ObjectiveTo explore the role of miR-34a in the proliferation, migration and invasion of hepatocellular carcinoma (HCC) cells.MethodsHepG2 and Huh-7 cells were transfected with miR-34a constructs by using lipofection. qPCR was applied to detect the expression of miR-34a. Cell proliferation assay, colony formation assay, cell migration and invasion assay were used to elucidate the invasive growth of transfected and un-transfected cells. The subcutaneous xenograft in nude mice was used to verified the anti-tumor effect of miR-34a in vivo. TargetScan, miR-WALK and luciferase assay were performed to explore the target of miR-34a in HCC.ResultsCompared with the overexpression control group and blank group, the expression of miR-34a in the miR-34a overexpression group cells were (2.727±0.173) and (4.042±0.104), respectively (P<0.05). The levels of miR-34a in overexpression control and blank groups were (0.003±0.030) and (0.005±0.027) (P>0.05). Compared with the overexpression control groups, cell proliferation, colony formation, migration and invasion of HCC cells overexpressing miR34a were all impeded with statistically significant differences (P<0.05). In vivo, tumor weight and tumor volume were significantly reduced in the miR-34a treatment group. The results of luciferase reporter gene indicated that c-Met was the direct target of miR-34a in HCC.ConclusionsUp-regulation of miR-34a in hepatocellular carcinoma can inhibit the proliferation, migration and invasion of HCC cells, and its repression may be benefited by declining c-Met signaling in HCC.

RESUMO

Chimeric antigen receptor T-cell immunotherapy (CAR-T) has gradually become a new therapeutic strategy for malignant tumor following more than 30 years of development. CAR-T therapy has achieved remarkable success in treating refractory hematological malignancies but less encouraging in solid tumors. There are many difficulties and challenges in the treatment of solid tumors such as low infiltration, immunosuppressive microenvironment, off-target effect. To overcome the limitation of CAR-T cells in the application of solid tumors, many new strategies were explored. Here, we will give a brief review on the recent progress about improvement approaches to improve the effectiveness of CAR-T cells in the treatment of solid tumors.

RESUMO

Primary liver cancer is the third fatal tumor in China, and 85% of them are Hepatocellular Carcinoma (HCC). Liver cancer progresses quickly and is easy to metastasize, which brings great difficulties for treatment. In recent years, great breakthroughs have been made in the treatment of advanced HCC by chimeric antigen receptor T cell (CAR-T) immunotherapy and flora treatment. This article reviewed the current status of cellular immunotherapy and intestinal probiotics therapy for HCC, aiming at providing new ideas for clinical treatment of liver carcinoma.

RESUMO

Faecalibacterium prausnitzii (F.prausnitzii) is the most abundant intestinal microorganisms in the intestine of healthy adults and has been considered as a biological indicator of human health. In recent years, many studies have shown that F. prausnitzii abundance is reduced in different intestinal diseases. Further more, scholars found F.prausnitzii plays an important role in intestinal mucosal barrier protection and inflammation suppression in intestinal diseases. Therefore, it has been proposed that the abundance of F. prausnitzii may serve as a biomarker to assist in gut diseases diagnostics, providing a new idea for the clinical diagnosis and treatment of intestinal diseases. This article reviews the research progress of F.prausnitziiin in intestinal diseases.

<p><b>OBJECTIVE</b>To compare the diagnostic value study the technique of ultrasonography, magnetic resonance cholangiopancreatography (MRCP) and endoscopic retrograde cholangiopancreatography (ERCP) in common duct stones.</p><p><b>METHODS</b>Three hundreds and eighty-four patients suspected common duct stones from August 2005 to October 2007 were undergone abdominal ultrasonography, MRCP and ERCP.</p><p><b>RESULTS</b>There was stone in common duct in 370 and no stones in 14 of 384 patients. Ultrasonography indicated stones 268 cases, 8 false positive result was among them. MRCP diagnosed stones in 362 cases and false positive result in 6 cases, ERCP diagnosed stones 370 cases. The diagnostic accuracy of ultrasonography, MRCP and ERCP was 70.3% (260/370), 96.2% (356/370) and 100% respectively.</p><p><b>CONCLUSIONS</b>The diagnostic accuracy of ultrasonography for common duct stone was little higher, US should be used as a primary checking method. There was higher concordance between MRCP and ERCP for common duct stone. Combination of MRCP and ERCP can improve diagnostic accuracy of common duct stone.</p>
