RESUMO

Leucine-rich alpha-2 glycoprotein 1 (LRG1) is a highly conserved member of the family of leucine-rich repeat (LRR) proteins. LRG1 has been reported to be closely related to the occurrence and development of malignant tumors. Recent studies have shown that LRG1 is abnormally expressed in many digestive system tumors, including esophageal cancer, gastric cancer, liver cancer, pancreatic cancer, and colorectal cancer. LRG1 has also been reported to be closely related to tumor cell proliferation, invasion, migration, apoptosis, and poor prognosis. This paper reviews the research progress of LRG1 in many digestive system tumors, providing new ideas for LRG1-based anti-tumor treatment.

RESUMO

Objective:To explore whether the limit of "before the equal-diameter period" is reasonable in the current criteria for "Diagnosis of Kashin-Beck Disease" (WS/T 207-2010) in children, and to provide basic data and technical support for revision of the criteria for "Diagnosis of Kashin-Beck Disease" (WS/T 207-2010).Methods:In 2018, the historical Kashin-Beck disease (KBD) areas in Heilongjiang Province were selected as the investigation sites. The right-hand X-ray films of all children aged 7-12 years old were taken. According to the different X-ray manifestations of the growth and development of the phalangeal epiphysis, they were divided into five periods: before the equal-diameter period, equal-diameter period, ultra-diameter period, pre-closure period and closure period. Firstly, after stratifying by basic data such as age and gender, the data were standardized and analyzed. Secondly, the detection rates of metaphysis-epiphysis (CRME) in each period were calculated and compared. Finally, based on the mean value of the detection rate of metaphyseal change in Linkou and Fuyu counties of Heilongjiang Province in 1990, the rates of expected detection and missed diagnosis of metaphyseal changes of KBD among investigated children were calculated and compared under the limitation of before the equal-diameter period, before the ultra-diameter period or age range reduction.Results:A total of 5 019 children were investigated. The proportion of children before the equal-diameter period was 53.94% (2 707/5 019), and that of before the ultra-diameter period was 77.92% (3 911/5 019). The results showed that the equal-diameter period mainly appeared in 7-10 years old, and showed a decreasing trend with the increase of age (χ 2trend = 390.10, P<0.05); the ultra-diameter period mainly occurred in 10-12 years old, showing a decreasing trend with the increase of age (χ 2trend = 65.39, P < 0.05); the pre-closure period mainly occurred in 10-12 years old, with an increasing trend with the increase of age (χ 2trend = 51.86, P<0.05); the closure period mainly occurred in 11-12 years old and increased with age (χ 2trend = 7.58, P<0.05). The CRME of children in ultra-diameter period was 14.78% (158/1 069), however CRME did not occur in children with equal-diameter period. Among children before equal-diameter period, before ultar-diameter period and aged 7-10 years old, the expected detection rates of metaphyseal changes of KBD were 5.90%, 8.53% and 7.42%, respectively. The expected missed diagnosis rates of metaphyseal changes of KBD were 5.06%, 2.45% and 3.52%, respectively. Conclusion:In order to improve the rate of expected detection and lower the rate of missed diagnosis of metaphyseal changes of KBD, children in "equal-diameter period" should be included in X-ray diagnosis and disease monitoring of KBD.

Kaschin-Beck disease (KBD) is an endemic and deforming osteochondropathy, and its pathogen and pathogenesis remain unclear up to now. In recent years, the results in the research of pathogenesis of KBD showed that oxidative stress, excessive chondrocyte apoptosis, imbalance of chondrocyte extracellular matrix metabolism and pro-inflammatory factors were all involved in the occurrence and development of KBD. And this paper summarized the important progress in the pathogenesis of KBD.

