BACKGROUND: Human skeletal muscle derived myoendothelial cells (MECs) are located in the vascular wall and co-express the markers of muscle stem cells and vascular endothelial cells (CD56+CD34+CD144+CD45-). Studies have shown that MECs are similar to mesenchymal stem cells, express the surface markers of mesenchymal stem cells and have the potential of multidirectional differentiation. OBJECTIVE: To establish an in vitro culture system for human umbilical cord blood CD34+ cells with MECs as trophoblastic layer, and to evaluate the in vitro supporting effect of human skeletal muscle MECs on hematopoietic stem/progenitor cells by measuring the changes in the number, immunophenotype and colony forming ability of CD34+ cells before and after culture. METHODS: There were three groups in the experiment. In experimental group, human umbilical cord blood CD34+ cells were co-cultured with MECs as the nourishing layer; in control group, human umbilical cord blood CD34+ cells were co-cultured with bone marrow mesenchymal stem cells as the nourishing layer; and in blank control group, human umbilical cord blood CD34+ cells were cultured alone without the nourishing layer. The main outcome measures, including the number of human umbilical cord blood CD34+ cells, immunophenotype of blood cells and colony formation ability of hematopoietic stem/progenitor cells were analyzed and compared at 1, 2, and 4 weeks after co-culture. No detection was conducted at 5 weeks due to the lack of survived cells. RESULTS AND CONCLUSION: (1) The number of human umbilical cord blood CD34+ cells increased by MECs as the nourishing layer compared with bone marrow mesenchymal stem cells as the nourishing layer at 1, 2, and 4 weeks; however, there was no significant difference between the two groups (P > 0.05). (2) The cell immunophenotype by flow cytometry analysis indicated that only in the 2nd week, the expression of CD34+CD33- in human umbilical cord blood CD34+ cells in the control group was significantly higher in the experimental group (P 0.05). (3) The colony formation capacity of hematopoietic stem/progenitor cells showed no significant difference between the experimental and control groups at 1, 2, and 4 weeks (P > 0.05). (4) Due to the non-nourishing layer culture system, the number of human umbilical cord blood CD34+ cells decreased significantly in the 1st week, and no cells survived in the 2nd week. Therefore, blood cell immunophenotype and colony analysis could not be performed. (5) To conclude, human skeletal muscle MECs as trophoblasts are the same as human bone marrow mesenchymal stem cells, which have a hematopoietic support in vitro.

RESUMO

Single-cell RNA sequencing (scRNA-seq) is a technique to analyze the transcriptome sequence at the single cell level. It can be used to discover new cell subtypes, reveal heterogeneity between cells, track the progression of diseases, and eventually improve the understanding of complexity of tissues, organs and organisms. At present, scRNA-seq is widely applied in the fields of tumor and embryonic development, and its application in dermatology has shown a gradual growth. This review briefly introduces technical procedures of scRNA-seq, and mainly elaborates relevant progress in dermatology.

Objective:To explore the relationship between the dark triad, basic psychological needs satisfaction and guilty proneness.Methods:Totally 595 college students were randomly selected from Zhengzhou University.They were tested with the dirty dozen(DD), the balanced measure of psychological needs scale(BMPN) and guilt-proneness-5(GP-5). SPSS 22.0 and AMOS 21.0 were used for data analysis, including descriptive analysis, correlation analysis and confirmatory factor analysis. Bootstrap measure was used for mediating effect test.Results:(1)The correlation analysis showed that the dark triad (29(21, 37)), Machiavellianism (4(4, 8)) and psychopathy(5(4, 9)) were negatively correlated with guilty proneness (20(16, 23)), and the correlation coefficients were -0.10, -0.22 and -0.18 respectively(all P<0.01). While narcissism (16(12, 21)) was not correlated with guilty proneness ( r=0.04, P>0.01). The satisfaction of basic psychological needs (56(54, 60)) was positively correlated with guilty proneness ( r=0.31, P<0.01). (2)The relationship between the dark triad and guilty proneness was partially mediated by basic psychological needs satisfaction, and the mediating effect accounted for 62.96%. Conclusion:The dark triad influences guilty proneness through basic psychological needs satisfaction.

We introduce the background of Shanghai medical purchasing service and supervision platform (later we call it "open platform") and the effect of its implementation. We also analyze the problems occurred by medical institutions in the management of supplies, explore how to use open platform to strengthen the management of medical supplies, further optimize the structure of supplies, standardize the clinical reasonable use and charges, and ensure the quality, safety and traceability of supplies.

BACKGROUND@#Tumor recurrence and drug resistance are the main causes of death in tumor patients. The family of acetaldehyde dehydrogenase (ALDH) is closely related to the proliferation, migration, invasion and resistance of tumor cells, and different ALDH subtypes are expressed in different tumor cells. The aim of this study is to elucidate the ALDH subtype in human lung adenocarcinoma HCC-827/GR cells, which resistant to the gefitinib.@*METHODS@#The human lung adenocarcinoma HCC-827 cells were used to generate the gefitinib-resistant HCC-827/GR cells; the expression of ALDH subtype in either HCC-827 or HCC-827/GR was detected by flow cytometry; The proliferative capacity and sensitivity to gefitinib of hcc-827/GR cells were analyzed by MTT assay before and after treatment with 100 μmol/L diethyllaminaldehyde (DEAB); Real-time quantitative PCR was used to detect the expression of ALDH subtypes at mRNA levels in hcc-827 cells and hcc-827/GR cells.@*RESULTS@#Compared with HCC-827 cells, the positive rate of ALDH in HCC-827/GR cells increased. The proliferation ability of HCC-827/GR cells decreased after treatment with 100 μmol/L DEAB. Compared with HCC-827 cells, the expression of ALDH1A1 and ALDH1L1 mRNA was increased in hcc-827/GR cells, but the ALDH3B2 expression was decreased.@*CONCLUSIONS@#ALDH might be used as a molecular biomarker to test the gefitinib-resistant to lung adenocarcinoma cancer cells, and the ALDH1A1 may play a role in gefitinib resistance in lung cancer.

AIM: To observe the efficacy and side effects of interferon ? 1b (interferon) combined with oxymatrine in the treatment of chronic hepatitis B. METHODS: 87 patients were randomly divided into two groups: 45 patients in treatment group were given interferon 30 ?g?d -1 (im) for 4 wk, then 30 ?g?d -1 (im, qod) combined with oxymatrine 600 mg?d -1 (im) for 3 mon; 42 patients in control group were given the same dose of interferon as above only. After completion of therapy, liver function and markers of HBVM were compared between the two groups.
