Objective: To understand the infection status of Enterovirus (EV) in cases of acute respiratory infections (ARIs) in Luohe City, Henan Province from 2017 to 2021, and analyze the prevalence and type composition of EV in ARIs. Methods: From October 2017 to May 2021, pharyngeal swab samples were collected from 1 828 patients with ARIs in Luohe Central Hospital and the clinical epidemiological data of these cases were also collected. EV-positive samples were identified by Quantitative Real-time Polymerase Chain Reaction (qPCR). The 5'-untranslated region (5'UTR) was amplified by Reverse Transcription-Polymerase Chain Reaction (RT-PCR). The results of 5'UTR region were initially typed by Enterovirus Genotyping Tool Version 1.0. Based on the typing results, the full-length of VP1 region was amplified by RT-PCR. The EV typing was identified again by VP1 region. Results: Among 1 828 cases of ARIs, 56.7% (1 036) were males. The median (Q1, Q3) age was about 3 (1, 5) years. Patients under 5 years old accounted for 71.6% (1 309 cases). Among all cases, a total of 71 EV-positive samples were identified by qPCR, with a detection rate of 3.88% (71/1 828). The EV detection rates for men and women were 3.28% (34/1 036) and 4.67% (37/792), without statistically significant differences (χ2=2.32, P=0.14). The EV detection rates for 2 to <6 years, 6 months to <2 years, 6 to <10 years, and <6 months were 6.29% (48/763), 3.00% (18/600), 2.52% (4/159), and 1.67% (1/60) (χ2=27.91, P<0.001). The EV detection rate was 0.92% (3/326) in autumn and winter of 2017. The EV detection rates were 1.18% (6/508), 2.47% (12/485) and 8.31% (34/409) in each year from 2018 to 2020, with an increasing trend year by year(χ2trend=29.76, P<0.001). The main prevalent seasons were summer and autumn. The detection rate in spring of 2021 was 4.00% (4/100). A total of 12 types were identified and classified as CVA2, CVA4, CVA5, CVA6, CVA10, CVB3, CVB5, E5, E11, E30, PV-1, and EV-D68. The types of CVA2, CVA10, CVA6, and CVB3 were the dominant phenotypes. In 59 sample of EV typing, the main clinical manifestation was upper respiratory tract infection (36/59, 61.01%). The dominant types detected in upper respiratory tract infections were CVA10 (10/36, 27.78%), CVA6 (9/36, 25.00%) and CVB3 (8/36, 22.22%). The dominant type detected in lower respiratory tract infections was CVA2 (7/19, 36.84%). Conclusion: In Luohe City, Henan Province from 2017 to 2021, EV infection in ARIs cases has clear seasonal and age-specific patterns, and the dominant types of upper and lower respiratory tract infections are different.

OBJECTIVE@#To investigate the inhibitory effects of levofloxacin (LEV) combined with cellulase against bacille CalmetteGuerin (BCG) biofilms in vitro.@*METHODS@#The mature growth cycle of BCG biofilms was determined using the XTT method and crystal violet staining. BCG planktonic bacteria and BCG biofilms were treated with different concentrations of LEV and cellulose alone or jointly, and the changes in biofilm biomass were quantified with crystal violet staining. The mature BCG biofilm was then treated with cellulase alone for 24 h, and after staining with SYTO 9 and Calcofluor White Stain, the number of viable bacteria and the change in cellulose content in the biofilm were observed with confocal laser scanning microscopy. The structural changes of the treated biofilm were observed under scanning electron microscopy.@*RESULTS@#The MIC, MBC and MBEC values of LEV determined by broth microdilution method were 4 μg/mL, 8 μg/mL and 1024 μg/mL, respectively. The combined treatment with 1/4×MIC LEV and 2.56, 5.12 or 10.24 U/mL cellulase resulted in a significant reduction in biofilm biomass (P < 0.001). Cellulase treatments at the concentrations of 10.24, 5.12 and 2.56 U/mL all produced significant dispersion effects on mature BCG biofilms (P < 0.001).@*CONCLUSION@#LEV combined with cellulose can effectively eradicate BCG biofilm infections, suggesting the potential of glycoside hydrolase therapy for improving the efficacy of antibiotics against biofilmassociated infections caused by Mycobacterium tuberculosis.

