Objective: To establish and evaluate a method of enriching bacteriophages in natural water based on ferric trichloride-polyvinylidene fluoride (FeCl3-PVDF)membrane filter. Methods: Based on the principle of flocculation concentration, the method of recovering bacteriophage from water sample was established by using iron ion flocculation combined with membrane filter. The titer of phage was determined by Agar double layer method. The recovery efficiency of phage was detected by phage fluorescence staining and real-time fluorescence PCR reaction. Water samples from different sources were collected for simulation experiment to evaluate the enrichment effect. At the same time, the sewage discharged from hospitals was taken as the actual water sample, and the common clinical drug-resistant bacteria were used as the host indicator bacteria to further analyze the enrichment effect of FeCl3-PVDF membrane filter rapid enrichment method on the bacteriophage in natural water samples. Results: The method of enrichment of bacteriophages in natural water by iron ion concentration 50 mg/L and PVDF membrane filter was established. The recovery rate of this method for bacteriophage was 93%-100%. Under the multi-functional microscope, it was found that the bacteriophage of the enriched water sample increased significantly and the fluorescence value of the enriched water sample determined by the enzyme labeling instrument was about 13 times as high as that before enrichment. After concentration of the actual water samples from the hospital drainage, the positive rate of bacteriophage isolation in the concentrated group and the non-concentrated group was 23% and 4%, and the fluorescence value in the concentrated group was 2-24 times as high as that of the non-concentrated group. Conclusion: The method of FeCl3-PVDF membrane filter is a simple, efficient and rapid method for enriching bacteriophages in different water samples.

Objective To stud)' the nerve repair effect of olanzapine on schizophrenia model rats through its effect on cyclic AMP response element binding protein (CREB)/brain-derived neurotrophic factor (BDNF)/receptor tyrosine kinase receptors B (TrkB) pathway. Methods Total 60 rats were divided into control group, model group, olanzapine low, middle and high dose group. The rats in the model group, olanzapine low, middle and high dose groups were injected intraperitoneally with MK-801[0. 2 mg/(kg-d) ], while the control injected with the same amount of normal saline. The low, middle and high dose olanzapine groups were perfused with olanzapine solution of 0. 5 mg/(kg-d),1. 0 mg/(kg-d) and 1. 5 mg/(kg-d) respectively. The behavior of rats was scored according to ataxia and stereotyped behavior standards, cognitive function and learning ability were evaluated by Moms water maze test, serum tumor necrosis factor-a (TNF-a) and interleukin-6 (IL-6) levels were detected by ELISA method, hippocampal histopathology was observed under microscope, and apoptosis and expression of CREB/BDNF/TrkB pathway related proteins in hippocampus were detected. Results Compared with the control group, the ataxia, the score of stereotyped behavior, the expression of TNF-a, IL-6 and the rate of apoptosis in the model group increased significantly (P < 0 . 01). Compared with the control group, the number of crossing the platform, the time of staying in the target quadrant and the relative expression of CREB, p-CREB, p-TrkB, TrkB and BDNF protein in the model group decreased significantly (P<0. 01), and those in the low and middle dose olanzapine groups decreased significantly (P < 0 . 05). Compared with the model group, the times of crossing the platform and the stay time in the target quadrant increased significantly in the low and middle dose olanzapine groups (P< 0. 05). In the model group and the low dose olanzapine group, the hippocampal cells were swollen obviously, the nucleus was broken and divided, pyknosis, and the tissue aiTangement was disorderly, while the phenomenon of fragmentation and nuclear pyknosis was rarely seen in the middle and high dose olanzapine groups. Conclusion The nerve repair mechanism of olanzapine on schizophrenic model rats is related to improving cognitive impainnent, protecting hippocampal neurons and activating the expression of CREB/BDNF/TrkB signal pathway in rats.

This study aimed to evaluate the ability of rete testis thickness (RTT) and testicular shear wave elastography (SWE) to differentiate obstructive azoospermia (OA) from nonobstructive azoospermia (NOA). We assessed 290 testes of 145 infertile males with azoospermia and 94 testes of 47 healthy volunteers at Shanghai General Hospital (Shanghai, China) between August 2019 and October 2021. The testicular volume (TV), SWE, and RTT were compared among patients with OA and NOA and healthy controls. The diagnostic performances of the three variables were evaluated using the receiver operating characteristic curve. The TV, SWE, and RTT in OA differed significantly from those in NOA (all P ≤ 0.001) but were similar to those in healthy controls. Males with OA and NOA were similar at TVs of 9-11 cm 3 ( P = 0.838), with sensitivity, specificity, Youden index, and area under the curve of 50.0%, 84.2%, 0.34, and 0.662 (95% confidence interval [CI]: 0.502-0.799), respectively, for SWE cut-off of 3.1 kPa; and 94.1%, 79.2%, 0.74, and 0.904 (95% CI: 0.811-0.996), respectively, for RTT cut-off of 1.6 mm. The results showed that RTT performed significantly better than SWE in differentiating OA from NOA in the TV overlap range. In conclusion, ultrasonographic RTT evaluation proved a promising diagnostic approach to differentiate OA from NOA, particularly in the TV overlap range.

