RESUMO

Objective:To analyze the curative effect and prognostic factors for comprehensive therapy in patients with high-grade glioma. Methods:Patients with high-grade glioma (WHO grade Ⅲ,grade Ⅳ) were chosen from July 2008 to May 2016 in the Hunan Cancer Hospital, and a retrospective analysis was performed in 64 patients with complete follow-up data. Results:The follow-up time was 3-111 (median 29.5) months, the median overall survival time was 36.00 (95% CI 22.85 to 49.16) months, the median progression-free survival time (PFS) was 21.00 (95% CI 9.72 to 32.28) months,The 1-year,2-year,3-year and 5-year survival rates of high-grade glioma patients were 87.50%,56.25%,40.63% and 17.19%,respectively.The univariate analysis of Log-Rank test and the Cox regression model analysis showed that the prognostic factors related to the prognosis of high-grade glioma patients were pathological grade, resection degree, and concurrent chemo-radiotherapy (P<0.05).Conclusion:The overall survival time,progression-free survival time and the 5-year survival rate of patients with high-grade glioma after comprehensive treatment is partially improved. The factors relevant to the prognosis of patients with high-grade glioma are pathological grade, resection degree, and concurrent chemo-radiotherapy,indicating that the glioma patients (WHO grade Ⅲ) received total resection of the tumor and concurrent chemo-radiotherapy have better clinical effect.

RESUMO

Objective To explore if the expression of follicle-stimulating hormone ( FSH ) and its receptor ( FSHR) exist in the rat submandibular gland tissue , which may provide a theoretic evidence for further studying on adjusting function of the FSH in submaxillary gland .Methods Twenty normal SD rats , with a mean body weight of (200 ±20)g, were adopted to collect out the submandibular gland after abdominal cavity anesthesia .The method of in situ hybridization was employed on cellular localization of FSH and FSHR .RT-PCR was used to detect if FSH and its receptor mRNA exist in the rat submandibular gland .RNA was extracted from the submandibular gland , then FSH and the core cDNA of its receptor gene were obtained with reverse transcription polymerase chain reaction and analyzed on its sequence . Results The epithelial cells of serous acinus and granular convoluted tubule in submaxillary gland of rats had FSH and FSHR mRNA hybridized signals , which were detected in cytoplasm with negative nuclei .Specific bands of FSH cDNA and FSHR cDNA were identical to be 193bp and 413bp respectively. Conclusion FSH and FSHR can be synthesized by the epithelial cells of serous acinus and granular convoluted tubule in submaxillary gland of rats , thus this study further proves that the submandibular gland is the target organ of FSH .

RESUMO

Three pSIREN-siRNA plasmids were constructed using a pSIREN-RetroQ vector to silence the expression of muhidrug resistance-associated protein (MRP1) gene, and subsequently characterized by restriction endonuclease digestion and DNA sequencing. A truncated MRP1 and a full-length MRP1 were cloned into pEGFP-N2 and PeDNA3.1 respectively as pEGFP-MRP1T and pcDNA-MRP1. The plasmid pEGFP-MRP1T was co-transferred with each of the three pSIREN-siRNAs into HEK293 cells for MRP1T-GFP targeted silencing, and pSIREN-siRNA1 was used as the negative control, pSIREN-siRNA2 and pSIREN-siRNA3 appeared to be more effective to silence MRP1T-GFP compared to pSIREN-siRNA1 as shown by fluorescence microscopy. For the silencing of full-length MRP1 expression, HEK293 ceils were co-transferred with pcDNA-MRP1 and either of the three pSIREN-siRNAs, then subjected for Western blot analysis and MTT assays, pSIREN-siRNA2 and pSIREN-siRNA3 were able to inhibit the expression of 190 kD MRP1, but not pSIREN-siRNA1. The MDR of MRP1-transfected HEK293 ceils was abolished with pSIREN-siRNA2 or pSIREN-siRNA3 transfections. RNA secondary structure predictions demonstrated that the mRNA local free energy (△G) of the siRNA1 targeted sequence was lower, as the GC content and Tm value of siRNA1 were higher than those of siRNA2 and siRNA3. These data suggest that the local structure siRNAs and target mRNA may influence the silencing efficiency of MRP1 expression.

RESUMO

Objective To investigate the distribution of nerve growth factor(NGF) and epidermal growth factor(EGF) and their colocalization with follicle-stimulating hormone(FSH) in rat submaxilary glands, and to study the effects of FSH on the secretion of NGF and EGF by submandibular gland tissue in vitro. Methods Immunohistochemical colocalization method was used in the experiment.Submandibular tissues of rats were incubated in vitro and different concentration of FSH was added.Enzyme link immunoassay was used to measure EGF and NGF in supernatant. Results The serous glandular cells, granular convoluted epithelial cells and all other duct epithelial cells in the submandibular gland showed FSH,NGF or EGF positive immunoreactivity. The positive substance was distributed in the cytoplasm with negative nuclei. Both FSH and NGF or EGF positive products were co-located in rat submandibular gland. When the concentration of FSH was more than 10-5-10-6 IU/L,the secretion of EGF and NGF lessened with the decreased concentration of FSH;when the concentration of FSH was less than 10-5-10-6 IU/L,the secretion of EGF and NGF increased with the decreased concentration of FSH.Conclusion FSH has the same bidirectional regulation effects on EGF and NGF in vitro.FSH may regulate the function of endocrine of rat submandibular gland.

RESUMO

In the present experiments, the effect of hypertonic NaCl infusion on the renal ultrastructure in rabbits with hypovolemia was examined by electron microscopy. Rabbits were anasthetized with pentobarbital sodium and submitted to control bleeding and held at a mean arterial pressure of 40 mm Hg for 45 minutes. They were then given 6 ml/kg i. v. of 1) isotonic NaCl (300 mosm/L; 2) hypertonic NaCl (2400mosm/L). Only animals receiving hypertonic NaCl had immediate and permanent resto ration of mean arterial pressure and nearly intact ultrastructure of nephron. However, the animals with isotonic NaCl had an ischemic changes of nephron ultrastructure and unrestored mean arterial pressure.

RESUMO

An ultrastructural analysis of distal nephron has been investigated using morphometric method for rats with hyperthyroidism and hypothyroidism. The cells analysed were distal convoluted tubule cells (DCT cells), distal straight tubule cells (DST cells), intercalated cells (I cells) and principal cells (P cells) of collecting duct. The cells were sampled from superficial cortex. The results revealed that in DCT, DST and P cells the profile area of cells, the surface density of plasma membrane (Sv), the boundary length of cell surface [B(A)], the volume density (Vv) of mitochondria, and the length of microvilli were significantly decreased in the rat with hypothyroidism. However, in the rat with hyperthyroidism there were an obvious increase of profile area of cell and Vv of mitochondria in I cells. The results suggest that thyroid hormone is an important factor for maintaining the normal cellular structure in the distal nephron. The morphometry shows that the changes of DCT, DST and P cells are distinct in the rat with hypothyroidism, and the ultrastructural changes of I cell are obvious in the rat with hyperthyroidism.
