RESUMO

Objective:To investigate the efficacy and adverse reactions of moderately hypofractionated intensity modulated radiation therapy (IMRT) combined with androgen deprivation therapy (ADT) for locally advanced prostate cancer (LAPC).Methods:This study retrospectively analyzed the medical records of 40 LAPC patients who were admitted in The Second Hospital of Dalian Medical University during 2014-2020. The planning gross target volume (PGTV) dose for prostate gland and seminal vesicle gland was 64.8-70.0 Gy/25-28 f, 2.4-2.8 Gy/f and the dose of PGTVnd in 20 cases with positive pelvic lymph nodes was 60.0-64.4 Gy/25-28 f, 2.3-2.4 Gy/f. The dose of planning target volume (PTV) for the drainage area of pelvic lymph nodes was 45.0-50.4 Gy/25-28 f. The enrolled patients were treated with long-term ADT, including neoadjuvant, simultaneous, and adjuvant therapies. The efficacy and adverse reactions were evaluated. The prognostic factors affecting the biochemical failure-free survival (BFFS) were analyzed.Results:The median follow-up time was 31 months. The 2- and 3-year overall survival (OS) rates were 100% and 96.9%, respectively. The 1-, 2-, and 3-year BFFS rates were 90%, 76.8% and 72%, respectively. The 1-, 2-, and 3-year distant metastasis-free survival (DMFS) rates were 92.2%, 82.8% and 75.1%, respectively. Gleason (GS) score ( χ2=10.00, P < 0.05) and adjacent tissue invasion ( χ2=8.85, P<0.05) were prognostic factors related to BFFS for LAPC. Adjacent tissue invasion and GS 9-10 were independent poor prognostic factors. The incidence of acute urinary adverse reaction and rectal injury (grade≥2) was 7.5% and 20%, respectively. The incidence of late urinary adverse reaction and rectal injury (grade≥2) was 12.5% and 17.5%, respectively. Adverse reactions at grade 3-4 did not occur. Conclusions:The moderately hypofractionated IMRT combined with ADT is feasible for LAPC treatment, achieving satisfactory survival effects. 70 Gy/25-28 f, 2.5-2.8 Gy/f is a safe and effective moderate hypofraction scheme. Adjacent tissue invasion and GS score are prognostic factors related to BFFS for LAPC.

Objective:To discuss strategies for building a high level research assistant team in research hospital.Methods:This paper illustrates and analyzes the construction experiences of the research assistant team and its positive impact on the construction of research hospitals, discusses reflections on the construction of high-level research assistant team from the aspects of talent introduction, management system, incentive measures and education and employment in combination with problems identified during the construction process.Results:Research hospitals can improve the stability and competitiveness of the research assistant team by improving the degree of attention, improving the management system, improving the evaluation and incentive system, and strengthening the training and cultivation.Conclusions:Accelerating the development of scientific research and innovation team with appropriate structure are the foundations for promoting the construction of research hospitals, so it is imperative to promote the construction of research assistant team.

Objective:To discuss the role of National Natural Science Foundation of China in promoting the innovation and development of Brain Science.Methods:Based on the data of Beijing Neurosurgical Institute for application and funding of NSFC from 2010 to 2019, statistically analyzed the number, category, grants, application code and SCI papers funded by NSFC.Results:The main research direction of Beijing Neurosurgical Institute is basic and clinical application of neurological diseases. The number of applications and funds for the institute has been increasing steadily, with 112 NSFC projects approved in the past ten years. General Programs and Young Scientists Funds are 83.04% of the total projects. And SCI papers funded by NSFC has increased significantly.Conclusions:With the guidance and guarantee of the NSFC, the institute has defined the development direction combined of basic and clinical research, promoted international cooperation and communication, strengthened the cooperation between hospital and institute, implemented fine management, optimized the talent team, improved the output and transformation of scientific and technological achievements, and promoted the innovation and development of Brain Science.

