OBJECTIVE@#To conduct qualitative and quantitative analyses of Tripterygium hypoglaucum in Yinning Tablets, a compound preparation of traditional Chinese herbal medicine.@*METHODS@#Thin-layer chromatography (TLC) was used for qualitative analysis of Tripterygium hypoglaucum in Yining Tablets and the analytical protocols were optimized. High-performance liquid chromatography (HPLC) was used to quantitatively analyze the content of triptolide (the main active ingredient of Tripterygium hypoglaucum) in Yinning Tablets.@*RESULTS@#The results of TLC analysis showed that the test sample of Yinning Tablets and the positive control samples both produced clear, well separated spots without obvious interference in the blank samples. Assessment of the influences of the thin-layer plates from different manufacturers, temperature and humidity on the test results demonstrated good durability of the test. HPLC analysis of triptolide showed a good linear relationship within the concentration range of 1-100 μg/mL (regression equation: A=22.219C-19.165, r=0.9999); the contents of triptolide in 3 batches of Yinning tablets were 0.34, 0.34, and 0.28 μg per tablet, all within the range of 0.28-0.34 μg per tablet. It was finally determined that each Yinning tablet should not contain more than 0.6 μg of triptolide.@*CONCLUSION@#TLC and HPLC are simple, accurate, durable and specific for qualitative and quantitative analyses of Tripterygium hypoglaucum in Yinning Tablets.

RESUMO

Conventional prediction model, as a static prediction model, can be only used to predict the probability of the occurrence of an event during the observation period using the information available at baseline survey. However, based on current clinical demands, dynamic prediction, which obtains prediction probabilities for both baseline survey and later time points given the history of the events and covariates up to that time, is gaining a growing attention. As a dynamic prediction model, the landmarking approach is simple, easy to use, computationally efficient and has a comparable performance of joint modeling, which makes it to be widely used in recent researches. Because of its limited application in China, this paper makes a brief introduction of its ideas and basic application to further promote its applications in clinical dynamic prediction.

Background Tinibs were a kind of important epidermal growth factor receptor (EGFR) inhibitors used aspotential therapeutic agents in treating non-small cell lung cancer (NSCLC) in clinic. The drug resistance ofclinical used tinibs made the development of more active tinib analogues an attractive field in research.Quinazoline ring was regarded as the key fragment in tinibs and quinazolinone was indispensible intermediatein the synthesis of quinazoline. Thus, synthesis of quinazolinone intermediates was a key step which wouldfurther limit the overall yield of final product of tinib analogues. However, the commonly used synthetic scheme was somewhat complicated and time consuming with relatively low yield in heterocylization ofquinazolinone and its derivatives.Aim In this work, we intended to explore an effective way to improve synthesis of heterocyclization of 6,7-bis(2-methoxyethoxy)quinazolin-4(3H)-one (compound 5), the key fragment of erlotinib analogues, in bothreaction procedure and yield, thus to provide reference to synthesis of other quinazolinone derivatives.Methods A simple microwave-assisted one-pot reaction was employed to improve the synthesis ofheterocyclization of quinazolinone ring. The reaction conditions, including microwave power, temperature andtime of reaction, were screened to achieve high yield under simple operation.Results 6,7-bis(2-methoxyethoxy)quinazolin-4(3H)-one (compound 5) was successfully synthesized fromstarting material of 4,5-bis(2-methoxyethoxy)-2-nitrobenzonitrile by microwave reaction, which was finished in1 hour just by one step. The yield of heterocyclization was increased from 29.8% of commonly used three-stepscheme to 50% herein.Conclusion Microwave reaction efficiently improved synthesis of heterocyclization of 6,7-bis(2-methoxyethoxy)quinazolin-4(3H)-one into "quinazolinone in both synthetic procedure and yield. The resultsmay provide valuable reference to synthesis of other quinazolinone derivatives.

Background Apocynin (4-hydroxy-3-methoxyacetophenone), an important active ingredient contained in root ofPicrorhiza kurroa Royle, has been widely investigated as an antioxidative and anti-inflammatory agent in kinds ofdisease models, and exerted certain efficacy deserving further research. However, little was known about thedisposal process of apocynin in vivo, which is important information required in drug research and development.Aim In this work, a tissue distribution study of apocynin was performed in Sprague-Dawley (SD) male rats to helpfurther understanding well the disposition of apocynin in vivo.Methods A simple HPLC-UV method was developed for measurement of apocynin concentration in rat tissues. Amixture of water-methanol (47:53, v/v) was used as solvent system, the flow rate was 1 mL/min, and the detectedwavelength was 279 nm. The method was validated and applied to the tissue distribution study of a single bolusintravenous administration of apocynin in SD male rats.Results The developed HPLC-UV method showed good specificity, precision, accuracy and extraction recovery. Thegood linearity was achieved within 0.8-32 μg/mL in tissues including heart, liver, spleen, lung, kidney and brain. Thelower limit of quantification (LLOQ) was 0.8 μg/mL. The method was well used in the study of tissue distribution ofapocynin. The results demonstrated that apocynin was distributed fast into the tested tissues and reached peakconcentration at 5 min after injection. Apocynin was mainly distributed in liver, kidney and lung within 15 minutesafter administration, and eliminated from the tissues with no sample be detected more than 2 h after dose