Objective:To analyze the health literacy level and influencing factors of Uyghur residents in Moyu County, Xinjiang between 2018 and 2020.Methods:A total of 5 211 and 5 207 Uyghur residents in Moyu County, Xinjiang were sampled using the multi-stage cluster method in 2018 and 2020, respectively. The health culture questionnaire for ethnic minority in Xinjiang was used. The survey included general demographic characteristics, overall health literacy levels, and the three aspects of health literacy levels (health knowledge and concepts, healthy lifestyles and behaviors, and health skills) and six dimensions (scientific health concept, health information literacy, infectious disease prevention and control literacy, chronic disease prevention literacy, safety and first aid literacy, and basic medical literacy). The influencing factors of residents′ health literacy in 2020 were analyzed using χ2 test and multivariate logistic regression. Results:In 2020, overall health literacy and its three aspects among Uyghur residents in Moyu County, Xinjiang were higher than in 2018 (overall 8.10% vs 5.07%, health knowledge and concepts 9.99% vs 7.60%, healthy lifestyle and behavior 6.88% vs 5.33%, health skills 40.12% vs 30.22%) (all P<0.05). In 2018, results for the six dimensions of health literacy were, from high to low, as follows: safety and first aid literacy (26.42%), basic medical literacy (14.14%), health information literacy (13.38%), scientific health concept (10.17%), infectious disease prevention and control literacy (8.96%), chronic disease prevention and treatment literacy (5.57%); In 2020, the results, from high to low, were: safety and first aid literacy (36.09%), basic medical literacy (24.08%), health information literacy (20.47%), infectious disease prevention and treatment literacy(15.19%), scientific health concept (13.85%), and chronic disease prevention and treatment literacy (10.10%). All values were higher than in 2018 (all P<0.05). Gender, age, education level, marital status, occupation, per capita monthly household income level, and area of residence influenced health literacy levels of local residents. Conclusions:The health literacy of Uyghur residents in Moyu County, Xinjiang improved from 2018 to 2020; however, the overall level remains low. Elderly people, people with low education levels, people with low incomes, business people & staff are the key groups requiring health education. Healthy lifestyle and behavior, chronic disease prevention and treatment literacy, scientific health concept, infectious disease prevention and control literacy are identified as areas necessitating health education and health promotion intervention.

@#Abstract: Objective To understand the prevalence and influencing factors of major chronic diseases among Kirgiz residents in Aheqi County, Xinjiang, and to provide theoretical basis for the prevention and control of chronic diseases in this area. Methods The data of residents in Aheqi County, Kizilsu Kirgiz Autonomous Prefecture, Xinjiang in 2020 for health checkup were selected, and the permanent residents of Kirgiz nationality aged ≥18 were selected as the survey objects, and the prevalence of major chronic diseases and their influencing factors were analyzed by Logistic regression model. Results A total of 21 935 patients were enrolled, and 10 602 patients suffered from chronic diseases with a prevalence rate of 48.33%, including 4 929 cases of hypertension with a prevalence rate of 22.47%, 4 238 cases of obesity with a prevalence rate of 19.32%, 3 610 cases of dyslipidemia with a prevalence rate of 16.46%, 1 632 cases of anemia with a prevalence of 7.44%, and 1 236 cases of type 2 diabetes with a prevalence of 5.63%. The prevalence of hypertension, obesity, dyslipidemia and type 2 diabetes increased with age, while the prevalence of anemia decreased with age (P<0.01). The prevalence of hypertension dyslipidemia and type 2 diabetes was higher in males than in females, and the prevalence of obesity and anemia was higher in females than in males, with statistical significance (P<0.01). Logistic regression analysis showed that 30-<40 years old (OR=1.836, 95%CI: 1.565-2.034), 40-<50 years old (OR=2.916, 95%CI: 2.613-3.255), 50-<60 years old (OR=5.244, 95%CI: 4.651-5.913), 60 years old and above (OR=10.866, 95%CI: 9.533-12.385), government personnel (OR=1.789, 95%CI: 1.405~2.279), professional and technical personnel (OR=1.774, 95%CI:1.372-2.295), the office staff (OR=1.923, 95%CI: 1.418-2.607) occasional alcohol consumption (OR=1.157, 95%CI: 1.055-1.270) and occasional exercise (OR=1.498, 95%CI: 1.238-1.812) were risk factors for chronic disease. Conclusion The prevalence of chronic diseases among residents in Aheqi County of Xinjiang is at a low level. Local health institutions have strengthened health education and publicity on multiple chronic diseases and the prevention and control of anemia among young and middle-aged women.

