RESUMO

OBJECTIVE:To explore the mo lecular mechanism of Chaihu shugan powder in the adjunctive treatment of major depressive disorder (MDD). METHODS :Network pharmacology was adopted. The microarray dataset GSE 76826 was downloaded from GEO common database ,and the limma package was used to screen the differently expressed genes in the samples of MDD patients. The active ingredients and corresponding targets of Chaihu shugan powder were screened out based on TCMSP platform. Through the online software venny 2.1.0,the differently expressed genes of MDD and the targets of Chaihu shugan powder were screened,and the common target genes of Chaihu shugan powder for MDD were obtained. The target genes were introduced into Cytoscape 3.6.1 software to construct the network diagram of “Chaihu shugan powder active ingredients-MDD target gene ”. Then , BisoGenet plug-in was used to construct the PPI network ,analyze the topological structure and screen the core target gene. KEGG pathway analysis was conducted. RESULTS :There were 139 active components of Chaihu shugan powder for the treatment of MDD,including quercetin ,kaempferol,luteolin,stigmasterol,etc. There were 8 core target genes ,including NR3C2,DPP4, ALOX5,GSTM1,CCND1,IGFBP3 and so on. They play an important role in the treatment of MDD via signaling pathway such as regulating cell receptor ,purine metabolism ,steroid biosynthesis ,aminoacyl biosynthesis ,etc. CONCLUSIONS :Chaihu shugan powder can play a role in the adjunctive treatment of MDD through multiple targets and pathways ,which can provide a theoretical basis for the follow-up study of its mechanism.

RESUMO

<p><b>OBJECTIVE</b>To study the structural integrity and continuity of the bilateral uncinate fasciculus (UF) in patients with temporal lobe epilepsy (TLE) using magnetic resonance diffusion tensor imaging (DTI) and evaluate the impact of structural damage of the UF on the executive function of the patients.</p><p><b>METHODS</b>Thirty patients with TLE (14 left, 16 right) and 15 healthy control subjects underwent DTI scanning between January, 2007 and July, 2011, and the left and right UF were analyzed for fractional anisotropy (FA) and fiber mean length.</p><p><b>RESULTS</b>In the control subjects, the average FA was significantly higher in the left than in the right UF (P<0.01). In patients with left TLE, the average FA of the left UF was significantly lower than that of the control subjects (P<0.01), but the FA of the right UF was comparable with that of the control group (P>0.05). In patients with right TLE, the average FA of the left and right UF was significantly lower than that of the control group (P<0.05 and P<0.01). In patients with unilateral TLE, the FA of their bilateral UF was similar. No significant difference was found in the mean length of UF fiber between these 3 groups.</p><p><b>CONCLUSIONS</b>FA is normally higher in the left UF than in the right UF, but inpatients with TLE, the left FA tends to have a lowered UF regardless of which hemisphere is involved, suggesting an early pathology in the microstructure of the left UF. This finding may help in the investigation of possible reasons for executive function damage in TLE patients.</p>

RESUMO

Objective To provide scientific evidence to identify and confirm severe acute respiratory syndrome (SARS) by laboratory detection.Methods Multiple clinical specimens were collected serially and systematically from the 4 suspected SARS patients, which occurred between Dec.2003 to Jan.2004 in Guangdong Province. The samples were tested by serologic and molecular methods.Results IgM or IgG antibodies against SARS-CoV were detectable after 6—8 days of the onset in four patients. The four-fold or greater rising in antibodies was clearly detected in three of the four patients, while the fourth patient’s seroconversion was from negative to positive. The results analysed by enzyme-linked immunosorbent assay( ELISA), immunoflourescence assay (IFA), and neutralization test were highly correlated. SARS-CoV RNA was just detected in 3 throat swab specimens from case 1 by real-time PCR. M, N and S genes were amplified by reverse transcriptase polymerase chain reaction (RT-PCR) from the positive samples. Sequencing results showed that they were SARS-CoV gene segments, and most closely matched SARS-CoV gene sequences were isolated from civet cats in Guangdong Province. Nevertheless, SARS-CoV was not isolated from any samples of the 4 patients.Conclusion Based on these results, the 4 reported cases were laboratorily confirmed as SARS cases.