To compare the power of dyslipidemia diagnosis by different sets of cut points in the prediction of cardiovascular metabolic risk factors and identify the appropriate cut points for the diagnosis of dyslipidemia in children and adolescents in China. Data were obtained from the baseline survey of 'School-based Cardiovascular and Bone Health Promotion Program' in Beijing in 2017. Dyslipidemia was diagnosed by using two set of cut points. Receiver operating characteristic curve analysis was conducted to assess the power of dyslipidemia diagnosis by the two set of cut points to predict the prevalence of hypertension, obesity, high fat mass percentage and impaired fasting glucose. A total of 14 390 children and adolescents were in included in the study. The prevalence rates of high TC, high LDL-C, low HDL-C, and high TG in the participants were 2.7, 2.7, 14.4, and 3.7 according to 'Chinese Reference Standard', and 5.0, 3.7, 13.3, and 3.5 according to 'China Expert Consensus'. Low HDL-C and high TG defined by the 'Chinese Reference Standard' had better performance for the prediction of high fat mass percentage and obesity in boys, but worse performance in girls (<0.001). Using 'China Reference Standard' can increase the true positive rate in the prediction of obesity or high fat mass percentage in boys, and reduce the false positive rate in girls. The cut points for the diagnosis of dyslipidemia in Chinese children and adolescents need to be further validated by using national representative sample and in longitudinal study.

OBJECTIVE@#To study the value of body fat mass measured by bioelectrical impedance analysis (BIA) in predicting abnormal blood pressure and abnormal glucose metabolism in children.@*METHODS@#Stratified cluster sampling was used to select the students aged 6-16 years, and a questionnaire survey and physical examination were performed. The BIA apparatus was used to measure body fat mass. Body mass index (BMI), body fat mass index (FMI), and fat mass percentage (FMP) were calculated. Fasting blood glucose level were measured.@*RESULTS@#A total of 14 293 children were enrolled, among whom boys accounted for 49.89%. In boys and girls, the percentile values (P, P, P, P, P, P, P, P) of FMI and FMP fitted by the LMS method were taken as the cut-off values. Based on the receiver operating characteristic curve analysis, the P values with a better value in predicting abnormal blood pressure and blood glucose metabolism were selected as the cut-off values for excessive body fat. When FMI or FMP was controlled below P, the incidence of abnormal blood pressure or abnormal glucose metabolism may be decreased in 8.25%-43.24% of the children.@*CONCLUSIONS@#The evaluation of obesity based on FMI and FMP has a certain value in screening for hypertension and hyperglycemia in children, which can be further verified in the future prevention and treatment of obesity and related chronic diseases in children.

The aim of this study was to identify some biomarkers for predicting lymph node metastasis and prognosis of human epidermal growth factor receptor 2 (Her-2)-positive breast cancer (BC). We analyzed correlations between microRNAs (miRNAs) and the prognosis of patients with BC based on data collected from The Cancer Genome Atlas (TCGA) database. The expression levels of miR-455, miR-143, and miR-99a were measured in clinical samples of Her-2-positive BC patients with different degrees of lymph node metastasis. We investigated the impacts of overexpressed miR-455 on the proliferation and invasiveness of MDA-MB-453 cells and measured its effects on the expression of long non-coding RNA (lncRNA) metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) by quantitative real-time polymerase chain reaction (qRT-PCR). The expression of miR-455 was significantly and positively correlated to the prognosis and overall survival (OS) of the BC (P=0.028), according to TCGA information. The expression level of miR-455 was positively correlated with OS and relapse-free survival (RFS) of patients with Her-2-positive BC, and was negatively correlated with the number of metastatic lymph nodes (P<0.05). Transwell assay suggested that MDA-MB-453 cells became much less invasive (P<0.01) after being transfected with miR-455 mimics. During the qRT-PCR, the expression level of MALAT1 declined significantly after transfection (P<0.01). Overexpressed miR-455 significantly inhibited the proliferation and migration of MDA-MB-453 cells and the expression of MALAT1. We conclude that miR-455 may be a useful potential biomarker for forecasting lymph node metastasis and the prognosis of Her-2-positive BC patients. miR-455 may play an important role in lymph node metastasis of BC by interacting with MALAT1.

