RESUMO

Objective@#To explore dynamics of parenting styles of adolescents from 1999 to 2019 from the perspective of intergenerational conflict, to provide support for family education and adolescent healthy development.@*Methods@#Using a multistage stratified cluster random sampling method, the unified questionnaire was administered to 2 590 students in the same sampling junior and senior high schools in 1999, 2009, and 2019 using the Egna Minnen av Barndoms Uppfostran own memories of parental rearing practices in childhood(EMBU).@*Results@#Overall there were differences in the nine factors of parenting styles across generations ( F = 12.07-72.52, P <0.01), with decreasing ratings of warmth and understanding of father and mother (F1, M1), over interference of father (F3) over generations(F1:46.72±9.41, 45.87±11.33, 43.61±11.27; M1:51.56±9.38, 51.03±11.59, 46.23± 12.27 ; F3:19.03±4.00, 18.29±4.32, 17.95±4.51), and all other parenting styles rated higher in 2019 than in 2009 and 1999(except for the over protection and over interference of mother, and punishment, firm control of mother). Parenting styles across generations (except for the rejection and denial of father among girls) showed gender difference.The overall gender trend coincided with the total population trend. Parenting styles across generations varied significantly among middle and high school students( F =3.92-47.27, P <0.05 ), changes in F1 and F3 factors coincided with the overall decreasing trend. Factor analysis showed that parenting styles could be classified into two dimensions, with varied factor loading across generation.@*Conclusion@#Intergenerational decreases in parental emotional warmth and paternal interfering are observed in a sex and grade specific manner. Based on the diversity of needs and population differentiation, optimal intervention for comprehensive health development of adolescents are in great need to keep pace with the times and promoting the high quality development of adolescents.

RESUMO

OBJECTIVE:To establish a method for the determination of dendrobine and its metabolites M-250 and M-280 in mice plasma for the first time. METHODS:Mice were given dendrobine 60 mg/kg by intragastric administration,1 h later plasma were collected and treated. Using pseudoephedrine hydrochloride as internal standard and dendrobine reference substance as control, the plasma concentrations of dendrobine and its metabolites M-250 and M-280 were determined by UPLC-MS combined with quantitative analysis of multi-components by single marker. The separation was performed on Hypersil Gold C18 column with 0.1%formic acid-acetonitrile(gradient elution)at the flow rate of 0.3 mL/min. The column temperature was set at 40℃,and sample size was 5 μL. Heatable electrospray ionization (HESI) source, scan/ESI + were applied and operated in positive ion mode with atomization temperature of 300℃,ion transmission tube temperature of 350℃,the sheath gas velocity of 35 arb,the auxiliary air velocity of 15 arb,the spray voltage of 3.5 kV,the collision voltage of 30,40,50 eV. The mass-to-charge ratio of detection range were 100-1500. RESULTS:The endogenous substances of mice plasma had no interference with the content determination of dendrobine and its metabolites M-250 and M-280. The linear range of dendrobine were 9.13-912.94 ng/mL(r=0.9996). The limit of quantitation was 3.04 ng/mL. RSDs of intra-day and inter-day were all less than 7.5%(n=5 or n=3). The accuracy were 96.8%-107.5%(n=5). Matrix effects were 97.1%-106.0%(RSD=1.8%-4.7%,n=5). RSDs of the content of sample at 15℃ for 24 h,at -70 ℃ after three times freeze-thaw,at -70 ℃ for 15 d were lower than 12.8% (n=3). The content of dendrobine in plasma sample of mice was (41.3 ± 5.7) ng/mL (n=12). The contents of its metabolites M-250 and M-280 were (493.0 ± 73.1) and (41.4 ± 3.0) ng/mL (n=12) with Relative correction factor of 1.0. CONCLUSIONS: The method is sensitive and accurate,and can be used for content determination of dendrobine and its metabolites M-250 and M-280 in mice plasma.

Objective To investigate the changes of CD4 + CD25 + Foxp3 + regulatory T cell (Treg) and relative cell factors in peripheral blood of steroid-resistant allergic rhinitis (AR) patients,and to study their functions in occurrence of steroid-resistant AR.Methods The CD4 + CD25 + Foxp3 + Treg cell in 30 cases general physical examination (The control group),30 cases steroid-sensitive AR patients (steroidsensitive group) and 30 cases steroid-resistant AR patients (steroid-resistant group) were detected by flow cytometry.The levels of serum interleukin 10 (IL-10) and transforming growth factor β1 (TGF-β1) in the three groups were detected by enzyme-linked immunoadsorbent assay (ELISA).Results Compare to the control group,proportion of CD4 + CD25 + Foxp3 + Treg cell and the levels of serum IL-10,TGF-β1 in steroid-sensitive group and steroid-resistant group were significantly reduced (P < 0.01).The proportion of CD4 + CD25 + Foxp3 + Treg cell and the levels of serum IL-10,TGF-β1 in steroid-resistant group were decreased significantly than steroid-sensitive group (P < 0.01).The proportion of CD4 + CD25 + Foxp3 + Treg cell were positively correlated with the serum IL-10,and TGF-β1 (P < 0.01).Conclusions The reduced proportion of CD4 + CD25 + Foxp3 + Treg cell and lower serum IL-10,TGF-β1 play an important role in the occurrence of steroid-resistant AR patients.