Objective:To study the dosimetric differences and short-term efficacy between intracavitary/interstitial brachytherapy (IC/ISBT) and conventional intracavitary brachytherapy (ICBT).Methods:Forty-five patients with locally advanced cervical cancer were treated with IC/ISBT and ICBT. Points A (A 1, A 2), D 90%, D 100%, organs at risk, and the doses of bladder, colon, rectum and small intestine were calculated and the short-term efficacy was observed between two groups. Results:Point A dose was significantly improved in IC/ISBT compared with ICBT ( P<0.05). The D 90% and D 100% in IC/ISBT were significantly higher than those in ICBT (both P<0.05). After brachytherapy, IC/ISBT could obtain a significantly larger increase in target dose when residual tumor diameter was ≥3 cm compared with ICBT ( P<0.05). The D 2cm 3 and D 0.1cm 3 of bladder, rectum, colon and small intestine did not significantly differ between IC/ISBT and ICBT (all P>0.05). The 1-, 3-and 6-month clinical efficacy did not significantly differ between two technologies (all P>0.05). Conclusion:During brachytherapy for locally advanced cervical cancer (residual tumor diameter ≥3 cm), IC/ISBT significantly increases the doses of target area and point A without increasing the dose of organs at risk or lowering the short-term clinical efficacy, which has significant dosimetric advantages.

OBJECTIVE: To explore current hot spots in drug research and application,and provide reference of the draft use standard for pharmacy journal editor. METHODS: Taking China Pharmacy (short for our magazine) as example,all literatures published by our magazine and included by CNKI were searched during inception (Jan. 1990) and Mar. 19th, 2019; top 150 literatures [top 100 non-review literatures (NRA) and top 50 review literatures (RA)] were collected according to comprehensive score of downloaded number and citied number as indicators, and then analyzed in respects of publication year,column of NRA,first author’s work unit, subject words; one literatures which the cited number was largest was analyzed separately. RESULTS: Totally 150 literatures were published between 1993 and 2016, while the literature amount reached the maximum value in 2010 (18 articles). Among NRA, the most were in the columns of “pharmacy administration” (44 articles, 44.00%), “TCM and ethnic medicine” (18 articles, 18.00%), “pharmacists and pharmaceutical care” (14 articles, 14.00%). Among 150 first authors,73 (48.67%) were from college, 62 (41.33%) were from hospital. Subject words of NRA were “status and development” (20 times), “active ingredient” (14 times), “international experience” (9 times), while those of RA were “pharmacological action” (25 times), “Chinese herbal medicine” (15 times), “active ingredient” (12 times). The Principles and Utilization of Sequential Analysis of DDD System of Drug in Hospitals was cited for 1 426 times, which was much higher than the second one (261 times). CONCLUSIONS: This study can show the good literatures origin and study direction,provide reference to the hot spots for readers. At the same time, high-frequency cited literatures also suggests that researchers can conduct in-depth analysis of controversial and unclear contents in academic circles.

Objective@#To analyze the prognostic factors and the value of radiotherapy (RT) for the early stage (T1-2N1M0) triple-negative breast cancer (TNBC) patients after modified radical mastectomy, and provide a basis for the selection of the clinical treatment strategy.@*Methods@#The retrospective analysis was performed in 87 TNBC patients at T1-2N1M0 stage. All patients were admitted to the Second Affiliated Hospital of Dalian Medical University from Jan 2006 to Oct 2011. Fifty-three cases received postoperative RT (RT group), and the other 34 cases without RT (non-RT group). Factors enrolled in Kaplan-Meier analysis were postoperative RT, age, menstruation, histological grade, vascular tumor thrombus, T staging, the number of positive lymph node and metastasis rate, surgery procedure, Ki-67 index. The endpoints were local regional recurrence rate (LRR), distant metastasis rate (DM), recurrence free survival (RFS), and overall survival (OS) rate for 5 years.@*Results@#The significant difference was found in the 5-year LRR (9.4% vs. 15.2%) and RFS (81.3% vs. 66.7%) between RT group and non-RT group (χ2=8.073, 12.789, P<0.05). No significant difference in the DM and OS was observed between the two groups (P>0.05). The univariate analysis showed that RT, lymph node metastasis, age, and Ki-67 index were the risk factors for 5-year LRR (P<0.05), while vascular thrombus and lymph node metastasis rate were risk factors for 5-year DM (P<0.05), RT, vascular tumor thrombus, lymph node metastasis rate, and Ki-67 index 5-year RFS (P<0.05). The multivariate analysis showed that RT and lymph node metastasis were the independent risk factors for 5-year LRR (HR=0.279, 5.277, P<0.05), vascular thrombus was an independent risk factor 5-year DM (HR=2.313, P<0.05), while RT, vascular tumor thrombus and lymph node metastasis rate were the independent risk factors for 5 years RFS (HR=0.378, 2.350, 5.084, P<0.05).@*Conclusions@#Postoperative RT might improve the local control rate of TNBC patients at T1-2N1M0 stage, while the effect on DM and OS in 5 years was little. Postoperative RT, lymph node metastasis rate, vascular tumor thrombus, Ki-67 index and age are related to patient′s prognosis of early stage TNBC.