Objective To observe the effect of excessive fluoride exposure on expression of tight junction protein zonula occludens-1 (ZO-1) in human vascular endothelial cells and to explore the mechanism.Methods Human umbilical vein endothelial cells (HUVECs) were cultured in vitro.Group design was adopted.Groups of 0.0 (control group) and 0.4,0.8,1.2 mg/L fluoride treatment groups (low,medium and high dose groups) and insulin-like growth factor-1 (IGF-1) group [before adding 1.2 mg/L fluoride ion (F-),100 nmol/L of IGF-1 was added in advance for 2 hours] were set up respectively.Each group of cells was cultured for 24 h to extract cellular proteins.The expression of ZO-1,phosphatidylinositol kinase (PI3K),protein kinase B (AKT) and phosphorylated AKT (p-AKT)were detected and analyzed by Western blotting.Results The expression of intracellular ZO-1 (0.063 ± 0.002,0.043 ± 0.007,0.039 ± 0.004,0.028 ± 0.007) decreased obviously with gradual increase of F-concentration in cultural medium (F =21.36,P < 0.01).And the expressions of ZO-1 in all three fluorine groups were clearly lower than that in the control group (P < 0.05).At the same time,the expression of ZO-1 in high fluorine group was significantly lower than those in other fluorine groups (P < 0.05).There were significant differences in the expression of PI3K (0.032 ± 0.004,0.031 ± 0.002,0.017 ± 0.001,0.017 ± 0.005) and p-AKT/AKT (0.745 ± 0.046,0.806 ±0.007,0.666 ± 0.058,0.641 ± 0.040) among all groups (F =20.38,9.57,P < 0.01 or < 0.05).The expressions of PI3K and p-AKT/AKT in high fluorine group were obviously lower than those of control and mild fluorine groups,and p-AKT/AKT in moderate fluorine group was clearly lower than that of mild fluorine group (P < 0.05).After pretreatment with IGF-1,the expression of ZO-1 (0.055 ± 0.015) increased significantly compared to that of high fluorine group (P < 0.05).Conclusion Excessive fluoride exposure has decreased the expression of ZO-1 in HUVECs through the PI3K/AKT signaling pathway.

Osteoarthritis is a chronic,degenerative joint disease characterized by destruction of articular cartilage,osteophyte formation and narrowing of the joint space,which seriously affects the life quality of patients.However,the pathogenesis of osteoarthritis is still not clear.Kashin-Beck disease (KBD) is an endemic,chronic and deformative osteoarthropathy that mainly occurs in children aged 5-15 years old.The etiology and pathogenesis of KBD are still unclear.Wnt/β-catenin signaling pathway is involved in the development of embryonic cartilage and maintaining the stability of adult bones,which is a key pathway regulating the growth,development and differentiation of chondrocytes.Therefore,the authors review the progress of Wnt/β-catenin signaling pathway in the pathogenesis of osteoarthritis or KBD.

Objective To explore the mechanism of pigment epithelium-derived factor (PEDF) on human liver cells in mitochondrial apoptosis induced by arsenic. Methods Human normal liver cell line (HHL-5) was exposed to NaAsO2 for 24 h, the dose was 0 (control), 2, 5, 8 and 10 μmol/L, respectively. The mitochondrial membrane potential was determined by using flow cytometry, and mRNA levels of PEDF, Bcl-2 and Bax were assayed by Real-time fluorescent quantitative PCR. Results The mitochondrial membrane potential in 10 μmol/L arsenic group (562.33 ± 31.50) was decreased markedly in comparison with that of control group (688.67 ± 47.50, P<0.01), the levels in other groups (5 μmol/L: 634.50 ± 49.03, 8 μmol/L: 655.67 ± 13.65) were not significantly changed (P > 0.05). Compared with control group [(86.69 ± 12.19)%], the mRNA levels of PEDF decreased significantly (P<0.01) when the HHL-5 cells were cultivated with 5 μmol/L [(59.35 ± 11.21)%], 8μmol/L [(58.46 ± 8.69)%] and 10μmol/L [(51.57 ± 7.58)%] arsenic, respectively, and the PEDF mRNA level in 10 μmol/L group was the lowest; but there was no significant change in PEDF mRNA level [(81.52 ± 13.93)%] when HHL-5 cell was cultivated with 2μmol/L arsenic. Similarly, mRNA level of Bcl-2 in 10 μmol/L arsenic group [(53.53 ± 21 . 49 )%] was significantly decreased compared with that of the control group [(96.48 ± 31.14)%, P < 0.05]. Whereas, Bax mRNA levels in arsenic exposure groups [(65.14 ± 32.34)%, (71.81 ± 12.61)%, (72.22 ± 28.40)%] were not significantly changed in comparison with that of control group [(84.47 ± 35.38)%, P > 0.05]. Conclusion PEDF may play a role in mitochondrial apoptosis of human liver cells induced by arsenic through regulating Bcl-2 level.