To analyze the frequency and spectrum of thalassemia mutations in amniotic fluid samples collected from Han and Li people in Hainan province of China. Methods: We carried out a retrospective analysis on prenatal diagnosis of amniotic fluid samples collected from pregnant women who may have next generation with high risks of medium or severe thalassemia between 2005 and 2016. Diverse fetal thalassemia genotypes and mutated alleles in Han and Li people were analyzed and cmpared. Results: We examined 536 amniotic fluid samples from Han people and 588 from Li people, among which 406 Han and 500 Li samples were found to carry at least one thalassemia gene mutation, with a detection rate of 75.75% and 85.03%, respectively. Among all - and β-thalassemia mutant alleles detected, the most frequently found mutations in Han and Li samples were SEA-type of -thalassemia and 41/42 (-CTTT) of β-thalassemia, respectively. A total of 75 severe thalassemia cases were identified in Han samples and 53 in Li samples. In most of these severe cases, parents chose to terminate pregnancy after being informed of thalassemia-related risks. Conclusions: The thalassemia mutations shows ethnic and area specificity, and that prenatal diagnosis for high-risk thalassemia carrier pregnant women is an efficient approach to prevent and control the occurrence of severe thalassemia in the high-prevalence areas.

Methods:Ten untrained healthy subjects were asked to image the movement guided by the picture accompanied with regular rhythm. The common spatial pattern feature extraction algorithm and Fisher classifier were used for classification, and compared with the traditional paradigm and the motor imagery paradigm accompanying with irregular rhythm. Results:More desynchronization was found as motor imagery with regular rhythm, while the classification accuracy improved. Conclusion:Motor imagery with regular rhythm gives a new idea for active rehabilitation training based on brain-computer interface.

AIM To study the influence and action mechanism of effective components in compatible solution of Sparganii Rhizoma-Curcumae Rhizoma on pelvic adhesions of model rats with chronic pelvic inflammatory disease.METHODS Laboratory rats were randomly assigned into six groups,a sham-operated group,a model group,a positive Fukeqianjin tablets group,compatible solution groups (40 g/kg high-dose group,20 g/kg medi um-dose group,and 10 g/kg low-dose group).Except rats of the sham-operated group,those in the other groups were induced into models of chronic pelvic inflammatory disease by mechanical bacterial strain implantation,after which a 20 d corresponding drug administration by oral gavage for each group was initiated.After completion of the last drug administration,Verco criterion was taken as the reference to evaluate pelvic adhesions,HE staining was performed on uterine tissues for pathological scoring,ELASA method was applied to measuring serum IL-1β and TNF-α levels,and Western blot method was used to detect the expression levels of proteins FGF-2 and IGF-1 in uterine tissues.RESULTS Compared with the model group,the high-dose compatible solution group had a much sharper fall in Verco scores (P < 0.05),and remarkably decreased serum IL-1β and TNF-α levels (P < 0.01);both high-dose and medium-dose groups manifested with notably alleviated degenerative necrosis and lowered scores of chronic inflammation infiltration and epithelial hyperproliferation (P < 0.01 or P < 0.05),and a distinct reduction in the expression of proteins FGF-2 and IGF-1 (P < 0.05,P < 0.01).CONCLUSION The significant improvement of the chronic pelvic inflammatory disease in model rats due to the effective components in the compatibile solution of Sparganii Rhizoma-Curcumae Rhizoma may be associated with inhibited release of inflammatory mediators IL-1β and TNF-α,and the reduced expression of proteins FGF-2 and IGF-1 as well.

