RESUMO

Objective To investigate the diagnosis, therapeutic monitoring and prognosis value of the total procollagen type 1 amino-terminal propeptide(tP1NP), beta-C-terminal telopeptide(β-CTx)and bone alkaline phosphatase(BAP)in the bone metastasis of lung cancer.Methods With the case-control study method, the serum levels of tP1NP, β-CTx and BAP in 196 lung cancer patients, including 109 patients with bone metastases,87 patients without bone metastases,and 106 healthy controls at the Shanghai Sixth People′s East Hospital affiliated to Shanghai Jiao Tong University between 2014 and 2015 were quantitatively detected by chemiluminescent immunoassay.Receiver operating characteristic(ROC)curve was calculated to assess the diagnostic value.Survival curve was performed by Kaplan-Meier method. Results The concentration of tP1NP,β-CTx and BAP in the lung cancer patients with bone metastasis were significantly higher than that in the lung cancer patients without bone metastasis(Z=-5.642,P<0.001;Z=-3.783,P<0.01;Z=-8.923,P<0.01).ROC curve analysis showed that the AUC of tP1NP, β-CTx and BAP were 0.874,0.776 and 0.678 respectively(P<0.05).The AUC of the combined three markers was 0.925(95%CI 0.867-0.963),with sensitivity of 77.11% and specificity of 98.11%.The levels of tP1NP and β-CTx were associated with the clinical response.The concentration of tP1NP,β-CTx were significantly decreased in patients achieved remission(t=4.607,P<0.05;t=5.355,P<0.05). Survival analysis showed that higher concentration of tP 1NP was correlated with poor prognosis[OR=3.287, 95%CI(1.118-9.661),P<0.05].Conclusions The levels of tP1NP,β-CTx and BAP cloud be used for the differential diagnosis of bone metastasis of lung cancer,and the combined usage was more effective. tP1NP and β-CTx cloud be used in therapeutic monitoring of lung cancer patients with bone metastasis. Moreover,tP1NP could be used as prognostic biomarker in lung cancer patients.

RESUMO

17β-estradiol (E2) treatment of cells results in an upregulation of SIRT1 and a down-regulation of PPARγ. The decrease in PPARγ expression is mediated by increased degradation of PPARγ. Here we report that PPARγ is ubiquitinated by HECT E3 ubiquitin ligase NEDD4-1 and degraded, along with PPARγ, in response to E2 stimulation. The PPARγ interacts with ubiquitin ligase NEDD4-1 through a conserved PPXY-WW binding motif. The WW3 domain in NEDD4-1 is critical for binding to PPARΓ. NEDD4-1 overexpression leads to PPARγ ubiquitination and reduced expression of PPARγ. Conversely, knockdown of NEDD4-1 by specific siRNAs abolishes PPARΓ ubiquitination. These data indicate that NEDD4-1 is the E3 ubiquitin ligase responsible for PPARγ ubiquitination. Here, we show that NEDD4-1 delays cellular senescence by degrading PPARΓ expression. Taken together, our data show that E2 could upregulate SIRT1 expression via promoting the PPARΓ ubiquitination-proteasome degradation pathway to delay the process of cell senescence.

Objective To investigate the relation of acupressure time and incidence rate of sub-cutaneous hemorrhage after abdominal injection of low-molecular-weight beparin calcium.At the same time to seek the most appropriate acupressure time to reduce incidence rate and area of subcutaneous hemor-rhage. Methods Patients(60 cases)undergoing abdominal injection of low-molecular-weight heparin cal-cium from October 2005 to August 2006 were selected.Each patient received 16 times of injection.They were divided into 8 groups according to acupressure time,the acupressure time for each group was 3,4,5,6,7,8,9,10 minutes. Incidence rate and area of subcutaneous hemorrhage was observed. Results Lo-cal acupressure time was related to the incidence rate and area of subcutaneous hemorrhage,P<0.01. The incidence rate and area of subcutaneous hemorrhage was statistically different between acupressure time of 6 minutes and 7 minutes,P<0.01,while other adjacent groups had no statistical difference. Conclusions Local acupressure time of 7 minutes can effectively reduce incidence rate and area of subcutaneous hemor-rhage after abdominal injection of low-molecular-weight heparin calcium.

Objective To clarify whether ethanol induced cytotoxicity of neurons was inhibited by proteasome peptidase activities. Methods A rat pheochromocytoma(PC12) was used in the present study. Cytotoxicity and proteasome peptidase activities were assessed in PC12 cells exposed to 50, 100, 150 mmol/L, 200 and 300 mmol/L ethanol. Results After 48 hours of treatment, ethanol inhibited proteasome peptidase activity in a dose-dependent manner at the concentration above 100 mmol/L. Moreover, the effect of ethanol on activities of proteasome was also in a time-dependent manner from 24 h. Cytotoxicity was still observed as ethanol concentrations increasing, started from 100 mmol/L, as assessed by MTT method. Conclusion Our results demonstrates ethanol may disturb the metabolism of protein by inhibiting proteasome peptidase activities, which is related to the toxic mechanism of ethanol on PC12 cells.