Objective To evaluate the application value of F wave by comparison with quantitative sensory testing(QST)and nerve conduction studies(NCS)in diabetes peripheral neuropathy (DPN). Methods Three hundred and fourteen diabetic patients with symptom of DPN were retrospectively analyzed, who were recruited and divided into four groups according the course of diabetes mellitus: ≤ 3 years group, 4-6 years group, 7-9 years group and >9 years group. The abnormal rates of QST, NCS and F wave were analyzed and compared. Results At earlier course of disease (≤ 3 years group, 4- 6 years group), the abnormal rate of QST [76.54%(62/81), 91.67% (88/96)]>F wave[(48.15%(39/81),64.58%(62/96)]>NCS[(32.10%(26/81),47.92%(46/96)], and there were significant differences(P<0.01).The abnormal rate of QST, F wave and NCS at 7-9 years group and>9 years group had no significant differences(P>0.05).The abnormal rate of QST in 4-6 years group[(91.67%(88/96)]was higher than that in≤3 years group[(76.54%(62/81)],and decreased in 7-9 years group and >9 years group. The abnormal rate of NCS was increased with course of disease, and the range was obviously between close together group. The abnormal rate of F wave was increased with course of disease,but in 4-6 years group the range was obvious and the range was slow down.At earlier course of disease (≤3 years group, 4-6 years group), the abnormal rate of only F wave significantly increased the abnormal rate of NCS + F wave from 30.86%(25/81), 44.79% (43/96)to 46.91%(38/81), 61.46%(59/96)respectively, but in 7-9 years group and>9 years group, the abnormal rate of only F wave increased the abnormal rate of NCS + F wave from 69.44%(50/72), 84.61%(55/65)to 72.22% (52/72),86.15%(56/65).Conclusions Although the abnormal rate of F wave is not as high as QST, it is still remarkable. Detection of F wave shows positive correlations with the course of disease, which can reflect the course of disease objectively and reliably.The added F wave detection can distinctly raise the abnormal rates on the base of NCS.

Objective To study the inhibitory effect of curcumin on the proliferation,migration and invasion of non-small cell lung cancer cell A549,and to discuss further if it is closely related to the expression of c-Jun N-terminal kinase (JNK) and relative protein p38.Methods A549 cells were cultured by conventional method,and then treated with different concentration of curcumin (10,20,40,80 μmol · L-1).The proliferation,migration and invasion of A549 cells were measured by real-time cellular analysis (RTCA).The expression levels of JNK,p-JNK,p38 and P-p38 were detected by real-time PCR and Western blotting.Results Curcumin showed an antiproliferation effect against A549 cells with IC50 =40 μmol · L-1,and curcumin exhibited obviously inhibitory effect on the migration and invasion of A549 cells.Additionally,compared with control group,curcumin suppressed the expression of JNK and p38 at the gene level,and significantly inhibited the expression of JNK,P-JNK,p38 and p38 (P<0.05) at the protein level.Conclusion These results demonstrated that curcumin can inhibit the proliferation,migration and invasion of A549 cells via reducing the level of JNK,p38 phosphorylation,and blocking JNK signal transduction pathway.

RESUMO

Objective To summarize the experience in nursing locally preserved skin of female patients for neurosurgical cranioctomy. Method The nursing key points were given to 66 female patients undergoing local preparation of skin for cranioctomy as follous preoperative preparation, observation over incision and head nursing. Results The time for skin preparation ranged between 3 to 6 minutes. Postoperatively, no drain tube was placed. No intracranial infections occurred. The incisions in all patients were healed at the first class. Conclusion The nursing measures of good preoperative preparation, close observation of incisions and care to the head for female patients undergoing local preparation of skin for cranioctomy can enhance their satisfaction and propel development of standardized nursing.