RESUMO

γδ T cells have been well recognized as a unique cell population that is actively involved in both innate and adoptive immunity of bodies. The features of γδ T cells, such as their major histocompatibility complex independent antigen recognition and their cytotoxic effects to tumor cells, make them as promising candidates used for cancer immunotherapy. There is a strong interest in developing γδ T cell-based immunotherapy for clinical application in treating cancer patients. This review discusses the progress of recent studies related to the γδ T cells and cancer immunotherapy, with an emphasis on the main characteristics of γδ T cells in several types of gynecologic tumors.

Objective To study the short-term efficacy of computed tomography (CT)-guided iodine-125(125I) radioactive seed implantation in the treatment of hypoxic tumors.Methods Twenty-two patients treated with 125I radioactive seed implantation in our department from 2014 to 2016 were divided into hypovascular tumor group (hypoxic group,n =12) and hypervascular tumor group (non-hypoxic group,n=10) based on the hemodynamics of solid tumor evaluated by color Doppler ultrasound.The enhanced CT images were loaded to the three-dimensional particle implantation planning system for preoperative planning.After 125I radioactive seed implantation,the D90 for target volume was verified to be 106-128 cGy.Treatment outcomes were evaluated according to the World Health Organization criteria at 1-3 months after surgery.Results In all the patients,the overall response rate was 82% at 3 months after surgery.There were no significant differences in response (complete response + partial response) rates at 1,2,or 3 months after surgery between the hypoxic group and the non-hypoxic group (P=0.840,0.696,0.840).Conclusions In the treatment of solid malignant tumor,125I radioactive seed implantation can overcome the resistance of hypoxic tumor to radiotherapy in vitro and achieve satisfactory short-term efficacy.