Klebsiella pneumoniae is one of the main pathogenic bacteria that causes nosocomial infections, such as pneumonia, urinary tract infection, and sepsis. Therefore, the rapid and accurate detection of K. pneumoniae is important for the timely treatment of infectious patients. This study aimed to establish a loop-mediated isothermal amplification (LAMP) method for the rapid and sensitive detection of K. pneumoniae-specific gene ureR_1 (Gene ID: 11847803). The ureR_1 gene was obtained through local and online BLAST, and the specific primers were designed for its detection. Positive reactions were observed on all 140 K. pneumoniae clinical isolates while all the 82 non-K. pneumoniae clinical isolates were negative. Plasmids with the specific gene and the mouse blood with K. pneumoniae were used for sensitivity analysis. The detection limit of the LAMP was 1 bacterium/reaction. The results showed that the LAMP targeted to ureR_1 is a fast, specific, sensitive, inexpensive, and suitable method for the detection of K. pneumoniae.

Objective @#To evaluate the clinical outcomes of all-ceramic crowns whose oral digital impressions were scanned by 3Shape Trios. @*Methods @#All the impressions were scanned by Trios and all the crowns were fabricated according to the standard procedure. The restoration quality were evaluated using modified United States Public Health Service (USPHS) criteria one year after treatment. Restoration integrity、 marginal adaptation、enamel wear、 color satisfaction and gingival index (GI) of the prostheses were evaluated according to the modified USPHS criteria. @*Results@#The clinical 1 year survival rate of 92 porcelain crowns fabricated by 3-shape Trios digital impression and made by CAD/CAM was 100%. The scored A rates of restoration integrity、 marginal adaptation、 enamel wear、 secondary caries、color satisfaction and gingival index of the prostheses were 100%、91.3%、 98%、95.6%、96.7% and 94.7% after 1 year, respectively.@*Conclusion@#The clinical outcomes of all-ceramic crowns whose oral digital impressions were scanned by 3Shape Trios and whose crowns were made by CAD-CAM are satisfying.

@#[Abstract] Objective: To investigate the effect of Tim-4 on invasion and migration of cervical cancer cells and its underlying mechanisms. Methods:The expression levels of Tim-4 in cervical cancer cell lines Siha, Hela and cervical epithelial immortalized cell line H8 were detected by Real-time PCR. The Tim-4 lentiviral vector was transfected into Siha cell line. The over-expression of Tim-4 in Siha cell line was detected by fluorescence microscopy. The effects of Tim-4 on the invasion and migration of cervical cancer cell line were detected by Transwell and scratches methods. The changes of MMP2, MMP9, E-cadherin and N-cadherin in Siha cells were detected by Western blotting. Results:The expression of Tim-4 was higher in Siha and Hela cell lines compared to that in the H8 cell line. The Siha cell line burdening Tim-4 lentiviral vector was successfully constructed. Over-expression of Tim-4 significantly inhibited the migration and invasion of cervical cancer cell line, and affected the expression of MMP2, MMP9, N-cadherin and E-cadherin. Conclusion:Over-expression of Tim-4 promotes the invasion and migration by regulating the EMT transformation in cervical cell carcinoma.

Objective@#The objective of this study was to compare the early clinical effects of zirconia all⁃ceramic crowns using two different impression methods. Zirconia ceramic crowns were produced using digital models based on either a silicone rubber impression perfusion model in vitro or a 3D mouth scanner.@*Methods @#A total of 50 patients with a planned restoration of the first permanent molar with zirconia all⁃ceramic crowns after root canal therapy were se⁃ lected and randomly divided into two groups: a digital impression by intraoral 3D scanning group and a digital impres⁃ sion by extraoral scanning after silicone rubber impression group. Zirconia all⁃ceramic crowns were created by CAD/ CAM in both groups. Marginal adaptation, proximal contact, and occlusal contact were compared between groups. @*Results@#There was no significant difference between the two groups in marginal adaptation (P > 0.05). For proximal con⁃ tact and occlusal contact, no significant differences regarding the number of cases for the criteria of level A and level B were found between two groups (P > 0.05), while the misfit value in level B was smaller in the first group than in the second group (P < 0.05). @*Conclusion@#Zirconia all⁃ceramic crowns with intraoral 3D scanning show excellent early clinical performance.