Objective To observe the effects of different levels of sodium arsenite ( NaAsO2) on mRNA expression of pigment epithelium-derived factor (PEDF) and apoptosis-related factors in PC12 cells ( rat neuron properties pheochromocytoma). Methods PC12 cells were treated with different levels of NaAsO2 [0 (control group), 2, 5, 10 μmol/L] for 24 hours. The mRNA expression of PEDF and apoptosis-related factors (Bax, Bcl-2) were detected by real-time quantitative PCR. Results There were significant differences in the mRNA expressions of PEDF between the 4 groups (F=102.28, P 0.05); there were significant differences in the mRNA expressions of Bcl-2 between the 4 groups (F=19.87, P<0.05), the mRNA expressions of Bcl-2 in the group of 2, 5, 10 μmol/L (0.65 ± 0.03, 0.49 ± 0.04, 0.57 ± 0.09) were lower than that of control group (0.95 ± 0.11, all P<0.05);there were significant differences in the mRNA expressions of Bax/Bcl-2 between the 4 groups (F=8.352, P<0.05), the mRNA expressions of in the group of 5, 10μmol/L (1.80 ± 0.72, 1.82 ± 0.36) were higher than that of control group (1.02 ± 0.24, all P<0.05). Conclusion NaAsO2 may increase the expression of apoptosis-related factorsBax/Bcl-2 mRNA by decreasing the expression of PEDF mRNA in PC12 cells, leading to apoptosis in PC12 cells.

Objective To analyze the radiographic characteristics of right hand X-ray film of adult patients with Kaschin-Beck disease (KBD) in Qinghai Province, to understand the most affected locations in adult KBD. Methods According to the criteria of KBD diagnose (WS/T 207-2010), 111 cases of patients were taken X-ray films of right hands. Joint space narrow, joint deformity, subchondral sclerosis, osteophyte, coarse and irregularity of joint, marginal retraction sign and capsule changes were chosen as the descriptive indexes, and these indexes were analyzed with SPSS 17.0. Results A total of 111 cases adult patients with KBD were examined right hand by X-ray, abnormality on X-ray film were 103 cases, the abnormal rate was 92.79%. The most affected fingers were Ⅱ- Ⅳphalanx bones, Ⅱphalanx bones accounted for about 92.23% (95/103), Ⅲ phalanx bones accounted for about 99.03% (102/103), and Ⅳ phalanx bones accounted for about 99.03% (102/103). There was significant difference of the abnormality between th e proximal phalanx and the middle phalanx among the Ⅱ - Ⅳ phalanx bones(χ2=79.33, P<0.05). Abnormal numbers of joint deformity, marginal retraction sign, coarse and irregularity of joint, osteophyte, capsule changes and joint space narrow in the proximal phalanx were 212, 7, 134, 47, 15 and 115 in Ⅱ - Ⅳ proximal phalanx, respectively; while the abnormal numbers of joint deformity, marginal retraction sign, coarse and irregularity of joint, osteophyte, capsule changes and joint space narrow in the middle phalanx were 77, 37, 137, 26, 19 and 126 in Ⅱ - Ⅳmiddle phalanx, respectively. Conclusion The Ⅱ - Ⅳ phalanx bones of right hand are the most affected locations in adult KBD.

Objective To explore effect of isoorientin on gastric cancer cell autophagy.Methods Isoorientin and autophagy activator and effect of inhibitors on proliferation of human gastric cancer cell line SGC-7901 by MTT assay.Cell apoptosis was detected by flow cytometry.Production of autophagy lysosomal in gastric cancer SGC-7901 cells was obseroved by AO and MDC staining.Characteristic expression of autophagy protein was analysed by Western blot.Results MTT assay indicated that isoorientin could inhibit gastric cancer cell growth, RAP has the same effects, but 3-MA inhibition of apoptosis.Flow cytometry was used to detect the apoptosis rate of isoorientin and RAP could promote cell apoptosis , while 3-MA had the opposite effect.In AO staining, the red light appeared in the cells, and the green fluorescence appeared in the cells of MDC staining, which showed that there was an autophagy in the cells.Western blot test found the isoorientin was cell autophagy specific protein LC-3II, Beclin-1 expression increased, 3-MA suppressed the expression of the two proteins, and RAP had the opposite effect.Conclusion Isoorientin could induce apoptosis in gastric cancer SGC-7901 cells, autophagy is involved in the process of death.