Objective To explore the effect of informationized time tracking management mode on the speed of treatment of acute ischemic stroke (AIS). Methods From Aug. 2016 to Jun. 2018, a total of 475 AIS patients receiving intravenous thrombolysis and/or intravascular treatment in Stroke Center of Changhai Hospital of Navy Medical University (Second Military Medical University) were divided into time tracking group (n=291) and non-time tracking group (n=184). The time indicators were compared between the two groups, including onset-to-door time (ODT), door-to-imaging time (DIT), imaging-to-needle time (INT), onset-to-needle time (ONT), imaging-to-puncture time (IPT), onset-to-puncture time (OPT), door-to-needle time (DNT) and door-to-puncture time (DPT). The correlations between ODT and DNT, and ODT and DPT were analyzed using Spearman correlation analysis in the non-time tracking group and the time tracking group. Results The DNT of the patients with intravenous thrombolysis was significantly shorter in the time tracking group than that in the non-time tracking group (27 [23, 36] min vs 33 [23, 43] min, Z=2.123, P<0.05). There was a significant negative correlation between ODT and DNT of the patients with intravenous thrombolysis in the non-time tracking group (r=–0.293, P=0.001), while there was no correlation in the time tracking group (r=–0.034, P=0.723). The ODT and OPT of the patients with endovascular treatment were significantly longer, and DIT was significantly shorter in the time tracking group than those in the non-time tracking group (246 [113, 431] min vs 166 [82, 299] min, 342 [211, 519] min vs 277 [142, 406] min and 18 [14, 23] min vs 26 [16, 34] min; Z=2.667, 2.182 and 4.077, all P<0.05). There was a significant negative correlation between ODT and DPT of the patients with endovascular treatment in the non-time tracking group (r=–0.234, P=0.024), while there was no correlation between ODT and DPT in the time tracking group. (r=–0.056, P=0.424). Conclusion The informationized time tracking management mode can improve the treatment efficiency of acute ischemic stroke and eliminate the influence of ODT on DNT and DPT.

Objective To explore the value of intelligent early warning system in pre- and in-hospital connection in the treatment of stroke. Methods The clinical data of the patients with suspected stroke, who received treatment and prehospital warning with intelligent early warning system in Stroke Center of Changhai Hospital of Navy Medical University (Second Military Medical University) between Nov. 2017 and Jun. 2018, were retrospectively analyzed. Pre-hospital warning was defined as 120 alerting stroke teams using software before transferring patients to the target hospital. According to the in-hospital treatment methods, the patients were divided into non-stroke group, conservative treatment group, thrombolysis group, thrombectomy group, bridging group and cerebral hemorrhage group. The efficiency in each link of the intelligent early warning system, the ability of emergency doctor diagnosing stroke, the ability of emergency doctor diagnosing severe stroke using gaze-face arm speech time (G-FAST) scale, the speed of first aid response of the attending physician and the door-in-to-door-out (DIDO) time in primary stroke center were evaluated in each group. Results A total of 248 patients were included in this study, including 24 non-stroke patients and 224 stroke patients (101 patients in the conservative treatment group, 23 patients in the thrombolysis group, 32 patients in the thrombectomy group, 22 patients in the bridging group and 46 patients in the cerebral hemorrhage group). The median primary screening time, early warning time and response time of 248 patients were 28.0 (13.0, 92.5), 11.0 (7.3, 15.3) and 19.0 (13.0, 35.0) s, respectively. The sensitivity, specificity, positive predictive value, negative predictive value and accuracy of 6 pre-hospital emergency physicians diagnosing severe stroke using G-FAST scale were 84%, 71%, 83%, 72% and 79%, respectively. Eight patients with severe stroke underwent whole-process monitoring of pre-hospital treatment, and had a median transport time of 113 (82, 142) min and a median DIDO time of 84 (12, 125) min. Conclusion Intelligent early warning system can realize the automation of data acquisition for transfer time points, making the data more comprehensive and reliable. It is helpful to analyze the efficiency of emergency physicians, drivers, stroke teams and primary stroke center in pre- and in-hospital connection in the treatment of stroke, so as to shorten the pre-hospital treatment time and improve the pre-hospital treatment efficiency.

