RESUMO

<p><b>OBJECTIVE</b>To study the inhibitory effect of wogonin on the growth and proliferation of breast cancer cells MDA-MB-23, and observe its effect on the adhesion, migration and invasion of MDA-MB-23 cells, in order to further study its molecular mechanism.</p><p><b>METHOD</b>MTT assay was used to detect the effect of wogonin on MDA-MB-23 cell growth. Ki-67 assay was adopted to test the effect of wogonin on cell proliferation. Scratch test, adherence test and invasion chamber assay were taken to detect the effect on the migration and invasion abilities of MDA-MB-231 cells. Proliferation and metastasis-related proteins and relevant signaling pathways were detected by Western blotting.</p><p><b>RESULT</b>Wogonin could remarkably inhibit the growth and proliferation of MDA-MB-231 cells, significantly inhibit migration, adhesion and invasion abilities of breast cancer cells at a low concentration, and effectively inhibit the expression of Survivin, Bcl-2, ICAM-1, MMP-2, MMP-9 proteins of MDA-MB-231 cells.</p><p><b>CONCLUSION</b>Wogonin could notably inhibit growth and proliferation of breast cancer cells, and inhibit migration, adhesion and invasion of MDA-MB-231 cells. Its invasive and adhesive effects on MDA-MB-231 cells may be related to the decrease in ICAM-1, MMP-2, MMP-9 expressions.</p>

Objective: To investigate the inhibition of gambogic acid on proliferation and metastasis ability of BGC-803 cells and to study their preliminary mechanism further. Methods: Methyl thiazolyl tetrazolium (MTT) and Hoechst staining were used to study the effects of gambogic acid-induced apoptosis in gastric cancer cell line BGC-803; Invasive methods were used to study the effects of invasion, adhesion, migration by gambogic acid; The influence of gambogic acid on the expression of bcl-2, bax, and ICAM-1 gene in gastric cancer cells was evaluated by RT-PCR and Western-blotting. Results: Gambogic acid was found not only to inhibit the growth of BGC-803 cells but also to induce the cell apoptosis in a dose- and time-dependent manner. The ability of adhesion, migration, and invasiveness was clearly inhibited by gambogic acid. With the increase of gambogic acid concentration, the expression of bcl-2, ICAM-1 gene, and protein were down-regulated and the expression of bax gene and protein were up-regulated. Conclusion: Gambogic acid could inhibit the growth of gastric cancer cells BGC-803 significantly, have a significant role in promoting tumor cell apoptosis, and inhibit tumor cell metastasis-associated properties. The mechanism may be related to decrease of bcl-2, ICAM-1 expression and increase bax expression.