Objective To investigate the prognostic factors affecting early breast cancer patients with conserving surgery and discuss the influences of chemoradiation sequence on treatment efficacy.Methods We retrospectively analyzed 191 cases of early breast cancer after breast conservation surgery in the second hospital of Dalian medical university,from January 1,2006 to December 31,2015.According to the treatment order,they were divided into radiotherapy group,chemotherapy group and sandwich group.There were 32 patients in the radiotherapy group,107 patients in the chemotherapy group,and 52 patients in the sandwich group.The sandwich group was treated with chemotherapy-radiotherapy-chemotherapy.The factors were analyzed independently from the patients' age,menopausal status,pathology type,tumor size (T1,T2),axillary lymph node,molecular type of breast cancer,the interval between surgery and radiation,vascular tumor emboli.Regression model was used to find the risk factors of early stage breast cancer patients with conserving surgery.The 5-year LRR and 5-year OS was compared among radiotherapy-first group,chemotherapy-first group,the sandwich group in patients with high risk factors,and the factors associated with the prognosis of patients were analysed.Results The Multivariate Cox Regression model showed that tumor size,axillary lymph node,vascular tumor emboli,the interval between surgery and radiation were independent risk factors associated with 5-year OS.Among patients with T1 and T2 stage in radiotherapy-first group,chemotherapy-first group,sandwich group,there was no statistical difference in 5-year LRR or 5-year OS (P > 0.05).Among patients with N0 stage in radiotherapy first group,chemotherapy first group,sandwich group,5-year LRR were not statistically different (P > 0.05).For patients with N1 stage in chemotherapy-first group and the sandwich group 5-year LRR were with statistical difference(x2 =4.813,P =0.028).OS were without statistical difference (P > 0.05).In patients with vascular tumor emboli in radiotherapy-first group,chemotherapy-first group,the sandwich group,5-year LRR were without statistical difference (5.3%,9.2%,vs.18.1%,P > 0.05),OS were statistically different (61.5%,90.1% vs.87.2%,x2 =6.282,P =0.043).For patients without vascular tumor emboli,there was no statistical difference in 5-year LRR (P > 0.05).For patients started radiation within 6 months after surgery in radiotherapy-first group,chemotherapy-first group,the sandwich group,5-year LRR were without statistical difference (P > 0.05).However,patients in chemotherapy-first group the time interval surgery and radiation ≤6 months vs.>6 months 5-year LRR was 5.2% and 14.1% (x2 =4.886,P=0.027),5-year OS were 96.9% and 85.7% (x2 =5.758,P =0.038).Conclusions Clinical treatment is based on individualized options.For patients with negative axillary lymph nodes,but high risk factors for local recurrence,radiotherapy may be performed firstly.For high-risk patients with axillary lymph node metastasis or vascular tumor embolus,chemotherapy should be started early and radiotherapy may be followed,but not more than 6 months.

Objective To evaluate the outcomes and prognostic factors of distant metastasis in patients with advanced cervical squamous cell carcinoma treated with concurrent chemoradiotherapy (CCRT). Methods A total of 118 patients with International Federation of Gynecology and Obstetrics (FIGO, 2009 version) stageⅡb-Ⅳa cervical squamous cell carcinoma treated with CCRT between 2006 and 2010 in the Second Affiliated Hospital of Dalian Medical University were analyzed. Their median age was 48 years (range, 23-70 years). FIGO stages were as follows:Ⅱb stage 56 cases,Ⅲa stage 6 cases,Ⅲb stage 52 cases, andⅣa stage 4 cases. Of the all patients, 55 cases showed bulkly tumor (tumor size>4 cm) and 35 cases were pelvic lymph node positive. Forty patients had pretreatment hemoglobin (Hb) levels no greater than 110 g/L. Patients with elevated squamous cell carcinoma antigen (SCC-Ag) >1.5 μg/L before CCRT and at one month after CCRT were 91 cases and 34 cases, respectively. Kaplan-Meier method was used to estimate survival. For the analysis of prognostic factors affecting distant metastasis, log-rank test was used for univariate analysis, and Cox proportional hazard model was used for multivariate analysis. Results Thirty-seven patients were diagnosed with recurrence, 19 cases of whom developed distant metastasis, 13 cases developed loco-regional recurrence and 5 cases had both distant and loco-regional recurrence. The 5-year overall survival rates and distant disease-free survival of all patients were 64.0% and 78.8%, respectively. Two patients had grade 3 acute gastrointestinal toxicity (mainly diarrhea) and 20 cases had grade 3 to 4 hematologic toxicity. Seven patients experienced grade 3 to 4 late toxicity, 5 cases of them were gastrointestinal and 2 cases were genitourinary toxicity. Univariate analysis showed that FIGO stages, SCC-Ag level at one month after treatment, pretreatment hemoglobin level, and pelvic lymph node metastasis were significantly correlated with distant metastasis (all P<0.05). Multivariate analysis showed that FIGO stage, SCC-Ag level at one month after treatment, and pelvic lymph node metastasis were independent prognostic factors for distant metastasis (all P<0.05). Conclusions For stageⅡb-Ⅳa cervical squamous cell carcinoma, the regimen of CCRT was efficacious and safe. The predictive factors for distant metastasis in patients withⅡb-Ⅳa stage squamous cell carcinoma of cervix treated with CCRT included FIGO stage, SCC-Ag level at one month after treatment, and pelvic lymph node metastasis. New treatment strategies should be considered to control distant metastasis for these patients.

