RESUMO

Objective To analysis TORCH pathogens infection of women in childbearing age in Beijing area,and to explore the relationship of TORCH infection with the level of TNF-α.Methods Using ELISA detect serum IgM and IgG antibody of TOX,RV,CMV,HSV-I and HSV-II from 970 cases of women during Jan.2015 to Dec.2015.TNF-αlevels of TORCH infection and control group were also determined by ELISA,the results were analyzed.Results Of 970 women,the IgM pos-itive rates of TOX,RV,CMV,HSV-I and HSV-II were 1.65%,2.16%,4.54%,17.42% and 6.08%,respectively.The IgG positive rates of them were 3.81%,93.40%,92.47%,64.02% and 14.64% respectively.The positive rates of CMV and HSV-I IgM for women <30 years old were higher than that of ≥30 years old (χ2=4.558,4.051;P<0.05).HSV-I IgM had statistically higher infection rate in summer than other seasons (χ2=5.356,P<0.05).TNF-αlevels of TORCH IgM positive group were elevated compared with control group (t=10.219,P<0.01).Conclusion Women planning pregnancy were easier infected by TORCH in Beijing area during 2015 with specific epidemiological features.TNF-αalso plays detri-mental role during reproduction of childbearing age women.

RESUMO

Objective To investigate inhibition effect of leptin on apoptosis of MCF-7 cells and its potential mechanism.Methods MCF-7 cells were cultured and devided into 5 groups:control group,tamoxifen (10-5 mol/L) group,tamoxifen(10-5 mol/L)-leptin(102 ng/mL)group,tamoxifen (10-5 mol/L)-leptin(103 ng/mL) group,tamoxifen(10-5 mol/L)-leptin(104 ng/mL)group.Fourty-eight hours after being treated with corresponding concentration of leptin and tamoxifen respectively,all groups of cells were quantatively tested for apoptosis by ELISA and were semi-quantatively detected for survivin,Bcl-2,Bax mRNA change by real-time PCR.Results Tamoxifen induced MCF-7 apoptosis were inhibited by leptin at a serial concentration of 103 ng/mL,104 ng/mL.Meanwhile,leptin largely enhanced survivin mRNA expression in MCF-7 cells,compared with control group.However,no significant increase of BCL-2,BAX mRNA in MCF-7 cells was observed after leptin treatment in either group.Conclusions Leptin could effectively inhibit tamoxifen induced MCF-7 apoptosis in a dose dependent manner.The potential mechanisrn of leptin' apoptosis inbition in MCF-7 cells may involve upregulation of survivin mRNA.

ObjectiveTo investigate the inhibitory effect of lentivirusly-mediated ObR-siRNA on transplanted MCF-7 human breast cancer cells by intratumoral injection.MethodsA model of subcutaneous implanted tumor was generated through injecting MCF-7 human breast cancer cells into the nude mice.Thirty established mice with MCF-7 breast cancer cells xenograft were divided into 3 groups randomly,and mice in the experimental group were intratumorally injected with ObR-siRNA lentivirus,while the negative control group and blank control group mice were injected with the same dose of negative lentivirus and normal saline.All mice were subcutaneously injected with recombinant human leptin around the tumor site once a day.Tumor size was blindly measured every other day and the mRNA expression and protein expression levels of ObR in each group were determined.ResultsKnockdown of ObR-treated xenografted nude mice with a high leptin microenvironment was successfully established.Local injection of ObR-siRNA lentivirus significantly suppressed the established tumor growth in nude mice(P < 0.01,P <0.01 ).Real time-PCR and Western blotting showed that the mRNA and protein expression of ObR was decreased in the ObR-siRNA lentivirus group( P < 0.01,P < 0.01 ).ConclusionsIntratumoral injection of recomhinant ObR-siRNA lentivirus inhibits the growth of MCF-7 cells xenografts in the nude mice,suggesting that ObR might represent a therapeutic target in the genotherapies of human breast cancer.