Objective To investigate the serum levels of biochemical indexes such as calcium,phosphorus,iron,zinc,and alkaline phosphatase of children in Kaschin-Beck disease areas and control areas in Qinghai Province.Methods According to the results of Kaschin-Beck disease monitoring in Qinghai Province,Tangnaihai Township of Xinghai County and Gandu Town of Hualong County were chosen as Kaschin-Beck disease areas and Qushian Township of Xinghai Country was chosen as a control.Children aged 7 to 15 in boarding schools in these areas were chosen as the study subjects in June 2014.Serum alkaline phosphatase level was determined by enzyme colorimetry and the serum iron,zinc,calcium and phosphorus levels were determined by colorimetry.Calcium to phosphate ratio was calculated at the same time.Results Fifty-nine qualified serum samples of children were sampled in Kaschin-Beck disease areas and 45 in control area.Serum alkaline phosphatase levels in Kaschin-Beck disease areas and control area were (311.34 ± 85.31) and (264.09 ± 73.44)U/L,respectively,and the difference was statistically significant (t =-2.97,P < 0.05).Children's serum phosphorus levels in Kaschin-Beck disease areas and control area were (1.62 ± 0.17) and (1.43 ± 0.13)mmol/L,respectively,and the difference was statistically significant (t =-6.29,P < 0.05).Calcium to phosphorus ratio were 1.62 ± 0.17 and 1.82 ± 0.21,respectively,and the difference was statistically significant (t =5.53,P < 0.05).Compared with the control area,the level of children's serum zinc was (19.96 ± 1.70) and (20.59 ± 2.45)μmol/L;the level of serum calcium was (2.59 ± 0.11) and (2.57 ± 0.11)mmol/L;and the level of serum iron was (15.06 ± 7.02) and (17.01 ± 6.70)μmol/L.The differences between these three biochemical indicators were not statistically significant between Kaschin-Beck disease areas and control area (t =1.39,-0.64,1.44,all P > 0.05).Conclusion In Kaschin-Beck disease areas in Qinghai Province,the level of children's serum alkaline phosphatase and phosphorus have increased markedly and the calcium to phosphate ratio has decreased obviously.

Objective To discuss the change of sympathetic skin response (SSR) in patients with cerebral thrombosis with no more than 60 years old. Method SSR was measured in four positions of the center of hand palm and foot palm of 40 normal controls ( control group ) and 56 cerebral thrombosis patients (cerebral thrombosis group) no more than 60 years old. Results SSR wave shape was all educed in control group,but 21 limbs (21/224,9.4%) were not educed in cerebral thrombosis group, SSR abnormality was 80 limbs (35.7% ). SSR's latency in cerebral thrombosis group was obviously prolonged as compared with control group,and the amplitudes of SSR of upper limps and lower limbs had significance between two groups (P< 0.05 or < 0.01). There were serious abnormity within 3 days to 2 months after cerebral thrombosis. Conclusion SSR is a sensitive diagnosis method for the autonomic neuropathy of cerebral thrombosis patients of no more than 60 years old.