Objective@#To investigate the prevalence of dyslipidemia among adults in Moyu County, Hotan Prefecture, Xinjiang Uygur Autonomous Region, so as to provide insights into the management of dyslipidemia.@*Methods@#The physical examination data of permanent residents at ages of 18 years and older were collected from Moyu County from 2018 to 2019, including demographic features, height, body weight, blood pressure and blood biochemical parameter measurements. The epidemiological characteristics of dyslipidemia were analyzed among residents.@*Results@#Totally 166 142 adults were investigated, with a mean age of (41.08±15.72) years. There were 77 744 men (46.76%), 34 728 obese adults (20.90%), 5 776 adults with diabetes (3.48%) and 26 294 adults with hypertension (15.83%). The levels of triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) were (1.38±0.98), (4.13±1.19), (1.31±0.41) and (2.25±0.77) mmol/L, respectively. The prevalence of dyslipidemia was 29.72%, and the detection rates of high TG, high TC, low-HDL-C and high LDL-C were 9.73%, 4.59%, 19.65% and 1.99%, respectively. The prevalence of dyslipidemia was higher in men than in women (33.97% vs. 25.98%; χ2=1 264.729, P<0.001), and the prevalence increased with age (χ2=539.382, P<0.001) and body mass index (χ2=3 601.833, P<0.001). The prevalence of dyslipidemia was 35.48% among patients with hypertension (χ2=497.251, P<0.001) and 46.07% among patients with diabetes (χ2=766.142, P<0.001). There were 9 059 adults with two and more abnormal blood lipid indexes (18.35%), and the detection rates of two and more abnormal blood lipid indexes were 26.50%, 24.14% and 33.15% among patients with obesity, hypertension and diabetes, respectively.@*Conclusions@#The prevalence of dyslipidemia among adults in Moyu County is lower than the national level, and low-HDL cholesterolemia is the most common type of dyslipidemia. The prevalence of dyslipidemia is high among patients with obesity, hypertension and diabetes mellitus, in whom abnormality of multiple blood lipid indexes is measured.

Objective@#To investigate the prevalence of metabolic diseases among residents in Moyu County, Hotan Prefecture, Xinjiang Uygur Autonomous Region, so as to provide the evidence for developing the control strategy for metabolic diseases.@*Methods@#The demographic features, laboratory examinations and B-mode ultrasonographic examinations of the liver and gall bladder were collected from residents at ages of 18 years and older in Moyu County through the hospital-based medical records system. The prevalence of metabolic diseases was estimated, including obesity, hypertension, diabetes, dyslipidemia and fatty liver, and the factors affecting the development of metabolic diseases were identified using a multinomial logistic regression model.@*Results@#Totally 281 803 residents were included in the study, including 129 549 men ( 49.97% ) and 152 254 women ( 54.03% ) and with a median age of 37.00 years ( interquartile range, 22.00 years ). The overall prevalence of metabolic diseases was 47.66%, and the prevalence rates of obesity, hypertension, dyslipidemia, fatty liver and diabetes were 25.72%, 24.77%, 12.30%, 12.05% and 5.49%, respectively, which appeared a tendency towards a rise with age ( χ2trend = 11 243.73, 14 086.41, 2 109.70, 6 631.22, and 2 011.15, all P<0.001 ). There were 73 811 residents suffering from one metabolic disease (26.19%), and 60 495 residents with two and more metabolic diseases ( 21.47% ). Multinomial logistic regression analysis showed that gender ( OR=0.870; 0.796 ), age ( OR: 1.703-8.701; 2.952-18.846 ), marital status ( OR: 1.397-1.845; 1.460-2.377 ) educational level ( OR: 0.891-0.937; 0.920-0.924 ), occupation ( OR: 1.177-1.270; 1.114-2.099), family history ( OR=1.136; 1.390), drinking frequency ( OR: 2.038; 1.395-2.574 ) and central obesity ( OR=3.448; 14.352 ) were factors affecting the development of one or more metabolic diseases.@*Conclusion@#The prevalence of metabolic diseases is high among residents in Moyu County. Men, the elderly, a low education level and unhealthy lifestyles may increase the risk of metabolic diseases.