Objective To evaluate the efficacy of trolamine cream in the prevention and treatment of acute radioactive dermatitis in patients with breast cancer after modified radical mastectomy.Methods 80 breast cancer patients who received postmastectomy chest-wall irradiation by 6-MeV electron beam were included.40 cases in the treatment group were given trolamine cream combined with the conventional nursing care during radiotherapy,while another 40 cases in the control group were treated with the conventional nursing care.Results In the treatment group,the rates of radiation dermatitis of grade 1,2 and 3 were 70.0 % (28/40),22.5 %(9/40),and 7.5 % (3/40),respectively,while they were 52.5 % (21/40),32.5 % (13/40) and 15.0 % (6/40),respectively,in control group (P < 0.05).Grade 1,2 and 3 radiation dermatitis appeared in treatment group when the radiation doses were (21.91±4.39) Gy,(37.43±6.50) Gy and (46.14±6.50)Gy,respectively,while in the control group,it was (12.67±2.16) Gy,(22.61±3.51) Gy,(42.71±8.11) Gy (P < 0.05),respectively.With cumulative doses of 10 Gy and 25 Gy,the rates of radiation dermatitis in the treatment group were 22.5 % (9/40) and 47.5 % (19/40),respectively,while they were 40.0 % (16/40) and 62.5 % (25/40),respectively,in the control group (P < 0.05).Conclusions Trolamine cream effectively alleviates radiation skin damage in breast cancer patients after modified mastectomy,thus it is suitable for use in patients with postoperative chest wall radiotherapy.

<p><b>OBJECTIVE</b>To study the absorption characteristics of alkaloids in Coptis chinensis in rat intestinal.</p><p><b>METHOD</b>In situ single-pass perfused rat intestinal model was used and the concentrations of berberine and palmatine were determined by HPLC, the drug absorption rate constant (K(a)) and apparent absorption coefficient (P(app)) were tested to evaluate their absorption characteristics.</p><p><b>RESULT</b>The absorption of berberine and palmatine in different regions of rat intestinals was significantly different. From bigness to smallness, the P(app) value of berberine at 50 mg x L(-1) were ileum, duodenum, jejunum and colon, of palmatine at 50 mg x L(-1) were ileum, colon, jejunum and duodenum. The concentration of berberine and palmatine had distinctive effect on the K(a) value of berberine and palmatine were ordered from larger to little as in Wujiwan compatibility, Coptis chinensis extractive and single compound.</p><p><b>CONCLUSION</b>The berberine and palmatine were absroped at all regions of rat intestinal. The absorption characteristics of 2 alkaloids were not passive diffusion. The other ingredients in Wujiwan compatibility and Coptis chinensis extractive could promote the absorption of alkaloids.</p>

<p><b>OBJECTIVE</b>To observe the effect of Tiangou Jiangya capsule (TJC) on blood pressure in renovascular hypertension rats and explore its possible mechanism.</p><p><b>METHOD</b>Seventy-two Wistar rats were randomly divided into normal control group, model group, captopril group, TJC small, medium and high dose groups. Non-invasive blood pressure measurement was used to detect the arterial blood pressure of rat tails. PRA, Ang II , ALD, 6-Keto-PGF1alpha, ET and TXB2 content in blood was measured by radioimmunoassay. NO content in blood was determined by method of nitrate reductase.</p><p><b>RESULT</b>The systolic, diastolic and mean pressure significantly increased, serum PRA, Ang II , ALD decreased, ET levels significantly increased in model group rats. TJC significantly reduced blood pressure, improved the plasma renin activity, decreased ET levels and increased NO content of model rats.</p><p><b>CONCLUSION</b>TJC can reduce blood pressure of renovascular hypertention rats, and the mechanism may be related to its regulating lower blood pressure regulation of the secretion of RAAS system and improving vascular endothelial function.</p>