Objective To investigate the different effect of exogenous leptin on estrogen receptor α,β mRNA in human breast tumor tissue in nude mice xenograft models. Methods We made nude mice xenograft models of MCF-7 human breast cancer cells cultured in vitro, then divided them into experimental group of]eptin( n = 30)and control group of normal saline( n = 30)randomly. The models of experimental group were injected subcutaneously the recombinant human leptin for 15 consecutive days, the models of control group were injected subcutaneously the same dose of normal saline. A real- time quantitative RT- PCR assay was developed to quantify the expression of estrogen receptor α, β mRNA in tumor tissue, using the relative quantitative analysis. Results The leptin-intervened nude mice xenograft models were safely established. The relative quantitation of estrogen receptor α mRNA was significantly higher in the leptin group than in the normal saline group ( P < 0.01 ), the relative quantitation of estrogen receptor β mRNA was significantly lower in the leptin group than in the normal saline group ( P < 0. 01 ). Conclusion The nude mice xenograft models can be safely intervened with human leptin by subcutaneous injection around tumor.Estrogen receptor is one of the targets of leptin in the progress of breast cancer. Exogenous human leptin can up- regulate the expression of estrogen receptor α and down- regulate the expression of the estrogen receptor βin nude mice xenograft models of human breast tumor.

RESUMO

Leptin,a 16 kD protein encoded by ob gene,has been shown to play a pivotal role in satiaty control and adipose metabolism.However,numemus recent studies indicme that leptin may also take part in mammary cells transformation,proliferation and angiogensis.Those evidences suggest that leptin has an important role in the mammary cell tumorigenesis and progession,but the undedine mechanism needs further exploration.Our aim is to review the current andvances on leptin's role in tumourigenesis and progression of breast cancer from perspectives of in vitro,aniamal model and breast cancer biopsy studies.

RESUMO

Objective To investigate whether HSP90α could be a sensitive and specific serum biomarker for the diagnosis and progression of lung cancer. Methods In the present study, different secretomic analy-ses on the two human lung adenocarcinoma cell lines CL1-0 and CL1-5 with low and high metastatic poten-tial, respectively, were performed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis and ma-trix-assisted laser desorption/ionization time-of-flight mass spectrometry. The candidate biomarker was con-firmed by Western blotting, and was further analyzed in 224 serum samples including 141 lung cancer, 37 benign pulmonary diseases, as well as 46 healthy individuals using ELISA assay. Results HSP90α was sig-nificantly upregulated in the CM of CL1-5 cells. It was found that the levels of HSP90α were specifically ele-vated in the sera of non-small cell lung cancer compared with other groups. At the cut-off point 0.535 on the receiver operating oharacteristie curve, HSP90α could comparatively discriminate lung cancer from benign lung disease and healthy control groups with sensitivity of 0. 817, specificity 0. 919 and total accuracy 80. 14%. Conclusion HSP90α may be a potential useful serum biomarker for discriminating lung cancer from benign lung diseases and healthy individuals and staging of non-small cell lung cancer.

Objective We established a model of human breast cancer in nude mice, to discuss the feature of pathology and biology of breast cancer,and give help to establish tools of pathogen research. Methods Human breast cancer cells MCF-7 were subcutaneously injected into the right armpit of nude mice to establish human breast cancer models.The mice were divided into composite group, estrogen group, leptin group and the blank group (30 in each). In the composite group,estrogen and leptin were injected into peripheral region of the tumor daily.In the estrogen group,estrogen was injected.In the leptin group, leptin was injected.In the blank group, physiological saline was injected.Tumor growth was observed, and the volume of the tumor was recorded.The tumor tissues obtained from the mice were pathologically examined. Results (1)The tumor-taking rate of leptin group and the blank group were 46.7% (14/30)and33.3% (10/30) ,and the mode is failure.In composite group and estrogen group they were 96.7% (29/30) and 70% (28/30).There was not significant difference between them ( P < 0.05).(2) The differences of average diameter and volume were statistically significant between the composite group and the estrogen group (P < 0.05).(3) Pathology diagnosis was invasive ductal carcinoma. Conclusions (1) Establishing human breast cancer model in nude mice need the stimulation of estrogen. The tumor-taking rate of nude mice has no relationship with leptin.(2) In the study in vivo, leptin as same as estrogen has stimulating effect on MCF-7 cells proliferation.(3) The transplanted cancer cell partly have the pathology and biology features of the human breast cancer cells and give help to establish tools of pathogen research.