Objective To explore the prevalence and characteristic of Non-alcoholic fatty liver disease (NAFLD) among the ethnic Kirghiz adult in Xinjiang Province. Methods The study was conducted based on the national health checkup in Xinjiang province during 2018. A total of 19 843 participants of Kirghiz adults in Aheqi County, Xinjiang Uygor Autonomous Region were selected for physical examination, questionnaires, blood biochemical indicators detection (FBG, TG, LDL-C, HDL-C, TC, ALT and AST) and abdominal ultrasonography. A total of 1 396 patients diagnosed with NAFLD by abdominal ultrasonography were selected as the case group, and 4 188 non-NAFLD subjects matched in age, sex and region were selected as the control group at a ratio of 1:3. The clinical characters and risk factors of NAFLD patients were analyzed. Results The overall prevalence of NALFD in Kirghiz adults was 7.0%, 7.8% in male, and 6.2% in female (standardized to 6.8%, 7.5%, and 6.0%, respectively). The prevalence in male was higher than that in female (P<0.01). The prevalence of NAFLD in the obesity, diabetes, and hypertension group was significantly increased, which was 19.0%, 20.6% and 18.5%, respectively. Body mass index (BMI), waist circumference (WC), triglycerides, AST (aspartate aminotransferase) and ALT (alanine transamilase) in the NAFLD group were all higher than those in the control group (all P < 0.01). In the NAFLD group, 17.2% had another one metabolic disorder and 62.0% had another two metabolic disorders. Logistic regression analysis showed that waist circumference (OR = 4.08, 95%CI = 3.04~5.10), FBG (OR = 3.76, 95%CI = 2.86~4.93) and BMI (OR = 3.32, 95%CI = 2.16~4.38) were the main independent risk factors for NAFLD. Literacy (OR = 0.42, 95%CI = 0.24~0.69) and physical activity (OR = 0.68, 95%CI = 0.45~0.96) were protective factors. Conclusion The prevalence of NAFLD in Kirgiz adults in Xinjiang is lower than the national average. More than half of NAFLD patients are comorbidity with at least two metabolic abnormalities. WC, FBG and BMI were the independent risk factors for NAFLD, however, literacy and physical activity were protective factors.

This study aims to investigate the potential mechanism of curcumin in mediating interleukin-6(IL-6)/signal transducer and activator of transcription 3(STAT3) signaling pathway to repair intestinal mucosal injury induced by 5-fluorouracil(5-FU) chemotherapy for colon cancer. SD rats were intraperitoneally injected with 60 mg·kg~(-1)·d~(-1) 5-FU for 4 days to establish a model of intestinal mucosal injury. Then the rats were randomly divided into model group(equal volume of normal saline), curcumin low, medium and high dose groups(50, 100, 200 mg·kg~(-1)), and normal SD rats were used as control group(equal volume of normal saline). Each group received gavage administration for 4 consecutive days, and the changes of body weight and feces were recorded every day. After administration, blood was collected from the heart, and jejunum tissues were collected. The levels of serum interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) were detected by ELISA, and at the same time, the concentration of Evans blue(EB) in jejunum was measured. Hematoxylin-eosin(HE) staining was used to observe the pathological state of jejunum, and the length of jejunum villi and the depth of crypt were measured. The positive expression levels of claudin, occludin and ZO-1 were detected by immunohistochemistry. Western blot was used to detect the protein expression of IL-6, p-STAT3, E-cadherin, vimentin and N-cadherin in jejunum tissues. The results showed that, curcumin significantly increased body weight and fecal weight(P<0.05 or P<0.01), decreased fecal score, EB concentration, IL-1β and TNF-α levels(P<0.05 or P<0.01) in rats. In addition, curcumin maintained the integrity of mucosal surface and villi structure of jejunum to a large extent, and reduced pathological changes in a dose-dependent manner. Meanwhile, curcumin could increase the positive expression of occludin, claudin and ZO-1(P<0.05 or P<0.01), repair intestinal barrier function, downregulate the protein expression of IL-6, p-STAT3, vimentin and N-cadherin in jejunum tissues(P<0.05 or P<0.01), and upregulate the protein expression of E-cadherin(P<0.05). Therefore, curcumin could repair the intestinal mucosal injury induced by 5-FU chemotherapy for colon cancer, and the mechanism may be related to the inhibition of IL-6/STAT3 signal and the inhibition of epithelial-mesenchymal transition(EMT) process.

OBJECTIVE@#To study the efficacy and safety of vitamin D as an adjuvant therapy for childhood pneumonia through a systematic review.@*METHODS@#Cochrane Library, PubMed, EMbase, CNKI, Wanfang Data, and Weipu Data were searched for randomized controlled trials (RCTs) of vitamin D as the adjuvant therapy for childhood pneumonia published up to August 2019. Literature screening, quality assessment, and data extraction were performed based on inclusion and exclusion criteria. Revman 5.3 was used to perform the Meta analysis of outcome indicators.@*RESULTS@#A total of 7 RCTs with 1 527 children were included, with 762 children in the vitamin D adjuvant therapy group and 765 children in the control group. The results of the Meta analysis showed that vitamin D adjuvant therapy had no effect on recovery time (P=0.67), length of hospital stay (P=0.73), and time to relief of fever (P=0.43). Furthermore, it did not reduce the recurrence rate (P=0.14), rate of adverse events (P=0.20), and mortality rate (P=0.98) of childhood pneumonia.@*CONCLUSIONS@#Current evidence shows that vitamin D adjuvant therapy has no marked efficacy in the treatment of childhood pneumonia.