Objective:To investigate the effect of hypoxia on the expression of forkhead box P3 (FOXP3) in human oral squamous cell carcinoma (OSCC) cells,and to clarify its possible epigenetic mechanism.Methods:Two kinds of OSCC cell lines,FaDu and OECM-1,were cultured under normoxic or hypoxic conditions for 18 h.The relative expression levels of FOXP3 mRNA and protein in the cells were detected by Real-time RT-PCR and Western blotting method.The histone modification levels on the FOXP3 gene promoter,including acetylation of histone 3 lysine 4 (H3K4ac),trimethylation of histone 3 lysine 4 (H3K4me3) and lysine 27 (H3K27me3),were analyzed by Chromatin Immunocipitation (ChIP) and quantitative PCR (ChIP-qPCR).The relative expression levels of histone deacetylase 3 (HDAC3) mRNA and inhibitory rates of FOXP3 mRNA expression in the HDAC3-knockdown FaDu cells were investigated by Real-time qPCR and ChIP-qPCR.Results:Compared with normoxic condition,the relative expression levels of FOXP3 mRNA in FaDu and OECM-1 cells under hypoxic condition were decreased by 65.6% and 75.7% (P<0.01).The Western blotting results indicated that compared with normoxic condition,the expression levels of FOXP3 protein in FaDu and OECM-1 cells under hypoxic condition were decreased.The ChIP experiment results showed that compared with normoxic condition,the levels of H3K4ac and H3K4me3 on FOXP3 gene promoter in FaDu cells were decreased under hypoxic condition (P<0.01),while the H3K27me3 level was not changed.In HDAC3-knockdown FaDu cells,compared with control cells,the inhibitory rates of the expressions of H3K4ac and H3K4me3 on FOXP3 gene promoter under hypoxia condition were decreased (P<0.05),so did expressions the FOXP3 mRNA expression (P<0.05).Conclusion:Hypoxia could suppress the expression of FOXP3 by HDAC3-mediated down-regulation of H3K4ac on FOXP3 gene promoter in the human OSCC cells.

ObjectiveTo investigate the risk factors for the complications of transumbilical single-port laparoscopic cholecystectomy (TUSPLC) and to explore the prophylactic methods. MethodsThe clinical data of 120 patients undergoing TUSPLC in our hospital from January 2012 to December 2014 were retrospectively analyzed. The association of different factors with the complications was analyzed, including sex, age, body mass index (BMI), height, underlying diseases (hypertension, diabetes, and hyperthyroidism), surgeon level, operation time, and intraoperative blood loss. The risk factors were determined by univariate and multivariate logistic regression analyses. ResultsThe incidence of the complications of TUSPLC was 8.3% (10/120), including surgical site infection (4 cases), umbilical hernia (2 cases), hemorrhage (2 cases), and intra-abdominal organ injury (2 cases). The univariate logistic regression analysis indicated that BMI, height, underlying diseases, surgeon level, operation time, and intra-operative blood loss were risk factors for the complications of TUSPLC (all P<0.05). The multivariate logistic regression analysis showed that underlying diseases and operation time were independent risk factors for the complications of TUSPLC (P<0.01). ConclusionThe underlying diseases and operation time are independent risk factors for the complications of TUSPLC, so appropriate intervention strategies for these factors could be developed to reduce the incidence of the complications of TUSPLC.

Objective To explore the feasibility and clinical value of transumbilical single port laparoscopic cholecystectomy . Methods In our hospital from 2010 October to 2013 August were selected with 120 patients were randomly divided into standard into two groups , 60 cases of transumbilical single port laparoscopic cholecystectomy ( Transumbilical Single Port Laparoscopic Cholecystec-tomy, TUSPLC) , 60 cases of the traditional four hole laparoscopic cholecystectomy ( Laparoscopic Cholecystectomy , LC) .Change rate were compared between the two groups , operation time , postoperative pain , postoperative intestinal function recovery time , postopera-tive drainage tube pulled out of time , postoperative hospitalization time , complications and wound condition index .Results The two groups in comparison , postoperative pain , postoperative drainage tube pulled out of time , postoperative hospitalization time of group TUSPLC and group LC were statistically different ( P 0.05 ) , TUSPLC group wound high sat-isfaction.During the follow-up of 1~3 months, no abdominal pain and other symptoms , TUSPLC group umbilical scar .Conclusions TUSPLC is safe and effective , more minimally invasive , beauty effect is good;the operation is relatively difficult , conditional hospi-tal can be carried out gradually and promotion .

<p><b>OBJECTIVE</b>To establish a kind of molecular biology clinical detective method to cariogenic S. mutans.</p><p><b>METHODS</b>Using the coamplification of target and reference genes. One pair of specific primers were designed according to a portion of the dextranase (dexA) gene of S. mutans. The reference gene was plasmid pET23b DNA. The saliva samples of 196 children were quantitative detected. The PCR method was compared with the routine culture method.</p><p><b>RESULTS</b>The rate of S. mutans counts >/= 10(8) CFU/L (colony-forming unit per millilitre) saliva by quantitative PCR was 91.3%. The results of coincidence rate between the new method and the routine way was 94.9%.</p><p><b>CONCLUSIONS</b>The new quantitative detective method is fast and provides with high scoincidence rate and high specificity, so have extensive clinical practice foreground.</p>