<p><b>OBJECTIVE</b>To investigate the effects of Tiangou Jiangya capsule on blood pressure of spontaneous hypertensive rats.</p><p><b>METHOD</b>The 13-14 week SPF rats were selected and randomly divided into model groups, the low, middle, high dose of Tiangou Jiangya capsule groups, positive control group administrated with captopril. Drugs were intragastric administrated once per day, lasting four weeks. The blood pressure, heart rate, heart ventricle indexes, urinary volume and the level of PRA,angiotensing II (Ang II), aldosterone (ALD) of rats were observed.</p><p><b>RESULT</b>The low, middle, high dose of Tiangou Jiangya capsule can remarkably reduce the systolic pressure, diastolic pressure and mean arterial pressure of spontaneous hypertensive rats (P < 0.05 or P < 0.01). The low, middle dose can reduce the heart rate of rats (P < 0.01). The low dose can effectively inhibit the left ventricle indexes (P < 0.05). The Tiangou Jiangya capsule has no markedly effects on the renin-angiotensin-aldosterone system (RAAS) activity and urinary output of rats.</p><p><b>CONCLUSION</b>The results indicate that the Tiangou Jiangya capsule has evident effect of lowering blood pressure of rats, which is related to reducing heart rate, heart ventricle indexes, and has no effect on the RAAS and diuresis.</p>

<p><b>OBJECTIVE</b>To study the acute and chronic toxicity of Tiangou Jiangya capsule.</p><p><b>METHOD</b>Tiangou Jiangya capsule was intragastrically administered to mice twice a day. The appearance, behavior, mental status, respiratory changes of mice and the number of poisoned and dead mice in each group were noted daily for 14 consecutive days. The amount of weight and feed of survived mice were recorded every day. The mice were divided into four groups: the treatment groups (minimum, middle, maximum dose of Tiangou Jiangya capsule) and the control group. After continuously orally administrated for 6 months, the rats' behavior, weight gain, food consumption, indications for hematology, blood biochemistry, urine analysis, electrocardiogram, systematic autopsy and histopathology were observed. The above physiological indexes were inspected again 1 month after cease of administration.</p><p><b>RESULT</b>The oral acute toxicity study of Tiangou Jiangya capsule in mice revealed that the maximum dose is 534.86 g x kg(-1), which was 534.86 times the recommended human maximum dose in clinical practice. Compared with normal control group, no significant differences were observed in rats' behaviors, food-intake, electrocardiogram and relative examination indexes among the treatment groups. There was no difference of hematology, biochemistry test, urine and histopathology.</p><p><b>CONCLUSION</b>The minimum dosage of Tiangou Jiangya capsule is relatively safe. It caused weight loss by administrated with the middle and maximum dose for 6 months, which should be paid attention in clinical studies.</p>

Objective To investigate the effect of elemene on the radiosensitivity of A549 cells and its possible molecular mechanism.Methods The effect of radiosensitivity was detected by colony forming assay.The protein expressions of DNA-PKcs,Bcl-2 and P53 were detected with Western blot.The correlation between the protein expression of DNA-PKcs and Bcl-2,DNA-PKcs and P53 was analyzed.Results Elemene had radiosensitizing effect on A549 cells,with the SERDo and SERDq 1.54 ± 0.20 and 1.43±0.15,respectively for 10 μg/ml elemene,and 1.63 ±0.32 and 1.75 ±0.19,respectively for 20 μg/ml elemene.Compared with irradiation group,the expression of DNA-PKcs was reduced significantly in 10,20 μg/ml elemene combined with radiation group ( t =7.52,8.33,P < 0.05 ),so was for Bcl-2(t =10.74,11.33,P <0.05).The expression of P53 protein increased significantly (t =-9.25,7.66,P <0.05).There was a remarkable negative correlation between the expression of DNA-PKcs and P53(r =-0.569,P <0.05),and a remarkable positive correlation between DNA-PKcs and Bcl-2 (r =0.755,P <0.05 ).Conclusions Elemene has radiosensitizing effect on A549 cells,which might be related to down-regulation of DNA-PKcs gene expression,up-regulation of P53 and down-regulation of Bcl-2.