Objective:To investigate health literacy level and its influencing factors of 7, 945 Kazakh residents in Yili Prefecture, Xinjiang, China.Methods:Participants were enrolled from rural or urban Kazakh permanent residents aged 18 to 75 years living within the city and four counties of Yili Prefecture, Xinjiang, from April to November 2019. Stratified random sampling was adopted to divided the sites into five layers: Yining city, Gongliu County, Nilk County, Xinyuan County and Zhaosu County. According to the calculation of actual sample size, a total of 7, 945 subjects were selected. A health literacy questionnaire was used to determine the different demographic characteristics and health literacy status of Kazakh residents.Results:A total of 7, 945 cases, with mean age of (40.6±11.6) years, including 3, 391 males (42.68%) and 4, 554 females (57.32%) were studied. The total health literacy score was (45.8±17.2). In this study, we aimed to identify health literacy status of at least 424 cases(5.34%). The number of people with safety and first aid literacy was the largest, with a total of 2, 437 cases (30.67%). The number of people with health information access literacy and infectious disease prevention literacy was relatively small, with 860 (10.82%) and 866 cases (10.90%), respectively. Univariate analysis results showed that there were statistically significant differences (all P<0.05) in the scores of marital status, age, education level, personal monthly income, occupation, and household health literacy level. The results of multiple linear regression analysis showed that the main influencing factors of Kazakh residents′ health literacy were marital status, educational level, personal monthly income, occupation, and household registration. Conclusion:The health literacy of Kazakh residents in Yili prefecture, Xinjiang is at a low level, among which literacy in health information access and infectious disease prevention and control were the lowest. Ultimately, marital status, education level, personal monthly income, occupation, household registration, and other factors can have an impact on the level of health literacy.

Objective:This study aimed to explore and analyze the correlation between pulse pressure index (PPI), body mass index (BMI), and cerebral hemodynamics and evaluate their significance in early screening for stroke.Methods:The subjects were selected from those who had completed the cerebral blood flow function test in the First Affiliated Hospital of Xinjiang Medical University from January to December 2018. First, basic information and disease history of the respondents were obtained through on-site questionnaire survey. Height, weight, and blood pressure were measured, and BMI, pulse pressure, PPI, and mean arterial pressure were calculated. Meanwhile, cerebral hemodynamic monitoring was completed. Individuals with cerebral hemodynamic indexes ≥75 were classified as normal cerebrovascular function group and those with cerebral hemodynamic indexes <75 as abnormal group. This study divided the PPI into three subgroups: high, medium, and low. According to the Chinese Adult Overweight and Obesity Prevention and Control Guidelines, the BMI was divided into too low, normal, overweight, and obese. The correlations of PPI and BMI with cerebral hemodynamics in different groups were calculated and compared.Results:A total of 12 375 subjects, aged (52.6±7.3) years, were included in the study, consisting of 7 275 men (58.79%) and 5 100 women (41.21%). There were 9 900 patients (80.00%) in the normal group and 2 475 patients (20.00%) in the abnormal group. The abnormal rate of cerebral hemodynamics was higher in the Han nationality; those with a history of hypertension, diabetes, and hyperlipidemia. Moreover, those with abnormal cerebral hemodynamic indicators had a higher BMI, systolic pressure, diastolic pressure, pulse pressure and PPI, mean arterial pressure was higher than the normal group (all P<0.001). In different PPI groups, the average blood flow, average velocity, maximum velocity, minimum velocity, and critical pressure level of the left and right sides showed a downward trend with the increase in PPI. Peripheral resistance, pulse wave velocity, characteristic impedance, dynamic resistance, and difference between diastolic and critical pressure all showed an upward trend with the increase in PPI. In different BMI groups, the average blood flow in the obese group on the left side was lower than that in the underweight, normal, and super recombination groups. The average blood flow in the obese group on the right side was lower than that in the normal and super recombination groups and higher than that in the subhypoxia group. The mean, maximum, and minimum velocities of the left and right sides decreased with the increase in BMI. The differences in peripheral resistance, pulse wave velocity, characteristic impedance, dynamic resistance, critical pressure level, diastolic blood pressure, and critical pressure showed an increasing trend with the increase in BMI, and the differences of the above indicators were statistically significant (all P <0.001).The results of unconditional multivariate logistic regression analysis showed that Han nationality, history of hypertension, history of diabetes, PPI, and BMI were the main risk factors for abnormal cerebral hemodynamics. The higher the PPI and BMI, the higher the risk of abnormal cerebral blood flow. Conclusion:The higher the PPI and BMI, the higher the possibility of abnormal cerebral hemodynamic indexes. Extremely high PPI and BMI should be paid greater attention.

