RESUMO

OBJECTIVE:To study the spectrum-ef fect relationship of anti-hepatoma effect of C Ⅱ-3 extract from Periplaneta americana, and to preliminarily clarify the anti-hepatoma active components of it. METHODS :Based on UHPLC fingerprints of 10 batches of C Ⅱ-3 samples, with the help of UHPL C-Q-TOF/MS,the compounds corresponding to each chromatographic peak were identified qualitatively by standard substances and related literatures. Using the IC 50 value of each batch of C Ⅱ-3 sample against human hepatoma cells HepG 2 as anti-hepatoma activity index ,the spectrum-effect relationship of fingerprint and anti-hepatoma effect was established and analyzed by the combination of grey relational analysis (GRA)and orthogonal partial least squares(OPLS). RESULTS :There were 25 common peaks in UHPLC fingerprints of 10 batches of C Ⅱ-3 samples, and 10 chemical compounds were identified ,which were cyclo- (Tyr-Pro)(peak 24),cyclo-(Gly-Phe)(peak 15),hypoxanthine(peak 3), adenine(peak 7),phenylalanine(peak 10),inosine(peak 11),N-acetyldopamine(peak 16),cyclo-(Pro-Ala)(peak 13),2-hydroxy propionyl(peak 22),cyclo-(Pro-Ser)(peak 6). The IC 50 of 10 batches of C Ⅱ-3 samples to HepG 2 cells was 70.550-200.303 μg/mL. Among 25 common peaks ,the order of GRA correlation (r)of anti-hepatoma activity was peak 20>23>24>15,all of r values were greater than 0.7;the order of variable importance projection (VIP)of OPLS analysis was peak 23>18>15>24>7>14>6> 2>20,all of VIP values were greater than 1. The standard regression coefficients of peak 7,15,20,23,24 were all greater than 0;while the standard regression coefficients of peak 2,6,14,18 were all less than 0. Conjoint analysis shows that the order of anti-hepatoma activity was peak 20>23>24>15. CONCLUSIONS:unknown chemical ingredients (peak 20, ),cyclo-(Tyr-Pro)(peak 24)and cyclo- (Gly-Phe)(peak 15) in C Ⅱ-3 may be main anti-hepatoma active components.

RESUMO

Objective To investigate the expression and clinical significance of macrophage colony stimulating factor ( M?CSF ) and tumor necrosis factor α ( TNF?α ) in placenta of early onset severe preeclampsia (PE)??Methods Immunohistochemical SP method was used to detect the expression of M?CSF and TNF?α in 77 cases paraffin specimens from Department of Pathology,First Affiliated Hospital of Henan University of Science and Technology from September 2015 to September 2017,including 35 cases of early?onset severe PE,42 cases of late?onset severe PE and 30 cases of normal pregnant women??Results (1)The positive expression rates of M?CSF in control group, late?onset severe PE group and early?onset severe PE group were 30%(9/30),61??90%(26/42),82??86%(29/35),respectively??The difference was statistically significant ( χ2 = 18??90, P<0??05)??The positive expression rates of early?onset severe PE group were significantly higher than those in control group (χ2=18??59,P<0??05),and the difference was statistically significant????The positive expression of early?onset severe PE group was higher than that of late?onset severe PE group, and the difference was statistically significant (χ2=4??017,P<0??05)??(2) The positive expression rates of TNF?α in control group, late?onset severe PE group and early?onset severe PE group were 33??33%(10/30),69??05%(29/42),91??43%(32/35),respectively??The difference was statistically significant (χ2=30??21,P<0??05)??The positive expression of TNF?α in early?onset severe PE group was significantly higher than that in control group (χ2=21??37,P<0??05),and the difference was statistically significant??The positive expression of M?CSF and TNF?a in early?onset severe PE group was higher than that in late?onset severe PE group, and the difference was statistically significant (χ2=4??529,P<0??05); (3) The expression of M?CSF and TNF?α was positively correlated in PE (r=0??441,P=0??000)??Conclusion Placental damage is higher in early?onset severe PE,and is related to the severity of the disease??The levels of M?CSF and TNF?alpha in placenta of PE patients may play a synergistic role in the occurrence and development of PE??

RESUMO

Objective To learn the current doctor-patient trust in contracted service of family physicians in suburban Beijing, and analyze its influencing factors from residents′ perspectives to propose countermeasures. Methods A total of 197 contracted residents were sampled from 12 community health service centers in 4 out of 10 suburb districts of Beijing, for a questionnaire survey using stratified random sampling method. Items of the questionnaire cover residents′knowledge of the family physicians′ contracted service, their trust of family physicians′ competence, and their trust level of the ethics and the influencing factors. Measurement data were calculated form the mean value, while the count data were subject to proportion analysis, frequency description and rank ordering. Results 175(88.8% )and 156(79.2% )of the contracted residents trust the ethics and competence of their family physicians. Whether the ethics trust exists between doctors and patients was decided by if residents′respect, care, and interests considerations taken into account by the physicians; the factors influencing the competence trust of the residents were the age, education of the residents, and whether the physicians display respect, care. Conclusions Upgrading the medical competence and ethical conduct is conducive to the establishment of doctor-patient trust. In the establishment of the family physicians contracted service, it is necessary to improve acceptance of the contracted residents. At the same time, medical ethics issues such as respect for care, privacy protection, and maximum interests of the residents in the process of medical treatment deserve further attention.

RESUMO

Objective This study aims to investigate the effect of rehabilitation skills training on suicide and relapse prevention of patients with depression. Methods Eighty patients were randomly divided into two groups. One group accepted depression rehabilitation skills training and the other group accepted general health education for 4 weeks. Both groups were followed up by 12 months, and the number of relapse and suicide and the score of Health-related quality of life made by Word Health Organization (WHOQOL-BREF) were recorded. Results The rate of relapse (10.0% vs.42.5%) and hospitalization (5.0% vs.20.0%) were lower in skills training group than in control group (P<0.05).Rate of seeking help of suicide was higher in skills training group than in control group (25.5% vs.7.5%) (P<0.05).The suicide mortality was insignificantly different between two groups (0.0% vs.2.5%) (P>0.05).The scores of WHOQOL-BREF were significantly higher in skills training group than in control group in follow-up (P<0.05).Conclusions Rehabilitation skills training program can not only reduce the rate of relapse and suicide but also improve the quality of life of patients with depression.