Objective To evaluate the association between aortic arterial stiffness and target organ damage, including renal function decline and left ventricular diastolic dysfunction among elderly diabetic patients and to provide theoretical basis for early prevention and intervention. Methods Brachial-ankle-artery pulse wave velocity(baPWV)reflecting aortic arterial stiffness was observed in 151 diabetic patients with(n=78)or without hypertension(n=73). Clinical data were collected, cardiac structure and function were assessed by echocardiography. Estimated glomerular filtration rate(eGFR)was calculated by CKD-EPI fomula. Results Of 151 subjects, the average age was(75.05±10.13)years and the average baPWV values was (1 803.85±491.04)mm/s. Patients with both diabetes and hypertension had significantly higher baPWV values[(2006.49± 536.70)mm/s]than that of diabetic patients[(1596.96 ±331.02)mm/s, P<0.01]. Multiple linear regression analysis demonstrated that baPWV was negatively associated with E/A ratio and eGFR(β'=-0.249, -0.225, P<0.05), but positively associated with hypertension(β'=2.008, P<0.05). By applying area under the receiver operator characteristic curve, baPWV had powerful predictive power for renal impairment(AUC=0.695, 95% CI: 0.548-0.842)and left ventricular diastolic dysfunction(AUC=0.687, 95%CI: 0.532-0.842). Conclusion Patients with both diabetes and hypertension had significantly higher baPWV values than that of diabetic patients. Higher baPWV was independently associated with greater burden of sub-clinical disease in renal impairment and left ventricular diastolic dysfunction.

Objective To studythe cephalocaudal relationship ofabdominal aortic bifurcation relative toumbilicus and iliac crest vertex and their correlations with abdominal adipose tissue thickness and age. Methods The vertical distances,cephalocaudal relationship and other related anatomic parameters of aortic bifurcation relative to umbilicus and iliac crest vertex in 108 patientswere measured by consecutive abdominal CT scanning. The correlations of the acquired data with abdominal adipose tissue thickness and age were analyzed using Pearson correlation coefficient. Results Umbilicus was located at cephalad to aortic bifurcation in 67 patients(62.0%), caudal in 34(31.4%)andthe same level in 7(6.5%),with the vertical distance of(4.53 ± 17.51)mm to the aortic bifurcation. No statistically significant relationship was found between abdominal adipose tissue thickness(P>0.05) or age(P>0.05). Iliac crest vertex relative to aortic bifurcationwas located at cephalad,caudal and the same level in 31,71 and 6 patients,taking up 28.7%,65.7%and 5.6%,respectively. Its vertical distance to the bifurcation was(-6.34 ± 14.49)mm,nonrelated with abdominal adipose tissue thickness(P>0.05),but positively correlated with age(P<0.01). The difference in the cephalocaudal relationship of aortic bifurcation relative to umbilicus and iliac crest vertex was statistically significant(P<0.01). Conclusion Compared with iliac crest vertex,umbilicus is an important landmark of locating abdominal aortic terminal occlusion position in vitro because it mostly lies cephalad to aortic bifurcation in the front of the body,not easy to vary with abdominal adipose tissue thickness and age.

Objective To observe the changes of esophageal pressure in patients with achalasia (AC) before and after peroral endoscopic myotomy (POEM), and to explore the value of high-resolution manometry (HRM) in evaluating the efficacy of POEM. Methods 38 cases were diagnosed as achalasia and treated with POEM from August 2015 to November 2016. Upper esophageal sphincter resting pressure (UESP), lower esophageal sphincter resting pressure (LESP), esophageal sphincter 4s integrated relaxation pressure (4sIRP) were detected by HRM before operation, at 1 week, 1 month after operation to evaluate the efficacy. Results A total of 38 patients with AC received POEM. At one week, one month after operation, the UESP were (45.34 ± 26.52) mmHg, (41.27 ± 20.09) mmHg, compared with that of before operation (49.58 ± 26.47) mmHg and the differences were not statistically significant. The differences in LESP, 4sIRP between before operation and at one week, one month after operation were statistically significant, P < 0.05. The differences in UESP, LESP and 4sIRP between 1 week and 1 month after operation were not statistically significant. Conclusion The pressure of lower esophageal sphincter was significantly reduced and the dysphagia was improved. HRM might play an important role in the assessment of AC received POEM.

At one week, one month after operation, the UESP were (45.34 ± 26.52) mmHg, (41.27 ± 20.09) mmHg, compared with that of before operation (49.58 ± 26.47) mmHg and the differences were not statistically significant. The differences in LESP, 4sIRP between before operation and at one week, one month after operation were statistically significant, P < 0.05. The differences in UESP, LESP and 4sIRP between 1 week and 1 month after operation were not statistically significant. Conclusion The pressure of lower esophageal sphincter was significantly reduced and the dysphagia was improved. HRM might play an important role in the assessment of AC received POEM.