OBJECTIVE@#To investigate the gene mutation occurved in AML patients with 29 kinds of fusion genes and 51 kinds of tumor gene.@*METHODS@#Next-generation sequencing (NGS) was used to detected the 49 kinds of targeted gene. FLT3 internal tandem duplication (FLT3-ITD), CALR, NPM1 and CEBPA mutation were detected by DNA-based PCR and Sanger sequencing. Twenty-nine kinds of fusion genes were dected by multiplex nested RT-PCR.@*RESULTS@#The total gene mutation rate was 91% (109/121) in all the 121 patients. On average, 2.1 mutated genes per patient were identified, among these 121 patients, coexistence of ≥ 3 mutations was frequent (34.7%). The most commonly mutated genes were NRAS (23.96%, n=29), followed by NPM1 (14.04%, n=17), CEBPA double mutations (14.04%, n=17), KRAS (11.57%, n=14),FLT3-ITD (10.74%, n=13), CSF3R (10.74%, n=13), TET2 (9.92%, n=12) and IDH1 (9.1%, n=11). Overall, fusion genes were detected in 47 (37.3%) patients, including AML/ETO (n=12), CBFβ/MYH11 (n=11), PML/RARa (n=12), MLL rearranagement realated mutation MLL-X (n=10). TLS/ERG (n=1) and DEK/CAN (n=1) in an order of decreasing frequency. Patients with normal karyotype (NK)- AML exhibited more mutations in CEBPA, NPM1, TET2, RUNX1 and IDH1, comparing with abnormal karyotype patients. KRAS mutation in abnormal kayotype patients was significantly higher than that in normal kayotype patients (P=0.014). TP53 mutations were predominantly associated with complex cytogenetics (P=0.199). KRAS mutations were more frequent in core binding factor (CBF) acute myeloid leukemia (AML) and 11q23/MLL rearrangement leukemia, compared with NK-AML (P=0.006 and 0.003, respectively). KIT mutations predominated in CBF-AML (P=0.006). JAK2V617F mutations were detected in two patients and co-occurred with AML-ETO fusions.@*CONCLUSION@#At least one mutation is observed in more than 90% patients. On average, more than 2 mutated genes per patient are identified. Some gene mutations are associated with gene rearrangement.

OBJECTIVE@#To study the association between maternal alcohol consumption and the risk of congenital heart disease (CHD) in offspring.@*METHODS@#PubMed, Cochrane Library, Web of Science, Google Scholar, China Biology Medicine disc, Wanfang Database, CNKI Database, and Weipu Database were searched for the articles on the association between maternal alcohol consumption and congenital heart disease in offspring. These articles were published up to November 30, 2019. A random effects model or a fixed effects model was used for the pooled analysis of the results of each study, and then the pooled effective value and its 95%CI were calculated. A subgroup analysis was performed to explore heterogeneous regulators. Funnel plots and an Egger's test were used to assess publication bias.@*RESULTS@#A total of 4 409 articles were searched, and 55 articles were finally included in this analysis, among which there were 6 cohort studies and 49 case-control studies. The Meta analysis showed heterogeneity across all studies (I=74%, P<0.01). The random effects model showed that maternal alcohol consumption was associated with CHD in offspring, with an OR of 1.18 (95%CI: 1.09-1.28). The Egger's test showed a certain degree of publication bias (P<0.05), and after adjustment, the pooled OR of CHD in offspring was 1.10 (95%CI: 1.01-1.21).@*CONCLUSIONS@#Maternal alcohol consumption may increase the risk of CHD in offspring.