Objective To explore the clinical characteristics of multiple primary carcinoma (MPC) in elderly patients and the effects of postoperative adjuvant chemotherapy on the prognosis and advanced cancer genesis. Methods The study population consisted of 220 elderly patients with malignant tumor. Thirty-seven elderly patients with MPC were selected in the study. The characteristics including onset age, carcinoma sites, effects of postoperative adjuvant chemotherapy, interval and survival time between first and advanced cancer were analyzed. Results of the 220 subjects,37(16.8%) patients were MPC, among whom 2 patients were synchronous multiple primary carcinomas (SMPC) and 35 patients were metachronous multiple primary carcinomas(MMPC). The median ages at onset were 70,77 and 77.5 years old for the first,second and third tumor respectively. Among a total of 82 tumor focus, the most predilection site of MPC was gastrointestinal tract, with a 34.2%(28 cases) in colorectum and a 13.4%(11 cases) in stomach. The postoperative patients were divided into two groups: 23 accepted adjuvant chemotherapy (chemotherapy group) and 14 accepted no adjuvant chemotherapy (no chemotherapy group). The survival time of chemotherapy group was significantly longer than no chemotherapy group (x2= 4.63, P = 0.0427 ). The median intervals between the first and the second cancer were 6 years and 4 years in chemotherapy group and no chemotherapy group, respectively(x2=1.63, P = 0.207), while the median intervals between the second and the third cancer were 7 years and 2 years, respectively(x2 = 4.255,P=0.043). The mediansurvival time of the third cancer postoperative patients were 3.5 years and 7 months in chemotherapy group and no chemotherapy group, respectively (x2 = 4.62, P = 0.0316 ) . ConclusionsPostoperative adjuvant chemotherapy may prolong the survival time and delay the advanced cancer genesis in elderly patients with MPC.

Numerous peptides that bind to a given target have been selected by phage display technology. However, some peptides isolated to date do not bind with high affinity to tumor or organ sites, even peptides were selected in vivo. Therefore, the biodistribution of 99mTc-labeled filamentous phage peptide library via MAG3 (mercaptoacetyltriglycine) were investigated to gain a better understanding of phage circulation in vivo. The experimental results showed that the liver and spleen were the organs of the greatest accumulation, while heart, muscle, pancreas and brain retained less radioactivity. In opposite to other tissues and organs, the radioactivity in stomach, intestine and bone gradually went up with time. The clearance of 99mTc-labeled phage in blood was very fast from 5 min to 30 min and then slowed down. When phage in vivo circulated at enough long period of time, some phage particles could extravasate in some organs or tissues and internalized there. In conclusion, the circulation time of phage in vivo should be experimentally determined beforehand according to the targeted organs and the specific location of target peptides in order to panning a peptide with high specificity and affinity to that target.

OBJECTIVE:To investigate the effects of newly decorated air-condition office to human health. DESIGN:A retrospective observational comparative study taking the office staff in newly decorated air-conditioned environment as the subjects and the office staff in natural ventilation environment with same decoration as the controls. SETTINGS:Occupational medicine department of a disease control and prevention center of a city. PARTICIPANTS:This study was conducted in the Department of Occupational Medicine of Dalian Centre for Disease Control and Prevention from April to May 2001.Inclusive criteria:office staff in a newly decorated and airconditioned telecommunication building of either sex;Exclusive criteria:people with acute or chronic disease history.Twenty-three office staff working in the airconditioned office was chosen as the observational group,the ratio of male to female was 12 to 11,aged from 24 to 51 with the mean age of 33.1 years old,average length of service of 14.2 years and average working length in air-conditioned office of 6.8 months.Other 23 office staff was randomly selected from natural ventilation office in the same building with the same decoration condition,the ratio of male to female was 10 to 13,aged from 22 to 58 with mean age of 36 years old, average length of service of 16.7 years and working length in this office for 8.2 months. INTERVENTIONS:To monitor the volatile organic compounds,hazard substance and microclimate in the decorated office with air-conditioner and conduct investigation to the health status of staff. Subjective symptoms between staff in different offices. RESULTS:The concentration of formaldehyde and ammonia exceeded the national hygienic standards in both air-conditioner and naturally ventilation offices,in which formaldehyde exceeded 6.1 and 2.3 times respectively while ammonia exceeded 8.5 and 2.3 times respectively.The illumination,assessment index of positive and negative ions and total bacteria in office with air-conditioner were lower than those of natural ventilation office (P< 0.01).The symptoms manifested in staff working in air-conditioned office such as dizziness,headache,chest pain,dry eye,decreased vision and menoxenia were more distinct than control group(P< 0.01).Dry mouth and hair losing were also more popular in air-conditioned group(P< 0.05). CONCLUSION:More attention should be paid to choosing environmental friendly building and decoration materials and office furniture when decorating.It is very important to make sure the ventilation of air-conditioner is normal and there is enough air change flow.