Objective To study if β-elemene can increase radiation-induced deoxyribonucleic acid (DNA) damage and decrease the damage repair.Methods Exponentially growing human lung adenocarcinoma cells (A549) were exposed to 10 or 20 μg/ml β-elemene for 24 h before irradiation.The effect of β-elemene on the in vitro radiosensitivity of A549 cells was evaluated using clonogenic assay.DNA damage and repair were evaluated using comet assay.Results Exposure to β-elemene before irradiation increased the radiosensitivity of A549 cells.The SERD0 for 10 μg/ml and 20 μg/ml β-elemene was 1.55 and 1.64, respectively.The SERDq for 10 μg/ml and 20 μg/ml β-elemene was 1.43 and 1.75, respectively.Combined treatment, comparing to irradiation or β-elemene treatment alone, induced higher levels of DNA damage and slower rate of damage repair.A549 cells exposed to 20 μg/ml β-elemene followed by irradiation showed a higher levels of tail moment (TM) than those exposed to irradiation or β-elemene alone at 0 h,2 h,6 h and 24 h after irradiation.The TM of the three groups at 0 h,2 h,6 h and 24 h after irradiation was 7.16±2.61,0.95±0.65 and 1.81±1.23(F=231.24,P<0.01), 3.65±2.06,0.11±0.07 and 1.58±1.40(F=90.22,P<0.01), 2.09±0.83,0.1±0.05 and 0.45±0.25(F=238.44,P<0.01), 1.45±1.37,0.11±0.08 and 0.60±0.40(F=38.94,P<0.01), respectively. Conclusions β-elemene can enhance the radiosensitivity of A549 cells through the enhancement of DNA damage and the inhibition of DNA damage repair.

Objective To screen radiosensitizing-related genes mediated by elemene in lung adenocarcinoma A549 cells by using gene chip. Methods MTT test was used to calculate the IC50 of elemene. ①The effect of radiosensitivity was detected by colony forming assay. A549 cells were divided into 2 groups: radiation group and radiation + elemene group. Oligonucleotide chip was used to screen the gene expression changes of A549 cells from these 2 groups. The up-regulated gene Egr-1 and the down-regulated gene CyclinDl were selected to undergo RT-PCR so as to confirm the reliability of the result. Results MTT test showed the elemene inhibited the proliferation of the A549 cells dose-dependently. The IC50 value of elemene on the A549 cells was 120 mg/L. ②10 mg/L elemene had radiosensiting effect on A549 cells.The values of SERDO and SERDq obtained from the survival curve were (1.54±0. 20) and (1.43±0. 15 )respectively. Gene chip screened 122 differentially-expressed genes, including 89 up-regulated genes and33 down-regulated genes. ③These altered genes could be related to cell structure, substance metabolism,cell proliferation, cell differentiation, signal transduction, material transport, DNA repair, apoptosis,immune response and so forth. The RT-PCR results of Egr-1 and Cyclin D1 were consistent with the genenchip analysis.Conclusions The mechanism of elemene enhancing the radiosensitivity of lung adenoearcinoma A549 cells is the result of participation and collaboration of multiple genes. Further study of the newly-discovered differentially-expressed gene helps find out new radiosensitizational targets of elemene.

Objective To examine the effect of voice cues on nonfluent aphasia patients'performance in reading idioms.Methods Very familiar idioms were presented to aphasia patients on a computer screen either with or without an accompanying voice cue.The patients were asked to say the last two words of each idiom.The number of errors with the last two words of the idioms wag used to compare the patients'performance under the two conditions.Results The mean correct score was 9.46±13.08 when the idioms were presented without a voice cue,but 13.61±6.06 with the voice cue.The difference was statistically significant.Conclusion Voice cues can improve the speaking of familiar idioms for nonfluent aphasia patients.