Objective@#This study aimed to explore and analyze the correlation between pulse pressure index (PPI), body mass index (BMI), and cerebral hemodynamics and evaluate their significance in early screening for stroke.@*Methods@#The subjects were selected from those who had completed the cerebral blood flow function test in the First Affiliated Hospital of Xinjiang Medical University from January to December 2018. First, basic information and disease history of the respondents were obtained through on-site questionnaire survey. Height, weight, and blood pressure were measured, and BMI, pulse pressure, PPI, and mean arterial pressure were calculated. Meanwhile, cerebral hemodynamic monitoring was completed. Individuals with cerebral hemodynamic indexes ≥75 were classified as normal cerebrovascular function group and those with cerebral hemodynamic indexes <75 as abnormal group. This study divided the PPI into three subgroups: high, medium, and low. According to the Chinese Adult Overweight and Obesity Prevention and Control Guidelines, the BMI was divided into too low, normal, overweight, and obese. The correlations of PPI and BMI with cerebral hemodynamics in different groups were calculated and compared.@*Results@#A total of 12 375 subjects, aged (52.6±7.3) years, were included in the study, consisting of 7 275 men (58.79%) and 5 100 women (41.21%). There were 9 900 patients (80.00%) in the normal group and 2 475 patients (20.00%) in the abnormal group. The abnormal rate of cerebral hemodynamics was higher in the Han nationality; those with a history of hypertension, diabetes, and hyperlipidemia. Moreover, those with abnormal cerebral hemodynamic indicators had a higher BMI, systolic pressure, diastolic pressure, pulse pressure and PPI, mean arterial pressure was higher than the normal group (all P<0.001). Objective To investigate the prevalence of complication and related risk factors in Uygur patients with type 2 diabetes mellitus (T2DM).Methods Total 1 507 Uygur patients aged above 20 with T2DM were recruited in the First Affiliated Hospital of Xinjiang Medical University from January 2010 to October 2015.Clinical characteristics,biochemical parameters and related complications were analyzed in different age groups of patients.Multivariate logistic regression was performed to determine associated factors for diabetic complications.Results Among 1 507 cases,there were 61.3% male patients (n=924),37.4% (n=564) with diabetes duration ≥5 years,12.5% (n=189) with HbA1c>10%,31.5% (n=475) with hypertension,28.8% (n=434) with smoking history and 37.4% (n=564) patients without antidiabetic drug treatment.The average body mass index (BMI) of patients was (27.4±5.2)kg/m2 with a obesity rate of 43.0% (648/1 507),and 65.0% (980/1 507) had physical exercise<150 min/week.The prevalence of diabetic complication was 70.6%(1 064/1 507),in which 28.7%,19.2% and 22.7% cases had 1,2 and 3 or more complications,respectively.The proportion of patients with coronary heart disease,stroke/transient ischemic attack and large artery atherosclerosis was 32.4%,10.4% and 35.8%,respectively.The proportion of patients with diabetic retinopathy,nephropathy and neuropathy were 23.4%,6.5% and 35.8%,respectively.Patients aged ≥60 years had highest prevalence of large artery atherosclerosis (38.2%,163/427) and diabetic retinopathy (27.2%,116/427);patients in age group 50-59 years had highest prevalence of diabetic neuropathy (40.3%,207/514).Multivariate logistic regression analysis showed that HbA1c>10% (OR=2.74,95%CI:1.40-5.34),obesity (OR=1.54,95%CI:1.07-2.21),systolic pressure≥140 mmHg (1 mmHg=0.133 kPa) (OR=1.42,95%CI:1.01-1.99),diastolic pressure≥90 mmHg (OR=0.37,95%CI:0.21-0.67),smoking (OR=1.53,95%CI:1.09-2.15) and physical aetivity<150 min/week (OR=1.42,95%CI:1.07-1.89) were the risk factors of diabetic complications.Conclusion Uygur patients with type 2 diabetes in Xinjiang show a relatively high prevalence of complications.Poor glycemic control,obesity,hypertension,smoking and physical inactivity may contribute to development of complications,while well-controlled blood glucose and hypertension and quitting smoking and increasing physical activity may reduce diabetic complications.

Objective The relationship between metabolic syndrome(MetS), metabolic syndrome score (MSS) and non-alcoholic fatty liver disease(NAFLD) in Urumqi was investigated by the MSS, reflecting the aggregation of MetS components. Methods The subjects were divided into non-NAFLD and NAFLD group by abdominal B ultrasound. The results of physical examination and blood biochemical examination were analyzed. The MSS was calculated and the relationship between the different aggregation of MSS and NAFLD was analyzed by Log-binomial regression. Results A total of 20 569 subjects were included in the study. The detection rate of MetS was 16.7%, the detection rate of NAFLD was 32.4%. Compared with non-NAFLD group, the systolic blood pressure, diastolic blood pressure,body mass index (BMI), waist circumference, fasting plasma glucose, triglyceride, total cholesterol, and low-density lipoprotein cholesterol were all increased in the NAFLD group, and the difference was statistically significant (P<0.05). After adjusting for gender, age, ethnicity and education level, Log-binomial regression analysis showed high BMI, hypertension and hyperglycemia. High triglycerides and low-density lipoproteins are the main risk factors for NAFLD (PR values were 3.194, 1.331, 1.623, 1.981, 1.254, respectively); the risk of NAFLD increased corresondingly when MSS, MSS0, MSS1, MSS2, MSS3, and MSS4 increased. The PR were 3.127, 4.983, 6.437, and 7.331, respectively. Conclusions The formation of NAFLD is not a single accumulation of liver fat, combined with abnormalities such as blood lipids, blood pressure, and blood sugar. The detection rate of male MetS and NAFLD was higher than that of female, but women with two abnormal metabolic indicators were more likely to develop into NAFLD. BMI as the obesity index has the strongest relationship with NAFLD, and NAFLD prevention should focus on obese people.

