RESUMO

OBJECTIVE To study the composition of chemical constituents of Sargassum fusiforme and its in vitro anti- neuroinflammatory activity ,and to provide reference for its development and utilization and the study of pharmacodynamic substances. METHODS UHPLC-QTOF-MS/MS analysis method and GC-MS/MS method were used to analyze the chemical constituents of S. fusiforme . The lipopolysaccharide (1 μg/mL)was adopted to establish the inflammatory model of neuromicroglia BV2. Using paroxetine (5 μg/mL)as positive control ,CCK-8 assay was used to detect the effects of the extracts of S. fusiforme (20,40,60,80,100 μg/mL)on the activity and morphology of neuromicroglia BV 2. The effects of the extracts of S. fusiforme (40,60,80 μg/mL)on the contents of tumor necrosis factor α(TNF-α)and interleukin- 6(IL-6)in cell supernatant were detected by ELISA. RESULTS A total of 103 non-volatile constituents were identified by UHPLC-QTOF-MS/MS ,and 60 volatile constituents were obtained by GC-MS/MS. The extracts of S. fusiforme (40,60,80 μ g/mL) could significantly reduce the abnormally increased activation of neuromicroglia BV 2 and the contents of TNF-α and IL-6 due to lipopolysaccharide (P<0.05 or P<0.01). CONCLUSIONS The study establish the full spectrum of chemical constituents of S. fusiforme ,and it is confirmed that fusiforme has certain in vitro anti-neuroinflammatory activity.

Objective:To analyze the difference in skin surface lipids (SSL) between facial skin lesions of neonates with acne and normal facial skin of healthy infants.Methods:SSL samples were collected from facial skin lesions of 36 neonates with acne (acne group) and normal facial skin of 36 healthy infants (control group) at the Department of Dermatology, Children′s Hospital, Capital Institute of Pediatrics from June 2018 to August 2018. A lipid map was constructed by ultra-performance liquid chromatography-quadrupole-time of flight-mass spectrometry (UPLC-QTOF-MS), and specific information about lipid components was obtained by multivariate data analysis. Independent-sample t test was performed with SPSS19.0 software for comparisons between 2 groups. Results:Compared with the control group, the acne group showed significantly increased relative content of glycerolipids and prenol lipids on the facial skin surface (both P < 0.001), but significantly decreased relative content of fatty acids, glycerophospholipids, sphingolipids, sterol lipids, saccharolipids and polyketides ( P < 0.01 or 0.05) .Further analysis showed that the average relative content of total lipids, diglycerides, wax esters and squalene was significantly higher in the acne group than in the control group ( P < 0.001 or 0.05), but the average relative content of linoleic acid was significantly lower in the acne group than in the control group ( P < 0.001) . Conclusion:Compared with the facial skin of healthy infants, the facial skin lesions of neonates with acne showed significantly increased total lipid content and different lipid components.

OBJECTIVE: To identify chemical components of Forsythia suspense. METHODS: UPLC-Q-TOF-MS technology was used for the chemical components analysis of F. suspense. The determination was performed on ACQUITY UPLC BEH C18 column with mobile phase consisted of 0.1% formic acid aqueous solution (A)- 0.1% formic acid acetonitrile solution (B) with  gradient elution, at the flow rate of 0.3 mL/min; the column temperature was set at 30 ℃; the sample size was 5 μL. Positive and negative ions were detected by electrospray ionization. The temperature of the ion source was 550 ℃; the atomizing gas was N2; the atomizing gas and the auxiliary pressure were 379.2 kPa; the air curtain pressure was 241.3 kPa; the decluster voltage was 80 V/-80 V; the collision energy was 35 eV/-35 eV; the mass scanning range was 80-1 500 Da. Peakview 2.0 software was used to screen the target components by the first-order mass spectrometry, and calculate the high-resolution and accurate molecular weight of each component, compare with the reference spectrum and related literature, or calculate the elemental composition of fragment ions in the second-order mass spectrometry, analyze their decomposition pathways, then infer the structure of compounds. RESULTS & CONCLUSIONS: 45 kinds of compounds were identified from F. suspense,which included 7 phenylethanol glycosides,5 lignans,5 terpenes, 12 flavonoids,7 organic acids,2 phenols,2 quinones,2 glycosides and 3 other components. There were 19 compounds identified for the first time in F. suspense. The study provides a reference for the in-depth study of the pharmacodynamic substance basis of F. suspense and the rapid qualitative and quantitative analysis of the components.

RESUMO

Objective To preliminarily discuss the effect of thrombectomy with the TREVO device in patients with acute ischemic stroke.Methods Six patients with acute ischemic stroke treated with TREVO device at the Department of Neurointervention,Beijing Fengtai You′anmen Hospital from April to June 2016 were analyzed retrospectively,including 2 patients with middle cerebral artery (MCA)occlusion (time window from 2. 7 to 5. 5 h),4 with basilar artery occlusion (time window from 4. 0 to 7. 0 h). The effect of thrombectomy was evaluated by the modified thrombolysis in cerebral infarction (mTICI ) scores. The National Institutes of Health Stroke Scale (NIHSS)scores were used to evaluate the neurological status of patients before and after treatment. The prognosis was evaluated by the modified Rankin scale. Results (1 )After 1 to 2 embolectomies,the occlusive arteries of 5 patients achieved recanalization (mTICI gradeⅡb-Ⅲ;posterior circulation in 4 cases,MCA in 1 case). (2)Compared with at admission,the NIHSS score in 1 of 2 patients with MCA occlusion was reduced by 8 points at discharge,and 1 was not any improvement;compared with at admission,the NIHSS scores in 3 of 4 patients with basilar artery occlusion were decreased by 8-26 points at discharge,and 1 was not any improvement;in 2 patients with MCA occlusion,the modified Rankin scale scores were 0 to 1 at 3 months after discharge (0 in 1 case,4 in 1 case);the Rankin scale scores in 4 patients with basilar artery occlusion at 3 months after discharge were 0 in 1 case,1 in 2 cases,and 5 in 1 case.Conclusion The preliminary experience of using TREVO device embolectomy for patients with acute ischemic stroke shows that it is more safe and effective.

RESUMO

The central nervous system (CNS) plays a key regulatory role in glucose homeostasis. In particular, the brain is important in initiating and coordinating protective counterregulatory responses when blood glucose levels fall. This may due to the metabolic dependency of the CNS on glucose, and protection of food supply to the brain. In healthy subjects, blood glucose is normally maintained within a relatively narrow range. Hypoglycemia in diabetic patients can increase the risk of complications, such as heart disease and diabetic peripheral neuropathy. The clinical research finds that the use of traditional Chinese medicine (TCM) has a positive effect on the treatment of hypoglycemia. Here the authors reviewed the current understanding of sensing and counterregulatory responses to hypoglycemia, and discuss combining traditional Chinese and Western medicine and the theory of iatrogenic hypoglycemia in diabetes treatment. Furthermore, the authors clarify the feasibility of treating hypoglycemia on the basis of TCM theory and CNS and have an insight on its clinical practice.
