RESUMO

Objective To employ high-throughput next generation sequencing (NGS) for analyzing the expression of lneRNAs and mRNAs in donor samples from pediatric living donor liver transplantation and search differentially expressed lncRNAs and drag metabolic gene for individualized guidance of immunosuppressive agents. Methods Between October 2016 and December 2017,10 liver tissue specimens from living donor liver transplantation children were collected and divided into fast and slow metabolic groups (n =5 each) according to the postoperative profiles of drug metabolism.Samples were assayed for high-throughput NGS.Target analysis was used for functional pathways and screening target genes prediction. Results There were differentially expressed 908 mRNAs and 1228 lncRNAs between slow metabolic and fast metabolic groups (P<0.05).According to the abundance and difference,22 up-regulated and 18 down-regulated mRNAs,13 up-regulated and 24 down-regulated lncRNAs were selected. In addition to CYP3A5,CYP2C19,CYP1A2 and UGT1A1 might affect the metabolism of tacrolimus. At the same time,NONHSAT108617.2 in differemially expressed lncRNAs might regulate the expression of CYP3A5 gene. Conclusions This study has comprehensively analyzed the expression of lncRNAs in donor liver from pediatric liver transplantation. Some differentially expressed drug metabolism related genes may affect tacrolimus metabolism in vivo and thus the postoperative use of immunosuppressive drugs.

RESUMO

Background:Cold storage and ischemia-reperfusion injury of liver grafts are critical for the prognosis of patients undergoing liver transplantation. Establishing a stable cold storage and reperfusion animal model is a fundamental measure for related studies. Aims:To establish a modified isolated perfused rat liver model for comprehensive assessment of liver grafts in studies on preservation of liver grafts before transplantation. Methods:Twelve Sprague-Dawley rats were randomly allocated into two groups. Livers of rats in control group were retrieved and perfused immediately for 90 minutes without preservation. In experimental group, liver grafts underwent a 30-minute warm ischemia followed by 24-hour cold storage before ex vivo perfusion. The perfusate was collected dynamically for monitoring the levels of transaminases, electrolytes and pH value;the portal vein pressure of liver grafts was measured by pressure sensor, and the hepatic hydrogen peroxide (HPO)level was assessed by microprobe for free radicals. The bile production was recorded after the ex vivo perfusion;meanwhile, the levels of malondialdehyde (MDA)and superoxide dismutase (Cu/ Zn SOD),the histopathological changes and apoptosis of hepatocytes of liver grafts were examined. Results:Compared with the control group, the levels of AST and ALT,the portal vein pressure and the HPO level of liver grafts in experimental group were obviously increased throughout the perfusion. Furthermore, the bile production and level of Cu/ Zn SOD of liver grafts in experimental group were significantly decreased. The histopathological injury and hepatocytes apoptosis of liver grafts were milder in control group. Conclusions:The liver function and antioxidant effect were reduced in warm ischemic and cold preserved liver grafts. The modified isolated perfused rat liver model established in this study is useful for monitoring and evaluation of the cold storage and reperfusion injury in liver grafts.

RESUMO

Objective To explore the efficacy and safety of three types of progesterone dosage forms in treatment of functional uterine bleeding, and compare their cost-effectiveness. Methods 180 cases with functional uterine bleeding were radomly divided into A,B,C three groups, each had 60 cases. On the basis of oral estradiol valerate,A,B and C group were given progesterone capsules, progesterone suppository and progesterone vaginal gel, respectively. The efficacy and cost-effectiveness were observed and compared. Results The efficacy rate of A,B and C group were 71.67%,86.67%and 93.33%,respectively. The adverse reaction ratio of A,B and C group were 5.00%,3.33% and 6.67%,respectively. The cost-effectiveness ratios were 336.26,98.80 and 1501.60,respectively. The increment of cost-effectiveness of A group and C group were-1035.80 and 19751.57.Conclusion Progesterone suppository(B group)has best effect in treatment of functional uterine bleeding.

RESUMO

Objective To investigate and understand the reference value range of hematological parameters for peripheral blood routine in preschool children from Shanghai. Methods The Sysmex XS-800i automated hematology analyzer and the original rea-gents were used to measure the hematological parameters in peripheral blood samples collected from 7 692 healthy preschool chil-dren aged 2-6 years in Shanghai, including white blood cells(WBC),red blood cells(RBC),platelet(PLT)count, hemoglobin(Hb), hematocrit(HCT),mean corpuscular volume(MCV),mean corpuscular hemoglobin(MCH)and mean corpuscular hemoglobin con-centration(MCHC);the various parameters in different age groups were collected and statistically processed to establish the refer-ence intervals for each parameter. Results In the blood routine among preschool children in Shanghai, there were statistically signif-icant differences in all parameters except WBC and PLT count between sexes among preschool children in Shanghai(P <0.05).The reference intervals of hematological parameters obtained in this investigation were obviously differed from those offered by the man-ufacturer.However,compared with those from the related reports, the difference existed in partial parameters, the upper limit of WBC count was highest compared with the results from some areas, while the reference value ranges of MCV,MCH and MCHC were higher than those of other area study results. Conclusion The independent blood routine medical reference value range for preschool children in Shanghai should be established and the influence of the factors of gender, instrument and reagents also should be taken into consideration in establishing the reference value range.

RESUMO

Objective To establish the pediatric reference intervals for Troponin Ⅰ and Creatine kinase-MB (CK-MB).Methods Healthy children (223 boys and 162 girls) aged from 7 months to 12 years old were studied.Blood specimens were collected and centrifuged, all serum samples were kept refrigerated below-70 ℃.Troponin Ⅰ and CK-MB were measured on the UniCel DxI 800 immunoassay system (Beckman Coulter) on the same day.Nonparametric statistics was used to estimate the 99th percentile reference intervals. Results The children were divided into three groups (group A 128 cases; group B 128 cases;group C 129 cases) according to age.The upper reference limits for CK-MB were dependent of age,and the group A (7 months-1 years) was shown to be 13.41 μg/L,the group B (2-3 years) was shown to be 9.35 μg/L,the group C (4-12 years) was shown to be 5.48 μg/L.The upper reference limit for Troponin Ⅰ was independent of age or gender and shown to be 0.01 μg/L among 385 children. Conclusions Pediatric reference intervals for Troponin Ⅰ and CK-MB have been defined in healthy children at a relevant age group (7 months-12 years). It is effective for clinical diagnosis and treatment of myocardial damage for children by determining reference intervals for Troponin Ⅰ and CK-MB.

RESUMO

Aim To study preparation of lutein from the extraction Marigold flowers by biotransformation. Methods Screen bacterium for lutein esters biotransforming by studying on the biotransformation of lutein esters by Penicillium on tabulate revulsive culture medium.Porule of and Aspergillus niger Penicillium could transform lutein esters into lutein, but biotransformation of Penicillium was better than Aspergillus niger.Porule of Pleurotus ostreatus could not grow on revulsive culture medium, but Pleurotus ostreatus could grow on the culture medium and transformed lutein esters into lutein.Research of shaking flash fermentation of Penicillium for trans-forming lutein esters. When pH is 6.0,quantity of incubation is 2 bacteria plugs and fermentation time is 12~24 hours, the transformation is completed and the content of lutein is 91.6%(HPLC)。Result Prolonging the transformation time will improve the percent of lutein but the content of lutein decreased. Conclusion Biological transrormation can be used from calendula officinacis extract preparation.