RESUMO

Recently, Geographic Information Systems (GIS) , as it could effectively combine space data (map) and social attributed information (database), has far-reaching impact the development of other disci-plines. Application of GIS as a tool to study epidemiology of breast cancer, can analyze the various factors affecting the distribution of breast cancer, determine their spatial distribution and spatial-temporal trends and spatial clustering analysis, the reconstruction of historical exposure to on-site to monitor the relationship be-tween living environment and breast cancer and thereby help the decision makers in targeted health policy. This paper discusses the application of GIS for breast cancer research progress .

RESUMO

Breast cancer is one of the most common cancers in women and has been increasing in the last few years. Tumor markers are usually substances occurring in blood or tissue which might be clinically usable in patients with cancer. Owing to a lack of sensitivity for early disease and lack of specificity, none of the a-vailable markers is of much value for the early detection of breast cancer. To identify potential early-stage markers for breast cancer is a difficult problem. Here we summarizes the application of tumor markers in the diagnosis and prognostic prediction in breast cancer, which include CA153 ,CEA, PR and AR, and recently defined markers such as miRNA, hMAM, Midkine.

RESUMO

Objective To observe the therapeutic effect of Dan-Shen Injection in the treatment of acute pancreatitis. Methods Forty-two cases of acute pancreatitis were classified into 2 groups randomly. The control group was given routine treatment. The therapeutic group was given with routine treatment combinat-ing with Dan-Shen Injection. Results The number of normal WBC cases within 7 days was more in the therapeutic group than that in the control group (P < 0. 05). The recover time of amylases in blood was shor-ter in the therapeutic group than that in the control group (P < 0. 05). The incidence rate of complication, the operative rate and the mortality in the therapeutic group were lower than that in the control group (P <0.05). Conclusion In the treatment of acute pancreatitis, Dan-Shen Injection can enhance the therapeutic efficacy, shorten the course of treatment and reduce the incidence rate of complication and the mortality.

RESUMO

As an increasing threat to women's health worldwide, breast cancer has many well-established risk factors, including genetic suscebility, high concentration of serum estrogen, early-age menarche, late-age menopause and postmenopansal obesity. However, these risk factors only account for about half of the in-cidence of breast cancer. Recent studies provided some clues that three primary hormone responsive virues-human papilloma virus (HPV), mouse mammary tumor virus (MMTV) and Epstein Barr virus (EBV)may act play roles in pathogenesis of human breast cancer. This review aims to have a close look at advances on the potential role, probable mechanism and relating hypothesis of Human papillomavirus in human breast cancer.

RESUMO

Since Bence-Jones protein was discovered and used in multiple myeloma, many tumor markers have been used in diagnosis and treatment monitoring of various cancers. However the lack of specificity and sensitivity of most tumor markers is always limiting their application in research and clinical diagnosis currently. It is predicted that the future development of tumor marker research may mainly focus on the innovation of research strategies, such as combined detection of a set of related tumors marker and comprehensive interpretation of testing results.

RESUMO

Objective To explore the diagnostic value of spiral CT for the intestinal obstruction of unknown causes.Methods The CT findings of 115 intestinal obstructive patients with unknown causes were analyzed retrospectively.Results Of the 115 patients,the diagnosis of intestinal obstruction was comfirmed by CT in 110 cases patients,the correct diagnostic rate was 95.7%.Conclusions Abdominal CT is a rapid、simple and effective means for diagnosis of the location、cause and degree of obstruction in cases of intestinal obstruction with unknown cause.It can act as an important basis for the treatment program by doctors.