Objective To analyze the incidence of gastric cancer in Qidong , Jiangsu Province from 1993 to 2012 and analyze the harmful levels and the clinical characteristics thereof , thus providing bases of implementing the prevention , research and control measures for gastric cancer . Methods The clinical data of gastric cancer from January 1993 to December 2012 for patients with census registration in Qidong were sorted out from the cancer registration center in Qidong .The annual percent change ( APC) model was used to analyze the trend over time for the incidence of gastric cancer .The incidence was standardized by utilizing the standard population age composition based on the nationwide census in 2000 . Results A total of 8 272 new cases of gastric cancer were diagnosed in Qidong from 1993 to 2012, and the median age for these patients was 67.92(57.50-76.00).Among them, 5 269 were male cases and the median age for these male patients was 67.25(57.75-74.75);3 003 were female cases and the median age for these female patients was 69.16(57.00-77.91).The annual average for the crude incidence rate of gastric cancer was 40.37/100 000, and the standardized incidence rate thereof was 25.22.37/100 000.The crude incidence rate of gastric cancer decreased significantly from 1993 to 2002(for the crude incidence rate thereof , APC=-1.59, t=-4.6, P<0.01);the crude incidence rate of gastric cancer increased significantly from 2002 to 2007(for the crude incidence rate thereof , APC=-8.88, t=-5.3, P<0.01).Both the standardized incidence rate of gastric cancer and the crude incidence rate thereof had the same trend for the period from 1993 to 2002 as well as the period from 2002 to 2007.The crude incidence rate of gastric cancer rose with the growth of ages(for male patients, APC=734.8, t=-511.2, P<0.01; for female patients, APC=43.6, t=17.9, P<0.01).The crude incidence rate of gastric cancer reached a climax for both males and females of the age group between 75 and 80. Conclusion The incidence of gastric cancer was relatively high in Qidong from 1993 to 2012;the male and the old-aged groups are high-risk groups .

To observe the protective effects of Lycium barbarum polysaccharides (LBP) on cerebral ischemia reperfusion injury in mice and explore its mechanism. Common carotid artery thread was used to cause middle cerebral artery ischemia, and the thread was taken out after 2 h ischemia to achieve cerebral ischemia reperfusion injury in mice. Therefore, the transient middle cerebral artery occlusion (tMCAO) models were established to observe the effects of LBP (25,50, 100 mg•kg⁻¹) on neurological outcome, infarct size and water contents. HE staining was used to observe its effects on neurocytes of cerebral tissues in mice. Western blotting was used to evaluate the protein expression levels of NF-κB p65. ELISA was used to evaluate the levels of TNF-α, IL-6 and IL-1β in the serum. According to the results, LBP markedly improved neurologic deficits, and decreased infarct size and water contents at 24 h after reperfusion in mice. Pathological section of brain tissues also proved its protective effects on neurocytes. Western blot analysis indicated that LBP markedly down-regulated the protein level of NF-κB p65. ELISA indicated that LBP decreased the levels of TNF-α, IL-6 and IL-1β in the serum 24 h after reperfusion.In conclusion, LBP has protective effects on cerebral ischemia reperfusion injury in mice, and this effect may be associated with inhibiting NF-κB and inflammatory reactions.

Objective To determine the hyperelastic constitutive equation of biological soft tissues and study the mechanical responses during the clamping process of biological tissues. Methods The destructive uniaxial tensile test was performed on fresh porcine liver and the uniaxial tension experiment was simulated in ABAQUS. The hyperelastic constitutive equation of porcine liver was determined by comparing the simulation results with the experimental data. Based on this equation, the sharp teeth-shaped and wave-shaped chucks were used to simulate the clamping process. Results The simulation results of the tensile experiment with the 4th-order Ogden model were in good agreement with the experimental data. The results of tissue clamping simulation showed that stress concentration was more likely to occur when the sharp teeth-shaped chuck was adopted. Conclusions The 4th-order Ogden model can be used to describe the hyperelasticity of porcine liver and determine the relevant para-meters. Using the sharp-shaped chuck is more likely to cause tissue clamping damage, and there is a linear relationship between tissue stress and clamping feed distance. These research findings provide references for the design of surgical clamp.
