RESUMO

OBJECTIVE To investigate the cytotoxicity of cyflumetofen for SH-SY5Y cells and the mechanism. METHODS SH-SY5Y cells treated with cyflumetofen 0.03, 0.06, 0.125, 0.25, 0.5, 1, 2, 2.6, 4, 6, 8 and 16 mmol·L-1 for 48 h. Cell survival was measured with MTT assay. The reactive oxygen species (ROS) was determined with the DCFH- DA probe, and mitochondrial membrane potential (MMP) was detected by JC-1 staining. The morphological changes in cell nuclei were observed with Hoechst33258 staining. Cell cycle and apoptosis were determined by flow cytometry. The protein levels of phosphorylated Jun Kinase (p-JNK) and p-P38 were measured by Western blotting. RESULTS Compared with solvent (DMSO) control group, cyflumetofen (≥0.06 mmol · L- 1) inhibited the proliferation of SH- SY5Y cells obviously (P<0.05), and the IC50 was 2.6 mmol·L-1. MMP declined and ROS levels increased significantly in cyflumetofen 1, 2, 4 and 6 mmol·L- 1 groups (P<0.01). Cyflumetofen 2, 4 and 6 mmol·L- 1 induced nucleic accumulation, nuclear shrinkage and disintegration in SH-SY5Y cells. Apoptosis rates of cyflu? metofen 2, 4 and 6 mmol·L- 1 groups increased from (0.7±0.1)% in DMSO control group to (6.7±0.1)%, (72.4±8.6)% and (90.7±3.2)% (P<0.01). Cyflumetofen 4 and 6 mmol·L- 1 induced G1 phase cell cycle arrest (P<0.01). In addition, Western blotting showed that cyflumetofen 4 and 6 mmol·L-1 up-regulated the expression of p-JNK (P<0.01), while the level of p-P38 in SH-SY5Y cells was increased in cyflumetofen 6 mmol · L- 1 group (P<0.01). CONCLUSION Cyflumetofen induces cell damage, apoptosis and G1 phase cell cycle arrest in SH- SY5Y cells. The mechanism may be associated with oxidative damage, and activation of P38 and JNK stress-response pathways.

RESUMO

Objective To explore the inhibitor of growth family member 4 (ING4) and hypoxia inducible factor-1α(HIF-1α)expression in colorectal cancer and the prognostic significance. Methods Immunohistochemistry was used to detect the ING4 and HIF-1α expression in 133 cases of colorectal cancer tissues and 76 cases of normal rectal tissues. Survival analysis was performed on the following data. Results ING4 in colorectal cancer tissues with positive rate (53.4%) was significantly lower than normal rectal tissue (85.5%) and the difference was statistically significant (P < 0.01); HIF-1α in colorectal cancer tissues with positive rate (69.9%) is higher than normal rectal tissue (42.1%), and the difference was statistically significant (P < 0.01); ING4 and HIF-1αexpression was related with tumor differentiation, Dukes stage and lymph node metastasis (P < 0.05); colorectal cancer tissues ING4 and HIF-1α expression was negatively correlated (r = -0.317, P < 0.001); By multivariate analysis, tumor differentiation, Dukes stage, lymph node metastasis, ING4 expression of HIF-1α expression has independent prognostic significance. Conclusion ING4 and HIF-1α may be involved in the occurrence and development of colorectal cancer , and combined detection could help determine the prognosis of colorectal cancer.

RESUMO

Objective To investigate the clinical significance of treadmill exercise test in the diagnosis of coronary artery disease. Methods137 patients with chest pain,chest distress and palmus but without the change of ECG were selected.Treadmill exercise test was used. Results78 patients were positive and the positive rate was 56.9%,while the negative rate was 43.1%. ConclusionTreadmill exercise test could improve the diagnosis rate and sensitivity rate of coronary artery disease.It plays an important clinical significance in the diagnosis and treatment of coronary artery disease.

RESUMO

Objective To investigate the effect of 24h Holter monitoring in patients with asymptomatic myocardial ischemia diagnosis. Methods 80 patients with asymptomatic myocardial ischemia in coronary heart disease(CHD)were detected by 24h Holter monitoring,and the results were recorded. Results In 80 patients of CHD myocardial ischemia occurred in 75 cases,accounting for 93.75%,and ischemic ST segment depression appeared total of 824 matrix times,and in each case there were 1 to 23 matrix times,an average of 8.3,and the average ST segment depression was(0.15±0.06)mV; Most of asymptomatic myocardial ischemia occurred in the daytime,and the peak onset occured in 6 to 12 hours; High and low heart rate was positively correlated with the degree of ischemia,and faster heart rate was,more obvious ST segment depression was down,more serious myocardial ischemia was induced. Conclusion 24h Holter monitoring of asymptomatic myocardial ischemia condition assessment and early prevention is important.
