RESUMO

<p><b>OBJECTIVE</b>To explore the isolation, purification and expansion of human umbilical cord blood mesenchymal stem cells (MSCs) into neuron-like cells in vitro.</p><p><b>METHODS</b>Human cord blood samples were obtained sterilely with 20 U/ml preservative-free heparin. MSCs were isolated by lymphocyte separation medium (density 1.077 g/ml), and purified and expanded with Mesencult trade mark medium. The surface antigen expression of MSCs was detected by flow cytometry. The passage 2, 5 and 8 of the expanded MSCs were induced to differentiate to neuron-like cells. Specific markers and structures were detected by immunohistochemistry and histochemistry methods.</p><p><b>RESULTS</b>The number of MSCs increased two- to three-fold with each expanded passage. 6.6 x 10(5) primary MSCs were expanded ten passages to reach a number of 9.9 x 10(8), and was increased about 1.5 x 10(3)-fold. Flow cytometry showed that MSCs did not express antigens CD(34), CD(11a) and CD(11b), but expressed strongly CD(29) and weakly CD(71), which was identical to human bone marrow-derived MSCs. 70% cells exhibited typical neuron-like phenotype after induction. Immunohistochemistry staining showed that all of the induced different-passage MSCs expressed neurofilament (NF) and neuron-specific enolase (NSE). Special Nissl body was found by histochemistry.</p><p><b>CONCLUSION</b>MSCs in human umbilical cord blood can expand in vitro and differentiate into non-mesenchymal cells.</p>

<p><b>OBJECTIVES</b>To study the tissue distribution of nodularin in mice and the cellular location of nodularin in the target organs.</p><p><b>METHODS</b>The nodularin was labeled with radioactive isotope (125)I and then was given to mice via oral, intraperitoneal and intravenous administration. The distribution of nodularin in target organs and the cellular location of nodularin were studied by radioisotope and autoradiography techniques.</p><p><b>RESULTS</b>The radioisotope study results showed that nodularin was mainly distributed in the kidney and liver in mice. Further autoradiography study indicated that nodularin was distributed in the renal cell nuclei and liver cell nuclei.</p><p><b>CONCLUSION</b>The kidney and liver are the two main target organs for nodularin in mice.</p>

RESUMO

<p><b>OBJECTIVE</b>To assess Microcystin LR (MCLR)-induced acute toxic effects in male Sprague-Dawley rats.</p><p><b>METHODS</b>The rats were injected with MCLR intraperitoneally in different doses for different days. The organs and serum with rats were collected at 1 and 7 days after injection, and 7 days after the final injection (total 14 days). Pathological and enzymatic changes were observed.</p><p><b>RESULTS</b>The rats injected with 122 microg/kg MCLR showed myocardial cells damage including pyknosis, plasma dissolve and myofibrilla (pls check with dictionary) necrosis in the heart muscles after 24 hours. At the same time, the activities of serum glutamate-oxaloacetate transaminase (GOT), lactate dehydrogenase (LDH) and creatine phosphonase (CPK) were higher than these in the other groups (P < 0.01). The kidney was also damaged, kidney cell degeneration, and the increase of blood creatine (BCr) and blood urea nitrogen (BUN) were also seen. In liver pathological study, liver cell hemorrhage, degeneration and/or necrosis was observed. In serum the activities of glutamate-pyruvate transaminase (GPT), alkaline phosphatase (LDH) and GOT were higher than these in the other groups (P < 0.01).</p><p><b>CONCLUSION</b>These results suggested that MCLR can injure the heart, kidney and the liver in SD rats, and there is a dose-response relationship between MCLR and the toxic effect.</p>

RESUMO

Objective To find out the appropriate bedbound period after Coronary Arteriography (CAG) in order to reduce the complications and uncomfort caused by prolonged Bedbound period and affected limb immobilization.Methods 121 cases of CAG patients were randomly divided into the reduced period group (experiment group) and the conventional group (the control group),two ways of nursing and observation respectively.Result the rate of complication occurred in the experiment group much less than that of in the control group and the bedbound period were different as well.The above items showed statistics significance (P<0.005~0.05).Conclusion The reduced bedbound period decreases the suffering of CAG patients and provides the reliable clinical evidence for proper nursing approach in this field.

RESUMO

This paper focus on conductive education approach during exercise therapy of cerbral palsy childrens, which emphasis the children active participation in the exercises and their phsycological reactions. Its main feature is using the rythmicl intention, task analysis and task series as the tools to perform the goal - directed activities in group setting and carry it out in the children' s daily life. The current studies on the motor control and motor learning are reviwed in this paper as well.

Purpose:The current study analyzed the polymorp hisms of DNA repair gene XRCC1 and relationship between genetic variations and s usceptibility to lung cancer Methods:A case-control study of 111 patients with lung cancer a nd 210 normal residents as controls was conducted to detect XRCC1 polymorphisms at Arg399Gln loci . Genotypes were analyzed by PCR-based restriction fragment le ngth polymorphism(RLFP) techniques. Results:XRCC1 codon 399 heterozygous genotype Arg/Gln might hav e weaker protection effect on squamous cell carcinoma and reduce the risk of lun g cancer on smokers. Homozygous genotype Gln/Gln of XRCC1 codon 399 might increa se the risk of lung cancer and have synergistic effect with smoking. Conclusions:The results demonstrated that genetic polymorphism of XRCC1 DNA repair gene might contribute to the susceptibility to lung cancer a nd have synergistic effect with smoking.