OBJECTIVE@#To investigate the gene mutation of patients with WAS gene defect and its correlation with clinical manifestations.@*METHODS@#Thirty-one patients consulted in Children's Hospital of Soochow University from January 2013 to February 2018 were enrolled in this study. The hot pot mutations of WAS gene in 31 patients were detected and related clinical phenotypes were analyzed retrospectively.@*RESULTS@#All patients were male. The median onset age was 1 month (range, 0-83 months). Nine mutants were reported as novel mutations among 25 mutants detected in 31 patients, including c.1234_1235dupCC, c.1093-1097delG, c.28-30dupC, c.436G>T, c.273 + 10_273 + 11dupCC, c.995_996insG, c.1010T>A, c.332_333delCC and c.683C>T mutations. There were 25 cases of classic WAS which mutations included missense mutation, deletion mutation, insertion mutation, splicing mutation and nonsense mutation, 2 cases of X-linked thrombocytopenia (XLT) were induced by missense mutation, 1 case of intermittent X-linked thrombocytopenia (IXLT) was induced by splicing mutation, 2 cases of X-linked pancytopenia were induced by missense mutation. Intravenous immunoglobulin (IVIG) and glucocorticoid therapy in IXLT patient was effective, and remission could be sustained, platelets could be increased in the short-term in treated XLT patients, but only a small part of classic WAS patients(8.0%) showed transient response to it, the IVIG and glucocorticoid therapy did not improve the status of platelet in XLP patients. Immune laboratory examination showed that CD3 was decreased in 60.0% patients, CD19 was decreased in 12.0% patients, and CD56CD16 in 4 patients was decreased, accounting for 16.0%. Out of 24 patients, 22 patients were alive after treated with hematopoietic stem cell transplantation (HSCT), 4 patients who were not given HSCT died of brain bleeding and severe infection, 1 patient diagnosed as IXLT got remission and survived.@*CONCLUSION@#WAS gene defect is an important basis for the diagnosis of WAS and related diseases. IVIG plus glucocorticoid therapy is less effective for fewer patients, the HSCT is an effective treatment for WAS.

Cordycepin exerts neuroprotective effects against excitotoxic neuronal death. However, its direct electrophysiological evidence in Alzheimer's disease (AD) remains unclear. This study aimed to explore the electrophysiological mechanisms underlying the protective effect of cordycepin against the excitotoxic neuronal insult in AD using whole-cell patch clamp techniques. β-Amyloid (Aβ) and ibotenic acid (IBO)-induced injury model in cultured hippocampal neurons was used for the purpose. The results revealed that cordycepin significantly delayed Aβ + IBO-induced excessive neuronal membrane depolarization. It increased the onset time/latency, extended the duration, and reduced the slope in both slow and rapid depolarization. Additionally, cordycepin reversed the neuronal hyperactivity in Aβ + IBO-induced evoked action potential (AP) firing, including increase in repetitive firing frequency, shortening of evoked AP latency, decrease in the amplitude of fast afterhyperpolarization, and increase in membrane depolarization. Further, the suppressive effect of cordycepin against Aβ + IBO-induced excessive neuronal membrane depolarization and neuronal hyperactivity was blocked by DPCPX (8-cyclopentyl-1,3-dipropylxanthine, an adenosine A₁ receptor-specific blocker). Collectively, these results revealed the suppressive effect of cordycepin against the Aβ + IBO-induced excitotoxic neuronal insult by attenuating excessive neuronal activity and membrane depolarization, and the mechanism through the activation of A₁R is strongly recommended, thus highlighting the therapeutic potential of cordycepin in AD.

Intestinal barrier dysfunction always accompanies cirrhosis in patients with advanced liver disease and is an important contributor facilitating bacterial translocation (BT), which has been involved in the pathogenesis of cirrhosis and its complications. Several studies have demonstrated the protective effect of Vitamin D on intestinal barrier function. However, severe cholestasis leads to vitamin D depletion. This study was designed to test whether vitamin D therapy improves intestinal dysfunction in cirrhosis. Rats were subcutaneously injected with 50% sterile CCl₄ (a mixture of pure CCl₄ and olive oil, 0.3 mL/100 g) twice a week for 6 weeks. Next, 1,25(OH)₂D₃(0.5 µg/100 g) and the vehicle were administered simultaneously with CCl₄ to compare the extent of intestinal histologic damage, tight junction protein expression, intestinal barrier function, BT, intestinal proliferation, apoptosis, and enterocyte turnover. Intestinal heme oxygenase-1 (HO-1) expression and oxidative stress were also assessed. We found that vitamin D could maintain intestinal epithelial proliferation and turnover, inhibit intestinal epithelial apoptosis, alleviate structural damage, and prevent BT and intestinal barrier dysfunction. These were achieved partly through restoration of HO-1 and inhibition of oxidative stress. Taken together, our results suggest that vitamin D ameliorated intestinal epithelial turnover and improved the integrity and function of intestinal barrier in CCl₄-induced liver cirrhotic rats. HO-1 signaling activation was involved in these above beneficial effects.
