate the effect of intensive rosuvastatin therapy on adhesion molecules in patients with peripheral atherosclerosis and explore the possible upstream mechanism.</p><p><b>METHODS</b>Twenty asymptomatic patients with peripheral atherosclerosis were enrolled and given 5-20 mg/day rosuvastatin for 3 months. Before and after the treatment, the lipid profile and plasma vascular cell adhesion molecule-1 (VCAM-1) levels were examined. The expression of intercellular adhesion molecule-1 (ICAM-1) in the mononuclear cells was measured using flow cytometry, and the mRNA and protein expressions of peroxisome proliferator-activated receptor γ (PPARγ) were detected using RT-PCR and Western blotting, respectively.</p><p><b>RESULTS</b>Compared with the baseline levels, ICAM-1 expression decreased and PPARγ protein expression increased in the lymphocytes. Rosuvastatin therapy did not produce obvious effects on plasma VCAM-1 level or ICAM-1 expression in the monocytes in these patients.</p><p><b>CONCLUSION</b>Rosuvastatin produces anti-inflammatory effects by decreasing the expression of ICAM-1 in mononuclear cells, and its upstream mechanism may involve the PPARγ pathway.</p>

OBJECTIVE@#To explore the change of orexin-A expression in hepatic reperfusion and their association with liver injury, and to find out the role of orexin-A in traumatic stress responses.@*METHODS@#A 70% hepatic reperfusion model of rats was established, setting groups of sham-operation and injury ones with different reperfusion time. A self-produced radioimmunoassay and relevant kits were used to detect the protein level of orexin-A in the plasma and the hypothalamus, serum glucose, total anti-oxidation capacity and alanine transaminase, HE staining and immunohistochemistry were used to investigate the pathological variation and protein expression of orexin-A in the liver, while RT-PCR was applied to observe mRNA expression of orexin-A in the hypothalamus and the liver.@*RESULTS@#Both the shape of standard curve and metrical results of the self-produced orexin-A radioimmunoassay were good. Protein levels of orexin-A in the plasma and the hypothalamus in each reperfusion group showed no significant change. Serum glucose and total anti-oxidation capacity increased significantly at the later phase of injury. There was significant and positive linear correlation between the plasma orexin-A and serum glucose and total anti-oxidation capacity; serum alanine transaminase in each reperfusion group was significantly higher, and liver damage was significantly alleviated at the later phase of the injury. Different extents of variation were observed in protein expression of orexin-A in the liver and its mRNA expression in the hypothalamus and the liver.@*CONCLUSION@#Orexin-A undergoes significant changes during hepatic reperfusion, indicating that orexin-A participates in the modulation of hepatic reperfusion-induced liver injury and internal disorders.

<p><b>OBJECTIVE</b>To construct and investigate the cell model of a novel mutation R675Q in the skeletal muscle Na channel type 4 alpha subunit gene (SCN4A) identified from a Chinese family with normokalemic periodic paralysis.</p><p><b>METHODS</b>cDNA encoding the adult isoform of SCN4A was used as a template for in vitro site-directed mutagenesis by PCR method. The mutated plasmid was transiently transfected into HEK-293 cells by calcium phosphate precipitation. Twenty four and 48 hours after transfection, the expression level of SCN4A was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. Whole cell voltage-clamp recording was used to study the current of sodium channels.</p><p><b>RESULTS</b>The site-mutagenesis of the plasmid was confirmed by sequencing. The expression of SCN4A gene was significantly elevated 24 h and 48 h after transfection. The relative current of R675Q is smaller than that of wide type before reaching peak current under the same test voltage, but larger than that of wild type current after reaching peak current. They both had the largest peak current under 0 mV test pulse.</p><p><b>CONCLUSION</b>A cell model of normokalemic periodic paralysis was successfully constructed. The R675Q mutation of the SCN4A gene enhances the activation and inactivation of the sodium channel, and the S4 transmembrane segment may have intimate relationship with the attack of weakness in normoKPP patients.</p>

RESUMO

Objective To observe the level of serum IFN-2? in patients with severe acute respiratory syndrome(SARS). Methods The level of serum IFN-2? in 66 patients with SARS were measured by RIA, and healthy subjects served as control. Results The results showed that level of serum IFN-2? in patients with SARS in active period were significantly higher than that in control, and level of serum IFN-2? in severe type of patients with SARS either rose or fell. Conclusions The change of serum IFN-2? level indicated that cellular immunity participated in immunoreaction to SARS virus. The change of IFN-2? level in severe type of patients with SARS reflected the difference of immunoreaction in the patients.

Apoptosis, or programmed cell death, is a conservation process essential for normal development and homeostasis of biologist. It's known that a number of factors and pathways can lead to apoptosis. Specific phosphorlipids and proteins of biomembrane could activate the signal cascades of apoptosis. The interaction of caspases, bcl-2 family and mitochondria play an essential role in regulation of apoptosis.

AIM: To detect the effect of sepsis on hepatic,renal functions and corresponding enzymes in intestine of mice,and to explore the role of leptin in acute inflammation.METHODS: A mice model of sepsis was made by cecum ligation and perforation,and 96-well spectrophotometry was used to detecte the levels of alanine aminotransferase(ALT),uric acid(UA) and activities of myeloperoxidase(MPO),glutathin-S-transferase(GST),xanthine oxidase(XOD),superoxide dismutase(SOD) in serum and intestinal homogenized fluids,respectively.Hematoxylin-eosin staining was used simultaneously to check the histopathologic changes of intestine.RESULTS: Compared with sham group(330.12 ?mol/L?94.15 ?mol/L),serum UA level(521.92 ?mol/L?91.86 ?mol/L) at 6 h after sepsis was significantly higher.12 h after sepsis,both serum ALT(83.55 U/L?40.44 U/L) and UA(474.03 ?mol/L?75.22 ?mol/L) were significantly higher than those in sham group(66.23 U/L?16.80 U/L and 320.95 ?mol/L?99.14 ?mol/L,respectively).12 h after leptin injection(0.1 mg/kg,ip) or indomethacin injection(2 mg/kg,ip),the serum ALT and UA levels significantly decreased(vs sepsis group,P

Objective:To investigate the effect of cPLA 2(cytosolic PLA 2, 85-kD PLA 2) on the signaling pathways of IL-1?、IL-6 release in LPS induced Hela cells.Methods:①LPS stimulated Hela cells were exposed to the complex of cPLA 2 initiation site-directed antisense oligonucleotide and liposome in the indicated doses and time, and then,analyzed the changes of cPLA 2 both in mRNA and in protein by RT-PCR and Western blot.②At the same time, the supernatants were collected to detect the level of IL-1? and IL-6 under radioimmunoassay(RIA).Results:①The transfection led to a dose-dependent decrease in cPLA 2 protein, while the change of mRNA is not notable.②The levels of IL-1?、IL-6 in the transfected cells were reduced following the repression of cPLA 2 compared with that of LPS added only.Conclusion:cPLA 2 probably plays an important role in the signaling mechanisms on IL-1?、IL-6 release in LPS-induced Hela cell.
